Abstract:Stem cell transplantation is a promising tool for the treatment of neurodegenerative disorders, including Parkinson's disease (PD); however, the therapeutic routes and mechanisms of mechanical approaches to stem cell transplantation must be explored. This study tests the therapeutic effect of transplantation of rat bone marrow mesenchymal stem cells (MSCs) into the substantia nigra (SN) of the PD rat. 5-Bromo-2-deoxyuridine-labeled rat MSCs were transplanted into the SN of the 6-hydroxydopamine-injected side of PD rat brains. The behavioral changes in PD rats were examined before and 4 and 8 weeks after MSC transplantation. The expression of tyrosine hydroxylase (TH) in the SN and the striatum and the survival and differentiation of MSCs were assessed by immunohistochemical and double immunofluorescence techniques. Abnormal behavior of PD rats was significantly improved by the administration of bone marrow MSCs, and the number of TH-positive cells in the SN and the optical density of TH-positive fibers in the striatum were markedly increased. Transplanted MSCs can survive and migrate in the brain and differentiate into nestin-, neuron-specific enolase-, and GFAP-positive cells. Our findings suggest that transplantation of rat bone marrow MSCs into the SN of PD rats may provide therapeutic effects. © 2016 Wiley Periodicals, Inc.

Abstract:Objective To explore the therapeutic effect of intracerebral transplantation of super-paramagnetic iron oxide ( SPIO) and enhanced fluorescence protein ( EGFP) double labeled embryonic mesencephalic neural stem cells (mNSCs) in 6-hydrodopamine (6-OHDA) rat model of Parkinson' s disease ( PD). Methods The rat models of PD were established by unilateral stereotaxic injection of 6-OHDA into rat ventral tegmental area and medial forebrain bundle region, then divided into control group ( n = 12) and transplantation group randomly (re = 12). The SPIO and EGFP double labeled mNSCs were ster-eotaxically injected into striatum of PD rats. Apomorphine ( APO) -induced rotational behavior test was performed to evaluate the therapeutic effect of cell transplantation. MR1 scan was carried out to track the survival and migration of transplanted cells in vivo, and immunofluorescence histochemistry was conducted to identify the distribution and differentiation of transplantated cells. Results Behavior test showed, as compared with control group, SPIO and EGFP double labeled mNSCs transplantation could significantly ameliorate abnormal rotational behavior induced by APO in PD rats (P <0.01). MRI images and immunofluorescence histochemistry revealed almost all cells were stayed in situ, not migrating out of transplantation area. Most mNSCs kept undifferentiation state (37 ± 6) % or differentiated into glial cells (36 ± 4) % , only very few cells could differentiate into DA neurons (6 ±2)%. Conclusion MRI detection of SPIO labeled cells is a promising method to analyze the cells following intracerebral transplantation. Intracerebral transplantation of SPIO and EGFP double labeled mNSCs can significantly improve abnormal behavior of PD rats.

Abstract:Objective To explore the curative effect of co-transplantation of Schwann cells (SCs) and neural stem cells (NSCs) in Parkinson's disease (PD) rats.Methods Eighteen hemiparkinsonian SD rats with right substantia nigra lesioned were selected by estimation of rotational behavior in response to apomorphine and randomly assigned to 3 groups:control group,NSCs group and co-graft group,with 6 rats in each group.PBS,NSCs,SCs plus NSCs were transplanted into the right 6-OHDA lesioned neostriatum of the PD rat respectively.The mesencephalic NSCs were obtained t~om the brain of SD rats at embryonic day 13～16 and SCs came from the sciatic nerves of neonatal rats at day 1-3.Results TH immunoreactive neurons emerged in the striatum of both NSCs group and co-graft group 10 weeks after transplantation.The volumes and nucleus of neurons in co-graft group were bigger than the ones of NSCs group and so do the number of the DA neurons (P＜0.05).Compared with control group,the percent ofapomorphine-induced rotation in both NSCs group and co-graft group was decreased significantly(P＜0.01),but no difference was found between them (P＞0.05).Conclusion Co-transplantation of SCs and NSCs is effective in treating PD rats.Not only dopaminergic neurons differentiated from NSCs functioned in the unilateral 6-OHDA lesioned neostriatum,but also SCs improved the survival of NSCs by secreting many neurotrophic factors.
Co-Transplantation of Gdnf-Overexpressing Neural Stem Cells and Fetal Dopaminergic Neurons Mitigates Motor Symptoms in A Rat Model of Parkinson'S Disease

