Abstract:A profiling analysis of the total isoflavone extract of the root of Pueraria lobata (Wild.) Ohwi was performed using HPLC coupled with ESI-ion trap mass spectrometry. A total of seven isoflavones were identified according to their retention times, UV, MS data, and comparing with the literature data. Among them, two proposed new compounds were isolated and their structures were determined to be 8-C--glucofuranosyl-7,4'-dihydroxyisoflavone and 8-C--glucofuranosyl-7,4'-dihydroxyisoflavone, named as neopuerarin A (7) and neopuerarin B (6), on the basis of chemical and spectral analyses.

Abstract:An HPLC/DAD/MSn method was established for the qualitative and quantitative analysis of the impurities in puerarin injection (PI), a widely used drug in China. The analytical HPLC was performed on an Agela RP-C18 column using 0.1% aqueous formic acid (v:v) and methanol as mobile phase. A total of nine impurities were detected and eight of them were identified as isoflavone-C-glycosides basing on their UV spectra and MSn spectra and comparing with the literature data. An HPLC method for the assay of two common impurities in the commercial PI samples, i.e., neopuerarin A and neopuerarin B, was then established. The validation of the method, including sensitivity, linearity, precision, accuracy, was carried out. The calibration curves showed good linearity of R2>0.9999 and LOQ (S/N=10) were less than 3.73ng. The precision was evaluated by intra- and inter-day assays and R.S.D. values were less than 0.94%. The average recovery rates were 97.0% and 99.5%, respectively, with R.S.D. less than 1.38%. The contents of neopuerarin A and neopuerarin B in various commercial brands of PI samples varied over the range of 0.30–1.16% and 0.42–1.66%, respectively. This is the first report on the impurities in PI.

Abstract:Pueraria lobata (Willd.) Ohwi (Gegen) is a nutritious medicine food homology plant that has been recorded in many ancient remedies for diabetes. However, its hypoglycemic active components and related mechanisms have not been fully elucidated. In this study, potential alpha-glucosidase inhibitors in Pueraria lobata were investigated by liquid-liquid refined extraction, high-resolution profiling and their hypoglycemic activity were evaluated. Ten compounds showing alpha-glucosidase inhibitory bioactivity were separated and identified, including two new compounds, 8-geranyl-7, 3', 4' -trihydroxy-isoflavone (4) and 8-geranyl-7, 2', 4'-trihydroxy-isoflavone (6), and eight known compounds. All screened compounds showed potential hypoglycemic activity, in which compounds 4, 6, 7, 8, 9 and 10 showed strong inhibitory activity (IC50 < 8 mu M). Furthermore, molecular docking simulation revealed binding energy and interacting amino acid residues of the screened compounds with alpha-glucosidase, strongly supporting their potential as alpha-glucosidase inhibitors.
Study on Enzymatic Extraction of Isoflavone from Pueraria (Pueraria lobata) Root Residue

Abstract:Pueraria lobata(Willd.)Ohwi is a kind of raw material to produce starch in China.The root residue of pueraria from(pueraria-starch) manufactory still contains some isoflavone which is a kind of phytoestrogen with important physiological effects.By(pretreating) pueraria root residue with cellulase or xylanase before ethanol extraction,the total yield of isoflavone could be efficiently(increased.) The optimal cellulase dosage was 10 FPIU/g pueraria root residue and the suitable cellulase treatment time was 12 h.Total yield of isoflavone was 1.36 times as that by normal ethanol extraction.For pretreatment of pueraria root residue with xylanase,the(optimal) xylanase dosage was 300 IU/g residue for 6 h.Total yield of isoflavone was 1.52 times as that by regular ethanol extraction.It was proved that cellulase and xylanase could cooperate with each other to enhance extraction efficiency.When pueraria root residue was pretreated with both cellulase(7.5 FPIU/g residue) and xylanase(300 IU/g residue) for 6 h,total yield of isoflavone reached(138) mg/10 g pueraria root residue,which was 1.64 times as that by regular ethanol extraction.

