Abstract:A series of experiments were conducted to determine the suitable freezing and thawing temperatures for the freezing of boar semen in 5 ml maxi-straws. The ultrastructure, in vitro fertilization (IVF) and artificial insemination (AI) of frozen-thawed semen were also be evaluated. The 5 cm freezing height gave the best results not only in post-thaw motility rate (54.00%), but also in normal acrosome morphology rate (NAR) (80.23%). There was no significant difference in the post-thaw motility between different thawing temperatures and corresponding thawing times (p>0.05); the group of 52°C and 25 s gave the highest motility rate (45.00%). As a whole, not only from the motility but also the NAR, thawing at 42°C was better than the other two treatments. In the freezing packages, 5 ml maxi-straw gave a little lower mobility (40%), viability rate (49.58%), plasma membrane integrity rate (53.91%) and NAR (52.65%) than the 0.25 ml straw, but there was no significant difference between the two straw volumes (p>0.05). The IVF capacity of frozen-thawed semen in this experiment was similar to fresh semen. From ultrastructure observation, the main damage to boar spermatozoa after freezing was seen in the acrosome, such as swelling and formation of vesicles. After AI in recipient Shanghai White sows, frozen-thawed semen from 5 ml maxi-straws and pellets produced 72.2% and 80% conception rate and 7.8 and 8 litter sizes, respectively, and there was no significant difference between the 5 ml maxi-straw and the pellet (p>0.05).

Abstract:Cryopreservation of few spermatozoa is still a major challenge for male fertility preservation. This study reports use a new micro-straw (LSL straw) for freezing few spermatozoa for intracytoplasmic sperm injection (ICSI). Semen samples from 22 fertile donors were collected, and each semen sample was diluted and mixed with cryoprotectant in a ratio of 1: 1, and then frozen using three different straws such as LSL straw (50-100 mu l), traditional 0.25 ml and 0.5 ml straws. For freezing, all straws were fumigated with liquid nitrogen, with temperature directly reducing to -130-140 degrees C. Sperm concentration, progressive motility, morphology, acrosome integrity, and DNA fragmentation index were evaluated before and after freezing. After freezing-thawing, LSL straw group had significantly higher percentage of sperm motility than traditional 0.25 ml and 0.5 ml straw groups (38.5% vs 27.4% and 25.6%, P < 0.003). Sperm motility and acrosomal integrity after freezing-thawing were significantly lower than that of before freezing. However, there was no significant difference in morphology, acrosome, and DNA integrity between the three types of straws (P > 0.05). As LSL straws were thinner and hold very small volume, the freezing rate of LSL straw was obviously faster than 0.25 ml straw and 0.5 ml straws. In conclusion, LSL micro-straws may be useful to store few motile spermatozoa with good recovery of motility for patients undergoing ICSI treatment.
Optimization of Procedure in Boar Semen Cryopreservation

Abstract:Four experiments were included in the present study.Five,two,three and three treatments were designed for experiment 1(plunging temperatures:-5,-30,-60,-90 and-120 ℃),experiment 2(fresh semen preprocessing: nonpre-extending and pre-extending),experiment 3(thawing temperatures: 37,50 ℃ and 70 ℃),and experiment 4(extended-spermatozoa concentration: 1.25×108,2.50×108 and 5.00×108 spz/mL) respectively.The results showed that(i) the differences of spermatozoa motility at different plunging temperatures were not significant,but motility of frozen-thawed spermatozoa revealed a tendency to improve with the drop of plunging temperature(ii) the preliminary dilution did not significantly increase motility of extended spermatozoa,but it made motility of equilibrated spermatozoa,motility,VAP,VSL and VCL of frozen-thawed spermatozoa significantly or very significantly higher,(iii) there were no significant differences among the three thawing temperatures in motility,VAP,VSL and VCL of frozen-thawed spermatozoa,however,the higher the thawing temperature was,the higher the motility of frozen-thawed spermatozoa was and(iv) three different concentrations of extended spermatozoa did not result in significantly different motility,VAP and VSL of frozen-thawed spermatozoa;nevertheless,the motility and VAP fell with the enhancement of extended-spermatozoa concentration,and the highest extended-spermatozoa concentration brought significantly lower VCL of frozen-thawed spermatozoa than the two other treatments.In conclusion,of the five plunging temperatures of the straw,the optimal one was-120 ℃,where the best sperm motility could be observed after thawing.And the preliminary dilution of fresh semen before extending was indispensable to cryopreservation of boar semen;besides,the highest motility of spermatozoa,thawed at 37,50 and 70 ℃ respectively,would be detected in the group 70 ℃,and moreover,high diluted-spermatozoa concentration could be disadvantageous to the cryopreservation of boar spermatozoa.