Abstract:Striatal transplantation of dopaminergic (DA) neurons or neural stem cells (NSCs) has been reported to improve the symptoms of Parkinson's disease (PD), but the low rate of cell survival, differentiation, and integration in the host brain limits the therapeutic efficacy. We investigated the therapeutic effects of intracranial co-transplantation of mesencephalic NSCs stably overexpressing human glial-derived neurotrophic factor (GDNF-mNSCs) together with fetal DA neurons in the 6-OHDA rat model of PD. Striatal injection of mNSCs labeled by the contrast enhancer superparamagnetic iron oxide (SPIO) resulted in a hypointense signal in the striatum on T2-weighted magnetic resonance images that lasted for at least 8 weeks post-injection, confirming the long-term survival of injected stem cells in vivo. Co-transplantation of GDNF-mNSCs with fetal DA neurons significantly reduced apomorphine-induced rotation, a behavioral endophenotype of PD, compared to sham-treated controls, rats injected with mNSCs expressing empty vector (control mNSCs) plus fetal DA neurons, or rats injected separately with either control mNSCs, GDNF-mNSCs, or fetal DA neurons. In addition, survival and differentiation of mNSCs into DA neurons was significantly greater following co-transplantation of GDNF-mNSCs plus fetal DA neurons compared to the other treatment groups as indicated by the greater number of cell expressing both the mNSCs lineage tracer enhanced green fluorescent protein (eGFP) and the DA neuron marker tyrosine hydroxylase. The success of cell-based therapies for PD may be greatly improved by co-transplantation of fetal DA neurons with mNSCs genetically modified to overexpress trophic factors such as GDNF that support differentiation into DA cells and their survival in vivo.

Abstract:Stem cell transplantation is a new therapeutic strategy in the treatment of neurodegenerative disorders such as Parkinson's disease (PD). Therefore, in this study, the therapeutic effects of Trabecular Meshwork Mesenchymal Stem Cells (TM-MSCs) transplantation, as a new source of mesenchymal stem cells, were evaluated in the animal model of PD. After the development and confirmation of hemi-parkinsonian rats by administration of 6-hydroxy dopamine (6-OHDA) and apomorphine-induced rotation test, green fluorescent protein (GFP) labeled TM-MSCs (normal and induced cells) were transplanted in the striatum of rats. Next, the rotation test, rotarod test, open field, passive avoidance memory tests and immunohistochemistry for tyrosine hydroxylase (TH) were done. The results showed that the number of turns significantly decreased and the improvement of motor performance was achieved after cell transplantation. However, there was no significant difference in passive avoidance memory of animals documented by shuttle box test. The number of GFP- labeled cells expressing TH significantly is increased compared to the vehicle group. Collectively, it seems that TM-MSCs and induced TM-MSCs cell transplantation have positive effects on some aspects of the animal model of PD. Other studies may reveal the potentially positive aspects of these cells in the laboratory and clinical studies.
Intravenous Transplantation of Adipose-Derived Mesenchymal Stem Cells Promoted The Production of Dopaminergic Neurons and Improved Spatial Memory in A Rat Model of Parkinson's Disease

Abstract:Objective: Parkinson's disease (PD) is a neurodegenerative disorder described by the dynamic decline of dopaminergic neurons in the substantia nigra pars compacta (SNpc). Stem cell transplantation is a new therapeutic strategy in the treatment of PD. The objective of the study was to assess the impact of intravenous infusion of adipose-derived mesenchymal stem cells (AD-MSCs) on memory disorder in Parkinsonian rats. Materials and methods: In this experimental study, male Wistar rats were randomly divided to four groups containing sham, cell treatment, control, and lesion. The cell treatment group received intravenous injection of AD-MSCs 12 days after PD induction by bilateral injection of 6-hydroxydopamine. Four weeks after lesion formation, spatial memory was examined using the Morris water maze (MWM) assessment. The rats' brains were removed and assessed by bromodeoxyuridine (BrdU), tyrosine hydroxylase (TH), and glial fibrillary acidic protein (Gfap) immunostaining. Results: Statistical analyses revealed a significant addition and reduction in time spent and escape latency in the target quadrant, respectively, in the cell group as compared to the lesion group. Also, BrdU-labeled cells were present in the substantia nigra (SN). The density of TH-positive cells was significantly increased in the AD-MSCs transplantation group as compared to the lesion group, and the density of astrocytes significantly diminished in the AD-MSCs transplantation group as compared to the lesion group. Conclusion: It appears that AD-MSCs treatment for Parkinson's could decrease the density of astrocytes and promote the density of TH-positive neurons. It appears that AD-MSCs could improve spatial memory impairment in PD.

