Abstract:Although the phylogenetic relationships of Apis species (Hymenoptera: Apidae) have been generally accepted, genetic variation within most species and some new possible species' status have not been studied in detail. In this study, we surveyed the genetic variation of mitochondrial DNA sequences in Apis species from China and investigated the phylogeography of Apis dorsata F. in China and neighboring Asian areas. The phylogeny of Chinese Apis species was consistent with the generally accepted Apis phylogeny, and a substructure within A. dorsata was suggested. Furthermore, results of analysis of molecular variance revealed significant genetic structuring of A. dorsata between Hainan Island and China mainland. When compared with neighboring Asian areas, Chinese A. dorsata grouped with one A. dorsata sample from Palawan Island of Philippines and two samples from Malaysia. Another two samples from Malaysia clustered with one Nepalese sample and the cluster from Thailand also was supported. These results support the hypothesis that glaciations and deglaciations during the Pleistocene could have greatly influenced the distribution and divergence of A. dorsata in China and Southeast Asia. In addition, the species status of Apis dorsata breviligula Maa and Apis dorsata binghami Cockerell were supported in our study and genetic variation may exist in Apis laboriosa Smith despite of limited natural distribution.

Abstract:Malate dehydrogenase(MDH) is an important enzyme in glycometabolism. MDH of Apis mellifera showed three enzyme active zones, MDHI,MDHIIand MDHIII. MDHIand MDHIII maintained relative stability in different castes and developmental phases,but MDHIIwas polymorphic,and controlled by three alleles,a,b and c.MDH of Apis cerana was coded by S and F alleles,but some authors reported it is monomorphic.MDH was applied to the studies of A. mellifera, which included several aspects as follows: the number of queen matings,labor division in honeybee societies, the analysis of genetic constitution in honeybee populations and so on. The combination of both MDH and molecular biology will certainly promote honeybee studies.

Abstract:The sequences of mitochondrial ND2, CO2, and 16S rRNA genes and nuclear ITPR gene were obtained from 22 samples of 5 Apis species from China. The characteristics of the sequences and the pairwise distances among species were analyzed. Phylogenetic trees were reconstructed for Apis species using maximum parsimony, neighbor-joining and maximum likelihood methods together with the sequences of the other 4 Apis species downloaded from GenBank. Results supported that Apis species were divided into three major clusters: dwarf bees (A. florea and A. andreniformis), giant bees (A. dorsata and A. laboriosa), and cavity-nesting bees (A. mellifera, A. cerana, A. koschevnikovi, A. nigrocinta, and A. nuluensis). The dwarf honey bees were confirmed as basal lineage. Our study also revealed a high level of genetic divergence between A. dorsata from Hainan Island and China mainland.

Abstract:Abstract Preproapamin genes were amplified by RT‐PCR from total RNA from the venom glands of 2 honeybee species, Apis mellifera, A. cerana cerana, and 4 wasp species, Vespa magnifica, V. velutina nigrothorax and Polistes hebraeus, respectively. Their PCR products were ligated into pGEM® ‐T easy vector and the nucleotide sequences analyzed. The six fragments were all 141 bp in length and contained an ORF coding the precursor of apamin. The apamin precursors of V. magnifica, V. velutina nigrothorax and P. hebraeus had 95% and 93% similarity with that of A. melliera in nucleotide and amino acid sequences, respectively. That of Vespula maculifrons was identical to that of A. mellifera in nucleotide and amino acid sequences. Apamin precursors of V. magnifica, P. hebraeus and V. velutina nigrothorax also had the same nucleotide sequences. The nucleotide sequences of preproapamin genes from the Chinese honeybee, A. cerana cerana and 4 wasp species are described for the first time. A notable discovery was that the wasps species had exactly same apamins as the honeybees despite the fact they belong to different insect families.

Abstract:Central China has a rich terrain with a temperate monsoon climate and varied natural environments for the Chinese honeybee (Apis cerana cerana). However, little comprehensive research on population genetic diversity has been done in this area. A population survey of the structure and genetic diversity of Apis cerana cerana in this area is deeply needed for understanding adaptation to variable environments and providing more references for the protection of honeybee biodiversity. In this study, we present a dataset of 72 populations of Chinese honeybees collected from nine sites by whole genome sequencing in Central China. We obtained 2,790,214,878 clean reads with an average covering a depth of 22×. A total of 27,361,052 single nucleotide polymorphisms (SNPs) were obtained by mapping to the reference genome with an average mapping rate of 93.03%. Genetic evolution analysis was presented via the population structure and genetic diversity based on the datasets of SNPs. It showed that Apis cerana cerana in plains exhibited higher genetic diversity than in mountain areas. The mantel test between Apis cerana cerana groups revealed that some physical obstacles, especially the overurbanization of the plains, contributed to the differentiation. This study is conducive to elucidating the evolution of Apis cerana in different environments and provides a theoretical basis for investigating and protecting the Chinese honeybee.

