Abstract:The monodisperse double emulsions obtained by microfluidic method can serve as ideal templates for preparing core-shell alginate microcapsules, which have attracted much attention in biological applications, such as drug delivery systems and cell encapsulation, tissue engineering. However, the formation behavior and dynamic analysis of double emulsion with an alginate shell is still unclear due to the complex rheological behavior of alginate solutions. Herein, we employ a dual-coaxial microfluidic device to generate the high-quality double emulsion droplets with alginate shell, focusing on the effects of the fluid properties of alginate solution in the middle phase (viscosity, μm) and the fluid flow rate on the droplet formation mechanism. As the viscosity of the middle fluid (μm) increased, the size of compound droplets (D2) increased and the size of inner droplets (D1) decreased, and the break-up regimes occurred a dripping-to-jetting transition when μm = 160 mPa s. The number of encapsulated inner droplets can be predicted and precisely controlled by regulating the generation frequency of inner (f1) and outer droplets (f2). The breakup dynamics of the alginate thread are also analyzed by using the volume-of-fluid/continuum-surface-force (VOF/CSF) method. The results show that the pressure and velocity in the neck of pinch-off is lower in the jetting than that in the dripping regime. This study will provide useful guidance for the rational design and controllable preparation of core-shell alginate microcapsules.
Fabrication and formation studies on single-walled CA/NaCS-WSC microcapsules

Abstract:The micron-sized calcium alginate/sodium cellulose sulfate-water soluble chitosan (CA/NaCS-WSC) microcapsules were prepared by membrane emulsification method using sodium alginate (NaAlg), NaCS and WSC as raw materials. The CA/NaCS microspheres prepared dispersed well and held spherical shape with an emulsifier volume ratio of 7:3 (Span 80:Tween 80) and a concentration of cross-linking agent of 1.5% (w/v) calcium chloride and 5% (w/v) sodium chloride. The CA/NaCS-WSC microcapsules had a spherical shape with average diameter of 62.36 +/- 13.87 pm. A fluorescent ring could be seen obviously on the surface of CA/NaCS-WSC microcapsules under confocal microscope, when WSC was labeled by fluorescein isothiocyanate. The discussion on the formation studies implied that Ca2+ could diffuse into the droplets of NaAlg/NaCS forming CA/NaCS microspheres, while NaCS.could react with WSC forming a polyelectrolyte complexes film. Abstract:This work presents a feasible method for controllable preparation of monodisperse spherical alginate microcapsules with structured multiphase oil cores, which have the potential to encapsulate multiple lipophilic actives that are not mutually soluble. Monodisperse (oil 1 +oil 2 )-in-water ((O 1 +O 2 )/W) droplets are generated using the assembled one-step co-flowing 3D-printed microfluidic chip. By cross-linking (O 1 +O 2 )/W droplets with the calcium chloride collection solution, alginate microcapsules are obtained. It is noticed that the multiphase oil cores inside the microcapsules display self-adjustment behavior. The size and shell thickness of the alginate microcapsules, as well as the number of the cores inside the microcapsules, can be precisely tailored. Meanwhile, the factors that influence the sphericity of the microcapsules are investigated. Furthermore, we explore the impact of different types of the collection solutions on the final structure of the microcapsules. The proposed method shows excellent controllability in the design of alginate microcapsules with multiphase oil cores.

Abstract:Here we report a novel strategy for controllable preparation monodisperse alginate microcapsules with oil cores, where the thickness of the alginate shells, as well as the number and diversity of the oil cores can be tailored precisely. Monodisperse oil-in-water-in-oil (O/W/O) emulsions are generated in a microfluidic device as templates, which contain alginate molecules and a water-soluble calcium complex in the middle aqueous phase. Alginate microcapsules are produced by gelling O/W/O emulsions in oil solution with acetic acid, where the pH decreasing will trigger the calcium ions being released from calcium complex and cross-linking with alginate molecules. Increasing the alginate molecule concentration in emulsion templates affects little on the thickness of the microcapsules but improves their stability in DI water. The strength of alginate microcapsules can be reinforced by post cross-linking in calcium chloride, polyetherimide, or chitosan solution. Typical payloads, such as thyme essential oil, lavender essential oil and W/O emulsions are encapsulated in alginate microcapsules successfully. Furthermore, tailoring the thickness of the alginate shells, as well as the number and the diversity of the oil cores precisely by manipulation the emulsion templates with microfluidics is also demonstrated. The proposed method shows excellent controllability in designing alginate microcapsules with oil cores.

