Abstract:Cutting propagation test of Jatropha curcas collected from Simao Port of Puer City were carried out.with hormones of different types and concentrations in the processing of different length and diameter of the ear.using comparative analysis,variance analysis and multiple comparison methods to analyze the test results.The results showed that the treatment combination with cutting lengths of 50 cm,spike bar diameter of 1.2 ~ 2.0 cm,hormone ABT1 100 mg / L has the highest rooting rate.

Abstract:Enhanced axillary branching and rooting induction of Jatropha curcas were researched,to save,exploit and optimize the qualities of J.curcas.The results showed that compounding of KT、2,4-D was good for callus inducement;axillary buds induced well in the medium of MS+1.0mg/L BA+0.01 mg/L IBA+(5.0mg/L AgNO_(3);)effect of pH was low;ratio of rooting could get to 92%, both by stimulated with high concentrationof auxin short time or cultured in the active carbon medium.

Abstract:The cutting experiment of Castanopsis hystrix including different media and different hormone of vary concentrations was conducted.The results showed that cutting medium affected the rooting percentage of cuttings significantly,the muddy loess was preferable for cuttings of Castanopsis hystrix.Treating with hormone could speed up the rooting of cuttings,different hormone and concentrations affected the rooting ratio and rooting speed significantly,the rooting ratio of the optimum treatment obtained 90 %.

Abstract:An efficient and reproducible protocol for induction of adventitious shoot buds and plant regeneration from petiole explant cultures of Jatropha curcas, an important biofuel crop, is described. Physiologically mature trees of three J. curcas genotypes were selected and explants were prepared from young petioles. Treating the explants with high concentrations (5 to 120 mg/L) of thidiazuron (TDZ) solution for short time periods (5 to 80 min) helped increase the regeneration frequency and improved the quality of the regenerated buds significantly. The age of the petioles and inoculation methods were found to influence the culture results. The best shoot buds induction (65.78%) and number of buds (6.77) per explant was seen in the second petiole explants of genotype M-1 treated with 20 mg/L TDZ solution for 20 min, followed by 35-day culture on hormone-free Murashige and Skoog medium. The regenerated buds could elongate to become shoots in a medium containing gibberellic acid. The elongated shoots initiated roots to become intact plantlets in rooting medium containing indole-3-butyric acid and L-glutamine (Gln), and supplementing 16 mg/L Gln into the rooting medium effectively stimulated the initiation and growth of roots, with the best rooting rate (51.72%). After acclimatization, these plantlets were transplanted to soil wherein normal growth was observed. Therefore, an intact plantlet could usually be obtained at 60 days of culture by using the culture protocol described in this study. This protocol can be used for mass production of true-to-type plants and the production of transgenic plants through Agrobacterium/biolistic-mediated transformation.

Abstract:To study cutting propagation techniques with one-year-old cuttings of C lematis finetiana and to in-vestigate the dynamic changes in activity of related oxidative enzymes [peroxidase (POD), indoleacetic acid oxidase (IAAO), and polyphenol oxidase (PPO)] as well as the content of soluble sugar, soluble protein, and starch, an L9 (34) orthogonal experiment was conducted with four treatments of exogenous hormones, hormone concentration, soaking time, and cutting matrix. Results indicated that rooting of C. finetiana was derived from the cortex. The effects of different exogenous hormones, hormone concentration, soaking time, and cutting ma-trix on rooting were notable by variance analysis. The best rooting formula was as follows:a matrix of river sand processed with Indole-3-butyric acid (1H-Indole-3-butanoic acid (IBA) 100 mg·L-1 for 2 h producing a rooting rate of 22.7%. In the rooting process, the content of soluble sugar increased during the induction and root growth periods, but decreased during the expression period and the late growth stage. Soluble protein content decreased gradually after cutting reaching a low point when most cuttings rooted. Starch content declined throughout the process. Also, the POD, IAAO, and PPO activities all increased first and then decreased reach-ing a peak in the early stages of expression. Additionally, greater nutrient content for the test group was found than the control, and peak oxidase activities of test groups arrived 10 d earlier than the control. This demon-strated that exogenous hormones could improve the nutrient utilization and oxidase activities of cuttings. [Ch, 3 fig. 4 tab. 20 ref.]
Factors Influencing Direct Shoot Regeneration from Mature Leaves of Jatropha Curcas, an Important Biofuel Plant

