Abstract:We describe herein an intriguing method for the synthesis of biaryl aldehydes bearing both axial and central chirality through a desymmetric [3 + 2] cycloaddition reaction of activated isocyanides with prochiral biaryl dialdehydes under silver catalysis. Abstract:We report here a Cu-catalyzed enantioselective acyloxylation of cyclic diaryliodonium salts. With readily available cyclic diaryliodonium salts and ubiquitous aliphatic or (hetero)aromatic carboxylic acids as the starting materials, various axially chiral acyloxylated 2-iodobiaryls were prepared in excellent yield and with excellent enantioselectivity (mostly 99% yield and 99% ee). Density functional theory calculations were conducted to reveal the stereo- and regioselectivities. This simple reaction protocol can be employed for the late-stage modification of some drug molecules. Finally, by diversity-oriented transformations, these acyloxylated 2-iodobiaryl products can be easily transformed into diverse valuable functionalized biaryls that could be used as chiral ligands or functional materials.

Abstract:Axially chiral tetrasubstituted alkenes are of increasing value and interest in chemistry‐related areas. However, their catalytic asymmetric synthesis remains elusive, due to the high steric repulsion and relatively low conformational stability. Herein, we disclose the straightforward construction of atropisomeric tetrasubstituted alkenes via an effective enantiocontrol of vinyl cations. This copper‐catalyzed enantioselective C(sp2)–H functionalization of sterically hindered (hetero)arenes with vinyl cations enables the efficient and atom‐economical preparation of axially chiral acyclic tetrasubstituted styrenes and pyrrolyl ethylenes with high atroposelectivities. Importantly, this reaction represents the first example for the assembly of axially chiral alkenes through vinyl cation approach. Computational mechanistic studies reveal the reaction mechanism, origin of regioselectivity, Z/E selectivity and enantioselectivity. The synthetic utility has been demonstrated by diverse product derivatizations, chiral organocatalyst synthesis, as well as further applications in asymmetric catalysis.

Abstract:C-N axially chiral skeletons are ubiquitous in bioactive natural products, pharmaceuticals, and chiral ligands. However, their atroposelective synthesis remains a formidable challenge because of their innate low configurational stability compared with that of well-developed C-C atropisomers. Herein, we report a general and efficient method for accessing C-N atropisomers through an axial-to-axial chirality transfer strategy based on palladium/chiral norbornene cooperative catalysis. The obtained C-N axial chirality originates from the preformed transient C-C axial chirality with high fidelity. A variety of C-N axially chiral phenanthridinones are obtained in excellent enantioselectivities (44 examples, up to > 99% ee). This method can be applied for the construction of two stereogenic axes via double atroposelective C-H arylation or further transformation of the products via axial-to-axial diastereoinduction. Additionally, the reaction mechanism and the chirality transfer process are elucidated by density functional theory calculations.

Abstract:The asymmetric cross-coupling reaction is developed as a straightforward strategy toward 1,1-diaryl alkanes, which are a key skeleton in a series of natural products and bioactive molecules in recent years. Here we report an enantioselective benzylic C(sp(3))-H bond arylation via photoredox/nickel dual catalysis. Sterically hindered chiral biimidazoline ligands are designed for this asymmetric cross-coupling reaction. Readily available alkyl benzenes and aryl bromides with various functional groups tolerance can be easily and directly transferred to useful chiral 1,1-diaryl alkanes including pharmaceutical intermediates and bioactive molecules. This reaction proceeds smoothly under mild conditions without the use of external redox reagents.

Abstract:Axially chiral cyclohexylidenes exhibit unique chirality because of the restricted rotation of the C=C bond. Compared with other axially chiral compounds, they aremuch less explored, and their efficient construction remains a synthetic challenge. Herein, we report the first palladium-catalyzed asymmetric coupling reaction of aryl iodides with 4-substituted cyclohexanol-derived alpha-diazocarbonyl compounds to give tetrasubstituted cyclohexylidenes in good yields (up to 92%) with excellent enantioselectivities (up to 96% ee) under mild conditions. This protocol provides a general access to axially chiral cyclohexylidenes. The utility of this unique scaffold was demonstrated by the carboxylic acid serving as a useful chiral ligand in transition-metal-catalyzed C-H bond activation, which showed great promise as a new type of olefin ligand.