Abstract:Glucose via acylation and bromization pathway gives acetobromo-α-D-glucose.The etherification of puerarin to 7-hydroxyl-4′-(2,3,4,6-tetraacetyl-β-D-glucopyranosyloxy) puerarin with acetobromo-α-D-glucose was carried out in acetone at the presence of potassium carbonate.The isolated yield was 61.4%(based on puerarin).Finally,7-hydroxyl-4′-(β-D-glucopyranosyloxy)puerarin was obtained via deacetylation in anhydrous methanol at the presence of potassium carbonate with the isolated yield of 90.5%.The structures of intermediates and final product were characterized by 1HNMR spectra and MS.Compared with puerarin,the water-solubility of 7-hydroxyl-4′-(β-D-glucopyranosyloxy) puerarin was increased by 14.2 times.

Abstract:Two new flavonoids, Alpinetin-7-O-[3''- O -galloyl-4'',6''-hexahydroxydiphenoyl]- β - D -glucose ( 1 ), Pinocembrin-7- O -[4''- O -galloyl]- β - D -glucose ( 2 ), and three known compound ( 3-5 ) were isolated from Penthorum chinense Pursh (Saxifragaceae). The structures of all compounds were deduced from their comprehensive spectroscopic analysis including IR, HR-ESI-MS, 1 H NMR, 13 C NMR, DEPT, COSY, HMBC and HMQC. Molecular docking model was used to test the anti-cholestatic liver activities of the isolated compounds, and compounds 1 and 2 showed higher docking scores (-9.90 and −11.27 kcal/mol, respectively) binding with FXR than 3 , 4 and 5 (-8.00, −9.77, −9.10 kcal/mol, respectively), suggesting 1 and 2 exhibited potential anti-cholestatic activities. The present results show that P. chinense is a potential source of 2 new lead compounds that can be utilised to produce therapeutic drugs for liver diseases upon further studies.

Abstract:Puerarin, a class of isoflavonoid compounds concentrated in the roots of Puerarias, has antipyretic, sedative, and coronary blood-flow-increasing properties. Although the biosynthetic pathways of puerarin have been investigated by previous researchers, studies focusing on the influence of different growth stages on the accumulation of metabolites in the puerarin pathway are not detailed, and it is still controversial at the last step of the 8-C-glycosylation reaction. In this study, we conducted a comprehensive analysis of the metabolomic and transcriptomic changes in Pueraria montana var. thomsonii during two growing years, focusing on the vigorous growth and dormant stages, to elucidate the underlying mechanisms governing the changes in metabolite and gene expression within the puerarin biosynthesis pathway. In a comparison of the two growth stages in the two groups, puerarin and daidzin, the main downstream metabolites in the puerarin biosynthesis pathway, were found to accumulate mainly during the vigorous growth stage. We also identified 67 common differentially expressed genes in this pathway based on gene expression differences at different growth stages. Furthermore, we identified four candidate 8-C-GT genes that potentially contribute to the conversion of daidzein into puerarin and eight candidate 7-O-GT genes that may be involved in the conversion of daidzein into daidzin. A co-expression network analysis of important UGTs and HIDs along with daidzein and puerarin was conducted. Overall, our study contributes to the knowledge of puerarin biosynthesis and offers information about the stage at which the 8-C-glycosylation reaction occurs in biosynthesis. These findings provide valuable insights into the cultivation and quality enhancement of Pueraria montana var. thomsonii.

Abstract:Kudzu (Pueraria montana lobata) is used as a traditional medicine in China and Southeast Asia but is a noxious weed in the Southeastern United States. It produces both O- and C-glycosylated isoflavones, with puerarin (C-glucosyl daidzein) as an important bioactive compound. Currently, the stage of the isoflavone pathway at which the C-glycosyl unit is added remains unclear, with a recent report of direct C-glycosylation of daidzein contradicting earlier labeling studies supporting C-glycosylation at the level of chalcone. We have employed comparative mRNA sequencing of the roots from two Pueraria species, one of which produces puerarin (field collected P. montana lobata) and one of which does not (commercial Pueraria phaseoloides), to identify candidate uridine diphosphate glycosyltransferase (UGT) enzymes involved in puerarin biosynthesis. Expression of recombinant UGTs in Escherichia coli and candidate C-glycosyltransferases in Medicago truncatula were used to explore substrate specificities, and gene silencing of UGT and key isoflavone biosynthetic genes in kudzu hairy roots employed to test hypotheses concerning the substrate(s) for C-glycosylation. Our results confirm UGT71T5 as a C-glycosyltransferase of isoflavone biosynthesis in kudzu. Enzymatic, isotope labeling, and genetic analyses suggest that puerarin arises both from the direct action of UGT71T5 on daidzein and via a second route in which the C-glycosidic linkage is introduced to the chalcone isoliquiritigenin.

