Abstract:We recently published details of an effective, reusable and benign heterogeneous amidation catalyst based on thermal treatment of amorphous K60 silica, however the loading of catalyst required was high for some reactions. We report herein our further development of a series of heterogeneous catalysts based on structured silica (SBA) that retain all the green credentials of the previously described amorphous silica, but with increased efficiency. These catalysts were then utilised in continuous flow systems, achieving excellent conversions with dramatically reduced reaction times.
Coated-wall microreactor for continuous biocatalytic transformations using immobilized enzymes

Abstract:Microstructured flow reactors are emerging tools for biocatalytic process development. A compelling design is that of the coated-wall reactor where enzyme is present as a surface layer attached to microchannel walls. However, preparation of a highly active wall biocatalyst remains a problem. Here, a stainless steel microreactor was developed where covalent immobilization of the enzyme in multiple linear flow channels of the reaction plate was supported by a macroporous wash-coat layer of gamma-aluminum oxide. Using surface functionalization with aminopropyl triethoxysilane followed by activation with glutardialdehyde, the thermophilic beta-glycosidase CelB from Pyrococcus furiosus was bound with retention of half of the specific activity of the free enzyme (800 U/mg), yielding a high catalyst loading of about 500 U/mL. This microreactor was employed for the continuous hydrolysis of lactose (100 mM) at 80 degrees C, providing a space-time yield of 500 mg glucose/(mL h) at a stable conversion of > or =70%. The immobilized enzyme displayed a half-life of 15 days under the operational conditions. Due to the absence of hydrophobic solute-material interactions, which limit the scope of microstructures fabricated from poly(dimethylsiloxane) for biocatalytic applications, the new microreactor was fully compatible with the alternate enzyme substrate 2-nitro-phenyl-beta-D-galactoside and the 2-nitro-phenol product resulting from its hydrolysis catalyzed by CelB.

Abstract:The catalytic properties of packed bed reactors filled with Y(OTf)3 immobilised onto functionalised silica have been tested in the Michael addition of indoles to benzylidene malonates under flow conditions, providing improved productivity over the batch conditions, a greater ease of recovery of the final products and a prolonged life of the catalyst. Noteworthy, the same catalytic reactor, after being used for five days, was employed to perform a different reaction, a stereoselective Diels Alder cycloaddition in flow, with a 24 times higher productivity compared to the in‐batch reaction.

Abstract:Solid bases prepared by derivatisation of amorphous silica and hexagonal mesoporous silica with dimethylaminopropyl groups are good catalysts for Michael addition reactions. Of the prepared catalysts, those based on hexagonal mesoporous silica show particularly high activities. The sol-gel method used in preparing the catalysts based on hexagonal mesoporous silica enables loading to be increased to more than twice that of those based on amorphous silica which are prepared by the post-modification method.

Abstract:The results on zirconia-amine bifunctional modification of hierarchically porous silica monoliths for continuous-flow processes ar presented. The study reports the synthesis and properties of the modified porous monoliths and their performance in the tandem process of deacetalization-Knoevenagel condensation reaction. The properties of the materials were studied by thermal analysis, FTIR spectroscopy, XRF and nitrogen adsorption. It was found that both active centres were uniformly distributed along the monolithic cores, and the antagonistic interaction of the acid and base centres was not observed. It was shown that hydrophobicity of the monolith surface has opposite effects on the efficiency of the studied reactions. The overall rate of the tandem process depends on the rate of the condensation reaction. The performance of the bifunctional microreactors was checked and compared with those of cascade-connected monofunctional microreactors and batch reactor. The yield of cyanocinnamate product obtained in both flow systems was ca. 78%; however, pressure drop in the bimodified reactor was only half of that in the cascade. An effective way of water supply to the reaction system has been proposed. The research demonstrates the benefits of using flow-through structured micro-/mesoreactors in tandem reaction processes.

Abstract:The rapid development of continuous flow processes is driving innovations in various chemical syntheses and industrial productions. Immobilizing catalysts in flow reactors allows transformations with high-efficiency and excludes the subsequent separation procedures. This concept outlines the approaches to incorporate catalysts within flow reactors, with particular focus on the application of additional supports including inorganic materials like silica, zeolite and reduced graphene oxide, polymeric materials like polymer packings, monoliths, cross-linked gels and polymer brushes, and other materials for specific conditions like transparent glass fibers and glass beads. Furthermore, advanced methods to develop ordered micro-/nanoarrays from internal walls of flow channels for immobilization of catalysts as well as application of innovative vortex fluidic devices are discussed to inspire new designs of supports for novel fluidic reactors with broad applications.