Abstract:Avian myelocytomatosis virus (MC29), a defective acute leukemia virus, has a broad oncogenic spectrum in vivo, and transforms fibroblasts and hematopoietic target cells in vitro, We have used recombinant DNA technology to isolate and characterize the sequences that are essential in the transformation process. Integrated MC29 proviral DNA was isolated from a library of recombinant phage containing DNA from the MC29-transformed nonproducer quail cell line Q5. The cloned DNA was analyzed by Southern blotting of restriction endonuclease digests and by electron microscopic visualization of R-loops formed between the cloned DNA and MC29 or helper virus RNA. It was found that the 9.2 kb cloned DNA insert contains approximately 4 kb of viral sequences and 5.2 kb of quail cellular sequences. The viral sequences contain all of the MC29 specific sequences and 5' helper related sequences as well as part of the envelope region. The size of the cloned EcoRI fragment is the same as that of the major band in EcoRI-cleaved Q5 DNA that hybridizes to viral sequences. Transfection of the cloned DNA into NIH 3T3 cells revealed that the MC29-specific sequences are functional in that they induce foci of transformed cells with high efficiency.

Abstract:Functional recovery after transplantation of brain-derived neurotrophic factor (BDNF)-modified neural stem cells (NSCs) was evaluated in a rat model of cerebral ischaemia damage induced by temporary middle cerebral artery occlusion (tMCAO). Western blotting and enzyme-linked immunosorbent assay demonstrated up-regulated BDNF protein expression by rat embryonic NSCs transfected with the human BDNF gene (BDNF-NSCs). BDNF-NSCs stimulated neurite outgrowth in co-cultured dorsal root ganglion neurons, suggesting that BDNF increased neurogenesis in vitro. In vivo, BDNF promoted recovery of tMCAO. Phosphate-buffered saline, untransformed NSCs or BDNF-NSCs were introduced into the penumbra zone of the right striatum of tMCAO rats and neurological function deficit was assessed for up to 12 weeks using the neurological severity score (NSS). The NSS was significantly lower in the BDNF-NSC transfected transplant group than in all the other groups from week 10. BDNF-NSCs recovered 1 week after transplantation expressed BDNF protein. Transplanted NSCs had differentiated into mature neurons 12 weeks after transplantation. Transgenic NSCs have potential as a therapeutic agent for brain ischaemia.

Abstract:Parkinson's disease (PD) is a ubiquitous brain cell degeneration disease and presents a significant therapeutic challenge. By injecting 6-hydroxydopamine (6-OHDA) into the left medial forebrain bundle, rats were made to exhibit PD-like symptoms and treated by intranasal administration of a low-dose (2 × 10 5 ) or high-dose (1 × 10 6 ) human neural stem cells (hNSCs). Apomorphine-induced rotation test, stepping test, and open field test were implemented to evaluate the motor behavior and high-performance liquid chromatography was carried out to detect dopamine (DA), 3,4-dihydroxyphenylacetic acid (DOPAC), serotonin, and 5-hydroxyindole-3-acetic acid in the striatum of rats. Animals injected with 6-OHDA showed significant motor function deficits and damaged dopaminergic system compared to the control group, which can be restored by hNSCs treatment. Treatment with hNSCs significantly increased the tyrosine hydroxylase-immunoreactive cell count in the substantia nigra of PD animals. Moreover, the levels of neurotransmitters exhibited a significant decline in the striatum tissue of animals injected with 6-OHDA when compared to that of the control group. However, transplantation of hNSCs significantly elevated the concentration of DA and DOPAC in the injured side of the striatum. Our study offered experimental evidence to support prospects of hNSCs for clinical application as a cell-based therapy for PD.

Abstract:Parkinson's disease (PD) is a common neurodegenerative disease. The main pathological changes are the loss of dopaminergic neurons and the formation of Lewy bodies. There is still no effective cure for PD, and cell replacement therapy has entered a bottleneck period due to tumorigenicity and rejection. Therefore, stem cell secretome has received widespread attention. However, the exploration of the secretome components of neural stem cells (NSCs) is still in its infancy. In this study, 6-hydroxydopamine (6-OHDA) was used to establish a PD rat model in vito and the PC12 cell-damaged model in vitro. The results indicated that the injection of neural stem cell-conditioned medium (NSC-CM) into the striatum and substantia nigra could improve the motor and non-motor deficits of PD rats and rescue the loss of dopaminergic neurons. In addition, NSC-CM alleviated 6-OHDA-induced apoptosis of PC12 cells, reduced the level of oxidative stress, and improved mitochondrial dysfunction in vitro. Parkinson disease protein 7 (Park7) was found in NSC-CM by Liquid chromatography-tandem mass spectrometry (LC-MS/MS), and it may be related to the protective effect of NSC-CM on 6-OHDA-injured neurons through Sirt1 pathway. In conclusion, NSC secretome might provide new ideas for the treatment of PD.

