Abstract:In this paper comparison of the four methods for isolating genomic DNA from cotton was studied. The results showed that both the modified SDS(sodium dodecyl sulphate) and CTAB(cetyl triethyl ammonium bromide) methods for isolating genomic DNA from cotton were better than the original SDS and CTAB methods. The Yield of genomic DNA was not only related to extraction methods, but also to the positions sampled from cotton plants. DNA yield of modified CTAB method was higher than that of modified SDS method. The yields of isolation of genomic DNA from the positions of cotton plants was in order: anther petalleave fiber. The cotton genomic DNA isolated by the modified methods could not only be digested by enzyme, but also to meet a demand for study of subsequent molecular such as SSR(simple sequence repeats) and AFLP(amplified fragment length polymorphism) analysis.

Abstract:To probe the adaptability of modified SDS method in apricot genomic DNA extraction and the optimal concentration, the genomic DNA samples of 6 apricot cultivars were isolated by 0.5%, 1%, 2%SDS extracted buffer. The results showed that good quality DNA would be obtained by modified SDS method and the 0.5% is better.

Abstract:Genomic DNA of Manilkara zapota was extracted using six different methods: CTAB,SDS and other improved methods,and compared in quality.The result showed that the improved CTAB in two way was the best,with high throughput,little degradation,less impurity and an OD260/OD280 ratio of around 1.8.

Abstract:Genomic DNA of leaves of mysterious fruit was extracted by means of six different methods of CTAB,SDS and all kinds of improved methods,The extraction DNA of these methods has been compared.The result showed that:the SDS improve method,increased the important content of extraction buffer and used method of two purification it was an effective way to solve all kinds of problem,degradation was little.impurity was little.That was the best method since the DNA extracted by it had the best OD260/OD280 ratio,it was 1.62 or so.

Abstract:Traditional CTAB method was improved by adding Vc in this experiment.The results showed that the best quality of DNA extracted from pear genome came from the method of adding 0.1 g/g of Vc when the sample was triturated.The DNA was optimal for RAPD analysis and other molecular biology researches.The results of the experiment also indicated that tender bud was the best material,which was followed by the shoot and mature leaf.

Abstract:The performances of three extraction methods of total RNA,Trizol method,modified SDS method and modified CTAB method,from‘Fuji’apple stem apical tissue were compared in this research. The yields,quality and integrity of RNA were tested by ultraviolet spectrophotometer analysis (OD260 /OD280) and 1. 2% agarose gel electrophoresis. The results indicated that good quality and high yields RNA would be obtained within 2 ～ 3 hours by changing extraction buffer of CTAB with less lithium chloride and absolute alcohol precipitation time. The RNA could be used for reverse transcription and PCR amplification efficiently. Therefore,the modified CTAB extraction method would be fast and efficient for total RNA isolation from apple tissues.

Abstract:Six methods included kit,high salt low pH,modified high salt low pH,column adsorption,SDS(Sodium dodecyl sulfate),modified CTAB(cetyltrimethyle ammonium bromide) were compared for extracting DNA from Agrostis varieties: Penncross,Seaside,PennA-4,Yuexuan No.1 and new strain Yuexuan No.10.The results showed that modified high salt low pH,modified CTAB and column adsorption methods could extract DNA of high-quality and high-integrity,as determined by agarose gel electrophoresis and UV-spectroscope.While the DNA productivity of column adsorption methods was lower than other two methods.The results of RAPD indicated that DNA extracted by these three methods satisfied the standard for gene molecular biological experiments.

Abstract:According to the high content of polysaccharides and polyphenols in apple fruits, three modified CTAB methods were used to extract genomic DNA of ‘Granny Smith’, with young leaves as control. The results showed that the method three was the best. This method integrated multiple purification steps, adding the interferent scavenger before cell lysis, increasing the concentration of β-mercaptoethanol and PVP to inhibite the oxidation of polyphenol, and adding 3 mol/L NaAC of 1/10 volume, which effectively removed the polysaccharides, polyphenols and other secondary metabolites. The results of ultraviolet spectrophotometer and agarose gel electrophoresis showed that DNA was high purity and good integrity. The results of ISSR primers amplification showed that the bands were clear. The results of the digestion of EcoR I restriction enzyme indicated that DNA could be digested by restriction enzyme. Therefore, the method three is the best for genomic DNA extraction. This method provides a reference for the removal of polysaccharides and polyphenols in the process of DNA extraction.