Abstract:Monodisperse hollow and core-shell calcium alginate microcapsules are successfully prepared via internal gelation in microfluidic-generated double emulsions. Microfluidic emulsification is introduced to generate monodisperse oil-in-water-in-oil (O/W/O) double emulsion templates, which contain Na-alginate, CaCO3 nanoparticles, and photoacid generator in the middle aqueous phase, for synthesizing Ca-alginate microcapsules. The internal gelation of the aqueous middle layer of O/W/O double emulsions is induced by crosslinking alginate polymers with Ca2+ ions that are released from CaCO3 nanoparticles upon UV exposure of the photoacid generator. The as-prepared hollow and core-shell calcium alginate microcapsules are highly monodisperse and spherical in water. Model proteins Bovine serum albumin (BSA) molecules can be encapsulated into the Ca-alginate microcapsules after the capsule preparation, which demonstrates an alternative route for loading active drugs or chemicals into carriers to avoid the inactivation during the carrier preparation. The proposed technique in this study provides an efficient approach for synthesis of monodisperse hollow or core-shell calcium alginate microcapsules with large cavity or encapsulated lipophilic drugs, chemicals, and nutrients. Abstract:A facile strategy for efficient and continuous fabrication of monodisperse gas-core microcapsules with controllable sizes and excellent ultrasound-induced burst performances is developed based on droplet microfluidics and interfacial polymerization. Monodisperse gas-in-oil-in-water (G/O/W) double emulsion droplets with a gas core and monomer-contained oil layer are fabricated in the upstream of a microfluidic device as templates, and then water-soluble monomers are added into the aqueous continuous phase in the downstream to initiate rapid interfacial polymerization at the O/W interfaces to prepare monodisperse gas-in-oil-in-solid (G/O/S) microcapsules with gas cores. The sizes of both microbubbles and G/O/W droplet templates can be precisely controlled by adjusting the gas supply pressure and the fluid flow rates. Due to the very thin shells of G/O/S microcapsules fabricated via interfacial polymerization, the sizes of the resultant G/O/S microcapsules are almost the same as those of the G/O/W droplet templates, and the microcapsules exhibit excellent deformable properties and ultrasound-induced burst performances. The proposed strategy provides a facile and efficient route for controllably and continuously fabricating monodisperse microcapsules with gas cores, which are highly desired for biomedical applications.

Abstract:This work describes a novel method for the preparation of monodispersed calcium alginate microbeads in a microfluidic device. A novel microfluidic device comprising of two cross-junction channels and a synthesizing channel was designed by controlling the mixing condition with laminar flow and two-phase droplet flow in microfluidic devices. At the first junction, the side two streams, sodium alginate aqueous solution and calcium chloride aqueous solution, were separated to prevent contact by the injection of deionized water in the middle channel. Then the aqueous mixture solution was induced into monodispersed droplets by a focusing sheath of immiscible oil phase at the second junction. The formed droplets were solidified in situ in the synthesizing channel with the rapid mixing of reactants in the droplets. The size of the Ca-alginate microbeads can be controlled from 50-300 mu m in diameter with a variation of less than 2 %, simply by changing the two-phase flow ratio. The proposed microfluidic platform is capable of generating uniform emulsions and has the advantages of in situ solidification and good control of particle size.

Abstract:A simple process is developed for the one-step preparation of dual-compartment alginate microcapsules with controlled size and structure from microfluid-generated water-in-water-in-oil (W/W/O) emulsion droplet. Unlike other methods that rely on transient W/W/O emulsion droplet, we introduce an aqueous two-phase system (ATPS) to form a stable W/W/O emulsion droplet as a template for preparing dual-compartment alginate microcapsules. Two different bioactive molecules are able to be spatially confined encapsulated in the shell and core of alginate microcapsules due to the partitioning effect of ATPS and the high viscosity of alginate solution. Moreover, an enzyme cascade reaction with a spatial confined glucose oxidase and horseradish peroxidase in the shell and core of alginate microcapsules confirms its excellent biocompatibility and high activity. This method provides a green platform for enzyme-catalyzed tandem reactions and controlled sequential release of multiple drugs based on alginate microcapsules.