Abstract:In order to establish a highly efficient and sustainable regeneration system, we systematically researched the key factors affecting direct shoot regeneration from Jatropha curcas leaves that were collected from Hainan (HN1-1), Lijiang (LJ3-1), and Yuxi (YX2-12) provinces in China. The L9(34) orthogonal test of thidiazuron (TDZ), kinetin (Kn), and gibberellic acid (GA3) were studied, and the explant type, growth age, and cultivar of leaves were subsequently investigated. Simultaneously, the combinations of plant growth regulators (PGRs) promoting shoot bud proliferation, elongation, and root establishment were examined. The results showed that the best medium for shoot bud induction was Murashige and Skoog (MS) medium supplemented with 1.0 mg/L TDZ, 0.5 mg/L Kn, and 0.5 mg/L GA3. TDZ was the key PGR, while Kn and GA3 played an important role in shoot bud elongation and the number of shoots per leaf disk, respectively. The induced shoot buds proliferated and readily elongated in MS medium with 0.3 mg/L 6-benzylaminopurine and 0.01 mg/L indole-3-butyric acid (IBA) and established roots in half-strength MS medium supplemented with 2.0 mg/L IBA. Using the previously described methods, the third to fifth leaves were found to be the best explant source for shoot bud induction, with a high induction rate, large shoot numbers per disk, excellent proliferation, and consistent rooting. With the use of this regeneration system, the shoot bud induction rate increased from the reported rate of 53.5% to more than 90% using different explants and cultivars, and the shoot number per leaf disk (shoot length ≥ 0.5 cm) increased from 1.6 to 3.5. Thus, this optimized regeneration system will effectively promote the propagation and genetic transformation of J. curcas.

Abstract:In order to improve the cutting propagation effects of cut rose,the authors carried on the cutting propagation experiment with L9(34) orthogonal test,including four factors(substrate,hormone types,shade,cutting depth) of Corolla.The results showed that substrate had the highest effects on rooting rate,root length and root coefficient than the other three factors.The rooting rate would be up to 93.33% under the optimum conditions,including substrate soil+vermiculite+saw dust(45∶35∶20),ABT 200 mg/L,shading 50%,cutting depth three eyes left.Meanwhile,the underground part had better growth vigor and higher indexes of root weight,leaf weight,root length and root activity.So the optimum cutting propagation substrate should be used in production extension.

Abstract:Studies on the effect for the explant regeneration and the quality of seedlings of Jatropha curcas showed that MS supplied with 6-BA 3 mg·L-1+IBA 0.5 mg·L-1 could obviously promote the explants' differentiation.The explants of 15-day-old seedlings have a higher regenerate rate.Studies on the best hormone density in promoting differentiation,using orthogonal experiment for the tests of 6-BA+IBA and 6-BA+NAA,indicated that the combination of 6-BA and IBA was better than that of 6-BA and NAA.The best cotyledon differentiated medium was MS+6-BA 3 mg·L-1+IBA 0.5 mg·L-1 and the differentiated rate was 82.9%.The best differentiated medium for hypocotyls bud was MS+6-BA 2 mg·L-1+IBA 0.2 mg·L-1,and the differentiate rate was 23.8%.Adding 1 mg·L-1 AgNO3 into the regeneration medium of cotyled and hypocotyls can promote the bud differentiation,and inhibit browning.The WPM medium is the best one in rooting of the regeneration buds.

