EFFECT OF COLLAGEN POLYPEPTIDES FROM APOSTICHOPUS JAPONICUS ON MELANOGENESIS IN B16 MELANOMA CELLS

Abstract:Objective:To study the effects of different molecular weight of collagen polypeptides from Apostichopus japonicus (A1:6 000U Mr10 000U, A2: Mr6 000U) on melanogenesis, trosinase activity and mRNA expression in B16 melanoma cells. Method: Cell proliferation rate was detected by MTT assay, the tyrosinase activity by dopa oxidase assay, the melanin content by NaOH assay, and the mRNA level of tyrosinase by RT-PCR assay. Results: The proliferation rate of B16 cells was stimulated significantly, and melanogenesis, tyrosinase activity were inhibited remarkably, and the tryrosinase mRNA level was decreased evidently in dose dependent manner by both A1 and A2 respectively. Conclusion: Different molecular weight of collagen polypeptides from Apostichopus japonicus can inhibit melanogenesis in B16 melanocytes significantly, A1 better than A2.