Abstract:Two fractions of polysaccharides, named PTPS-1 and PTPS-2, was extracted and purified from puerh tea by Sephacryl S-300 column chromatography.The physicochemical properties of these two polysaccharides were investigated by high-performance liquid chromatography (HPLC), Fourier Transform IR spectra, scanning electron microscope (SEM) and atomic force microscope (AFM).Analysis of chemical compositions (protein, neutral sugars, uronic acid and monosaccharide composition) suggested that they were both kinds of acid heteropolysaccharides bound with protein, and contained seven monosaccharides with different molar ratio.Meanwhile, evaluation of antioxidant activities by in vitro assays of DPPH, ABTS and FRAP showed that PTPS-1 demonstrated stronger antioxidant ability than PTPS-2.Similarly, PTPS-1 exhibited remarkable inhibitory potential on α-glycosidase in vitro, significantly stronger than that of PTPS-2 and acarbose.Moreover, the results from animal test indicated PTPS-1 possessed significant inhibition on postprandial hyperglycemia in diabetic mice compared with the model group.

Abstract:Puerarin is an isoflavonoid extracted from the roots of Pueraria lobata in the Pueraria thomsonii family. Puerarin displays a broad range of pharmacological activities and has been widely used in clinics. However, puerarin's low solubility and permeability result in poor gastrointestinal absorption and low bioavailability. Simply increasing the dose does not effectively increase bioavailability and might lead to toxicity and side effects. Considerable progress has been achieved in the recent years on puerarin structural biotransformation using microorganisms and free enzymes. Increasingly numerous new puerarin derivatives with high solubility and bioactivity have been developed. This paper reviews recent progress in the biotransformation of puerarin.

Abstract:A mass spectrometry method that combines electrospray with an ion trap time-of-flight mass analyzer has been used to characterize puerarin (7,4'-dihydroxyisoflavone-8-C-beta-D-glucoside). MS' spectra (n <= 6) were obtained in positive and negative ion mode. The combination of accurate mass measurement in MS2 spectra and sequential MSn experiments enabled fragmentation pathways to be elucidated in detail. A novel structure for [M+H-150]+ has been found. Abstract:A new guaiane-type sesquiterpene glycoside, torilolone 8-O-beta-D-glucopyranoside (1), together with a known analogue compound, torilolone 11-O-beta-D-glucopyranoside (2), was isolated from the fruits of Daucus carota L. Their chemical structures were elucidated on the basis of MS, NMR spectroscopic analyses coupled with chemical degradation. The cytotoxic activities of two isolated compounds were evaluated against two human gastric cancer cell lines BGC-823 and AGS using MTT assay.
Simultaneous screening and isolation of activated constituents from Puerariae Flos by ultrafiltration with liquid chromatography and mass spectrometry combined with high-speed counter-current chromatography

