Abstract:Although the phylogenetic relationships of Apis species (Hymenoptera: Apidae) have been generally accepted, genetic variation within most species and some new possible species' status have not been studied in detail. In this study, we surveyed the genetic variation of mitochondrial DNA sequences in Apis species from China and investigated the phylogeography of Apis dorsata F. in China and neighboring Asian areas. The phylogeny of Chinese Apis species was consistent with the generally accepted Apis phylogeny, and a substructure within A. dorsata was suggested. Furthermore, results of analysis of molecular variance revealed significant genetic structuring of A. dorsata between Hainan Island and China mainland. When compared with neighboring Asian areas, Chinese A. dorsata grouped with one A. dorsata sample from Palawan Island of Philippines and two samples from Malaysia. Another two samples from Malaysia clustered with one Nepalese sample and the cluster from Thailand also was supported. These results support the hypothesis that glaciations and deglaciations during the Pleistocene could have greatly influenced the distribution and divergence of A. dorsata in China and Southeast Asia. In addition, the species status of Apis dorsata breviligula Maa and Apis dorsata binghami Cockerell were supported in our study and genetic variation may exist in Apis laboriosa Smith despite of limited natural distribution.

Abstract:An odorant-binding protein cDNA (Acer-ASP2) was cloned and characterized from antennae of adult workers of an Asian honey bee, Apis cerana cerana F. (Hymenoptera: Apidae). The full-length open reading frame of Acer-ASP2 cDNA was 429 bp, encoding 142 amino acids. Protein signature analyses revealed that it contained six conserved cysteines with an N-terminal signal sequence of 19 amino acids. The deduced protein sequence shares high homology with Amel-ASP2 from the honey bee, Apis mellifera L., and low similarity with odorant-binding proteins from other species of insects. Immunocytochemical localization showed that Acer-ASP2 was concentrated in the lymph of olfactory sensilla, such as sensilla placodea and sensilla trichodea A. Real-time polymerase chain reaction of Acer-ASP2 transcripts showed that Acer-ASP2 was expressed on antennae but not in other general anatomical regions of the body. Temporally, Acer-ASP2 was expressed at a relatively high level in adults during two periods (9 and 27 vs. 1, 15, and 30 days). This timing is correlated with the production of beeswax and searching behavior for nectar/pollen, respectively. Thus, Acer-ASP2 is a species-specific gene that we propose to be involved in the acquisition of odorant molecules from nectar, pollen, and other general odorant sources.

Abstract:The spatio-temporal expressed profiles of two kinds of chemical communication-related protein genes, the odorant-binding protein of Ac-ASP2 and chemosensory protein of Ac-ASP3, were identified by real-time polymerase chain reaction (PCR). Results obtained using the 2 -D DCt method showed that Ac-ASP2 was a gene coding antenna-specific protein that did not express in larvae and pupae, but had discontinuous high abundance periods at 1, 9, 15, 27 and 30 days. The expressing abundance at such periods was at least ten times higher than that at other periods. From the distribution ofAc-ASP3 mRNA observed in different tissues, the transcript levels seemed to be higher in the wings, abdomen and thorax (of order � 10 6 ), and lower in the legs, antennae and head (of order � 10 5 ). From highest to lowest, the original copy number was found in the various body parts in the following order: wings, abdomen, thorax, legs, antenna, and head. The results suggest that Ac-ASP3 has an intimate relation with the chemosensory behaviour of wings and abdomen in Apis cerana cerana.

Abstract:Pheromone binding proteins(PBPs)play an important role in recognition,transmission and processing of insect pheromones.In this study,a cDNA named Ac-ASP1 encoding a PBP was cloned(from the Chinese native honeybee,Apis cerana cerana.The predicted protein has six conservative cysteines in mature peptide,a typical symbol of odorant binding proteins(OBPs).The sequence had been submitted into GenBank and registered is the accession no.DQ449670.The profile of spatio-temporal expression of Ac-ASP1 was identified by real-time PCR.The results of absolute quantitation showed that Ac-ASP1 was expressed at a higher level in antenna(2.07×106 copy number/μg),but at a less level in other tissues including head,thorax,abdomen,wings and legs(102 copy number/μg);while the results of relative quantitation revealed that Ac-ASP1 was expressed in two high abundance periods:from larva to the 6th day of adult and around the 21st day of adult.These results may provide the reference to ascertain the function of Ac-ASP1 involved in signal recognition and transmission of queen pheromone.

