Abstract:We investigated the fermentation of arachidonic acid by Mortieralla alpina.Some optimum para-meters were obtained,such as temperature,pH,carbon source,nitrgon source and so on.The optimum medium contained(g/L): glucose 60,yeast extract 5,peptone 12,peanut protein powder 10,corn oil 10,glutamate 5.Under the optimum cultrue conditions,the biomass,lipid yield and arachidonic acid were 36.95 g/L,13.38 g/L and 4.86 g/L,respectively.

Abstract:[Objective] To study and optimize the ingredients of fermented liquid for synthetic culture medium for producing arachidonic acid with Mortierella alpine and increase the yield of ARA.[Method] The carbon sources in the culture medium were optimized,single carbon source and composite carbon source were screened and their initial concentrations were optimized.Many kinds of inorganic nitrogen sources were screened out by using single factor test.After steepest ascent test on four significant factors(glucose,glycerol,yeast powder and amino acid mixture)with significant effects,significant factors were optimized by using response surface center combination design.[Result] The optimum carbon source combination was 80 g/L glucose + 20 g/L glycerol.Taking yeast powder as nitrogen source,amino acid mixture added in the culture medium as a growth factor could promote the accumulation of ARA in the fermentation liquid of M.alpine.The optimized synthetic medium was composed of 80 g/L glucose,12 g/L glycerol,20 g/L yeast powder and 0.3 g/L amino acid mixture.The optimized synthetic medium was verified and the yield was detected after shake flask fermentation culture 7 days.The average yield of ARA was 7.084 1 g/L,which was close to the predicted value.[Conclusion] The research results can provide research basis for improving the yield of ARA in the industrial production of ARA further.

Abstract:Arachidonic acid (ARA, C20:4) is a typical ω-6 polyunsaturated fatty acid with special functions. Using Yarrowia lipolytica as an unconventional chassis, we previously showed the performance of the Δ-6 pathway in ARA production. However, a significant increase in the Δ-9 pathway has rarely been reported. Herein, the Δ-9 pathway from Isochrysis galbana was constructed via pathway engineering, allowing us to synthesize ARA at 91.5 mg L-1. To further improve the ARA titer, novel enzyme fusions of Δ-9 elongase and Δ-8 desaturase were redesigned in special combinations containing different linkers. Finally, with the integrated pathway engineering and synthetic enzyme fusion, a 29% increase in the ARA titer, up to 118.1 mg/L, was achieved using the reconstructed strain RH-4 that harbors the rigid linker (GGGGS). The results show that the combined pathway and protein engineering can significantly facilitate applications of Y. lipolytica.

Abstract:Arachidonic acid (ARA) is an important polyunsaturated fatty acid having various beneficial physiological effects on the human body. The aging of Mortierella alpina has long been known to significantly improve ARA yield, but the exact mechanism is still elusive. Herein, multiple approaches including large-scale label-free comparative proteomics were employed to systematically investigate the mechanism mentioned above. Upon ultrastructural observation, abnormal mitochondria were found to aggregate around shrunken lipid droplets. Proteomics analysis revealed a total of 171 proteins with significant alterations of expression during aging. Pathway analysis suggested that reactive oxygen species (ROS) were accumulated and stimulated the activation of the malate/pyruvate cycle and isocitrate dehydrogenase, which might provide additional NADPH for ARA synthesis. EC 4.2.1.17-hydratase might be a key player in ARA accumulation during aging. These findings provide a valuable resource for efforts to further improve the ARA content in the oil produced by aging M. alpina.

Abstract:Matierella is rich in a ranchidonic acid is a kind of healthy oil resource having great exploitable, lipid containing arachidonic acid is extracted by a organic solvent. The results indicate that extraction with ethanol and nhexane can achieve better lipid yield (34.8 %). After chemical saponification, arachidonic acid was perified by urea inclusion treatment of the lipid, resulting an increase of arachidonic acid content 23.4 % of the crude oil to 60.6%.

Abstract:The oleaginous fungus Mortierella alpina has distinct advantages in long-chain PUFAs production, and it is the only source for dietary arachidonic acid (ARA) certificated by FDA and European Commission. This review provides an overall introduction to M. alpina, including its major research methods, key factors governing lipid biosynthesis, metabolic engineering and omics studies. The current research interests in M. alpina focus on improving lipid yield and fatty acid desaturation degree by enhancing fatty acid precursors and the reducing power NADPH, and genetic manipulation on PUFAs synthetic pathways is carried to optimise fatty acid composition. Besides, multi-omics studies have been applied to elucidate the global regulatory mechanism of lipogenesis in M. alpina. Current research challenges lie in strain breeding and cost control due to the coenocytic mycelium, long fermentation period and insufficient conversion rate from carbon to lipid. In this review, we also proposed future research goals based on a multidimensional engineering: obtaining ideal chassis by directional evolution and high-throughput screening; rewiring central carbon metabolism and inhibiting competitive pathways by multi-gene manipulation system to enhance carbon-lipid conversion rate; optimization of protein function based on post-translational modification; application of dynamic fermentation strategies suitable for different fermentation phases. By reviewing the comprehensive research progress of this oleaginous fungus, we aim to further comprehend the fungal lipid metabolism and provide reference information and guidelines for the exploration of microbial oils from the perspectives of fundamental research to industrial application.