Abstract:Puerariae Flos is the flower of Puerariae Radix, which is a common Chinese herb containing numerous isoflavones in all parts of the flower. Standard methods for screening and isolating isoflavones are typically labor intensive and time consuming. In this study, a new assay based on ultrafiltration with liquid chromatography and mass spectrometry was developed for the rapid screening and identification of ligands for α-glucosidase, xanthine oxidase and lactate dehydrogenase in the extract of Puerariae Flos. Three isoflavones were identified as α-glucosidase inhibitors, three isoflavones were identified as lactate dehydrogenase inhibitors, and no specific binding ligands were identified for xanthine oxidase in the extract. Subsequently, specific binding ligands, puerarin, genistin, and tectorigenin (purities were 90, 60, 99, and 91.73%, respectively), were separated by high-speed counter-current chromatography. The partition coefficient values of the target compounds and resolutions of peaks were employed as indicators and the solvent system and mobile phase flow rate were optimized for two-stage separation. An optimized two-phase solvent system comprised of ethyl acetate/ethanol/water (4:0.5:3, v/v/v) was successfully used to isolate the three compounds from Puerariae Flos. The monomer compounds isolated, collected, and purified by high-speed counter-current chromatography were analyzed by high-performance liquid chromatography, resulting in the isolation of three targeted compounds. The chemical structures of all three targeted compounds were individually identified by ultra high performance liquid chromatography with high-resolution mass spectrometry. The results demonstrate that ultrafiltration with liquid chromatography and mass spectrometry combined with high-speed counter-current chromatography is not only a powerful tool for screening and isolating α-glucosidase and lactate dehydrogenase inhibitors in complex samples, but also a useful platform for identifying bioactive compounds for preventing and treating diabetes and stroke.
Enzymatic Synthesis of Novel and Highly Soluble Puerarin Glucoside by Deinococcus geothermalis Amylosucrase

Abstract:Puerarin (daidzein-8-C-glucoside) is an isoflavone isolated from several leguminous plants of the genus Pueraria. Puerarin possesses several pharmacological properties; however, the poor solubility of puerarin limits its applications. To resolve this poor solubility, Deinococcus geothermalis amylosucrase (DgAS) was used to modify puerarin into more soluble derivatives. The results showed that DgAS could biotransform puerarin into a novel compound: puerarin-4′-O-α-glucoside. The biotransformation reaction was manipulated at different temperatures, pH values, sucrose concentrations, reaction times, and enzyme concentrations. The results showed that the optimal reaction condition was biotransformed by 200 μg/mL DgAS with 20% (w/v) sucrose at pH 6 and incubated at 40 °C for 48 h, and the optimal production yield was 35.1%. Puerarin-4′-O-α-glucoside showed 129-fold higher solubility than that of puerarin and, thus, could be further applied for pharmacological use in the future.

Abstract:An ultra performance liquid chromatography (UPLC) coupled with quadrupole time-of-flight mass spectrometry (QTOF/MS) method is established for the rapid analysis of isoflavones, saponins and flavones in 16 samples originated from the flowers of Pueraria lobata and P. thomsonii. A total of 25 isoflavones, 13 saponins and 3 flavones were identified by co-chromatography of sample extract with authentic standards and comparing the retention time, UV spectra, characteristic molecular ions and fragment ions with those of authentic standards, or tentatively identified by MS/MS determination along with Mass Fragment software. Moreover, the method was validated for the simultaneous quantification of 29 components. The samples from two Pueraria flowers significantly differed in the quality and quantity of isoflavones, saponins and flavones, which allows the possibility of showing their chemical distinctness, and may be useful in their standardization and quality control. Dataset obtained from UPLC-MS was processed with principal component analysis (PCA) and orthogonal partial least squared discriminant analysis (OPLS-DA) to holistically compare the difference between both Pueraria flowers.

Abstract:Two new guaianolides (1 and 2) were isolated from the aerial parts of Ajania fruticulosa along with a triterpene (alpha-amyrin), two plant sterols (beta-sitosterol and daucosterol), four flavonols [santin (3), axillarin (4), centaureidin, and 5,7,4'-trihydroxy-3, 3'-dimethoxyflavone], and five sesquiterpenes [ketoplenolide B, 9beta-hydroxyeudesma-4,11(13)-dien-12-oic acid, 9beta-acetoxyeudesma-4,11(13)-dien-12-oic acid, 1alpha,4alpha, 10alpha-trihydroxy-9alpha-angeloyloxyguaia-2,11(13)-dien-12, 6alpha-olide, and 3beta,4alpha-dihydroxyguaia-11(13),10(14)-dien-12, 6alpha-olide]. The structures of the new guaianolides were established as 1alpha-hydroperoxy-4beta,8alpha,10alpha, 13-tetrahydroxyguaia-2-en-12,6alpha-olide (1) and 1alpha-hydroperoxy-4alpha, 10alpha-dihydroxy-9alpha-angeloyloxyguaia-2,11(13)-dien-12, 6alpha-olide (2), respectively. Xanthine oxidase assays of all isolates revealed that santin (3) and axillarin (4) inhibited the enzyme with IC(50) values of 36.5 and 36.0 &mgr;M (that of allopurinol used as a positive control in the study was 24.2 &mgr;M), respectively.
Determination of puerarin in rat cortex by high-performance liquid chromatography after intravenous administration of Puerariae flavonoids.