Abstract:By using diazonaphthoquinones and anilines as key reagents and through a point-to-axis chiral transfer strategy, the atroposelective synthesis via asymmetric C(sp2 )-H bond insertion reaction of arenes has been realized under rhodium catalysis, providing the resulting biaryl atropisomers in moderate to excellent yields with good enantiomeric ratios (up to 99:1). Further elaboration indicates this type of axially biaryl scaffold may have promising potentials in developing novel chiral ligands.

Abstract:Metal-catalyzed enantioselective construction of the loosening axial allene chirality spreading over three carbon atoms using a chiral ligand is still a significant challenge. In the literature, steric effect of the substrates is the major strategy applied for such a purpose. Herein, we present a general palladium-catalyzed asymmetrization of readily available racemic 2,3-allenylic carbonates with different types of non-substituted and 2-substituted malonates using (R)-(-)-DTBM-SEGPHOS as the preferred ligand to afford 1,3-disubstituted chiral allenes with 90-96% ee. This protocol has been applied to the first enantioselective synthesis of natural product, (R)-traumatic lactone. Control experiments showed that in addition to the chiral ligand, conducting this transformation via Procedure C, which excludes the extensive prior coordination of the allene unit in the starting allene with Pd forming a species without the influence of the chiral ligand, is crucial for the observed high enantioselectivity.

Abstract:Axially chiral styrenes, which exhibit a chiral axis between a substituted alkene and an aromatic ring, have been largely overlooked. The hurdle is the lower barriers to rotation compared with that of their biaryl counterparts, rendering their asymmetric synthesis more difficult. We report herein the highly atroposelective synthesis via a C-H functionalization strategy of axially chiral styrenes with an open-chained alkene. Various axially chiral styrenes were produced by Pd(II)-catalyzed C-H alkenylation and alkynylation in good yields (up to 99%) and enantioselectivities (up to 99% ee) by using L-pyroglutamic acid as an inexpensive chiral ligand. The potent application of the styrene atropisomers is demonstrated by a Co(III)-catalyzed enantioselective C-H amidation of ferrocene with axially chiral styrene-type acid as chiral ligand. Experimental and computational studies were conducted to elucidate the reaction mechanism. The chiral induction model of the enantioselectivity-determining C-H bond activation step was also provided based on DFT calculations.

Abstract:Abstract N-N axially chiral biaryls represent a rarely explored class of atropisomers. Reported herein is construction of diverse classes of diaxially chiral biaryls containing N-N and C-N/C-C diaxes in distal positions in excellent enantioselectivity and diastereoselectivity. The N-N chiral axis in the products provides a handle toward solvent-driven diastereodivergence, as has been realized in the coupling of a large scope of benzamides and sterically hindered alkynes, affording diaxes in complementary diastereoselectivity. The diastereodivergence has been elucidated by computational studies which revealed that the hexafluoroisopropanol (HFIP) solvent molecule participated in an unusual manner as a solvent as well as a ligand and switched the sequence of two competing elementary steps, resulting in switch of the stereoselectivity of the alkyne insertion and inversion of the configuration of the C-C axis. Further cleavage of the N-directing group in the diaxial chiral products transforms the diastereodivergence to enantiodivergence.

Abstract:The merger of photoredox and transition-metal catalysis has evolved as a robust platform in organic synthesis over the past decade. The stereoselective 1,4-functionalization of 1,3-enynes, a prevalent synthon in synthetic chemistry, could afford valuable chiral allene derivatives. However, tremendous efforts have been focused on the ionic reaction pathway. The radical-involved asymmetric 1,4-functionalization of 1,3-enynes remains a prominent challenge. Herein, we describe the asymmetric three-component 1,4-dialkylation of 1,3-enynes via dual photoredox and chromium catalysis to provide chiral allenols. This method features readily available starting materials, broad substrate scope, good functional group compatibility, high regioselectivity, and simultaneous control of axial and central chiralities. Mechanistic studies suggest that this reaction proceeds through a radical-involved redox-neutral pathway.

