Studies on Apoptosis and the Expression of PCNA、P16 in Gastric Cancer

Abstract:Objective: The apoptosis and the expression of proliferating cell nuclear antigen(PCNA) and P16 gene in gastric carcinoma were examined were ,the biological and clinicpathologic significance were explained .Methods:The measures of terminal deoxynucleotidy transferasemediated dUTPbiotin nick end labeling(TUNEL)and streptavidin peroxidase immunohistochemical were used. Results:Among well or moderately differentiated adenocarcinomas,the AI(apoptoticindex)was (10.4±3.1%),While the poorly differentiated adenocarcinomas and mucous carcinomas was (4.12±1.4%) and (3.5±1.2%),respectively ,The AI of specimens with invasion depth exceeding muscularis(4.3±2.1%) was significantly lower than that of specimens with invasion depth not exceeding muscularis(imcluding muscularis).The subgroup with lymph node metastasis AI was (3.7±1.5%),while the subgroup without lymph node metastasis was (9.2±4.1%),The percentage of PCNA positive cells in well or moderately differentiated specimens was 41.7%.Among poorly differentiated specimens of carcinomas and mucos carcinomas ,the percentage of PCNA positive cells was 60% and 77.3%,respectively .The percentage of PCNA positive cells in the subgroup of specimens with lymph node metastasis was 72%,while the subgroup withou tlymphnode metastasis was 27.3%,The subgroup of specimens with invasion depth exceeding muscularis was 28.6% and the subgroup not exceeding muscularis was 77.2%. Among the PCNA positive subgroup,the AI was (3.4±1.2%),while in the PCNA negative subgroup was (7.6±3.1%).In the subgroup of well or moderately differentiated adenocarcinonas ,the positive percentage of P16 was 66.7%,while in the subgroup of poorly differentiated adenocarcinomas and mucous carcinomas was 33.3% and 44.4%,respectively .Conclusions:Apoptosis in gastric carcinoma was positively associated with the differentiation degree while negatively associated with invasion depth and lymph noed metastasis ,The expression of PCNA was positively associated with the invasion depth and lymph node metastasis ,while negatively associated with differentiation degree.Apoptosis index and expression of PCNA being examined at the same time can be useful to know the biological characteristics .In the gastric carcinomas tissues ,the express of P16 was positively associated with the differentiation degree.
