Abstract:A series of rigid backbone 1,8-anthracene-linked bis-oxazolines (AnBOXes) have been designed, synthesized, and evaluated in the catalytic asymmetric aziridination with [N-(p-toluenesulfonyl)imino]phenyliodinane (PhINTs) as a nitrene source. The results indicate that highly enantioselective aziridination of chalcones catalyzed by an AnBOX and CuOTf complex with up to >99% ee and the opposite enantioselectivity, compared with the ligands of Evans et al., can be achieved. The enantioselectivity is substituent dependent with respect to chalcones. Chalcones with electron-donating substituents show higher enantioselectivities due to the stronger Lewis basicity of the oxygen of their carbonyl groups than those with electron-withdrawing substituents. The results also indicate that the coordination between the oxygen of the carbonyl group in chalcones and the ether group in alkenes with the copper in the catalyst is essential for high enantioselectivity, while the π–π stacking interaction between two reactants plays an importantly additional role for high enantioselectivity in asymmetric aziridination. An excellent backbone-controlled stereoselectivity was observed for the AnBOX ligands in asymmetric aziridination, as this will provide very important information for designing novel ligands.

Abstract:[reaction: see text] Chalcones were asymmetrically aziridinated with [N-(p-toluenesulfonyl)imino]phenyliodinane (PhI=NTs) as a nitrene source under catalysis of CuOTf and a series of cyclohexane-linked bis-oxazolines (cHBOXes), which are chelate size rationally tuned bis-oxazolines. The results indicate that highly enantioselective aziridination of chalcones with up to >99% ee have been achieved under catalysis of (S,S)-1,2-bis[(S)-(4-phenyl)oxazolin-2-yl]cyclohexane, which is the most-matched stereoisomer among cyclohexane-linked bis-oxazolines. It was also found that the enantioselectivity is not substituent-dependent with respect to chalcones in the present case, unlike with 1,8-anthracene-linked bis-oxazolines (AnBOXes).

Abstract:A fast and highly enantioselective chlorination of 4-pyrazolones catalyzed by bis(oxazoline)–Cu(ClO4)2·6H2O complexes has been developed. Under the optimized conditions, a series of pyrazolones bearing stereogenic chlorine-attached carbon centers were obtained in moderate to high yields (up to 98%) and with enantioselectivities of up to 98% ee.

Abstract:Chiral cyclopentadienyl-rhodium(III) CpxRh(III) catalysis has been demonstrated to be competent for catalyzing highly enantioselective aziridination of challenging unactivated terminal alkenes and nitrene sources. The chiral CpxRh(III) catalysis system exhibited outstanding catalytic performance and wide functional group tolerance, yielding synthetically important and highly valuable chiral aziridines with good to excellent yields and enantioselectivities (up to 99% yield, 93% ee). This protocol presents a novel and effective strategy for synthesizing enantioenriched aziridines from simple alkenes. Various transformations were performed on the aziridine products, illustrating the versatility and synthetic potential of this protocol for constructing highly functionalized compounds.

Abstract:A rapid construction of enantioenriched benzofurochromanes was developed by SaBOX/copper(II) catalyzed enantioselective [3 + 2] annulation of γ-chromenes with quinones. This process takes advantage of the simple starting materials and a highly efficient chiral SaBOX/copper(II) catalyst system, leading to a variety of benzofurochromanes in up to 96% yield with up to 97% ee.

Abstract:In the past two decades, substantial advances have been made on the asymmetric alkyne functionalization by the activation of inert alkynes. However, these asymmetric transformations have so far been mostly limited to transition metal catalysis, and chiral Bronsted acid-catalyzed examples are rarely explored. Here, we report a chiral Bronsted acid-catalyzed dearomatization reaction of phenol- and indole-tethered homopropargyl amines, allowing the practical and atom-economical synthesis of a diverse array of valuable fused polycyclic enones and indolines bearing a chiral quaternary carbon stereocenter and two contiguous stereogenic centers in moderate to good yields with excellent diastereoselectivities and generally excellent enantioselectivities (up to >99% enantiomeric excess). This protocol demonstrates Bronsted acid-catalyzed asymmetric dearomatizations via vinylidene-quinone methides.