Abstract:The seedling leaves grown in greenhouse were used as explants to study the effects of different concentrations of 6-BA and IBA added in MS media on the regeneration of adventitious shoots.Cultured in the dark condition for 60 days,it is found that the optimal hormone combination for callus induction and adventitious shoots induction is 6-BA 5 mg·L-1 and IBA 0.5 mg·L-1.The induction rate of adventitious shoots is as high as 75.8%.Based on the optimal combination,the effect of gibberellic acid(GA3) on the regeneration of adventitious shoots was studied.The regeneration frequency of leaf-discs could reach to 90.9% and the average number of adventitious shoots was up to 4.6 when the concentration of GA3 was 0.05 mg·L-1.
Study on Raising Seedlings of Jatropha Curcas L. by Grafting, in Panxi Region.

Abstract:Raising seedlings by grafting in Jatropha curcas L. (JC)could quickly fix the fine properties ofimproved variety and shorten its breeding period.5 months seedlings were selected as parental rootstocks and 1-2a branches on 3a saplings as scions,the impact of different grafting methods including whip grafting (cleft grafting and cut grafting),budding (plate budding and single bud cut grafting)and grafting time on grafting survival rate were conducted,Meanwhile,high grafting was used in the first order branching of 4-5a saplings.The results showed that the survival rate of JC in Panxi region all can be up to 100 % in period of germination (dry season) by using all aboved grafting methods.There was not obviously different in survival rate between whip grafting and budding grafting after 30 days.The rate of scion germinate both grafted exceeded 90 % after 20 days,which indicated that stages in germination of JC concentrated in 10-20 days grafted.The growth increment in the later of scion by budding was obviously higher than that by means of whip grafting.The survival rate exceeded 90 % by high grafting using budding.There was strong linear regression relationship between thickness of rootstocks and the length of tassel,which equation of regression was:y=11.342+19.753x(P=0.001).There was relatively strong linear regression relationship between the thickness of scion and the length of tassel that equation of regression was:y=11.973+35.442x(P=0.001).This grafting technique was appropriate for clone propagation of the improved variety and reforming low efficient stands.

Abstract:The microspores at late uninucleate stage were used as explants in anther culture of Jatropha curcas L..The processes include callus induction,callus differentiation,shoots growth,and rooting. The most efficient callus induction medium is N6+2,4-D 1.0 mg/L+KT 0.4 mg/L+10 % sucrose and the cultures should be kept in darkness.The best differentiation medium is MS+TDZ 2.0 mg/L+IAA 2.0 mg/L+CH 1g/L+ 3% sucrose and the TDZ plays an important role during the shoots differentiation. The optimal culture medium for shoots growth is MS+GA3 1.0 mg/L+6-BA 0.5 mg/L+IBA 1. 0 mg/L+3% sucrose.The best rooting medium is 1/2 MS+IBA 0.2 mg/L+IAA 0.2 mg/L+3% sucrose and the rooting rate is 40 %.

Abstract:Fertilization is one of the important measures to promote the growth,yield and quality of tree.The effect of various levels of fertilizers on the growth and fruiting of Jatropha curcas L.was studied in this paper.The results showed that the height,new shoot increment,stem diameter,fruiting number and thousand fruit weight increased with increase of fertilizer rate;the fruiting branches rate,fruiting rate and kernel rate first increased and then decreased.When the fertilizer rate was 0.4 kg/strain/year,the fruiting branches rate and kernel rate reached the highest,were 81.54 % and 64.18 % respectively;When the fertilizer rate was 0.2 kg/strain/year,the fruitingrate hit 95.82 %.

Abstract:It was proved by our experiment that the subsoil was the best and the perlite was the better and the chaff of coconut was the worst medium for the coppice sprouts from the superior tissue culture shoots of Acacia crassicarpa with different concentration of NAA、ABT、IBA.The best rooting rate was 87% when the NAA was used as the growth regulator,and ABT the better and IBA the worst one .Different period of rooted cutting have different effect on rooting rate,July is better than September.

