Abstract:Recently, we have developed highly regio- and stereoselective carbometalation of 2-alkynols and 2,3-allenols. The organometallacyclic intermediates may be trapped with I(2) to afford 3-iodoalk-2-en-1-ols. These 3-iodoalk-2-en-1-ols may readily undergo the Sonogashira coupling with terminal propargyl alcohols to form 4-alkyn-2-ene-1,6-diols. Subsequent cycloisomerization in DMA or CH(2)Cl(2) with Au(PPh(3))Cl and AgOTf as the catalyst would afford polysubstituted 2-(1-alkenyl)furans; with PdCl(2) as the catalyst and the reaction in DMA in the presence of allylic bromides, the same substrates afforded polysubstituted 2-(1,4-alkadienyl)furans. In both types of catalyzed cyclization reactions, the elimination of H(2)O promoted the aromatization to form the furan ring. Different alkyl or aryl groups could be introduced into different positions of furans due to the substituent-loading capability of 3-iodoalkenols and diversity of the terminal propargyl alcohols and allylic bromides.

Abstract:A gold-catalyzed regioselective [3 + 2] cycloaddition of alkynyl triazenes with 1,2,4-dioxazoles was developed. The triazene group in the products could be replaced to obtain iodo-oxazoles, providing potential transformations to diverse oxazole structures. This protocol features readily available starting materials, mild reaction conditions and scalability. A plausible mechanism involving a nitrene transfer process was proposed.

Abstract:Cu(NO3)2/acetonitrile was proved to be an efficient oxidation system in the oxidation of 5-HMF to DFF in aqueous solution. After optimization of the reaction conditions, a DFF yield of 97.0% was obtained under the condition of 1.5 mmol 5-HMF, 1.5 mmol Cu(NO3)2, 0.48 ml acetonitrile, 10 ml H2O, 80 °C, 18h. The oxidation system could be recycled at least three times by adding HNO3, which avoids the risk of producing Cu-containing waste. The formation of Cu(CH3CN)4+ in aqueous solution plays an important role in enabling the oxidation of 5-HMF by the redox cycle of Cu2+/Cu+. In order to further improve the reaction efficiency, a reactive-extraction process was developed by introducing toluene as the organic phase for its excellent extraction capacity of DFF, through which the reaction time was shorten to 8 h and a highest DFF yield of 98.9% was achieved.

Abstract:An efficient route to 1,3,4-alkatrien-2-yldihydrofurans via a highly chemo- and regioselective palladium(0)-catalyzed coupling-cyclization reaction of 2-(2',3'-allenyl)acetylacetates with propargylic carbonates was reported. The reaction proceeded smoothly under neutral conditions, affording the O-attacked five-membered products with a 1,3,4-trienyl substituent exclusively in good to excellent yields. The products can be efficiently applied to the synthesis of polysubstituted 2-(dihydrofuryl)cyclopentenone derivatives via a catalytic Pauson-Khand reaction under ambient conditions. Abstract:Comprehensive Summary Furans bearing alkynyl substituents are highly useful in organic synthesis. However, the methodologies to access these important furan derivatives are rather limited. We herein report an efficient synthesis of alkynylated furan derivatives based on Pd‐catalyzed oxidative cross‐coupling reaction between allenyl ketones and terminal alkynes. This novel synthesis of alkynylated furans with wide substrate scope is operationally simple and tolerates various functional groups. Mechanistically, the formation of the palladium carbene through cycloisomerization and the subsequent alkynyl migratory insertion are proposed as the key steps in the transformation. The reaction reported in this manuscript further demonstrates the generality of the carbene‐based cross coupling. This article is protected by copyright. All rights reserved.

Abstract:A convenient synthesis of monosubstituted 3-alkynylfurans by Suzuki coupling reaction of 3-furanylboronic acid with substituted acetylene bromides is described. The internal alkyne products were attainable in 50-89% yields from substrates free of relatively acidic protons. Our protocol is expected to find applications in the synthesis of structurally related natural products.