Abstract:This study aimed at comparing the different methods of DNA extraction from wheat. The results showed that the DNA quality extracted by standard operation was good, but it spent so much time and money. The DNA concentration and purity extracted by improved method were not as good as standard, but it also met the DNA requirements of major molecular research . The improved CTAB extraction method which was designed by our research group is suitable to extract the high quality DNA from wheat. Facile operation can save time and be economical. The quality of DNA extracted by improved CTAB method is so good that it fits for AFLP experiment.

Abstract:High quality genomic DNA from microorganism is the precondition of genetic manipulation.Now many methods for the extraction of genomic DNA are developed according to different research object and intention.Three kinds of methods widely applied are compared and reformed in isolation of genomic DNA from microorganism.And effective methods for extracting high pure genomic DNA are established considering different cell wall components.Genomic DNA gained by three different methods above is specifically amplified and clear band is observed by agarose gel electrophoresis,which is convenient to genetic manipulation later.

Abstract:Gastrodia elata B1 is rich in polysaccharides,phenol and other secondary metabolites.It is difficult to obtain high quality of genomic DNA from tissues by conventional methods such as CTAB(cety-trimethylammonium bromide)method and SDS(sodium dodecyl sulfate)method.Based on the research results of comparative analysis of several extraction methods for total genomic DNA of Gastrodia elata B1 in Orchidaceae, SDS-CTAB method was found to be optimal to produce high quality DNA templates which were suitable for polymerase chain reaction(PCR). The results also showed that among the different tissues of Gastrodia elata B1, the young floral shoots were the best position to extract good genomic DNA.

Abstract:Modern plant breeding studies are largely based on plant genetic engineering programs. Extraction of DNA with high quality plays as a key factor in most of plant genetic Studies, therefore two different DNA extraction protocols based on CTAB buffer and SDS buffer were tested for the purpose of selecting the best DNA extraction protocol from dry seeds of nine variety of barely plant . Barley seeds were taken from Libyan seeds gene bank .After Barely samples were prepared DNA was extracted directly using CTAB and SDS solutions .The quality of extracted DNA was assessed by spectrophotometric measurements and gel electrophoresis system. The results of this study showed that purity of extracted DNA by CTAB method was clearly batter compared with SDS method. CTAB method seemed to be more effective for extracting DNA from barely dry seeds. High quality DNA obtained through use of CTAB method, while CTAB had overall better A260/A280 ratio (1.736-1.932). SDS method seemed to be not suitable for DNA extraction from dry seeds of barely crop.

Abstract:The sodium dodecyl sulfate(SDS) and improved cetyltriethyl ammnonium bromide(CTAB) methods were used for the genomic DNA extraction of two kinds of Zanthoxylum plants in order to explore an efficient method for extracting genomic DNA from Zanthoxylum L.The results showed that the improved CTAB method was more suitable for the DNA extraction of Zanthoxylum plant.The improved CTAB method of extracting genomic DNA from Zanthoxylum plants was better than the SDS method,and the DNA purity of the former was higher than that of the latter.The OD260 /OD280 ratio of the extracted DNA with the improved CTAB method was in the range from 1.8 to 2.0,and distinct bands were found in the agarose gel electrophoresis map of ISSR.

Abstract:The high quality DNA required for construction of AFLP analysis system was isolated with an improved method of CTAB from young leaves of apricot.A fluorescent-AFLP reaction system was constructed for genetic analysis of apricot by optimizing DNA restriction-ligase reaction,pre-amplification and selective amplification.The clear and stable amplification patterns could be obtained from 12 pairs primer combinations with good polymorphism,which were selected from 64 pairs AFLP primer combinations.These results might provide a method based on the fluorescent-AFLP system for the further study in the field of molecular marker in cultivar identification,genetic diversity and genetic relationship in apricot.

Abstract:To find out a better one from three malt dextrin DNA extraction methods for the molecular marker analysis of high quality and sufficient samples.With malt dextrin purchased from the market as the tested material,the genomic DNA was extracted from malt dextrin using three different methods,such as CTAB method,SDS method and GuSCN method.The extraction effects of different methods were compared.The results of optical density detection showed that extraction amount and purity with GuSCN method were better than SDS method and CTAB method.The results of agarose gel electrophoresis determination showed that genome DNA band extracted by SDS method was clear and stable and genome DNA extracted by SDS method and CTAB method were not banded.The genomic DNA extracted with these methods all could be used in PCR reaction.The results showed that DNA could be obtained with three of all methods from malt dextrin,but the GuSCN method was the best extraction method.