Abstract:The Yangtze sturgeon (Acipenser dabryanus) is extinct in the wild; however, artificial propagation in captivity under fully controlled conditions has been successful. Sperm cryopreservation is an effective method for germplasm conservation and aquaculture. Sperm concentration affects the quality of cryopreserved spermatozoa. Thus, in this study, we optimized the concentration (0.25 × 109, 0.5 × 109, 1.0 × 109, and 2.0 × 109 spermatozoa [spz]/mL) for Yangtze sturgeon sperm cryopreservation in a 0.5 mL plastic straw. The greatest value for the kinetic parameters was achieved when sperm were cryopreserved at 0.5 × 109 spz/mL. In comparison to other concentrations, there was a notable increase in both acrosomal and plasma membrane integrity when sperm was cryopreserved at a concentration of 0.5 × 109 spz/mL. No significant difference of superoxide dismutase and glutathione peroxidase enzyme activity was present in post-thaw sperm, except in sperm cryopreserved at a concentration of 2.0 × 109 spz/mL. Catalase activity was significantly higher in post-thaw sperm at a concentration of 0.5 × 109 spz/mL than at other concentrations. Excluding sperm cryopreserved at 2.0 × 109 spz/mL, the fertilization rate of cryopreserved sperm was not significantly different among the different concentrations. This concentration optimization improves cryopreserved sperm quality and standardizes sperm cryopreservation procedures for the Yangtze sturgeon. These findings are especially beneficial for the establishment of a sperm cryobank.
Comparison of rapid freezing versus vitrification for human sperm cryopreservation using sucrose in closed straw systems

Abstract:Rapid freezing and vitrification using sucrose are two simple and cost-effective sperm cryopreservation methods. However, it is still unclear which method is better and what the optimal concentration of sucrose is. This study aimed to determine the optimal sucrose concentration for human sperm cryopreservation and compare the cryoprotective effects of rapid freezing versus vitrification using different closed straw systems in terms of sperm motility and DNA integrity. Our data showed that: (1) The optimal sucrose concentration for vitrification was 0.25 mol/l among the tested 0, 0.125, 0.25 and 0.5 mol/l concentrations; (2) Sperm total motility and progressive motility were cryopreserved significantly better by rapid freezing than vitrification in standard 0.5 ml cryostraws (<i>P</i> < 0.05); and (3) Sperm total motility and progressive motility were cryopreserved significantly better by vitrification in the straw-in-straw system than rapid freezing in the standard 0.5 cryostraw (<i>P</i> < 0.05), but no difference was found in sperm nuclear DNA fragmentation level between the two cryopreservation methods (<i>P</i> > 0.05). It was concluded that sucrose at 0.25 mol/l concentration is suitable for human sperm rapid freezing and vitrification, and sperm cryopreservation can be achieved by rapid freezing using closed standard 0.5 ml straws or by vitrification using the novel straw-in-straw system made of standard 0.25 and 0.5 ml straws.

Abstract:The aim of this study was to investigate the effect of supplementation of caffeine in freezing extender on frozen-thawed semen quality.Optimize freezing and thawing protocol in 0.5 mL straws.Supplementation of caffeine directly into preliminary design freezing extender,the concentration of caffeine was 0,0.2,0.4,0.6 mg/mL,determined the best concentration of caffeine.Then compared with the traditional TCG extender and the AndrohepCryoGuardTM extender,comparison of three different freezing curves on the frozen-thawed sperm quality.The frozen sperm samples were thawed by warming them at 37 ℃ water bath for 30 s and 50 ℃ water bath for 13 s.The results showed that there was a significantly higher percentage of progressive motility,membrane integrity and acrosome integrity in caffeine supplemented groups than control group(P0.05),the best concentration was 0.2 mg/mL.The preliminary design was better than the TCG(P0.05),but it had a significant variance with AndrohepCryoGuardTM extender(P0.05).Three different freezing curve had different influence on sperm quality parameters.Freezing curve A had a higher percentages of TMS,PMI and NAR of spermatozoa than B and C(P0.05),thawed at 50 ℃ for 13 s was better than 38 ℃ for 30 s.