Abstract:The explants of Jatropha curcas, hypocotyl, blade and petiole, were used for the test on MS with BA and IBA. The induction of shoot bud was recorded highest on MS medium with 0. 5 mg/L BA and 1 mg/L IBA from blade. Under the condition with the same BA concentration, if IBA concentration was reduced, it would be obviously effective for budding from hypocotyl callus. The petioles required much lower concentrations of the two growth regulators (0. 1 mg/L BA + 0. 1 mg/L IBA) . Regenerated shoots were rooted on growth regulator-free MS medium and could be transplanted in soil after simply hardening for several days. Fig 1 , Tab 2, Ref 13
Establishment of an Agrobacteriuim -Mediated Cotyledon Disc Transformation Method for Jatropha Curcas

Abstract:Jatropha curcas contains high amounts of oil in its seed and has been considered for bio-diesel production. A transformation procedure for J. curcas has been established for the first time via Agrobacterium tumefaciens infection of cotyledon disc explants. The results indicated that the efficiency of transformation using the strain LBA4404 and phosphinothricin for selection was an improvement over that with the strain EHA105 and hygromycin. About 55% of the cotyledon explants produced phosphinothricin-resistant calluses on Murashige and Skoog (MS) medium supplemented with 1.5 mg l−1 benzyladenine (BA), 0.05 mg l−1 3–indolebutyric acid (IBA), 1 mg l−1 phosphinothricin and 500 mg l−1 cefotaxime after 4 weeks. Shoots were regenerated following transfer of the resistant calli to shoot induction medium containing 1.5 mg l−1 BA, 0.05 mg l−1 IBA, 0.5 mg l−1 gibberellic acid (GA3), 1 mg l−1 phosphinothricin and 250 mg l−1 cefotaxime, and about 33% of the resistant calli differentiated into shoots. Finally, the resistant shoots were rooted on 1/2 MS media supplemented with 0.3 mg l−1 IBA at a rate of 78%. The transgenic nature of the transformants was demonstrated by the detection of β-glucuronidase activity in the primary transformants and by PCR and Southern hybridization analysis. 13% of the total inoculated explants produced transgenic plants after approximately 4 months. The procedure described will be useful for both, the introduction of desired genes into J. curcas and the molecular analysis of gene function.
Effects of mixed substrates of different agricultural and forestry residues on the cutting seedlings of Thuja sutchuenensis

Abstract:To screen environment-friendly seedling cultivation substrates which could replace peat and with less cost, we compared the effects of different agricultural and forestry residue mixed substrates on cutting propagation of Thuja sutchuenensis, in an experiment following randomized block design. There were five types of mixed substrates, including peat + vermiculite + perlite (T1), edible mushroom residue (EMR) + vermiculite + perlite (T2), carbo-nized rice husk (CRH) + vermiculite + perlite (T3), EMR + slag + sawdust (T4) and CRH + EMR + slag (T5). The results showed that the bulk density of T3 was the lowest, followed by T2, which significantly differed from other mixed substrates. The non-capillary porosity of T2 was significantly greater than that of T1, while the capillary porosity and the total porosity of T2 was lower than T1 and T3, respectively. T2 had the highest contents of total nitrogen, total phosphorus, total potassium, alkali-hydrolyzed nitrogen, available phosphorus, substrate moisture and the highest pH, which differed significantly from other mixed substrates in most chemical indicators. The membership function values of rooting rate and growth indicators of cuttings with different mixed substrates were in order of T2 > T3 > T1> T5 > T4. Most indicators with larger grey relation values were physical indicators. The top five indicators were capillary water capacity, total potassium, field water capacity, maximum water capacity, and total porosity, with both capillary water capacity and total potassium content ranking first. In general, the physicochemical properties, rooting rate, and growth characteristics of cuttings under T2 were better than those of other mixed substrates. The capillary water capacity and total potassium were the main factors affecting rooting and growth of cuttings. At the early stage of cutting, the physical properties of mixed substrate had greater effect on rooting rate and growth of cuttings than the chemical properties. Overall, our results suggested that T2 should be preferred in the cutting propagation of T. sutchuenensis.