Multivariate Morphometric Analyses of the Giant Honey Bees, Apis Dorsata F. and Apis Laboriosa F. in China

Abstract:Multivariate morphometric analyses were performed on the giant honey bees Apis dorsata F. and Apis laboriosa F. of China. The two giant honey bee species were readily separated by principal components, cluster and discriminant analyses. Apis laboriosa showed higher values for the variables of size and cubital index than A. dorsata (ranging respectively as the width of the wax mirror 2.83 similar to 2.99 mm and 2.36 similar to 2.56 mm, width of sternum 6 3.61 similar to 3.86 mm and 3.03 similar to 3.26 mm, cubital index 10.36 similar to 14.56 and 6.38 similar to 9.61). Most of the wing venation angles were also significantly different in the two giant honey bees (angle J10 were 38.2 degrees similar to 40.7 degrees and 34.7 degrees similar to 37.7 degrees respectively). A. dorsata is distributed widely across China, and cluster and discrimination analyses indicated that they could be subdivided into two morphometric subclusters: one from the Chinese mainland and another from Hainan Island which reflects their relative geographic isolation. Few characters differed in the two A. dorsata morphometric subclusters, however, which suggests that they possibly diverged from each other very recently.
Morphometric Characters of Apis cerana cerana in Zhejiang Province

Abstract:Samples of Apis cerena cerana were collected from 36 colonies in 12 locations of Zhejiang Province.Twentyseven morphological indicators of 15 worker bees in each colony were measured.And the factor analysis,cluster analysis and variation analysis of the morphological data were analyzed by SPSS16.0.The results showed that high degrees of morphology differentiation and population differentiation existed among the honeybees in Zhejiang.Factor analysis revealed that three high eigenvalues(>3) were extracted.And they comprised 63.798% of variations data.Cluster analysis showed that the main factors could be divided into 4 groups.The first group was Yongkang,Yiwu,Hangzhou and Lishui populations.The second group was Changxing,Huangyan and Cixi populations.The third group was Jiangshan,Shengzhou and Zhoushan populations.Cangnan population which had the biggest genetic distances was classified as the fourth group.Variation analysis showed that the differences among the morphological indicators of A.cerana cerana in Zhejiang Province were significant except the number of hind wing hook.Zhoushan population had the biggest values in the length and width of the wing,the length of tergum 3 and 4,the length of sterna 4 and 7,the leg indicators.There were significant differences between Zhoushan and other populations.Cangnan population,which was significant difference with others,had the biggest values in the length of wax mirror,the distance between wax mirror and the diagonal of wax mirror.

Abstract:We investigated the genetic structure of Chinese Apis dorsata population based on DNA microsatellites. The results show that populations from different geographical regions are significantly differentiated. Furthermore, the A. dorsata population from Hainan Island very probably diverged from that of the China mainland and also exhibits a lower level of genetic diversity, which is probably the result of founder effects and genetic drift. The results provide useful information for effective management and conservation of A. dorsata in China, particularly with regard to the population of Hainan Island.

Abstract:Central China has a rich terrain with a temperate monsoon climate and varied natural environments for the Chinese honeybee (Apis cerana cerana). However, little comprehensive research on population genetic diversity has been done in this area. A population survey of the structure and genetic diversity of Apis cerana cerana in this area is deeply needed for understanding adaptation to variable environments and providing more references for the protection of honeybee biodiversity. In this study, we present a dataset of 72 populations of Chinese honeybees collected from nine sites by whole genome sequencing in Central China. We obtained 2,790,214,878 clean reads with an average covering a depth of 22×. A total of 27,361,052 single nucleotide polymorphisms (SNPs) were obtained by mapping to the reference genome with an average mapping rate of 93.03%. Genetic evolution analysis was presented via the population structure and genetic diversity based on the datasets of SNPs. It showed that Apis cerana cerana in plains exhibited higher genetic diversity than in mountain areas. The mantel test between Apis cerana cerana groups revealed that some physical obstacles, especially the overurbanization of the plains, contributed to the differentiation. This study is conducive to elucidating the evolution of Apis cerana in different environments and provides a theoretical basis for investigating and protecting the Chinese honeybee.

Abstract:The sequences of mitochondrial ND2, CO2, and 16S rRNA genes and nuclear ITPR gene were obtained from 22 samples of 5 Apis species from China. The characteristics of the sequences and the pairwise distances among species were analyzed. Phylogenetic trees were reconstructed for Apis species using maximum parsimony, neighbor-joining and maximum likelihood methods together with the sequences of the other 4 Apis species downloaded from GenBank. Results supported that Apis species were divided into three major clusters: dwarf bees (A. florea and A. andreniformis), giant bees (A. dorsata and A. laboriosa), and cavity-nesting bees (A. mellifera, A. cerana, A. koschevnikovi, A. nigrocinta, and A. nuluensis). The dwarf honey bees were confirmed as basal lineage. Our study also revealed a high level of genetic divergence between A. dorsata from Hainan Island and China mainland.