Abstract:Objective To study the effects of two cryopreservation methods ( direct vapor nitrogen method and program freezing method) on the motility of post-thaw human sperm.Methods The 25 semen samples were taken from 25 donors from Shanghai Human Sperm Bank during May to July 2011,using straw as a frozen carrier, the semen samples contained modified glycerol-yolk-citrate ( GYC ) ( 1 ∶1 ) as cryoprotectant,after the liquefaction, each semen sample was divided into 2 groups,and separately cryopreserved by direct vapor nitrogen method and program freezing method.According to the recommended standard of the World Health Organization, semen samples were analyzed before and after cryopreserving.Density,total motility,PR,motility parameters ( including VAL,VSL,VCL,BCF) were determinated by the IVOS sperm analyzer.Semen samples were stained by modified papanicolaou stain and 200 sperms of each sample were counted under microscope by manual analysis to calculate the rate of normal morphology sperm.The paired t test was used to compare the above indictors before and after cryopreserving in one method and after thawing between two methods.Results Before cryopreserving,the total motility,PR,VAL,VSL,VCL,BCF were(91.36±4.96)％,(81.28 ±6.41)％,(59.29 ±9.30) μm/s,(49.80±8.40) μm/s, (83.39 ± 15.70) μm/s, (16.68 ± 3.36) Hz respectively, and after cryopreserving by direct vapor nitrogen method the results were (46.04 ± 20.14)％,(40.22 ± 18.42 )％,(48.00± 11.60)μm/s,(41.11 ±8.50) μm/s,(71.55 ±17.19) μm/s,(15.94 ± 14.30) Hz respectively,the differences of the indictors above were observed with statistic significance except BCF ( t =11.324,11.958,4.266,3.964,3.620,0.258,P of BCF ＞0.05,P of others ＜0.05); and after cryopreserving by program freezing method,the results were(48.05 ± 18.27)％,(42.47 ± 15.62)％,(45.30 ± 6.51) μm/s,(38.08 ± 4.99) μm/s,(64.23 ± 11.75) μm/s,(14.00 ± 2.99) Hz respectively,the differences of the indictors above showed statistic significance (t =10.337,10.874,7.154,5.510,6.494,3.244,P ＜0.05).No significant differences were observed in the revival rate,total motility,PR and normal morphology sperm rate by using different methods (t =0.425,0.364,0.622,1.106,P ＞0.05).The results of VAL,VSL,VCL,BCF in direct vapor nitrogen method were higher than that in program freezing method,when the differences of VSL and VCL were observed with statistic significance(t =2.489,2.822,P ＜ 0.05 ).Conclusions Using straw as carrier is practicable.Direct vapor nitrogen method is easier to operate than program freezing method.Post-thaw sperm cryopreseved by direct vapor nitrogen method has higher motility parameters,thus making this method an ideal one for fast freezing.
Research on Compatibility Effect Between Pretreatment Extenders and Cryoprotective Extenders in Frozen Boar Semen

Abstract:The purpose of this study was to research the effect of pretreatment extenders and cryoprotective extenders on frozen boar semen and their co-effect.Six home-temp extenders were used in pretreatment and three cryoprotective extenders were used in cryo-preservation in this experiment.The results were as follows:there was significant difference in the frozen-thawed sperm motility among six pretreatment extenders (P0.05),however,there was no significant difference in the frozen-thawed sperm motility among three cryoprotective extenders (P0.05).Significant difference existed in compatibility effect between pretreatment and cryoprotective extenders (P0.01).Different balancing time led to different compatibility effects.The motility and acrosomal integrity of frozen-thawed sperm could be significantly improved by removing the seminal plasma.
Study on the Effect of Seminal Plasma in Porcine Sperms Cryopreservation

Abstract:In order to improve the frozen porcine sperms motility and fertilizing ability after thawed,sperms were extended in basic cryoprotection extender with 0,10%,20%,30% seminal plasma respectively before freezing.Sperm motility,membrane integrity,fertilization rate and blastocyst rate of IVF embryos and artificial insemination results were chosen as criteria for assessing the sperm quality.The results showed that the motility,membrane integrity of sperms extended in cryoprotection extender containing 10% seminal plasma were improved significantly(P<0.05) after thawed compared with the control group.The blastocyst rate of IVF embryos derived from these sperms was also increased significantly.Artificial insemination with the thawed sperms from the group extended in cryoprotection extender containing 10% seminal plasma resulted in a significantly lower pregnancy rates,litter size and number of live littermate compared with fresh sperm insemination(P<0.05).These results indicated that cryoprotectant with 10% seminal plasma could increase the motility of thawed porcine sperm after cryopreservation and the blastocyst rate after IVF,but for artificial insemination it was still need to be improved.