Abstract:In this paper, plumules, cotyledons, hypocotyls, leaf blades, petioles and stalks of physic nut （Jatropha curcas L.） were used as explants, and callus induction and plant regeneration were studied on MS medium contained different concentrations of 6-BA and NAA. The results showed that the MS medium with 5.0 mg/L BA and 1.0 mg/L NAA was the best for callus induction, and with 5.0 mg/L BA and 0.1 mg/L NAA, for formation of adventitious buds, and with 1.0 mg/L BA and 1.0 mg/L NAA, for bud growth, and that 1/2 MS medium with 1.0 mg/L NAA was the best for formation and growth of adventitious roots.
Effects of different substrates, growth regulators, grades of cuttings and metabolism regulator on cutting propagation of Juniperus squamata 'Blue Star'

Abstract:Effects of substrates,growth regulators,grades of cuttings and metabolism regulators on the cutting propagation of Juniperus squamata ‘Blue Star’ were researched,and five different grades of cuttings were compared between rooting process and rooting type. Results showed that the rooting rate of the cuttings with the substrate of perlite increased significantly. ABT~1# was superior to other growth regulators on the rooting rate,rooting quality and one-side rooting rate of both softwood and wood cuttings. The optimized combinations were the softwood cuttings treated by perlite+ABT~1# 1 000 mg/L(quickly dipped for 2-3 seconds),and wood cuttings treated by perlite+ABT~1# 500 mg/L (soaked for 1 hour). The rooting rate reached 66.7% and 60.17% separately,and for wood cuttings,the one-side rooting rate was only 28.0%.The rooting ability of cutting grade Ⅰ was the best with 80% rooting rate and better rooting qualities compared with others,the lower rooting ability followed was grade Ⅳ,Ⅱ,Ⅲ,and that of grade Ⅴ was the worst. Metabolism regulators(MS+NaHSO_3) had obvious effects on the rooting ability. The rooting rate of gradeⅤrose by 34.9% compared with the control,when treated by 1/4MS+NaHSO_3(200 mg/L).The same rooting process and different rooting characteristics of five grades of cuttings were found. The adventitious roots of cuttings from grade Ⅰ to Ⅳ mostly originated from cortex of cuttings,but grade Ⅴ from callus tissue of the cuttings' bottom section.

Abstract:With river sand as cutting medium and annual shoots from 2 -year -old mother tree of Ptero-celtis tatarinowii as materials,the influences of different concentrations and treating times of 4 kinds of hor-mones,ABT1,ABT2,special -effect ABT and IBA,and solution of rooting raw powder on cutting effects were studied using cutting woods without any treatment as control.The results showed that the survival rate of Ptero-celtis tatarinowii treated with 250 mol /L solution of rooting raw powder was the highest,which reached to 85%;followed by control treatment with the survival rate of 60%;the survival rates of hormone treatments were rela-tively lower,which ranged from 10% to 50%.The average root number of cutting woods treated with solution of rooting raw powder reached to 18 and the average root length was 7.25 cm,which were superior to other treat-ments.

Abstract:Abstract Background and objective A simple, rapid, efficient, and reproducible protocol for callus induction and regeneration of plantlets from callus of Jatropha curcas plant was established. Materials and methods Randomly amplified polymorphic DNA (RAPD) and inter simple sequence repeat (ISSR) analyses were used to determine the genetic variation between the regenerated, micropropagated, and mother plants. Results The highest callus induction percentage from leaf explant was recorded with MS medium containing 2.5 mg/l BA ( 6-benzylaminopurine ) + 1.0 mg/l NAA (1-naphthaleneacetic acid). Leaf-derived callus was grown on medium containing 2.0 mg/l BA + 0.2 mg/l IBA (indole-3-butyric acid) for adventitious shoot regeneration. In addition, using five random RAPD primers with the tested samples produced 117 amplified products out of which 25 were polymorphic bands resulting in 21.37% polymorphism, whereas the five ISSR primers used yielded 116 scorable bands out of which 22 were polymorphic bands producing a polymorphism pecentage of 18.96. Conclusion An optimized protocol for large-scale production of J. curcas plants using plant biotechnology tools was achieved. RAPD and ISSR techniques would introduce an alternative system for large-scale production and establishment of genetically stable plants.
