Abstract:Cyclodepsipeptides are a large family of peptide-related natural products consisting of hydroxy and amino acids linked by amide and ester bonds. A number of cyclodepsipeptides have been isolated and characterized from fungi and bacteria. Most of them showed antitumor, antifungal, antiviral, antimalarial, and antitrypanosomal properties. Herein, this review summarizes the recent literatures (2010-2022) on the progress of cyclodepsipeptides from fungi and bacteria except for those of marine origin, in order to enrich our knowledge about their structural features and biological sources.

Abstract:Marine organisms are an important source of antitumour active substances. Thus, pharmaceutical research in recent years has focused on exploring new antitumour drugs derived from marine organisms, and, many peptide drugs with strong antitumour activities have been successfully extracted. Based on different mechanisms, this paper reviews the research on several typical antitumour bioactive peptides in marine drugs and the latest progress therein. Additionally, the development prospects for these antitumour bioactive peptide-based drugs are discussed so as to provide a reference for future research in this field.

Abstract:The biodiversity of the marine environment and the associated chemical diversity constitute a practically unlimited resource of new antitumor agents in the field of the development of marine bioactive substances. In this review, the progress on studies of antitumor peptides from marine sources is provided. The biological properties and mechanisms of action of different marine peptides are described; information about their molecular diversity is also presented. Novel peptides that induce apoptosis signal pathway, affect the tubulin-microtubule equilibrium and inhibit angiogenesis are presented in association with their pharmacological properties. It is intended to provide useful information for further research in the fields of marine antitumor peptides.

Abstract:Abstract: Cyclodepsipeptides, mainly derived from marine organisms and soil microorganisms, are amphiphilic molecules consisting of short oligopeptides with fatty acid tails attached to form a macrocyclic structure. Studies on the activity of cyclodepsipeptides have shown that they have cytotoxicity, antibacterial and anthelmintic effects, and are widely used in biological control, drug development, environmental remediation and disease treatment. Cyclodepsipeptides play a prominent role in the development of new drugs and drug lead compounds, especially as antibiotics with great medicinal potentiall, and are slowly seeping into the public consciousness. The biosynthesis of cyclodepsipeptides is mainly based on the synthesis of non-ribosomal peptide synthases, and selection of key regulatory enzymes for homologue regulation and biosynthetic strategies using genetic engineering and metabolic engineering approaches. The biosynthesis method is miniaturised, recyclable, and safer. The total synthesis methods of cyclodepsipeptides are mainly combined solid-liquid phase methods, which synthesise cyclodepsipeptides faster and are easy to purify. This paper reviews the biological activities of cyclodepsipeptides, their biosynthesis, and total synthesis.

Recent Patents on Anti-Cancer Drug Discovery

Abstract:BACKGROUND:Marine environment constitutes an almost infinite resource for novel anticancer drug discovery. The biodiversity of marine organisms provides a rich source for the discovery and development of novel anticancer peptides in the treatment of human cancer. Marine peptides represent a new opportunity to obtain lead compounds in biomedical field, particularly for cancer therapy.OBJECTIVE:Providing an insight of the recent progress of patented marine peptides and presenting information about the structures and mechanistic mode of anticancer activities of these marine peptides.METHODS:We reviewed recent progress on the patented anticancer peptides from marine organisms according to their targets on different signal pathways. This work focuses on relevant recent patents (2010-2018) that entail the anticancer activity with associated mechanism and related molecular diversity of marine peptides. The related cellular signaling pathways for novel peptides that induce apoptosis and affect tubulin-microtubule equilibrium, angiogenesis and kinase activity that are related to the anticancer and related pharmacological properties are also discussed.RESULTS:The recent patents (2010-2018) of marine peptides with anticancer activity were reviewed, and the anticancer activity of marine peptides with associated mechanism and related molecular diversity of marine peptides were also discussed.CONCLUSION:Marine peptides possess chemical diversity and display potent anticancer activity via targeting different signal pathways. Some of the marine peptides are promising to be developed as novel anticancer agents.

Abstract:Cancer is one of the leading causes of death in the world, and antineoplastic drug research continues to be a major field in medicine development. The marine milieu has thousands of biological species that are a valuable source of novel functional proteins and peptides, which have been used in the treatment of many diseases, including cancer. In contrast with proteins and polypeptides, small peptides (with a molecular weight of less than 1000 Da) have overwhelming advantages, such as preferential and fast absorption, which can decrease the burden on human gastrointestinal function. Besides, these peptides are only connected by a few peptide bonds, and their small molecular weight makes it easy to modify and synthesize them. Specifically, small peptides can deliver nutrients and drugs to cells and tissues in the body. These characteristics make them stand out in relation to targeted drug therapy. Nowadays, the anticancer mechanisms of the small marine peptides are still largely not well understood; however, several marine peptides have been applied in preclinical treatment. This paper highlights the anticancer linear and cyclic small peptides in marine resources and presents a review of peptides and the derivatives and their mechanisms.

Abstract:Depsipeptides, an important group of polypeptides containing residues of hydroxy acids and amino acids linked together by amide and ester bonds, have potential applications in agriculture and medicine. A growing body of evidence demonstrates that marine organisms are prolific sources of depsipeptides, such as marine cyanobacteria, sponges, mollusks, microorganisms and algae. However, these substances have not yet been comprehensively summarized. In order to enrich our knowledge about marine depsipeptides, their biological sources and structural features, as well as bioactivities, are highlighted in this review after an extensive literature search and data analysis.

Abstract:Marine environment has demonstrated to be an interesting source of compounds with uncommon and unique chemical features on which the molecular modeling and chemical synthesis of new drugs can be based with greater efficacy and specificity for the therapeutics. Cancer is a growing public health threat, and despite the advances in biomedical research and technology, there is an urgent need for the development of new anticancer drugs. In this field, it is estimated that more than 60% of commercially available anticancer drugs are natural biomimetic inspired. Among the marine organisms, algae have revealed to be one of the major sources of new compounds of marine origin, including those exhibiting antitumor and cytotoxic potential. These compounds demonstrated ability to mediate specific inhibitory activities on a number of key cellular processes, including apoptosis pathways, angiogenesis, migration and invasion, in both in vitro and in vivo models, revealing their potential to be used as anticancer drugs. This review will focus on the bioactive molecules from algae with antitumor potential, from their origin to their potential uses, with special emphasis to the alga Sphaerococcus coronopifolius as a producer of cytotoxic compounds.

Abstract:Marine sponge is a kind of symbionts with many species. It grows in a very complicated environment with a very special route of metabolism so there are abundant metabolites. The study on bioactive peptides from marine sponge is up and doing now. The analysis of the structure of bioactive peptide from marine sponge, the bioactivities of marine sponge, such as anti-tumour, anti-virus, anti-bacteria and so on, were reviewed. It was pointed out that marine sponge is an infinite storeroom of medicine. And the prospect of the application of bioactive peptides form marine sponge was viewed.

Abstract:Most biological resources of the earth exist in the ocean and there are abundant new bioactive peptides among them. It has been showed that many marine polypeptides have obvious pharmacological activities, such as antitumor, anti-AIDS, antifungus, prevention and cure of cardiovascular diseases, immunomodulation, etc. This paper reviews the progress on studies of bioactive polypeptides from marine products in the past five years and gives a prospective outline on the study of this field in the near future.
