Abstract:The palladium-catalyzed direct coupling reactions of 4-hydroxycoumarins with various alkynes under copper-free conditions are described. This transformation is highly effective under mild conditions and the desired products are obtained in good yields.

Abstract:Sonogashira coupling is one of the cornerstone Palladium mediated cross coupling reactions, giving a direct and high yield access to symmetric and non‐symmetric alkynes with high tolerance of substituents. The method generally requires the use of CuI as a cocatalyst, an inert atmosphere to protect palladium catalyst, somewhat harsh conditions like prolonged heating and the use of organic solvents due to the hydrophobicity of reagents and products. Leveraging on the opportunities offered by micellar catalysis, we here present a general methodology to perform Sonogashira coupling in water, with moderate or no heating, under air, without copper co‐catalysts and at very low catalyst loading. The method offers the same generality and scope of organic solvents based couplings, but strongly improves sustainability while offering excellent yields.

Abstract:A one-pot two-step reaction of 4-aminocoumarin with arylglyoxal monohydrates and p-toluenesulfonates has been developed, which offers 3-alkoxy-substituted chromeno[4,3-b]pyrrol-4(1H)-ones in moderate to good yields. At first, arylglyoxal monohydrates and 4-aminocoumarin were converted into 3-hydroxychromeno[4,3-b]pyrrol-4(1H)-one intermediates in AcOH under reflux. Then the title compounds in an in situ reaction of the intermediates with p-toluenesulfonates catalyzed by DBU in refluxing toluene were obtained.

Abstract:The exploration of inexpensive and stable heterogeneous catalysts and application of green solvents for Ullmann C-N coupling reaction remain challenging. We present a facile fabrication of copper nanoparticles on hydrochar as prepared from natural, inexpensive and renewable chitosan together with in-situ reduction of copper salt in a one-pot hydrothermal carbonization process. The copper nanoparticles were uniformly dispersed on hydrochar by choosing block copolymer F127 as surfactant. Moreover, maleic acid was introduced to improve the hydrophilicity of hydrochar. The most active copper nanocomposite catalyst, that is, Cu/HCS-MA-F127, exhibited excellent catalytic activity for Ullmann C-N coupling reaction in water. The nature of the Cu/HCS-MA-F127 was characterized by FTIR spectroscopy, TG, XRD, SEM and XPS. Moderate to excellent yields of aimed products were gained by using this catalytic strategy. Moreover, the Cu/HCS-MA-F127 catalyst can be reused by simple centrifugal recovery with a stable performance.

Abstract:An efficient palladium-catalyzed copper-free Sonogashira reaction in water and acetone has been developed under mild conditions. The results showed that the aryl iodides could carry out the cross-coupling reaction with a variety of terminal alkynes in high yields in water-acetone in the absence of amine, copper(l) salts, or phosphine ligands at 60 degrees C for one hour, and good yields were obtained for aryl bromides at 60 degrees C for 12-24 hours in the presence of triphenylphosphine and piperidine. The method could be used to synthesize polyethynyl aromatic compounds in a one-pot reaction.

Abstract:An efficient copper-catalyzed method has been developed for Sonogashira couplings of aryl halides with terminal alkynes in water. The protocol uses inexpensive CuBr as the catalyst, 1,10-phenanthroline as the ligand, tetrabutylammonium bromide (TBAB) as the phase-transfer catalyst, environmentally friendly water as the solvent, and various internal alkynes were synthesized in good to excellent yields.

Abstract:The Pd-catalyzed copper-free carbonylative Sonogashira coupling reaction to synthesize alkynyl ketones from terminal alkynes and aryl iodides was achieved by using water as a solvent. The reaction was carried out at room temperature under balloon pressure of CO with Et(3)N as a base. The developed method was successfully applied to the synthesis of flavones.

Abstract:We describe a copper-catalyzed Sonogashira coupling reaction of alkyl halides with terminal alkynes under ambient conditions, efficiently providing a versatile tool for the construction of substituted alkynes. A new proline-based N,N,Pligand is utilized to promote the transformation under a mild reaction condition. Diverse alkyl halides, such as primary and secondary (hetero) benzyl chlorides and bromides, secondary and tertiary α-bromo amides and propargylic bromide, are applicable to provide a wide array of alkynes.

Abstract:A new copper-catalyzed regioselective cross-dehydrogenative coupling of coumarins with benzylic C-sp3-H bonds has been developed. This reaction provides direct access to a wide range of 3-benzylcoumarins in moderate to good yields. The protocol was also extended successfully to other heterocyclic compounds, such as quinolinones.

Abstract:In the absence of Cu(I), using Bu2NH as base, PdCl2(PPh3)2 showed highly catalytic activity for cross-coupling reactions of a variety of terminal alkynes with both electron-rich (unactivated) and electron-poor aryl bromides in DMF at 60°C affording internal arylated alkynes in good to excellent yields.

Abstract:We studied copper-free Sonogashira reaction using 7-chloro camptothecins, and determined that rac-BINAP/Pd(OAc)2 was an efficient catalyst for the coupling reaction. With this process, a number of 7-substituted camptothecins with a wide range of functional groups are potentially accessible. Besides, two drugs, SN-38 and BNP-1350, could be prepared by this method.

Abstract:An efficient approach for the direct synthesis of alkylated 4-hydroxycoumarin derivatives via a Cu-catalyzed cascade dehydrogenation/conjugate addition sequence starting from simple saturated ketones and 4-hydroxycoumarins has been developed. This protocol features excellent functional-group tolerance, easy scale-up, and a broad substrate scope including bioactive molecules. More importantly, a series of marketed drugs, such as warfarin, acenocoumarol, coumachlor, and coumafuryl, can be obtained by this method.

Abstract:Both carbon dioxide- (CO2) and biomass-based molecules have recently been regarded as renewable, abundant, environmentally benign, and attractive carbon feedstocks for organic synthesis. Therefore, a mild and efficient method for the synthesis of 1,3-oxazolidin-2-ones was developed through a green four-component coupling reaction by using CO2, biomass-based aldehydes such as furfural and 5-hydroxymethylfurfural (HMF), terminal aromatic alkynes, and primary aliphatic amines with copper iodide as a catalyst. 24 Furyl-substituted 1,3-oxazolidin-2-ones were obtained in 29-84% yield from the four-component coupling reaction. Moreover, the coupling reaction was applicable to the syntheses of furyl-containing bisoxazolidinone and polyoxazolidinone under mild conditions.