Abstract:Low density lipoprotein was added at concentrations of 6%, 7%, 8%, 9% and 10% to the diluents used to freeze boar semen and its effect on the quality of cryopreservation was assessed The results indicated that 9% low density lipoprotein supplementation significantly improved the total motile sperm (p<0.05). The sperm straight-line velocity increased with the concentration of low density lipoprotein except at the concentration of 10%, at which concentration the sperm velocity declined (p<0.05). Supplementation at 8% and 9% low density lipoprotein significantly increased plasma membrane integrity (p<0.05). Compared with control, the low density lipoprotein supplementation significantly improved the percentage of acrosome integrity (p<0.05). With all parameters measured, the concentration of 9% low density lipoprotein showed a better effect on the quality of boar cryopreservation semen. The sperm DNA was more seriously damaged in the species of Yorkshire than in Duroc.

Abstract:Effects of several extenders,thawing diluents and freezing-thawing protocols on boar sperm viability,motility,plasma membrane integrity and acrosome integrity were studied in this experiment and the best extender and protecol were modified to improve boar semen freezing efficiency.The results were as follows:(1)The number Ⅳ extender was the best among the extenders tested and its sperm motility (32.4±7.3)%,vaibility (42.2±3.2)%,acrosomal integrity (62.3±0.8)% and plasma membrane integrity (42.6±7.5)% after freezing and thawing were significantly(P0.05) higher than those obtained with number Ⅰ,Ⅱ and Ⅲ extenders. (2)When 0.5%,1% or 2% O.E.P. were added to the number Ⅳ extender,sperm viability,plasma membrane integrity and acrosomal integrity increased significantly(P0.05) by 5%,6% and 10%,respectively,over those of the controls. (3)When boar semen was frozen in straws with modified extender(number Ⅳ plus O.E.P.),the sperm motility and viability were increased by 7% and 6% over those when semen was frozen in pellets with the same extender(P0.05). (4)Pre-equiliberation for 4 hours at room temperature increased both sperm viability and motility significantly(P0.05) in comparison with those in the controls(0 hours of pre-equilibrium). (5)The sperm motility obtained with thawing diluent number Ⅲ(42.9±2.6)% was significantly higher than those obtained with thawing diluents number Ⅰ,Ⅱ and Ⅳ. (6)The sperm motility after 50℃ water bath thawing(46.3%) was significantly higher than those obtained with 39℃(42.9±2.6)% and 70℃ water bath thawing(41.1±5.7)%. (7)When the frozen semen was thawed in semen plasma,the sperm motility,viability,plasma membrane integrity and acrosomal integrity was not significantly different from those when semen was thawed in thawing diluent number Ⅲ. (8)All of the 6 sows inseminated with the frozen-thawed semen with a motility of around 46%(which was processed with modified extender,pre-equiliberated for 4 hours and thawed in thawing diluent number Ⅲ at 50℃) got pregnant and farrowed at an average liter size of 9.
Effect of Egg Yolk Concentrations on Cryopreservation of Boar Semen

Abstract:The present experiment was conducted to investigate the effects of egg yolk on cryopreservation of boar semen by screening the best concentration of egg yolk in freezing boar semen.Different percentages(20,25 and 30%) of egg yolk were added to freeze diluted boar semen,and the quality of semen before diluting,after balancing and freezing were compared using sperm viability,motility,velocity of activity path,velocity of circle locus,velocity of straight locus,etc.The results showed that there were no significant differences in five velocity indices of sperm on adding different concentrations of egg yolk in boar semen freezing dilution after diluting,balancing and refreezing.Therefore,increasing concentration of egg yolk for boar semen freezing dilution could not enhance the quality of semen,and the 20% concentration of egg yolk in boar semen freezing dilution was found to be the best.
A Study on Optimizing the Cryopreservation Methods for Bama Miniature Pig Semen

