Natural Products Ribosomally synthesized and post-translationally modified peptides (RiPPs) are a rapidly growing class of natural products with a high degree of structural and functional diversity. These compounds are typically biosynthesized from a larger precursor peptide that consists of a leader and a core peptide, which is sometimes flanked by recognition sequences (RS). The post-translational modifications take place in the core peptide, but the enzymes that carry out these modifications usually recognize the leader peptide and/or the RS. The current understanding regarding leader peptide recognition and enzyme activation is discussed in the Concept article by W. A. van der Donk and X. Yang on page 7662 ff.1 Organocatalysis The Mannich reaction is one of the most powerful methods for the synthesis of nitrogen-containing molecules and the asymmetric catalytic Mannich reaction of functionalized imines can afford synthetically useful chiral building blocks. On page 7678 ff., Y. Hayashi and co-workers report the domino reaction of desulfonylative formation of α-keto N-Boc imines and asymmetric Mannich reaction catalyzed by a Jørgensen–Hayashi catalyst, such as diarylprolinol silyl ether, to afford the Mannich product containing several functional groups with excellent anti selectivity and enantioselectivity.1 Tetrahydroisoquinolines Monitoring progress in organic reactions is possible by in situ IR spectroscopy. This is particularly suited to organometallic chemistry in which the metal coordinates to a carbonyl group, changing the carbonyl stretching frequency on reaction. In their Full Paper on page 7724 ff., I. Coldham and co-workers show how this technique was used to optimize the lithiation of N-Boc-tetrahydroisoquinolines.1