Abstract:Odorant binding proteins(OBPs) play an important role in detection and recognition of semiochemicals for insects. In this study,the gene sequences of odorant binding protein OBP5 were cloned from five Chinese native honeybee species. Combining the sequence of Apis mellifera downloaded from GenBank,evolution of the 0BP5 in the genus Apis was analyzed. The results showed that the 0BP5 gene is relatively conserved in the genus Apis.The difference of 0BP5 gene sequences between A.cerana and A.mellifera possibly related to the different geographic environment.

Abstract:We investigated the genetic structure of Chinese Apis dorsata population based on DNA microsatellites. The results show that populations from different geographical regions are significantly differentiated. Furthermore, the A. dorsata population from Hainan Island very probably diverged from that of the China mainland and also exhibits a lower level of genetic diversity, which is probably the result of founder effects and genetic drift. The results provide useful information for effective management and conservation of A. dorsata in China, particularly with regard to the population of Hainan Island.

Abstract:Zhejiang is the traditional feeding area of Apis cerana cerana.In this paper,the genetic diversity of microsatellite DNA for Apis cerana cerana in Zhejiang was analyzed.The results showed that the genetic diversity of Apis cerana cerana in Zhejiang was relatively low.Therefore,more effective measures should be taken to protect Apis cerana cerana in Zhejiang in the future.

Abstract:Abstract Background Exploration of adaptive evolution of organisms in response to environmental change can help to understand the evolutionary history of species and the underlying mechanisms of adaptation to local environments, thus guiding future conservation programmes. Before the introduction of Apis mellifera in China, eastern honey bees ( Apis cerana ) were the only species used for beekeeping in this region. In the mountains of Changbai, populations of A. cerana are considered a distinct ecotype of the species which formed through the distinct selective pressures in this area over time. Result We performed a measure of 300 wing specimens of eastern honey bees and obtained the geometric morphological variation in the wing of A. cerana in Changbai Mountain. A total of 3,859,573 high-quality SNP loci were yielded via the whole-genome resequencing of 130 individuals in 5 geographic regions. Conclusion Corresponding geometric morphology and population genomics confirmed the particularity of the A. cerana in Changbai Mountain. Genetic differentiation at the subspecies level exists between populations in Changbai Mountain and remaining geographic regions, and a significant reduction in the effective population size and an excessive degree of inbreeding may be responsible for a substantial loss of population genetic diversity. Candidate genes potentially associated with cold environmental adaptations in populations under natural selection were identified, which may represent local adaptations in populations. Our study provided insights into the evolutionary history and adaptation of A. cerana in Changbai Mountain, as well as its conservation.

Abstract:Apis cerana is widely distributed in China and, prior to the introduction of western honeybees, was the only bee species kept in China. During the long-term natural evolutionary process, many unique phenotypic variations have occurred among A. cerana populations in different geographical regions under varied climates. Understanding the molecular genetic basis and the effects of climate change on the adaptive evolution of A. cerana can promote A. cerana conservation in face of climate change and allow for the effective utilization of its genetic resources.

Abstract:The Asian honeybee Apis cerana is one of two bee species that have been commercially kept with immense economic value. Here we present the analysis of genomic sequence and transcriptomic exploration for A. cerana as well as the comparative genomic analysis of the Asian honeybee and the European honeybee A. mellifera . The genome and RNA-seq data yield new insights into the behavioral and physiological resistance to the parasitic mite Varroa the evolution of antimicrobial peptides, and the genetic basis for labor division in A. cerana . Comparison of genes between the two sister species revealed genes specific to A. cerana , 54.5% of which have no homology to any known proteins. The observation that A. cerana displayed significantly more vigilant grooming behaviors to the presence of Varroa than A. mellifera in conjunction with gene expression analysis suggests that parasite-defensive grooming in A. cerana is likely triggered not only by exogenous stimuli through visual and olfactory detection of the parasite, but also by genetically endogenous processes that periodically activates a bout of grooming to remove the ectoparasite. This information provides a valuable platform to facilitate the traits unique to A. cerana as well as those shared with other social bees for health improvement.

