Effect of Carboxymethyl Chitosan Modification to Paclitaxel Self-assembled Proliposome in vivo Biodistribution to Rats

Abstract:AIM:To study in vivo biodistribution of paclitaxel self-assembled proliposome modified by carboxymethyl chitosan (CM-PSAP) in rats by LC-MS/MS and evaluate its targeting performance in contrast to unmodified paclitaxel self-assembled proliposome (PSAP). METHOD:We have developed a LC-MS/MS method based on positive electrospray ionization (ESI + -MS/MS) for quantifying paclitaxel in rat tissue homogenates. CM-PSAP and PSAP were administered by tail vein at dose of 5 mg·kg -1 body weight and tissues has been excised and handled properly at different time interval to quantify PTX by LC-MS/MS. The biodistribution characterization and targeting performance of PSAP after being modified were evaluated. RESULT: By comparison with PSAP group, PTX distribution in the liver, spleen and lung were significantly higher in the CM-PSAP group than those of in PSAP (P<0.05). t_ max and MRT were lengthened. Targeting efficiency (t_ e ) was larger than that of PSAP. Especially PTX distribution in the lung has been increased by 2 times and significant affinity and targeting to lung could be found. PTX distribution in the ovary and uterus was also increased significantly (P<0.05). Obvious sustained release performance could be found in the ovary and uterus with lower C_ max and longer MRT. No significant improvement for PTX distribution in the heart and kidney could be obtained after PSAP was modified by carboxymethyl chitosan (P> 0.05 ). CONCLUSION: Significant change of drug disposition in vivo could be found when iv CM-PSAP to rats using PSAP as control. CM-PSAP could improve the drug distribution in the liver, spleen, lung, ovary, and uterus especially in the lung. No significant improvement for PTX distribution in the heart and kidney could be got after PSAP being modified by carboxymethyl chitosan.