Abstract:In this study, the cryopreservation methods for Bama miniature pig semen were investigated and optimized. First, using an orthogonal experimental design, the semen freezing-thawing procedure for Bama miniature boars was optimized based on analysis of the effects of concentrations of LDL (LC, parameter A), trehalose (TC, parameter B) and glycerol (GC, parameter C), the equilibration time at 15°C (ET, parameter D), and the thawing method (TM, parameter E) on sperm motility. The results showed that the effects of the parameters could be arranged as A>C>B>D>E. The LDL concentration and final glycerol concentration had exceedingly significant effects on the motility of thawed spermatozoa (P<0.01), and the effects of the trehalose concentration, equilibration time at 15°C, and the thawing method were not significant (P>0.05). Scheme 2 (A(3)B(4)C(2)D(3)E(1)) gave a motility of 52.26% after thawing. Then, using sperm motility, acrosome integrity, plasma membrane integrity, and DNA injury rate as indicators, four combinations, on the basis of scheme 2, were designed to analyze the protective effects of different combinations of LDL, glycerol, and trehalose; the results showed that combination of 9% LDL, 200 mM trehalose, and 2% glycerol (i.e., combination 4) demonstrated significantly better protective effects than the other combinations (P<0.05), further verifying that scheme 2 was the best for cryopreservation of Bama miniature boar semen. In this way, a method with favorable performance was established for cryopreservation of semen of Bama miniature boars.
Effect of Real-Time Sperm Separation System on the Quality of Boar Sperm Frozen

Abstract:【Objective】 The study was to explore the effect of real-time sperm separation system on the quality of boar semen frozen.【Method】 Original semen was separated with real-time sperm separation system and tubule-froze with liquid nitrogen.The original semen was looked as control group,and the effect of real-time sperm separation system on the quality of boar semen frozen was detected.【Result】 After the separated semen was thawed,the sperm motility,the rate of spermatozoa with intact acrosome,the rate of spermatozoa with normal plasma membrane,the rate of spermatozoa with normal chromatin of the separated group were higher than those of the control group(P0.01);For the rate of spermatozoa with normal morphology,the separated group was better than that of the control group(P0.01),but it was not affected by the freeze-thaw process(P0.01);The spermatozoa density was not significantly affected by the separation process(P0.01).【Conclusion】 The real-time sperm separation system can significantly improve the quality of boar frozen semen.
Boar Seminal Plasma Improves Sperm Quality by Enhancing Its Antioxidant Capacity During Liquid Storage at 17°C

Abstract:The objective of this study was to investigate the effects of different levels of seminal plasma (SP) on boar sperm quality, antioxidant capacity and bacterial concentrations during liquid storage at 17°C. Boar sperm was diluted with Beltsville Thawing Solution (BTS) consisting of 0, 25, 50 and 75% (v/v) of SP. Total motility, progressive motility and dynamic parameters were assessed by the computer assisted sperm analysis (CASA) system. Acrosome and plasma membrane integrity were measured by FITC-PNA/DAPI and SYBR-14/PI staining, respectively. In addition, total antioxidant capacity (T-AOC), malondialdehyde (MDA) content, and reactive oxygen species (ROS) levels were detected using commercial assay kits. Bacterial concentrations were assessed by turbidimetric assay. Our results showed that 25% SP markedly improved total motility, progressive motility, sperm dynamic parameters, acrosome integrity compared with 0, 50 and 75% SP (P < 0.05). In addition, 25% SP significantly increased T-AOC but decreased MDA content and ROS levels compared with 0, and 75% SP (P < 0.05). Moreover, 25% SP significantly decreased the bacterial concentrations in extended semen compared with 50% and 75% SP, however, which was higher than with 0% SP (P < 0.05). These results suggest that 25% SP can promote boar sperm quality through enhancing its antioxidant capacity during liquid storage.
Effect of Different Thawing Methods on Post-thaw Motility of Cow Semen in Cryopreservation Straws

Abstract:The study was to investigate the effects of different thawing temperatures(5,15,40,75,90 ℃) and times(1- 120 s) on properties of post-thaw cow semen by detecting frozen-thawed semen motility,acrosome integrity and tail membrane integrity,further obtaining the optimal thawing method of straw frozen semen from dairy cow. The results showed that(1) Thawing of the straw frozen semen of dairy cow at 75 ℃ for 3 s yielded the highest semen motility,followed by 40 ℃for 20 s,and the least by low temperature 5 ℃ and room temperature 15 ℃ with a semen motility of 0. 3,moreover thawing at high temperature 90 ℃ was not suitable for large scale production due to the difficult control of the temperature;(2) The acrosome intact rate and plasma membrane integrity rate of semens thawed at90 ℃ were remarkably lower than that at 40 ℃ and 75 ℃ significantly(P 75 ℃ > 90 ℃. In practice,the thawing method of straw frozen semen of dairy cow should be selected according to the specific circumstance and inseminated immediately,with the recommended condition of thawing at 75 ℃ for 3 s. If the thawed semen could not be inseminated immediately,the thawing should be performed at 20 s for 40 ℃to maintain the motility for a longer term.

