Abstract:An efficient and selective method for the deprotection of aryl tert-butyldimethysilyl (TBS) ethers is described. The protecting group TBS could be cleaved from aryl silyl ethers in the prensence of alkyl TBS ethers using KOH in ethanol at room temperature to give the corresponding phenols in excellent yields (87-99%).

Abstract:We report a mild protocol for removal of the p-methoxybenzyl ether protecting group under acidic conditions with eosin Y combined with LEDs as a photoredox catalysis system and hydrogen peroxide as the terminal oxidant. This protocol is compatible with ethers derived from primary, secondary, and tertiary alcohols, as well as with various functional groups. The protocol showed unusual selectivity for a tertiary ether over a primary ether. The scale up to gram scale is also explored and identical reactivity is observed.

Abstract:A tetrabutylammonium peroxydisulfate-mediated oxidative procedure for the mild selective deprotection of allyl ethers to form the corresponding alcohols is described. Abstract:A mild and efficient protocol for the deprotection of silyl ethers using KF in tetraethylene glycol is reported. A wide range of alcoholic silyl ethers can be selectively cleaved in high yield in the presence of certain acid- and base-labile functional groups. Moreover, the phenolic silyl ethers were cleaved exclusively, without affecting the alcoholic silyl ethers, at room temperature.

Abstract:A novel isomerization of O-allyl glycosides into prop-1-enyl glycosides was observed instead of cross-metathesis during an olefin metathesis reaction using Grubbs' ruthenium benzylidene catalyst (Cy3P)2RuCl2=CHPh (1), N-allyltritylamine, and N,N-diisopropylethylamine as necessary auxiliary reagents. In the search for a better catalytic system, it has been found that dichlorotris(triphenylphosphine)ruthenium(II), [(C6H5)3P]3RuCl2, (2) was much more efficient for the isomerization of allylic ethers. The labile prop-1-enyl group was easily hydrolyzed using HgCl2-HgO and the hemiacetals (25-32) were isolated in excellent yields (ca. 90%).Key words: allyl ether, carbohydrate, Grubbs' catalyst, isomerization, metathesis, deprotection.

Abstract:Dealkylation of alkyl aryl ethers has been extensively explored. In comparison to the reported methods, selective methods that work under mild reaction conditions remain to be developed. Herein we report the development of a method that is selective, dealkylating only one of the methyl groups in polymethylated aryl ether system. This selective dealkylative method is applied to the synthesis of a few advanced intermediates for drug synthesis. Detailed mechanistic studies have shown that this reaction probably proceeds via a single‐electron transfer process.

Abstract:Primary alcohols were selectively tetrahydropyranylated in good to excellent yields at room temperature using PdCl2(CH3CN)(2) as catalyst in tetrahydrofuran (THF) in the presence of phenols, secondary, and tertiary alcohols. The tetrahydropyranyl (THP) group could be efficiently removed using PdCl2(CH3CN)(2) as catalyst in CH3CN, while other protection groups such as p-toluenesulfonyl (Ts), tert-butyldiphenylsilyl (TBDPS), benzyloxycarbonyl (Cbz), allyl, benzyl (Bn), and benzoyl (Bz) remained intact under these conditions. Abstract:A mild and efficient method to selectively cleave p-methoxybenzyl (PMB) ether with a catalytic amount of Co2(CO)8, hydrosilane and CO (1atm) is presented. The cleavage reaction shows regioselectivity to primary O-PMB of a variety of permethoxybenzylated saccharides and chemoselectivity to O-Bn, sulfur-containing group and common ester protecting groups.

Abstract:An efficient, chemoselective, and environment-friendly method for the deprotection of tert-butyldiphenylsilyl ethers mediated by triflic acid supported on silica gel is reported. A wide range of tert-butyldiphenylsilyl ethers derived from carbohydrate and saponin residues can be smoothly cleaved in the presence of various types of other protecting groups in good to excellent yields in acetonitrile. This heterogeneous reaction does not require aqueous workup, and the supported catalyst can be readily recycled.

Abstract:An efficient and practical demethylation of ortho substituted aryl methyl ethers using AlCl3 has been developed. This method gives a high conversion, is simple to operate and is cost-effective. A mechanism involving the complexation of AlCl3 with the OMe and the adjacent electron withdrawing group is proposed. Many functional groups can be tolerated in the demethylation process, and 29 examples gave a demethylated product in a yield of 90-98%.

Abstract:Commercially available zinc dust in the presence of ammonium chloride in acetonitrile at reflux removes the 2,2,2-trichloroethyl (TCE) group at anomeric centers with excellent yields (>95%) in short reaction times. This present method is easily implemented on substrates containing acyl and benzyl groups and large-scale reactions also proceed in high yield.

Abstract:An organo-photocatalyzed approach for the chemoselective dealkylation and de-esterification of phenols is developed by employing trimethyl silyl chloride as the C‒O bond activator and 9-Mes-10-MeAcr+ClO4‒ as the photoredox catalyst. This method demonstrates an exceptional selectivity towards the cleavage of phenolic ethers and esters over equivalent aliphatic scaffolds, presenting a broad range of functional group sustainability. This strategy also enables selective debenzylation of phenols in the presence of reduction-sensitive functional groups. The photocatalytic efficiency was further extrapolated in the fragmentation of lignin models and synthetic modification in manufacturing a FLAP inhibitor. Mechanistic studies, photophysical experiments and computations provide evidence for the involvement of an oxonium intermediate, generated through the photo-oxidation of the arene ring, which ultimately leads to the selective disintegration of C‒O bond, facilitated by the oxophilicity of silicon.

Abstract:The first aerobic protocol of direct transformation of p‐methoxybenzyl (PMB) ethers to carboxylic acids efficiently with Fe(NO3)3•9H2O and TEMPO as catalysts at room temperature has been developed. The reaction accommodates C‐Br bond, terminal/non‐terminal C‐C triple bond, amide, cyano, nitro, ester, and trifluoromethyl groups, etc. Even highly selective oxidative deprotection of different benzylic PMB ethers has been realized. The reaction has been successfully applied to the total synthesis of natural product, (R)‐6‐hydroxy‐7,9‐octadecadiynoic acid, demonstrating the practicality of the method. Based on experimental studies, a possible mechanism involving oxygen‐stabilized benzylic cation has been proposed.