Abstract:Abstract Background Exploration of adaptive evolution of organisms in response to environmental change can help to understand the evolutionary history of species and the underlying mechanisms of adaptation to local environments, thus guiding future conservation programmes. Before the introduction of Apis mellifera in China, eastern honey bees ( Apis cerana ) were the only species used for beekeeping in this region. In the mountains of Changbai, populations of A. cerana are considered a distinct ecotype of the species which formed through the distinct selective pressures in this area over time. Result We performed a measure of 300 wing specimens of eastern honey bees and obtained the geometric morphological variation in the wing of A. cerana in Changbai Mountain. A total of 3,859,573 high-quality SNP loci were yielded via the whole-genome resequencing of 130 individuals in 5 geographic regions. Conclusion Corresponding geometric morphology and population genomics confirmed the particularity of the A. cerana in Changbai Mountain. Genetic differentiation at the subspecies level exists between populations in Changbai Mountain and remaining geographic regions, and a significant reduction in the effective population size and an excessive degree of inbreeding may be responsible for a substantial loss of population genetic diversity. Candidate genes potentially associated with cold environmental adaptations in populations under natural selection were identified, which may represent local adaptations in populations. Our study provided insights into the evolutionary history and adaptation of A. cerana in Changbai Mountain, as well as its conservation.

Abstract:The eastern honey bee ( Apis cerana ) is of great importance for ecosystems and agriculture in Asia, whose diversity has yet to be explored. Previous studies have found a high diversity of A. cerana in mountainous areas or islands. To better understand the morphological diversity of A. cerana , we focused on the eastern Qinghai-Tibet Plateau (QTP), a region with high mountains, Weizhou Island, Yongxing Island and Hainan Island, much of which lacks systematic investigation of honey bee diversity. Five sampling sites in the plain were also selected as references. A total of 140 colonies of A. cerana were collected from 9 localities in QTP, 3 localities in islands, and 5 localities in plain. We used standard morphometry and geometric morphometrics to assess the level of diversity, in which principal component analysis and discriminant analysis were conducted. The results revealed the A. cerana populations from QTP and islands were differentiated from the plain populations. Moreover, two populations from QTP and one from Yongxing Island showed significant divergence. Our study provided insights into the A. cerana diversity and provides information for the effective conservation of A. cerana genetic resources in China.

Old Comb for Nesting Site Recognition by Apis Dorsata? Field Experiments in China

Abstract:The Asian giant honey bee, Apis dorsata, often conducts seasonal, long-distance migrations in southern China, between a preferred tree (having more than one nest) and alternate sites. Although worker bees cannot make a round-trip journey, colonies re-utilize preferred trees after an absence of several months. We performed comb experiments in which bases and all abandoned combs were entirely scraped off trees and their sites covered with plastic, or comb was moved to trees of the same species. Swarms of giant honey bees investigated trees where combs were removed and continued to nest on the same trees. In contrast, placing combs in nets on previously used trees, or on nearby trees of the same species, did not attract more swarms. The same number of colonies that left them returned to previously occupied trees. Our findings suggest that direct olfactory or sensory contact with old comb bases might regulate nest establishment, but individual trees, lacking normal visual or chemical cues of old nests, are relocated using behavioral devices that remain to be elucidated.

Abstract:The sensilla on the antenna of the worker of the' Chinese honey bee, Apis cerana, are studied with scanning electron microscope., The types of sensilla present on the antenna include sensillum placqdeum, s. coelonconicum, s. ampullaceum, s. basiconicum, s. campaniformium, s. trichodeum A, B, C and D, and several forms of setae. The distribution and external morphology of the antenhal sensilla of this species differ from Ithat of the Italian honey bee Apis mel-Jifera.
Morphological and Molecular Identification of Chalkbrood Disease Pathogen Ascosphaera Apis in Apis Cerana Cerana

Abstract:Chalkbrood is a fungal disease of honey bees and leads to serious apicultural loss worldwide. Ascosphaera apis, the pathogen of chalkbrood, has been found in Apis mellifera, Apis cerana, Xylocopa californica arizonensis larvae as well as adult bumble bees. Here, the fungal pathogen of Apis cerana cerana drone mummies was isolated and identified using morphological and molecular methods. Morphological observation indicated that the sizes of the fruiting bodies, spore balls and ascospores of the fungus isolated from A. c. cerana drone mummies were similar to those of A. apis. Phylogenetic analysis suggested that the fungus is identical to A. apis. Furthermore, the results of a cross infection assay demonstrated that the isolated fungus is capable of infecting eastern and western honey bee larvae and results in chalkbrood. These results confirmed that the isolated fungal pathogen is A. apis. This is the first documentation of morphological and molecular identification of A. apis in A. c. cerana. Our results not only provide novel insight to better understand the pathology of A. apis, but also lay a solid foundation for further investigations of host responses and pathogen-host interactions during chalkbrood of eastern honey bee larvae.