Abstract:This work reports a novel and facile route to generate spherical and bullet-like alginate microcapsules with a core–shell structure based on the deformation of double-emulsion droplets. Here, monodisperse double-emulsion templates are synthesized from a dual-coaxial capillary microfluidic device and solidified via internal gelation in microchannel to achieve one-step preparation of the bullet-like and spherical microcapsules. The deformation degrees and internal structures of droplets and corresponding microcapsules can be flexibly controlled by simply adjusting the fluid flow rates. In addition, the spherical and bullet-like microcapsules can be successfully prepared by varying the concentrations of reactants. And the shell thickness and core diameter of the spherical microcapsules can be precisely tailored by modulation of the flow rate of fluids. The release performance of as-fabricated spherical and bullet-like microcapsules is investigated in detail by using α-tocopherol as a model drug. The results show that the bullet-like microcapsules with one inner core exhibit a faster release rate than that of spherical microcapsules in phosphate buffer solution (PBS, pH 7.4). In addition, one by one release of the inner cores can be observed in bullet-like microcapsules with two inner cores, which is different from spherical microcapsules that the two inner cores are first fused and then released. Our study provides an easy and effective strategy for the one-step synthesis of spherical and non-spherical microcapsules, which can be potentially used for drug delivery, cell encapsulation, microstructure control, and rheology.

Abstract:We demonstrate a method to create surfactant-free core–shell microcapsules in a hydrophilic polydimethylsiloxane microfluidic device. An ultraviolet light curable polymer was used to encapsulate an oil core. These microcapsules ensure contamination-free compartmentation of the core material without any surfactant, while maintaining the monodispersed generation at a rate of 100 microcapsules per second. The device fabrication process is greatly simplified without the alignment of microchannels and hydrophobic/hydrophilic surface treatment. After drying, physically shaking the collection chamber can crack the capsule to release the liquid core material. Such solid microcapsules with a liquid core are ideal for the storage and delivery of oil-based materials in skincare products or reagents for biochemical assays.

Abstract:In this study, we present a two-stage method for fabricating monodisperse alginate hydrogel microspheres using a symmetrically designed flow-focusing microfluidic device. One of the flow-focusing junctions generates alginate hydrogel droplets without the addition of surfactants, while the other junction introduces corn oil with acetic acid, which facilitates the solidification of the homogeneous alginate hydrogel droplets and prevents coalescence. These hydrogel microspheres can be easily separated from the oil phase using an oscillation state, eliminating the need for a demulsifier. This microfluidic system for hydrogel microsphere formation is notable for its simplicity, ease of fabrication, and user-friendliness.

Abstract:Many active ingredients in food products are destroyed by gastric acids and enzymes as they pass through the stomach, thereby reducing the desired effect of the target ingredient. In addition, traditional microcapsule preparation methods usually break up the dispersion by mechanical stirring or oscillation, which generates droplets of uneven size and inconsistent inclusion content, and mechanical external forces may cause denaturation and inactivation of active substances. This study aimed to use microfluidics to generate Oil-in-water-in-oil (O/W/O) droplet structures with good monodispersity and highly tunable properties, and to further prepare microcapsules with high encapsulation rate and targeted intestinal release by taking advantage of the cross-linking properties of alginate that reacts with Ca 2+ under acidic conditions. Finally, the particle size distribution, release effect, encapsulation effect and oxidative stability of the core-shell microcapsules were evaluated. In this study, two types of microcapsules, homogeneous and core-shell microcapsules, were prepared by selecting tea polyphenols and vitamin E as models for hydrophilic and lipophilic bioactive compounds, respectively. Thin shell microcapsules with a shell thickness of about 25 μm were prepared using PDMS chip conditioning. Large microcapsules with a particle size of 230 μm were generated by using capillary glass tube chips, which had an excellent antioxidant property with a retention rate of 88.66 ± 1.25% of VE after 20 days of storage at 4 °C, while all of the VE was oxidized in the bare environment. In addition, in simulated gastric fluid, the release of VE was only about 20%, while in artificial intestinal fluid, the release was close to 100%, so the microcapsules also had intestinal targeting properties.