Abstract:A method for the synthesis of tri-substituted furans has been developed based on Cu(I)-catalyzed reaction of terminal alkynes with β-keto α-diazoesters. This method for the synthesis of 2,3,5-trisubstituted furans is operationally simple and applicable to wide substrate scope. Moreover, this synthesis employs cheap Cu(MeCN)4PF6 as the catalyst and no additional ligand is needed. Similar reaction has also been applied to ethyl (E)-2-diazo-3-(methoxyimino)butanoate for the synthesis of 2,3,5-trisubstituted N-methoxypyrroles with limited success.

Abstract:In the absence of a base, palladium(II) catalysts, such as PdCl(2), PdCl(2)(CH(3)CN)(2), Pd(OAc)(2), and [(pi-C(3)H(5))PdCl](2), can catalyze the cyclizative coupling reaction of 2,3- or 3,4-allenols with allylic halides in DMA at room temperature to provide 2,5-dihydrofurans and 5,6-dihydro-2H-pyrans, respectively, in moderate to good yields. Under similar reaction conditions, nonsubstituted 2,3-allenol 1s affords bimolecular cyclizative coupling product 5s as the major product. The scope of the reaction and its mechanism have been studied briefly. On the basis of the experimental results, the transformation was believed to proceed via a divalent palladium-catalyzed pathway.

Abstract:We have developed a facile and effective synthesis of 3-iodofurans from 3-monosubstituted 1,2-allenyl perfluoroalkyl ketones or 2-hydroxy-4-iodo-2,5-dihydrofurans from 3,3-disubstituted 1,2-allenyl perfluoroalkyl ketones. The perfluoroalkyl substituent and the amount of water in the solvent are important for the success of this electrophilic cyclization.

Abstract:We report here a general synthetic methodology for palladium-catalyzed carbonylative annulation of o-alkynylphenol to construct 2-substituted-3-aroyl-benzo[b]furan. On the basis of the results, this methodology could be applied to a wider selection of iodide substrates to generate desired products. In accordance with mechanistic studies, this process involves coordination of cationic and less hindered acyl palladium complexes with o-alkynylphenols to create a desired cascade triad (coordination, nucleophilic addition, and reductive elimination). Consistent with this mechanism, addition of 1 equiv of AgBF4 to palladium catalyst Pd(Ph3P)(4) generates an ideal candidate for this unique transformation.

Abstract:A novel silver-mediated highly selective oxidative C-H/C-H functionalization of 1,3-dicarbonyl compounds with terminal alkynes for the creation of polysubstituted furans and pyrroles in one step has been demonstrated. Promoted by the crucial silver species, perfect selectivity and good to excellent yields could be achieved. This protocol represents an extremely simple and atom-economic way to construct polysubstituted furans and pyrroles from basic starting materials under mild conditions.

Abstract:Differently substituted 2-alkoxyfurans (2,3,4- or 2,3,5-trisubstituted furans) were highly regioselectively synthesized by means of the ring-opening cycloisomerization of the same cyclopropenyl carboxylates with good yields in different solvents and excellent regioselectivity by using [Cu(acac)(2)] (acac=acetylacetonate) or [RuCl(2)(PPh(3))(3)] as the catalyst, respectively. The structures of these two different types of furans were established by X-ray diffraction studies. A rationale has been proposed.

Abstract:: A straightforward and atom-economical approach to the synthesis of dihydrofurans employing electro-oxidative [3+2] annulation of 1,3-dicarbonyl compound and alkenes has been developed. Neither metal catalysts nor chemical oxidants are required to promote the dehydrogenation process. Moreover, the use of base to decrease the oxidative potential and 1,1,1,3,3,3-hexafluoro-2-propanol to stabilize the reactive intermediates are key to the success of the cyclization.

Abstract:Herein, we devised a synthetic approach for skeletally diversified furfuryl sulfides and 4‐alkylthiopyridines involving the heterocyclization of flexible α‐acyl ketene dithioacetals, enabled by cooperative Pd/Cu catalysis. The overall transformation was initiated by acyl‐directed desulfurative Sonogashira coupling, which was proposed as the key step in surmounting the competitive carbothiolation pathway. This step was followed by 1,6‐addition of either a S‐ or N‐nucleophile and intramolecular cyclization. Notably, the two different annulation reactions featured readily accessible starting materials, selective cleavage and reassembly of multiple chemical bonds, broad functional group tolerance, pharmacologically significant heterocyclic products with high regio‐ and chemoselectivities, and high atom economy.