Abstract:The successful application of catalytic chain transfer polymerisation (CCTP) by adopting an 'in situ' catalyst preparation methodology in several polymerisation media is described. More specifically, this study is focused on reporting the development of 'in situ' CCTP within a CO2 expanded phase polymerisation process, which achieved high yields of polymer whilst minimising both VOC footprint and CO2 compression costs. The 'in situ' method is shown to be effective in controlling polymerisations conducted in both conventional solvents and bulk under inert atmosphere, delivering molecular weight reductions and a Cs value of appropriate similar magnitude to those achieved by the benchmark, commercially sourced CoPhBF catalyst. The 'in situ' effect has been achieved with equal efficiency when both using catalysts with different axial ligands and where the complex is required to undergo a facile ligand dissociation in order to create the required catalyst necessary to achieve CCTP control. Furthermore, both catalysts are shown to effectively control polymerisations in a CO2 expanded phase process, in which a small amount of compressed CO2 is introduced to reduce the viscosity of the reaction mixture, allowing for easy heat transfer and good catalyst diffusion during reaction. In this way, yield limitations imposed to avoid the Trommsdorff effect required in bulk processing and the need for post precipitation have been successfully overcome. Both of these factors further improve the sustainability of such a polymerisation process. However, the 'in situ', high pressure expanded phase environment was observed to retard the ligand dissociation required for catalyst activation.

Abstract:We present an approach to harnessing the tuneable catalytic properties of complex nanomaterials for continuous flow heterogeneous catalysis by combining them with the scalable and industrially implementable properties of carbon pelleted supports. This approach, in turn, will enable these catalytic materials, which largely currently exist in forms unsuitable for this application (e.g. powders), to be fully integrated into large scale, chemical processes. A composite heterogeneous catalyst consisting of a metal-organic framework-based Lewis acid, MIL-100(Sc), immobilised onto polymer-based spherical activated carbon (PBSAC) support has been developed. The material was characterised by focused ion beam-scanning electron microscopy-energy dispersive X-ray analysis, powder X-ray diffraction, N2 adsorption, thermogravimetric analysis, atomic absorption spectroscopy, light scattering and crush testing with the catalytic activity studied in continuous flow. The mechanically robust spherical geometry makes the composite material ideal for application in packed-bed reactors. The catalyst was observed to operate without any loss in activity at steady state for 9 hours when utilised as a Lewis acid catalyst for the intramolecular cyclisation of (±)-citronellal as a model reaction. This work paves the way for further development into the exploitation of MOF-based continuous flow heterogeneous catalysis.

Abstract:Silica rod monoliths of 6mm diameter and 1–5cm length were obtained by phase separation between silica gel and water in presence of aqueous soluble polyethylene oxide (spinodal decomposition). They exhibit a three-dimensional network of uniform flow-through macropores with adjustable size between 5 and 25μm. The skeleton of the monolith has a thickness comparable to the size of the macropores and is composed of amorphous silica with a mesoporosity adjustable between 7 and 35nm. These monolith rods, called MonoSil, were cladded and used as continuous flow catalytic microreactors after introduction of the active sites, via grafting of organic functions, as sulfonic acid groups (HSO3-MonoSil) and inorganic species, as aluminium oxide (Al-MonoSil); or via the transformation of the amorphous silica skeleton into MCM-41, ZSM-5 and zeolite A (LTA-MonoSil) by a pseudomorphic transformation process with the preservation of the macroporous network. LTA-MonoSil was used in catalysis after cationic exchange of the former Na+ cations with K+ cations to yield to K-LTA-MonoSil exhibiting strong basic sites. The catalytic behavior of these modified monoliths was evaluated as continuous flow microreactors for the triacetine transesterification, the Diels–Alder reaction and the Knoevenagel condensation. These monolithic reactors demonstrated higher productivities than batch or packed-bed reactors for the described reactions.

Abstract:A general, green chemistry approach is developed for the immobilization of inorganic catalysts in monolithic porous flow-reactors for continuous flow reactions, in which pyrogallol (PG) acts as an assistant layer for the in situ immobilization of various catalysts on the inner pore surface of the flow-reactor. Au nanoparticles (AuNPs), Al2O3, and phosphotungstic acid (HPW) are selected as examples of noble metals, metal oxides, and polyoxometalate (POM) type catalysts, respectively, and the corresponding flow-reactors show high catalytic performance in continuous flow hydrogenation reactions, esterification reactions, and Knoevenagel reactions, respectively. These reactions are important in the treatment of environmental contaminants and in the chemical industry. In addition, multiple monolithic flow-reactors can be assembled together for efficient performance of multi-step cascade reactions.

Abstract:Flow reactors are appealing alternatives to conventional batch reactors for heterogeneous catalysis. However, it remains a key challenge to firmly immobilize the catalysts in a facile and flexible manner and to simultaneously maintain a high catalytic efficiency and throughput. Herein, we introduce a dense cylindrical micelle brush support in glass capillary flow reactors through a living crystallization-driven self-assembly process initiated by pre-immobilized short micelle seeds. The active hairy corona of these micellar brushes allows the flexible decoration of a diverse array of nanocatalysts, either through a direct capture process or an in situ growth method. The resulting flow reactors reveal excellent catalytic efficiency for a broad range of frequently utilized transformations, including organic reductions, Suzuki couplings, photolytic degradations, and multistep cascade reactions, and the system was both recyclable and durable. Significantly, this approach is readily applicable to long capillaries, which enables the construction of flow reactors with remarkably higher throughput.