Abstract:A modified protocol of neutral comet assay was utilized to assess the effect of low density lipoprotein (LDL) on the DNA integrity of boar freezing-thawing semen. The results demonstrated that the method was high sensitive and easier manipulation and LDL significantly protected sperm DNA integrity (p<0.05) from the damage caused by cryopreservation except TD at the concentration of 6%, 7% and TM at 6%, the optimal LDL concentration in diluents was 9%. Moreover, LDL showed better protection in 0.25 ml than in 0.5 ml types of straw (p<0.05) and no difference was observed in the same volume straw at the concentration of 9% and 10%. It was just the same for LDL effect on boar sperm DNA in cryopreservation 0 day and 30 days (p>0.05).

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Objective</h3><p>To evaluate a novel micro-straw as an efficient, simple method for freezing a small number of human spermatozoa for intracytoplasmic sperm injection (ICSI).</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Design</h3><p>Prospective cohort study.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Setting</h3><p>Sperm bank.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Patient(s)</h3><p>Men with severe oligozoospermia or azoospermia undergoing a total of 143 ICSI cycles at the CITIC-Xiangya Hospital of Reproduction and Genetics from June 1, 2015, to June 31, 2019, and 20 donors at the Hunan Province Human Sperm Bank from 2001 to 2016.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Intervention(s)</h3><p>Analysis of sperm samples and clinical outcomes after sperm use.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Main Outcome Measure(s)</h3><p>Clinical information, including number of motile sperm before and after freezing, freeze–thaw survival rates, two-pronuclear fertilization rates, clinical pregnancy, and early pregnancy loss rates after sperm use.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Result(s)</h3><p>In the feasibility experiment using the micro-straw, we found a freeze–thaw survival rate of 73% ± 8.3% and no difference in normal sperm morphology, normal acrosome integrity, or DNA fragmentation index between the micro-straw and 1.8-mL cryotubes. The prospective cohort included 1,325 cases, and we collected sperm from testicular, epididymis, and ejaculation sources. We observed motile sperm in 1,294 (97.6%) of 1,325 frozen–thawed samples. Postthaw sperm were available for ICSI in 140 (97.9%) of 143 of cycles. The fertilization, cleavage, and high-quality embryo rates were 1,007 (81.7%) of 1,233; 995 (98.8%) of 1,007; and 537 (53.9%) of 995, respectively. Sixty-nine (49%) clinical pregnancies were achieved, and the miscarriage rate was 6 (8.6%) of 69.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusion(s)</h3><p>The micro-straw is suitable and clinically useful for the cryopreservation of small numbers of spermatozoa.</p>

Abstract:In order to simplify the source of semen of in vitro fertilization(IVF),and avoid the effects by different boars and different semen collection time,we used frozen semen by 5 mL-straw to do the IVF.The results showed that there were no significant differences in the polyspermy rates among the 2-8 h oocyte-sperm incubation time(P0.05).The 2-cell rate was improved along with the increasing of oocyte-sperm incubation time,and 8 h group(72.8%) was much higher than the 2 h group(60.5%,P0.05).The morula and blastocysts rates of the 6 h group were the highest and got 46.1% and 3.48%,respectively.Polyspermy rate was also increased with the increasing of sperm concentration in the IVF(P0.05).The 2-cell rate of 108 group(81.2%) was the highest,which was much higher than the 105 group.The 107 and 106 group got 8.11% and 3.13% blastocyst rate,respectively.Using NCSU-23 as the in vitro maturation(IVM) medium and matured oocytes partly denuded cumulus cells could get the highest cleavage and blastocyst rates(77.1%,7.33%) among the four groups.The comparation between the frozen semen and fresh semen in the IVF showed that there were no significant differences in the cleavage,morula and blastocyst rates(P0.05).

Abstract:The sperm being removed seminal plasma and not being removed seminal plasma by centrifuga-tion were cryopreservated.1.The percentages of the frozen-thawed sperm of motility, normal acrosome and abnormal spermatozoa were significant difference between the two treatments being removed seminal plasma and the other two treatments not being removed seminal plasma (P<0.05); 2.From diluting to post-thawing, the concentrations of GOT and LDH of seminal plasma all rose. But there were significant difference between the preceding two treatments and the latter ones (P<0.05). After post- thawing, the four aliquots had no significant difference; 3.The acrosome of frozen-thawed sperm of not being removed seminal plasma was damaged more badly than the one of seminal plasma; 4.
