Migration and Differentiation of Mesenchymal Stem Cells Labeled with Enhanced Green Fluorescent Protein in the Parkinson's Disease Rat Brain

Abstract:Objective To study curative effects,survival,migration and differentiation of mesenchymal stem cells(MSCs) transplanted into the Parkinson's Disease(PD) rats.MethodsThe 6-hydroxydopamine(6-OHDA)-induced hemiparkinson rats were selected and allocated randomly into 2 groups.MSCs labeled with enhanced green fluorescent protein(EGFP) or PBS were injected into the striatums of the PD rats respectively.The rotation tests were conducted every week after transplantation.Immunohistochemical staining of Neun,GFAP and TH was carried out at the first and ninth week after transplantation.The expression of TH was detected by Western blot at the 9th week.ResultsThe rotations per minute(RPM) in the MSCs and PBS groups were(8.2±0.6) and(8.3±0.6)r/min respectively before transplantation,and there was no significant difference between them(P>0.05).The RPM were(7.5±0.5) and(8.1±0.6)r/min respectively at the third week.There was significant difference in RPM between two groups from the third week to the ninth week after transplantation(P<0.05).MSCs were noted along the injection tract mostly at the first week,while the cells migrated to the cerebral hemispheres at the ninth week.But most of MSCs were still in the injected hemisphere and some could reach the termination of corpus callosum.No labeled cells were observed in the cerebellum and brain stem.Furthermore,dysembryoma was not found in brain.Immunohistochemical staining of Neun and GFAP was observed at the first and ninth week after transplantation,but the expression of GFAP was predominant.And at the ninth week the expression of TH in the grafted striatum was observed by Western blot and immunohistochemical analysis only in the MSCs group.The result demonstrates that MSCs can differentiate into neuron,gliocyte and dopaminergic neurons(DN).None was detected in the PBS group.ConclusionIt is probably safe and effective to transplant MSCs into striatum of PD rats to correct behaviors of Parkinsonian rats by their differentiation into DN.