Comparative analysis of C-terminal repetitive sequences of MRJP2 among six species of the genus Apis

Abstract:The cDNA library was constructed with heads of 8-day old workers of Apis cerana cerana. cDNA sequence of major royal jelly protein 2 (MRJP2), which includes 1 605 bp and an open reading frame (ORF) encoding 468 amino acid residues, was screened and obtained from this library. A repetitive fragment of variable numbers of tandem repeat (VNTR) was found at the C-terminal sequence of A. cerana cerana MRJP2 for the first time. C-terminal repetitive sequences of MRJP2 were also cloned and sequenced for other five species of Apis. The results showed that in the other five species the sequences of the C-terminal repetitive fragment performed individual polymorphism based on core bases mutation instead of different duplications, while the lengths of the repetitive sequences were almost the same among different individuals belonging to one species. A. cerana cerana and A. mellifera, A. dorsata and A. laboriosa, and A. florea and A. andreniformis formed three evolutionary branches, respectively. The branch of A. cerana and A. mellifera showed a closer relationship with the branch of A. dorsata and A. laboriosa than that with the branch of A. florea and A. andreniformis.

Abstract:The eastern honey bee ( Apis cerana ) is of great importance for ecosystems and agriculture in Asia, whose diversity has yet to be explored. Previous studies have found a high diversity of A. cerana in mountainous areas or islands. To better understand the morphological diversity of A. cerana , we focused on the eastern Qinghai-Tibet Plateau (QTP), a region with high mountains, Weizhou Island, Yongxing Island and Hainan Island, much of which lacks systematic investigation of honey bee diversity. Five sampling sites in the plain were also selected as references. A total of 140 colonies of A. cerana were collected from 9 localities in QTP, 3 localities in islands, and 5 localities in plain. We used standard morphometry and geometric morphometrics to assess the level of diversity, in which principal component analysis and discriminant analysis were conducted. The results revealed the A. cerana populations from QTP and islands were differentiated from the plain populations. Moreover, two populations from QTP and one from Yongxing Island showed significant divergence. Our study provided insights into the A. cerana diversity and provides information for the effective conservation of A. cerana genetic resources in China.

Abstract:There have been many population-based genomic studies on human-managed honeybees (Apis mellifera and Apis cerana), but there has been a notable lack of analysis with regard to wild honeybees, particularly in relation to their evolutionary history. Nevertheless, giant honeybees have been found to occupy distinct habitats and display remarkable characteristics, which are attracting an increased amount of attention. In this study, we de novo sequenced and then assembled the draft genome sequence of the Himalayan giant honeybee, Apis laboriosa. Phylogenetic analysis based on genomic information indicated that A. laboriosa and its tropical sister species Apis dorsata diverged ∼2.61 Ma, which supports the speciation hypothesis that links A. laboriosa to geological changes throughout history. Furthermore, we re-sequenced A. laboriosa and A. dorsata samples from five and six regions, respectively, across their population ranges in China. These analyses highlighted major genetic differences for Tibetan A. laboriosa as well as the Hainan Island A. dorsata. The demographic history of most giant honeybee populations has mirrored glacial cycles. More importantly, contrary to what has occurred among human-managed honeybees, the demographic history of these two wild honeybee species indicates a rapid decline in effective population size in the recent past, reflecting their differences in evolutionary histories. Several genes were found to be subject to selection, which may help giant honeybees to adapt to specific local conditions. In summary, our study sheds light on the evolutionary and adaptational characteristics of two wild giant honeybee species, which was useful for giant honeybee conservation.

Abstract:The mitochondrial DNA tRNAleu～COⅡgene in 31Apis ceranacolonies from Jinfo Mountain of Chongqing was amplified and sequenced.6Apis ceranacolonies from other places of Chongqing were also determined using the same method.Furthermore,10 mtDNA tRNAleu～COⅡgene sequences ofApis cera-nafrom other areas in China were downloaded from GenBank for comparative analysis.The alignments were performed using related softwares.The results showed that there were 4 haplotypes inApis cerana colonies from Jinfo Mountain of Chongqing,one of them was main haplotype and same as that from many other places in China,the other 3 haplotypes were different from the otherApis ceranain China.By cluster analysis,haplotypeⅡfrom Jinfo Mountain was clustered with theApis ceranafrom Haikou of Hainan Province.Apis ceranafrom Jinfo Mountain of Chongqing has the similar genetic background as theApis ceranafrom other areas in China,meanwhile,some haplotypes are unique.

Abstract:In recent years,the honeybee population is being threatened by a variety of diseases. Honeybee colonies have drastically reduced. These diseases definitely hinder the healthy development of apiculture. Essential oil is a kind of secondary metabolism in plants. It bears multiple biological activities including the insecticidal,antibacterial and antifungal activities,and so on. Compared with other chemical drugs,it has the advantage of security,pollution-free,and is not easy to produce drug resistance. It has a wide application prospect in the future. This paper reviews the recent advances of essential oils for controlling bee disease(Varroa destructor,American foulbrood,chalkbrood,stonebrood,nosema disease).