Abstract:We report a novel strategy for the synthesis of chiral 2-oxazolidinones via a dinuclear copper-catalyzed asymmetric propargylic amination-carboxylative cyclization sequence of propargylic esters with nucleophilic alkyl amines under ambient pressure of carbon dioxide. A variety of chiral 2-oxazolidinones featuring an exocyclic methylene moiety was obtained in good yields with high enantioselectivities via a one-pot operation.

Abstract:The asymmetric aziridination of 1,3-dienes catalyzed by bisoxazoline-CuOTf complexes with PhI=NTs as a nitrene precursor has been achieved in moderate yields with up to >99:1 regioselectivity, up to >99% diastereoselectivity, and up to 80% enantioselectivity. alpha,beta,gamma,delta-Unsaturated ketones usually produced cis-gamma,delta-aziridinated products, while 1,4-diphenyl-1,3-butadiene afforded both of cis- and trans-aziridine derivatives as major products by the use of different bisoxazoline ligands. The configuration of cis-aziridine derivatives was proposed on the basis of the reaction mechanism.

Abstract:A general and efficient protocol for the synthesis of 9,10-phenanthraquinone derivatives has been successfully developed involving a copper(0)/Selectfluor system-promoted oxidative carbon-carbon bond cleavage/annulation of o-aryl chalcones. A variety of substituted 9,10-phenanthraquinones were synthesized in moderate to good yields under mild reaction conditions.

Abstract:An axially chiral phenanthroline ligand bearing two BINOL units was found to be capable of promoting the copper-catalyzed enantioselective arylation of 2-azonaphthalenes with arylboronic acids. Abstract:We report a tandem asymmetric Cu-catalyzed propargylic animation (ACPA)/Ag-catalyzed carboxylative cyclization (SCC) with CO 2 to 5-methylidene-2-oxazolidinones, even with tetrasubstituted stereocenters. By varying pyridine bisoxazoline (PYBOX) ligands, a general and highly enantioselective ACPA of unprecedentedly broad scope of secondary propargylic acetates and primary amines is achieved. Both α-aryl and α-aliphatic propargylic acetates could react with either aromatic or aliphatic primary amines to give the corresponding N -aryl or N -aliphatic ethynylamines with α-aliphatic or α-aryl groups in over 90% ee for use in the next step. The key to developing this one-pot sequence is to use a chelator triethylenetetramine (TETA) to mask the copper ion, to avoid its negative effect on Ag-catalyzed cyclization, whilst releasing PYBOX to activate the silver catalyst. With the methylidene moiety, these oxazolidinones can be readily elaborated. The value of the sequence is further shown by the catalytic enantioselective total synthesis of (−)-cytoxazone and the potent ezetimibe analogue.

Abstract:An enantioselective [5 + 1] annulation of α-hydroxyl aryl ketones with cyclohexadienone-tethered enones via chiral dinuclear zinc catalysts is reported. A Brønsted base and Lewis acid cooperative activation model is the critical factor for the chirality transfer process, giving access to a wide array of enantioenriched bicyclic pyrans scaffolds bearing two oxa-quaternary carbon centers with good yields and high optical purities. Notably, this reaction offers a new reaction mode of α-hydroxyl ketones as C,C-bisnucleophiles in catalytic asymmetric transformation.

Abstract:The first catalytic enantioselective C(sp)―C(sp3) cross‐coupling reaction between N‐tosylhydrazones and trialkylsilylethynes in the presence of Cu(I) salts and chiral phosphoramidite ligands was developed. Abstract:Anthracenylidene is an intriguing structural unit with potential in various fields. The study presents a novel approach to introducing axial chirality into this all-carbon core skeleton through a remotely controlled desymmetrization strategy. A palladium-catalyzed enantioselective Heck arylation of exocyclic double bond of anthracene with two distinct substituents at the C10 position is harnessed to realize such a transformation. The judicious identification of the P-centrally chiral ligand is pivotal to ensure the competitive competence in reactivity and stereocontrol when the heteroatom handle is absent from the anthracenylidene skeleton. Both C10 mono- and disubstituted substrates were compatible for the established catalytic system, and structurally diverse anthracenylidene-based frameworks were forged with good-to-high enantiocontrol. The subsequent derivatization of the obtained products yielded a valuable array of centrally and axially chiral molecules, thus emphasizing the practicality of this chemistry. DFT calculations shed light on the catalytic mechanism and provided insights into the origin of the experimentally observed enantioselectivity for this reaction.