Abstract:In the present study, a rapid and simple high-performance liquid chromatographic (HPLC) assay for determination of puerarin in rat cortex was developed. The analysis was carried out on a Zorbax SB-C-18 column with mobile phase acetonitrile-0.5% aqueous phosphoric acid (11:89, v/v). The detection was by UV at 252 nm. The calibration curve for puerarin was linear (r = 0.9999) over the concentration range 0.516-206.250 mu g/mL. The limit of detection was 0.206 mu g/mL (signal-to-noise ratio 3) and the limit of quantification (signal-to-noise ratio 10) was 0.516 mu g/mL. Stability studies showed that puerarin was stable at temperatures of 4 degrees C in methanol for at least 30 days. The intra- and inter-day assays of puerarin from rat cortex were less than 2.5% at concentration range 0.516-206.250 mu g/mL and good overall recoveries (97.4-101.7%) were found at same concentrations The method was applied to determine the pharmacokinetic parameters and the time course of puerarin in rat cortex, following a single dosage of intravenous administration of flavonoids from Puerariae radix at 32 mg/kg of puerarin to male Wistar rats. Abstract:Isoimperatorin (IP) and phellopterin (PP) are two furocoumarins existing in Angelicae Dahuricae Radix. There is an isopentenyloxyl substituted at C-5 in IP, and an isopentenyloxyl and a methoxyl substituted at C-8 and C-5, respectively, in PP. To elucidate the in vivo metabolic characteristics of PP and IP, HPLC coupled with diode array detector and electrospray ionization ion trap time-of-flight mass spectrometry technique was used. In total, 111 metabolites, including 53 new ones, were identified from the urine and plasma samples of rats after oral administration of IP and PP, respectively. The metabolites were formed through eight reactions on IP and PP: oxidation, hydroxylation-hydrogenation, carboxylation on the isopentenyloxyl, O-dealkylation, hydroxylation on the furocoumarin nucleus, ring-opening reaction on the furan ring and reduction or ring-opening reaction on the lactone ring. Among these, hydroxylation on the furocoumarin nucleus was found for the first time for in vivo metabolites of PP and IP, and the ring-opening reaction on the furan ring or lactone ring was found for the first time for in vivo metabolites of isopentenyloxyl furocoumarins. The research gave us a new insight into the in vivo metabolic profiles of IP and PP, which could help us better understand their important roles as two active constituents of Angelicae Dahuricae Radix.

Abstract:Eight unknown impurities in xinfujunsu and its injection were characterized by liquid chromatography coupled with ion trap/time-of-flight mass spectrometry (LC-IT-TOF MS). In order to determine the m/z values of the molecular ions and predict the formulas of all detected impurities, full scan LC-MS in positive ion mode was firstly executed to obtain the m/z value of the molecules. Then LC-MS2, LC-MS3 and LC-MS4 were carried out on target compounds to obtain as much structural information as possible. Based on MSn spectral data and exact mass measurements, the chemical structures of eight unknown impurities were characterized, among which three impurities were degradation impurities and five impurities were process impurities. In addition, the source of impurities and the correlation between process and impurities were also studied. The production of degraded impurities was caused in the high pressure sterilization process of xinfujunsu injection. Based on characterization of impurities, this study revealed the cause of impurity production and provided guidance for enterprises to improve the process to reduce impurity content. Furthermore, this study also provided scientific basis for the further improvement of official monographs in pharmacopoeias.