Abstract:The optimum medium and culture conditions for arachiaonic acid (AA) production by martierella alpina M20 was investigated. The optimum medium contained (g/L): glucose 150; yeast flour 15; KH2PO4 3, NaNO3 3; MgSO4·7H2O 0.5; pH6.5. The optimum culture conditions were as follows: volume 50mL/500mL; shaking speed 150 r/min; before three days at 25℃ , then at 20℃ . Under these conditions, the biomass, lipid and AA were 35.5 g/L, 13.20 g/L and 2.20 g/L respectively. In 15L and 1000L fermentors AA yield reached 1.86 g/L and 1.70 g/L respectively.

Abstract:BackgroundThe organic nitrogen source is one of the key factors affecting Mortierella alpina cell growth as well as arachidonic acid (ARA) production. The aim of the present work is to achieve an optimized recipe of organic nitrogen source for ARA production by M. alpina by testing four organic nitrogen sources.ResultsIn the flasks, the results showed yeast extract or corn steep liquor were the most suitable sole nitrogen sources for biomass and ARA yield. In the bioreactor, a biomass of 17.5 g L−1 and an ARA yield of 2.7 g L−1 were achieved when using sole yeast extract, while cell autolysis was induced when using sole corn steep liquor; When using the combined nitrogen source with corn steep liquor and yeast extract at a 3:7 weight ratio, the biomass and ARA yield were significantly improved to 37 and 7.8 g L−1, respectively.ConclusionsThis work evaluated whether using a mix of organic nitrogen sources could improve ARA production when scaling up from a flask to a bioreactor culture. The 3:7 ratio of corn steep liquor to yeast extract was quite favourable for large-scale ARA production, and as a result, this combination has great potential for improving fungal cultures.

Abstract:Effects of various concentration of oil from Mucor on Mortierella alpina producing arachidonic acid (AA) were studied 0.3% oil from Mucor resulted in the greatest AA production. Therefore, 0.3% oil from Mucor was selected to further study the effect of oil from Mucor on AA biosynthesis during the whole process. The results shoed that: supplementation of oil from Mucor had no significant effect on Mortierella growth and total lipid yield. Oil from Mucor increasing AA yield was mainly due to its stimulatory effect on AA content in total lipids.

An Inexpensive Medium for Production of Arachidonic Acid by Mortierella Alpina

Abstract:The production of arachidonic acid was studied in the fungus Mortierella alpina using an inexpensive medium. Glucose derived from maize starch hydrolysate was the sole carbon source and defatted soybean meal and sodium nitrate were the nitrogen sources. Optimal arachidonic acid yield (1.47 g l(-1)) was observed at a glucose concentration of 100 g l(-1). Various treatments of defatted soybean meal to extract soluble nitrogen nutrients were evaluated. Alkali extract was the most effective for arachidonic acid production. A mixture of soybean alkali-extract protein and sodium nitrate was an excellent nitrogen source for fungal growth, lipid accumulation, and arachidonic acid production. A maximum yield of 1.87 g arachidonic acid l(-1) was obtained with a soybean protein concentration of 4.6 g l(-1) and a sodium nitrate concentration of 2.3 g l(-1).

Abstract:Microbes can produce not only commodity fatty acids, such as palmitic acid (16:0) and stearic acid (18:0), but also high-value fatty acids (essential fatty acids). Most high value fatty acids belong to long chain polyunsaturated fatty acids (PUFA), such as omega-3 fatty acids (e.g., eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA)) and omega-6 fatty acids (e.g., arachidonic acid (ARA) and γ-linolenic acid (GLA)). EPA (20:5n-3) is a 20-carbon fatty acid with five double bonds, and the first double bond is in the n-3 position. DHA (22:6n-3) is a 22-carbon fatty acid with 6 double bonds and the first double bond is also in the n-3 position. Both EPA and DHA play an essential role in cardiovascular health including prevention of atherosclerotic disease development (Zehr and Walker, Prostaglandins Other Lipid Mediat 134:131-140, 2018). ARA (20:4n-6) is a 20-carbon fatty acid with four double bonds, and the first double bond is in the n-6 position. GLA (18:3n-6) is an 18-carbon fatty acid with three double bonds, and the first double bond is in the n-6 position. ARA and GLA have multiple biological effects, such as lowering blood cholesterol, and lowering cardiovascular mortality (Poli and Visioli, Eur J Lipid Sci Technol 117(11):1847-1852, 2015). Isolation and identification of the marine diatom Entomoneis sp. MMOGRB 0374S (Bacillariophyta): a strain with high arachidonic acid composition