Abstract:Monodisperse hollow microcapsules have potential applicationsin the fields of chemical,medicaland biological engineering.In the present work,monodisperse oil-in-water(O/W) single emulsions containing hydroxyethyl methacrylate(HEMA) and methyl methacrylate(MMA)comonomers generated in microfluidic device were used as templates for synthesis of monodisperse poly(HEMA-MMA) hollow microcapsules,withthe aid of porogen inside emulsions and the UV-initiatingpolymerization.The prepared microcapsules were characterized with optical microscope and scanning electron microscope,and the results showed that thediameter of microcapsule is about 40 μm with a narrowsize distribution,and the microcapsule has a smooth exteriorsurface and rough interiorsurface,thin shell and hollow structure.The method developed in this study provides a new way for preparing monodisperse hollow microcapsules.

Abstract:This work investigates the molecular interaction of hydrocolloids (xanthan gum (XG), hydroxyethyl cellulose (HEC), carbomer (CBM) and hymagic™-4D (HA)) with sodium alginate (SA) in microspheres in detail. The molecular interaction of hydrocolloids with SA are demonstrated by the rheological property analysis of the mixed solutions as well as the morphology structure and texture characteristics studies of the microspheres. It is found that the hydrocolloids (XG, HEC and CBM) with branches or capable to coil are able to form complex networks with SA through molecular interactions which hinders the free diffusion of calcium ions and changes the texture characteristics of microspheres. In addition, the mixed solutions (SA-XG and SA-HEC) with complex networks and do not have a chelating effect on calcium ions are used to form the shell of the microcapsules through droplet microfluidic technology, and stable with soft microcapsules encapsulating multiphase oil cores have been successfully prepared. At the same time, the textural properties of microcapsules are quantized, which are related to human sensory properties. The developed stable and soft microcapsules which have the properties of sensory comfort are expected to be applied in the personal care industry and a variety of fields.
Microfluidic generation of monodispersed Janus alginate hydrogel microparticles using water-in-oil emulsion reactant.

Abstract:Microparticles with uniform anisotropic structures are widely used in physical, chemical, and biological fields owing to their ability to combine multiple functions on a micro-scale. Here, a microfluidic emulsion-based external gelation method was demonstrated for the first time to produce monodisperse Janus calcium alginate (Ca-alginate) hydrogel microparticles consisting of two compartments. This approach provided a fast reaction condition under which we could prepare magnetic Janus Ca-alginate microparticles with diameters ranging from 148 to 179 μm and a coefficient of variation (CV) less than 4%. Moreover, the boundaries between the two compartments were clear. In addition, the volume fraction of each compartment could be adjusted by varying the flow rate ratio between two dispersed phases. Next, we produced fluorescent Janus beads and magnetic-fluorescent Janus beads with an average diameter of ∼150 μm (CV < 4.0%). The magnetic Janus hydrogel microparticles we produced could be manipulated by applying a magnetic field to achieve self-assembly, rotation, and accumulation. Magnetic Janus hydrogel microparticles are also capable of mammalian cell encapsulation with good cell viability. This article presents a simple and stable approach for producing monodisperse bi-compartmental Janus hydrogel microparticles that could have great potential for application in physical, biochemical, and biomedical fields.

Microencapsulation of Solid Cores to Prepare Double Emulsion Droplets by Microfluidics

