Value of Tumor Marker Alpha-methylacyl-CoA Racemase in Diagnosis of Prostatic Diseases

Abstract:Objective To observe the changes of Alpha-methylacyl-CoA racemase(P504S)in diseased prostatic tissues,so as to assess its role in diagnosis of prostatic tissues.Methods A total of 200 prostatic tissues,including 77 with prostatic cancer(PCa),1 with small cell carcinoma of the prostate(SCCP),2 with atypical small acinar proliferation(ASAP),118 with benign prostatic hypertrophy(BPH)and 2 with high grade prostatic intraepithelial neoplasia(HGPIN).Immunocytochemical examinations(En Vision staining)were performed to detect the expression of P504S in the above tissues.Some tissues also underwent examination for high-molecular-weight cytokeratin 34βE12 and p63(a marker that locates at the nuclei of prostate basal cells). All the data obtained were analyzed by SPSS 11.5 software.The relationship between Gleason score and P504 expression was analyzed;the influence of operation methods on P504S expression was also assessed.Results Seventy-three out of 80(91.2%)PCa tissue in our group were P504S positive,no BPH 0/118 tissue was positive of P504S;weak staining of P504S was noticed in the 2 HGPIN tissues.PCa tissues with different Gleason scores had significantly different expression of P504S expression(x~2=35.052,P0.01),and the expression was also different between PCa tissues obtained with different operation methods.(x~2=18.882,P0.05).The positive rate of 34βE12 was 98.30% in BPH tissues.Seventy PCa tissues had typical absence of basal lamina,10 had incomplete basal lamina.The basal lamina of 2 GPIN tissues was positive of 34βE12,but was weaker than that in the BPH tissues.All the BPH tissues labeled were positive of p63;39 of the 44 labeled PCa tissues were negative of p63.Conclusion Detection of P504S protein can be helpful in the diagnosis of PCa, especially when comes to small needle biopsy specimens for benign and malignant identification.Combined application of P504S with 341βE12,p63 is of greater help in the diagnosis.
