Abstract:An improved synthetic approach to tenatoprazole 1 is described. It started from 2,3,5-trimethyl-4-nitropyridine-N-oxide 2 with acetic anhydride via rearrangement and hydrolysis to give 3, Chlorination with SOCl2 yielded 2-chloromethyl-3,5-dimethyl-4-nitropyridine hydrochloride 4, then 4 condensed with 2-mercapto-5-methoxy imidazole [4,5-b]pyridine 5 to give 5-methoxy-2-[(4-nitro-3,5-dimethyl-2-pyridinyl)methylthio]imidazole[4,5-b]pyridine 6. At last the title compound 1 was produced by two methods: 6 was oxidized with MCPBA and then methoxylated with CH3ONa to give 1 and 6 was first methoxylated with CH3ONa and then oxidized with MCPBA to give 1. The overall yield is around 26% for both five-step syntheses.

Abstract:5-[2-(ethylthio)propyl]-1,3-cyclohexanedione, the title compound of this study, is an important intermediate for preparation of 3-hydroxy-cyclohex-2-enones herbicides, such as clethodim, sethoxydim, cloproxydim, etc.. The optimization of the synthesis route and the preparation method of the title compound were studied and the results were described concisely as follows. Firstly, 3-ethylthiobutanal was prepared with high yield by reacting crotonaldehyde with ethanethiol in triethylamine. Secondly, methyl acetoacetate was converted to 6-ethylthiohept-3-en-2-one via three steps, hydrolysis, condensation with 3-ethylthiobutanal and dehydration, then the 6-ethylthiohept-3-en-2-one was cyclocondensed with di-Me malonate by Michael addition and Clasisen condensation, and further via saponification and decarboxylation , the title compound was obtained. The products of each steps and the final title compound obtained were compared with each its own standerd samples and were confirmed by LC/MS.

Abstract:A new synthesis method of 2,3,4,5-tetramethoxytoluene was devised by using 4-methyltoluene which is easily available in industry.4-methyltoluene being catalyzed by Fe powder,gave high yields of 2,3,6-tribromic4-methyl-phenol,which when being added into sodium methoxide solution and catalyzed by cuprous chloride resulted in 2,3,4,5-tetramethoxytoluene with a yield of 95%.The yield can be optimized by varying the reaction temperature and the sodium methoxide concentration.

Abstract:2,3-Dimethoxy-5-methyl-1,4-benzoquinone called coenzyme Q0,a key fragment in coenzyme Qn series and a key intermediate for synthesizing coenzyme Q10 was prepared from 3,4,5-trimethoxy toluene using polyethylene glycol (PEG-1000) as phase transfer catalyst(PTC), as well as supported heteropolyacid as acid catalyst, acetic acid as medium, hydrogen peroxide as oxidant. The suitable condition was found that 40mL acetic acid, 40mL 50% hydrogen peroxide, 20mL supported heteropolyacid catalyst and 0.4g PEG-1000 are required for 24g raw material 3,4,5-trimethoxy-toluene and the reacting temperature is 30~35 ℃. Via washing reaction medium with water, extraction with dichloromethane and removal of solvent, the crystal of product was prepared with the purity and yield of 97% and 90% respectively.

Abstract:Methyl 3-oxo-2,6,6-trimethylcyclohex-1-ene-1-carboxylate was synthesised over eight steps from the starting material of citral instead of α-ionone. KMnO4, HC(OEt)3 and O2/TEMPO/CuCl were substituted for NaIO4, CH3I and pyridinium chlorochromate, respectively. Every step was simplified and gave a 48% overall yield. The procedure is suitable for large scale production.

Abstract:A parallel process for the new synthesis of 2,7-dimethyl-2,4,6-octatriene-1,8-dial was investigated.3-(5,5-Dimethyl-1,3-dioxan-2-yl)-2-butenal was obtained by a process involving aldehyde protection,ester exchange and catalytic oxidation from 4-acetoxy-2-methyl-2-butenal.A new catalytic oxidation process was proposed to get 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-butenal by oxidating 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-buten-1-ol with copper(Ⅰ) chloride、2,2,6,6-tetramethyl-1-piperidinyloxy and oxygen.3-(5,5-Dimethyl-1,3-dioxan-2-yl)-2-buten-1-ol was chlorizated with SOCl_2 in DMF to get 2-(3-chloro-1-methyl-1-propenyl)-5,5-dimethyl-1,3-dioxane.2,7-Dimethyl-2,4,6-octatriene-1,8-dial was acquired in an overall yield of 48% by one-pot synthesis method from 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-butenal,2-(3-chloro-1-methyl-1-propenyl)-5,5-dimethyl-1,3-dioxane and triethylphosphite.

Abstract:A procedure for synthesis of 2-methoxyphenothiazine (1) has been developed, starting from resorcinol and aniline by condensation, following methylation and cyclization. p-Toluenesulfonic acid and polysubstituted aromatics were employed as the catalyst of condensation and the solvent of cyclization, respectively, to improve the yield. The use of parallel experiments, statistical experimental design, and multivariate modeling made the total yield of the procedure as high as 74.2%. J. Heterocyclic Chem., (2012).