Abstract:We herein present an effective microfluidic method based on facile glass capillary assembly for encapsulating solid cores to prepare double emulsions. Visualization experiments are conducted to verify the effectiveness of the presented microfluidic device. It is indicated that, via the presented microfluidic device, solid cores can be controllably encapsulated into every generated double emulsion droplets under wide operating range of flow rate. The number of the encapsulated solid cores can be well manipulated by adjusting the flow rate of different phases. Particularly, by employing multiple injection microchannels, multiple kinds of solid cores can be encapsulated into a single droplet in different combinations. This microfluidic methods show good applicability to generate both solid-in-oil-in-water (s/o/w) and solid-in-water-in-oil (s/w/o) double emulsion droplets. We believe that the current microfluidic methods are capable to extend the application of microencapsulation in microcapsule preparation, drug detection, food delivery, single cell analysis, and synthesis of nanomaterials, etc. (C) 2019 Elsevier Ltd. Abstract:Core-shell hydrogel microcapsules have sparked great interest due to their unique characteristics and prospective applications in the medical, pharmaceutical, and cosmetic fields. However, complex synthetic procedures and expensive costs have limited their practical application. Herein, we designed and prepared several multichannel and multijunctional droplet microfluidic devices based on soft lithography for the effective synthesis of core-shell hydrogel microcapsules for different purposes. Additionally, two different cross-linking processes (ultraviolet (UV) exposure and interfacial polymerization) were used to synthesize different types of core-shell structured hydrogel microcapsules. Hydrogel microcapsules with gelatin methacryloyl (GelMA) as the core and polyacrylamide (PAM) as the thin shell were synthesized using UV cross-linking. Using an interfacial polymerization process, another core-shell structured microcapsule with GelMA as the core and Ca2+ cross-linked alginate with polyethylenimine (PEI) as the shell was constructed, and the core diameter and total droplet diameter were flexibly controlled by carving. Noteworthy, these hydrogel microcapsules exhibit stimuli-responsiveness and controlled release ability. Overall, a novel technique was developed to successfully synthesize various hydrogel microcapsules with core-shell microstructures. The hydrogel microcapsules possess a multilayered structure that facilitates the coassembly of cells and drugs, as well as the layered assembly of multiple drugs, to develop synergistic therapeutic regimens. These adaptable and controllable hydrogel microdroplets shall held great promise for multicell or multidrug administration as well as for high-throughput drug screening.
Non-Spherical Hollow Microgels with Uniform Sizes and Tunable Shapes from Microfluidic-Assisted Approach

Abstract:We conducted microfluidic technique to generate uniform double emulsion droplets with alginate aqueous solution forming the shell phase. Size and shell thickness of the droplets can be tailored by changing the property of each fluid and flow ratio of the fluids. The viscoelastic ionic cross-linkable emulsion droplets (e.g., sodium alginate aqueous droplets) could experience a deformation process when dropping down into a viscous gelation bath containing Ba(Ac)(2) or Ca(Ac)(2). The deformed non-spherical structures of the droplets can be fixed by ionic cross-linking of the alginate during the dropping down process, triggering the formation of hollow microgels with varied shapes in the viscous gelation bath. By manipulating the concentration of sodium alginate in the aqueous solution, viscosity of the gelation bath, and properties of the core fluids, microgels or hollow microgels with varied shapes (e.g., tear-like, rod-like and pepper-like shapes) can be systemically generated. We show that functional species (quantum dots and/or fluorescence dyes) can be successfully encapsulated into the microgels, demonstrating the potential applications of the non-spherical microgels in the area of bioimaging, drug delivery and release, barcoding, single cell analysis and cell culture.

Abstract:Due to the benefits involving brain development and cardiovascular health, algae oil which is rich in omega-3 fatty acids has become a popular dietary supplement in recent years. However, the incompatibility with water and poor oxidative stability limits its addition in foods and beverages. In this work, the formation and characterization of zein-based core-shell microcapsules with tunable shell thicknesses for the potential delivery of algae oil were reported. They were prepared based on the in situ precipitation of zein from the continuous phase onto the surface of oil droplets by self-assembly at a refrigerated temperature without any need for surfactants and cross-linkers. The shell thickness could be controlled by the ratio of algae oil to zein (O/Z), which was indicated by their characterization including static light scattering, confocal laser scanning microscopy and scanning electron microscopy. Evidence from the peroxide value and the hexanal level of the emulsions in a thermally accelerated storage test suggested that the oxidation degree of the loading algae oil was negatively correlated with the shell thickness of the capsules. Furthermore, the release profiles of vanillin, which was used as a model volatile lipophilic compound and masking agent for the off-flavor from algae oil, could be tuned by manipulating their shell thickness. This suggested that the all-natural edible core-shell microcapsule might further be used as a model system for the delivery of lipophilic compounds. They may further promote the sustainable use of underutilized water-insoluble proteins as functional biomaterials in functional foods and pharmaceutical applications.