Abstract:A neutral amine templating route was used to prepare dimethylaminopropyl-derivatised micelle templated silicas with various loadings. The resultant materials were characterised by various techniques and were also screened as catalysts in model Michael addition reactions. Results obtained show that materials with surface areas and organic group loading as high as 800 m2g-1 and 3.4 mmol per g of silica, respectively, are obtainable. The materials show moderate to high catalytic activity as well as selectivity in a range of substrates. The activities and selectivities,however, are affected by catalyst loading and type of solvent.

Abstract:A highly efficient and enantioselective organocatalyst, imidazolidin-4-one, has been successfully immobilized on siliceous MCF and polymer-coated MCF The resulting heterogenized catalyst demonstrated excellent catalytic performance and recyclability for Friedel-Crafts alkylation and Diels-Alder cycloaddition. The performance of the supported catalysts in relation to the surface environment of siliceous MCF was examined. It was found that partially pre-capping the MCF with TMS groups enabled us to attain well-dispersed catalysts on the siliceous support with optimal performance. We have also developed a polymer-coated MCF, which retained the porous structure of MCF without the surface silanol groups. High reactivity and excellent recyclability were achieved by the organocatalyst immobilized on polymer-coated MCF.

Abstract:In this study, we present an improved packed-bed system designed for continuous-flow reactions using platinum (Pt)-black powder and silica gel (SiO 2 ). The Pt-leaching from the reaction column is suppressed via compression-molding of the Pt and SiO 2 . Scanning electron microscopy results and particle-size distribution analysis demonstrate that crushed and downsized SiO 2 is effective in suppressing outflow. Furthermore, we successfully conducted a scaled-up experiment of the flow reaction using a large column, achieving excellent productivity.

Abstract:In this work, porous silica monoliths that have been functionalized for organocatalysis were investigated using time‐of‐flight secondary ion mass spectrometry to reveal the radial catalyst distribution inside the reactor which is usually not obtainable by conventional methods. Hierarchically porous monolithic silica shows promise as a carrier material for immobilized organocatalysts. Conventional analysis usually includes physisorption, infrared spectroscopy and elemental analysis, among others, to elucidate the pore space and degree of functionalization of the material. However, these methods do not yield information about the spatial distribution of the organic species inside the monolithic reactor. In this work, time‐of‐flight secondary ion mass spectrometry has been applied to characterize the surface of organically functionalized silica monoliths. Cross sections of a silica monolith functionalized with 4‐dimethylaminopyridine were analyzed and the results were compared with physisorption and elemental analysis experiments of the same material. This way, insight into the radial distribution of the catalyst could be achieved, which might assist in interpreting the performance of such reactors in heterogeneous flow catalysis.

Abstract:Biomaterial‐functionalized microreactors are pivotal in basic research, sensor technology, and catalysis. This study delves into the flow‐induced deposition of functional biocatalytic materials, investigating transitions from DNA–hydrogel to DNA–protein hybrids and protocols for immobilization of enzyme materials. The research culminates in stable reactors through the flow‐induced deposition of living cells, showcasing exceptional biocatalytic performance and promising prospects for future innovations. Exploring the potential of microfluidic systems, this study presents a groundbreaking approach harnessing energy in microfluidic flows within a purpose‐built microreactor, enabling precise deposition of functional biomaterials. Upon optimizing reactor dimensions and integrating it into a microfluidic system, sequentially flow‐induced deposition of DNA hydrogels and transformation into DNA‐protein hybrid materials with SpyTag/SpyCatcher technology is investigated. However, limited functionalization rates restrict its viability for targeted biocatalytic processes. Therefore, the direct deposition of a phenolic acid decarboxylase is investigated, which is efficiently deposited but shows limited biocatalytic performance due to shear‐induced denaturation. This challenge is overcome by a two‐step immobilization process, resulting in microfluidic bioreactors demonstrating initial high space‐time yields of up to 7000 g L−1 d−1, but whose process stability proves unsatisfactory. However, by exploiting the principle of flow‐induced deposition to immobilize recombinant E. coli cells as functional living materials overexpressing biocatalytically relevant enzymes, bioreactors are produced that show equally high space‐time yields in continuous whole‐cell catalysis which remain constant over periods of up to 10 days. The insights gained offer optimization strategies for advanced functional materials and innovative reactor systems holding promise for applications in fundamental materials science, biosensing, and scalable production of microreactors for biocatalysis and bioremediation.