Abstract:A unicellular marine diatom, Entomoneis sp. MMOGRB 0374S, was isolated as a candidate strain for high production of a polyunsaturated fatty acid (PUFA), arachidonic acid (ARA, 20: 4 omega 6). Its morphological characters were typical for representatives of the genus Entomoneis, e. g. junction line, sigmoid keel and panduriform girdle view. This species was differentiated from other Entomoneis species by its smaller size and denser stria. Phylogenetically, it was distinct from other taxa with a long branch. This relationship was also emphasized by only 94% nucleotide similarity between Entomoneis sp. MMOGRB 0374S and other related species. The biomass concentration reached 182 mg l(-1) at the stationary phase and 229 mg l(-1) after 3 days of nitrogen-limited culture. Lipids were extracted using four methods, and the dichloromethane (DCM)/methanol method was the most effective, yielding 36% of dry biomass at the stationary phase and 40% after 3 days of nitrogen-free culture. Gas chromatography revealed 17 types of fatty acids. ARA was the major component of PUFA and represented between 16% and 19% of the total fatty acids using the four lipid extraction methods, among which DCM/methanol yielded the highest proportion of ARA. Considering the yields of both total lipids and ARA, the DCM/methanol method was the most effective for ARA extraction.

Abstract:An arachidonic acid (ARA)-producing strain Mortierella alpina LU166 ,which is independently isolated by our lab,was studied in the present work.Firstly,it's found that the mycelial morphology can be steadily kept as fluffy small pellets when cultured in baffled flask,which is more favorable for the accumulation of total lipids and ARA than that by filaments and big pellets morpho-logy.Then,the concentration of B-group vitamins (VB1 ,VB12 ,biotin and calcium pantothenate)was optimized to enhance the lipid production.When adding the combination of vitamins with the optimized concentration into the culture,the biomass,lipids and ARA yield reached 24.74,12.66 and 6.46 g/L,respectively,which increased by 10％,30％ and 16％,compared to the control group.

Abstract:Mortierella alpina is known as an arachidonic acid (AA) producing oleaginous fungus. Extraction of lipids from wet and dry M. alpina biomass was compared. Lipids yield of extraction from dry cells was higher than that of extraction from wet. Wet extraction mainly extracted lipid bodies and lipids in membranes did not extract effectively. Enrichment of AA from the fungal lipids by a urea inclusion method was studied. Most of the saturated and monounsaturated fatty acids, 93.0% and 84.6%, respectively, were removed by forming urea inclusion compounds. AA was concentrated after urea inclusion. Its content in total fatty acids increased 6.2-folds and reached 57.1% with a recovery of 81.9%.

Abstract:The microalga Porphyridium purpureum within Rhodophyta abundantly produces several valuable proteins, polysaccharides, pigments and long-chain polyunsaturated fatty acid; it is especially effective in accumulating arachidonic acid (ARA). However, this high ARA yield is always achieved in conditions unfavourable for cell growth. In this study, we present a method for obtaining desirable ARA levels from P. purpureum while simultaneously promoting cell growth using appropriate concentrations of the growth hormone 5-Aminolevulinic acid (5-ALA).

Abstract:Abstract Background Arachidonic acid (ArA) is important for human health because it is one of the major components of mammalian brain membrane phospholipids. The interest in ArA inspired the search for a new sustainable source, and the green microalga Myrmecia incisa Reisigl H4301 has been found a potential ArA-producer due to a high content of intracellular ArA. To gain more molecular information about metabolism pathways, including the biosynthesis of ArA in the non-model microalga, a transcriptomic analysis was performed. Results The 454 pyrosequencing generated 371,740 high-quality reads, which were assembled into 51,908 unique sequences consisting of 22,749 contigs and 29,159 singletons. A total of 11,873 unique sequences were annotated through BLAST analysis, and 3,733 were assigned to Gene Ontology (GO) categories. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis uncovered a C4-like photosynthesis pathway in M . incisa . The biosynthesis pathways of lipid particularly those of ArA and triacylglycerol (TAG) were analyzed in detail, and TAG was proposed to be accumulated in oil bodies in the cytosol with the help of caleosin or oil globule-associated proteins. In addition, the carotenoid biosynthesis pathways are discussed. Conclusion This transcriptomic analysis of M . incisa enabled a global understanding of mechanisms involved in photosynthesis, de novo biosynthesis of ArA, metabolism of carotenoids, and accumulation of TAG in M. incisa . These findings provided a molecular basis for the research and possibly economic exploitation of this ArA-rich microalga.