Abstract:Apis cerana is widely distributed in China and, prior to the introduction of western honeybees, was the only bee species kept in China. During the long-term natural evolutionary process, many unique phenotypic variations have occurred among A. cerana populations in different geographical regions under varied climates. Understanding the molecular genetic basis and the effects of climate change on the adaptive evolution of A. cerana can promote A. cerana conservation in face of climate change and allow for the effective utilization of its genetic resources.

Abstract:There have been many population-based genomic studies on human-managed honeybees (Apis mellifera and Apis cerana), but there has been a notable lack of analysis with regard to wild honeybees, particularly in relation to their evolutionary history. Nevertheless, giant honeybees have been found to occupy distinct habitats and display remarkable characteristics, which are attracting an increased amount of attention. In this study, we de novo sequenced and then assembled the draft genome sequence of the Himalayan giant honeybee, Apis laboriosa. Phylogenetic analysis based on genomic information indicated that A. laboriosa and its tropical sister species Apis dorsata diverged ∼2.61 Ma, which supports the speciation hypothesis that links A. laboriosa to geological changes throughout history. Furthermore, we re-sequenced A. laboriosa and A. dorsata samples from five and six regions, respectively, across their population ranges in China. These analyses highlighted major genetic differences for Tibetan A. laboriosa as well as the Hainan Island A. dorsata. The demographic history of most giant honeybee populations has mirrored glacial cycles. More importantly, contrary to what has occurred among human-managed honeybees, the demographic history of these two wild honeybee species indicates a rapid decline in effective population size in the recent past, reflecting their differences in evolutionary histories. Several genes were found to be subject to selection, which may help giant honeybees to adapt to specific local conditions. In summary, our study sheds light on the evolutionary and adaptational characteristics of two wild giant honeybee species, which was useful for giant honeybee conservation.
Diversity of Plants Foraged by Apis cerana Fabricius Around Qinling Mountains (Central China) Based on Honey Pollen Samples

Abstract:To illustrate the essential role of Apis cerana Fabricius in plant pollination, a survey was conducted using PCR, DNA barcoding, and high-throughput sequencing technologies to assess the diversity of plants foraged by A. cerana at five sites around Qinling Mountains. A fragment of the rbcL gene in pollen isolated from honey collected at each site was amplified and sequenced. The results show that samples BJ and TC, samples FP and AK have a high correlation, respectively. According to the information of a total of 64 plant species identified in the five sites, belonging to 53 genera, 37 families, and 23 orders, it is found that there is a relatively high diversity and complex composition of nectar and pollen plants and that most of the plants are sporadically available. The identified species show higher occurrence in some orders, such as Sapindales, Rosales, etc. There are significantly more plants of temperate origin than those of tropical origin, 58.5% and 34.0%, respectively. Some species of Rosales or Sapindales are more visited by A. cerana , with Toxicodendron vernicifluum (Stokes) being the dominant species. These results provide a basis for assessing the role of A. cerana in plant pollination and maintaining biodiversity.

Bees can pollinate in various environments and climates and play important roles in maintaining the ecological balance of the earth. Results: We performed an analysis of 462 Apis cerana (A. cerana) specimens from 31 populations in 11 regions and obtained 39 representative morphological features. We selected 8 A. cerana samples from each population and performed 2b-RAD simplified genome sequencing. A total of 11,506 high-quality single nucleotide polymorphism (SNP) loci were obtained. For these SNPs, the minor allele frequency (MAF) was > 1%, the average number of unique labels for each sample was 49,055, and the average depth was 72.61x. The ratios of the unique labels of all samples were 64.27-86.33%. Conclusions: Using 39 morphological characteristics as the data set, we proposed a method for the rapid classification of A. cerana. Using genomics to assess population structure and genetic diversity, we found that A. cerana has a large genetic difference at the ecotype level. A comparison of A. cerana in North China revealed that some physical obstacles, especially the overurbanization of the plains, have isolated the populations of this species. We identified several migration events in North China and Central China. By comparing the differences in the environmental changes in different regions, we found that A. cerana has strong potential for climate change and provides a theoretical basis for investigating and protecting A. cerana.