Abstract:Parkinson's disease (PD) is a neurodegenerative disorder due to reduced dopamine (DA) in the striatum and loss of DA neurons in the substantia nigra pars compacta. Embryonic stem cells (ESCs) are an optimal source for cell therapy for PD. We recently developed a fast (one-week) protocol using small molecules that effectively induces human ESCs to become primitive neural stem cells (pNSCs), which are then differentiated into DA-like neurons in vitro. As pNSCs are infinitely expandable, this approach offers a strategy to readily generate DA neurons on a large scale. But whether these pNSC-differentiated DA (pNSC-DA) neurons can functionally integrate into the damaged brain is unknown. Here, we transplanted pNSC-DA neurons into 6-hydroxydopamine-induced rat models of PD and monitored their behavior for 16 weeks. In striatum in vivo, we first determined that the grafted pNSC-DA neurons secreted DA(using HPLC with a 200-μm microdialysis probe at a sampling rate of 0.002 Hz); then we recorded DA release by electrochemical amperometry (using a 7-μm carbon-fiber electrode at a sampling rate of 1000 Hz), showing that the grafted pNSC-DA neurons significantly rescued DA release in slices (7%) and in vivo (16%). Furthermore, the transplanted cells survived for at least 16 weeks and dramatically rescued the apomorphine-induced asymmetric rotation behavior in PD rats. Thus, we draw the conclusion that transplanted pNSC-DA neurons can functionally integrate into the brain and partially relieve Parkinsonian symptoms in rats. Our work provides direct evidence for a therapeutic role of pNSC-DA neurons in treating Parkinsonian syndrome.

Abstract:Earlier published research showed that cord blood-derived multipotent stem cells (CB-SCs) exhibited the intrinsic expression of specific transcription factors (e.g., En1, Nurr1 and Wnt1) and seems to be induced to form dopamine neurons in vitro. In this research, we further investigated the therapeutic potential of CB-SCs in 6-hydroxydopamine lesioned Parkinson's disease (PD) rats. The results of PCR analysis showed that CB-SCs could express transcription factors associated with pluripotentiality and dopaminergic differentiation (e.g., Klf4, c-Myc, Nanog, Sox2, Ngn2, and Nurr1). After being transplanted into the striatum and substantia nigra of PD rats, most of CB-SCs (>90%) developed a fate commitment to dopaminergic differentiation, expressed as the expression of tyrosine hydroxylase (TH) and dopamine transporter (DAT). The improvement effect of cell transplantation on dyskinesia in PD rats was better than that in sham control group. Moreover, higher levels of TH protein in brain homogenates further demonstrated that there were more surviving dopamine neurons in the brain of transplanted PD rats. Study concluds, CB SCS transplantation could promote the regeneration of dopamine neurons and behavioral recovery of PD rats.

Abstract:Neural stem cells(NSCs) is one specific type of multipotential stem cells that have the ability to proliferate for a long time,to differentiate into multiple lineages,including neurons,astrocytes and oligodendrocytes,and to migrate toward damaged sites.In all mammalian species,these NSCs mainly exist in most regions of embryonic brain and at least two regions of the adult brain,the subgranular zone(SGZ) of the hippocampal dentate gyrus,and the subventricular zone(SVZ) of the lateral ventricle wall.Because of their properties,NSCs are currently considered to be promising components of cell-replacement strategies aimed at treating Parkinson's disease(PD).There are two therapeutic channels,exogenous and endogenous.For the former,cell is transplanted directly or as a gene carrier for cell replacement and gene therapy.For the latter,neurogenesis is induced by regulating growth factors and cytokines.Exogenous and endogenous NSCs undergo proliferation,migration,and differentiation into dopaminergic neuron in brain,and improve the motor function in animal models of PD.Moreover,the both channels are synergistic.The transplantation of exogenous NSCs can enhance the response of endogenous NSCs in PD models,and the both NSCs are correlated with a host microenvironment which supports to repair and protect damaged neuron.

Abstract:Abstract Bone marrow-derived neural stem cells (BM-NSCs) have shed light on novel therapeutic approaches for PD with the potential to halt or even reverse disease progression. Various strategies have been developed to promote therapeutic efficacy via optimizing implanted cells and the microenvironment of transplantation in the central nervous system (CNS). This current study further proved that the combination of fasudil, a Rho-kinase inhibitor, and BM-NSCs exhibited a synergetic effect on restoring neuron loss in the MPTP-PD mice model. It simultaneously unveiled cellular mechanisms underlying synergistic neuron-protection effects of fasudil and NSCs, which included promoting the proliferation, differentiation, and maturation of exogenous and endogenous NSCs, and contributing to microglia shift into the M2 phenotype. Corresponding molecular mechanisms were observed, including the inhibition of inflammatory responses, the elevation of neurotrophic factors, and the induction of WNT/β-catenin and PI3K/Akt/mTOR signaling pathways. Our study provides evidence for the co-intervention of BM-NSCs and fasudil as a promising therapeutic method with enhanced efficacy in treating neurodegenerative diseases.
