Cell Adhesion Behavior, Fabrication and Surface-Modification of Chitosan Scaffolds

Abstract:The chitosan scaffolds were fabricated by lyophilization technique,and the prepared scaffolds were surface-modified by the gelatin to biomimetically construct a degradable composite for biomedical application.The density,porosity and degradation of the fabricated and modified scaffolds were determined;the cell adhesion behavior and biocompatibility of the treated scaffolds were detected by inoculation of the bone marrow mesenchymal stem cells(BMSCs) derived from New Zealand rabbits,and the MSCs were inoculated onto the untreated scaffold too,which was used as the control sample group.The MSCs-chitosan composites were stained with Hoechst method and observed under fluorescence microscopes and scanning electron microscope(SEM) to study the cell growth,adhesion and distribution of the MSCs.The results show that the porosities of the scaffolds prepared with two different chitosan concentrations are(68.85±2.42)% and(71.50±3.29)%,respectively;the pore size is 90~130 μm,and the MSCs inoculated onto the treated surfaces present good adhesion and spreading ability.Besides,the effect of the crosslinking agent 1-ethyl-(3-dimethylanimopropyl) carbodiimide(EDC) on the performance of the scaffolds was initially investigated in this study.It can be concluded that the gelatin surface-modified biomimetic scaffolds have excellent biocompatibility and are appropriate for cell adhesion and proliferation.Therefore,it would be a promising scaffold for tissue engineering application.
