Abstract:BcMF2r gene,homologous with the BcMF2 gene encoding pollen-specific polygalacturonase of Chinese cabbage-pak-choi,was cloned from turnip (Brassica campestris L.spp.rapifera).'Wenzhoupancai' by PCR amplification,with two pairs of primers designed according to the sequence of BcMF2.The largest opening reading frame of BcMF2r gene was 1263 bp in length and encoded a protein of 420 amino acids.The molecular mass and isoelectric points of the putative protein,calculated by computer analysis,was 43.8 kDa and 8.108,respectively.Components of amino acids encoded by BcMF2r contained 36 basic amino acids(K,R),32 acidic amino acids(D,E),152 hydrophobic amino acids(A,I,L,F,W,V) and 113 polar amino acids(N,C,Q,S,T,Y).Blast result shows it shared 87% overall amino acid similarity to PGA3 of Arabidopsis.Sequence analysis revealed that it had three potential N-glycosylation sites,four protein kinase C phosphorylation sites,six casein kinase II phosphorylation sites,twelve N-myristoylation sites,one polygalacturonase active position(()~(257)RVTCGPGHGLSVGS),and so on.The predicted secondary structure composition for the protein had 7.86% helix,46.90% sheet and 45.24% loop.Four domains which are highly conserved in the all plants and fungal PGs were present in BcMF2r.Phylogenetic analysis showed that BcMF2r falled into the category of clade-C,which includes PG related to pollen.All results test that BcMF2r might act as polygalacturonase related to pollen development.