Abstract:Axially chiral C2-arylquinoline has been successfully constructed via asymmetric heteroannulation of alkynes catalyzed by chiral phosphoric acid with high yield and high enantioselectivity. Inspired by this intriguing work, theoretical calculations have been carried out, and the detailed reaction mechanism has been elaborated, in which the whole reaction can be divided into steps including hydrogen transfer, C-N bonding, annulation reaction and the final dehydration processes. The initial hydrogen-transfer reaction has two possible pathways, while the subsequent C-N bonding process has eight possible pathways. Then, after the annulation reaction and the final dehydration processes, the major product and byproduct were formed. QTAIM and IGMH analyses were used to illustrate the role of weak intermolecular interactions in the catalytic process, and the distortion/interaction and EDA analyses provided a deeper understanding of the origin of enantioselectivity. The calculated results are consistent with the experimental results. This work would provide valuable insights into asymmetric reactions catalyzed by chiral phosphoric acid.

Abstract:Catalytic asymmetric hydrofunctionalization of alkenes is a well-established method with which to construct complex C(sp3)-enriched molecules with central chirality. In contrast, the use of catalytic asymmetric hydrofunctionalization of abundant alkyne substrates to produce valuable multisubstituted alkenes with axial chirality remains largely unexplored. Here, we report a general procedure of this type catalyzed by abundant Ni(II) salts and employing a structurally simple chiral PyrOx ligand. A wide variety of structurally diverse atropisomeric chiral styrenes have been obtained from hydroarylation of alkynes with high efficiency, complete Z-selectivity, and excellent enantioselectivity.

Abstract:We present herein an unprecedented stereoselective synthesis of triaryl-2-pyrones with monoaxial or contiguous diaxes from readily available starting materials. This N-heterocyclic carbene catalysis method adopts an atroposelective annulation of 2-aryketones with ynals under oxidative conditions. The annulation includes the construction of one or two axes in a single operation, achieves step economy, and affords axially chiral triaryl-2-pyrones in moderate to good yields, with high to excellent enantioselectivities. DFT calculations of the relative energies of stereoisomers and rotational barriers were performed.

Abstract:Comprehensive Summary Herein we report an asymmetric two‐component alkenyl Catellani reaction for the construction of C–N axial chirality through a palladium/chiral norbornene cooperative catalysis and an axial‐to‐axial chirality transfer process. Various partially aromatic iodinated 2‐pyridones, quinolones, coumarin and uracil substrates react with 2,6‐disubstituted aryl bromides with a tethered amide group, to afford a wide variety of polycyclic C–N atropisomers (38 examples, up to 97% e.e.). The obtained C–N axial chirality is originated from the preformed transient C–C axial chirality with high fidelity. The synthetic utility of this chemistry is demonstrated by facile preparation of complex quinoline and pyridine based C–N atropisomers through a N‐deprotection and aromatization sequence. In addition, a remote axial‐to‐central diastereoinduction process dictated by C–N axial chirality is observed with excellent diastereocontrol. This article is protected by copyright. Abstract:Axially chiral molecular scaffolds are widely found in pharmaceutical molecules, functionalized materials, and chiral ligands. The synthesis of these compounds has garnered considerable interest from both academia and industry. The construction of such molecules, enabled by transition metal catalysis and organocatalysis under thermodynamic conditions, has been extensively studied and well-reviewed. In recent years, photoinduced enantioselective reactions have emerged as powerful methods for the catalytic construction of axial chirality. In this review, we provide an overview of various synthetic strategies for the photoinduced construction of axial chirality, with a specific focus on reaction design and catalytic mechanisms. Additionally, we discuss the limitations of current methods and highlight future directions in this field.