Abstract:3-[(E)-5-(2,3,4,5-Tetr~ethoxy-6-methylphenyl)-3-methylpent-3-enyl]-2,2-dimethyloxirane (8) is a key intermediate for total synthesis of coenzyme QlO (1) and has previously been synthesized from p-cresol via bromination, methylation, Grignard reaction and epoxidation.'S2 However, this procedure is not attractive for the large-scale preparation of 8 due to drawbacks such as low yield (ca. 30% overall yield), low reaction temperature (-78°C). chromatographic separation and the use of several expensive and hazardous reagents. Therefore, a practical method for synthesis of 8 is desirable. Herein, we report a new efficient and convenient method for the preparation of 8 from commercially available starting material 2.

Abstract:sulfomethyltetrazole 5 thiol disodium salt, a side chain of cefonicid,was synthesized from sodium bisulfite,ammonium hydroxide and formaldehyde through five steps of reactions. The yield of product was 10.8% in the optimum condition. The process was more economic in which the iodomethane is substituted to the bromoethane in the second reaction.

Abstract:2-(9-Hydroxynonyl)-5,6-dimethoxy-3-methyl-1,4-benzoquinone was readily synthesized from commercially available 3,4,5-trimethoxytoluene in two steps. First, 2,3-dimethoxy-5-methyl-1,4-benzoquinone (coenzyme Q0) was obtained in one step by treatment of 3,4,5-trimethoxytoluene with hydrogen peroxide under metal-free conditions, followed by free-radical alkylation with 10-hydroxydecanoic acid in the presence of potassium peroxodisulfate and silver nitrate in a mixed solvent (MeCN–H2O, 1:1) to afford the title compound in good yields (60%, based on coenzyme Q0).

Abstract:The title compound,a key intermediate for synthesis of coenzyme Q_(10),was simply but efficiently prepared via Wolff-Kishner-Huang reduction and hydrogen peroxide oxidation with 3,4,5-trimethoxybenzadehyde as a starting material in a high yield.The important influencing factors of the reaction were studied.The results showed that using three times superfluous hydrazine hydrate,with 50% of hydrogen peroxide,the phosphomolybdic acid as catalyst(a 2.5%～3.0% mass ratio of TMT) and the formic acid as a medium,and reacting for 45 min could be the proper conditions.

Abstract:The titled compound (1), which is an important intermediate of metoprolol, was synthesized in overall 29% yield from cheap p-nitrotoluene and paraformaldehyde via condensation, alkylation, reduction, diazotisation and hydrolysis. The key step (p-nitrotoluene condensation with paraformaldehyde) was investigated in detail, including effects of catalysts, solvent, and reactants mole ratio on yield. Thus, 2 mol p-nitrotoluenes reacted with 1 mol paraformaldehyde in DMSO, catalyzed by potassium phenol to give p-nitrophenyl ethanol (4) with the conversion of p-nitrotoluene of 31% and selectivity of 72%. The remaining p-nitrotoluene could be recycled.

Abstract:2,4,4′,6-tetramethoxydeoxybenzoin was prepared from 1,3,5-trimethoxybenzene and 2-(4-methoxy phenyl)acetonitrile as materials,and toluene as solvent instead of diethyl ether.Through Hoesch reaction,the target compound 2,4,4′,6-tetramethoxydeoxybenzoin was got.It was characterized by H1NMR.Because of the protective effect of ether bond,the side reactions were inhibited,and the yield of Hoesch reaction was reached to 82.2% after the synthetic method was improved.Reaction time and the amount of solvent on the yield were researched.Optimization of reaction conditions were reaction time 28h and the amount of solvent 150mL,under the other conditions constant.

Abstract:An efficient and stereoselective approach to the synthesis of coenzyme Q(10) is described (Scheme). The MeOCH2-protected p-hydroquinone 4 containing the C-5 (E)-allyl (tert-butyl)dimethylsilyl ether moiety Was obtained via a halogen-lithium exchange of the MeOCH2-proctected 2-bromo-5,6-dimethoxy-3-methylhydroquinone 2 and subsequent addition to (E)-('BUMe2Si)-OCH2C(Nle) CHCH2Br (3). The reductive desulfonylation of compound 8, obtained from 4 via 5 - 7, was successfully carried out by employing Li/EtNH2.

Abstract:We developed a novel method for the synthesis of 2‐substituted‐4,5‐dihydrothiazol‐4‐ols compounds through cyclization of thioamides and 1,4‐dithiane‐2,5‐diol in the presence of Et3N. Stable hydroxy‐substituted thiazole compounds were isolated of substituted thioamides. The reaction conditions are simple, the substrate range is wide, and the target compounds were obtained in up to 82% yield.

Abstract:We report a concise synthesis of tanikolide 1, which was obtained from ethyl 2-oxocyclopentanecarboxylate in four steps: alkylation, Baeyer–Villiger reaction, saponification, and reduction/lactonization, in 70% overall yield. Our strategy should be suitable for the preparation of 1 in multigram or larger quantities. The net result of the last two steps (i.e. saponification and reduction/lactonization) is an efficient reduction of the ethoxycarbonyl of 3 while keeping the lactone carbonyl intact.

Abstract:Based on the oxidation of 1,5-naphthalenediol (4) and 6-bromo-2-naphthol (9) via Teuber reaction, an efficient synthesis of 5,6-dimethoxy-1-naphthol (1) and 5,6-dimethoxy-2-naphthol (2) was achieved with high overall yield (16% for 1 and 25% for 2). The key steps of the synthetic strategy involved the oxidation of naphthols (4 and 9) to the corresponding naphthoquinones (5 and 10) and the conversion of 5,6-dimethoxy-2-naphthaldehyde to 5,6-dimethoxy-2-naphthol formate through Baeyer-Villiger oxidation-rearrangement.
