Abstract:Fucoidan was isolated from the brown seaweed by multienzyme enzymolysis method,and the method was optimized by orthogonal experimental design.After removing the protein in the crude fucoidan using Sevage method,we got two components from crude fucoidan by DEAE-Sepharose FF ion exchange chromatogram,and analyzed their structure and antioxidation.The total sulfate and polysaccharide reached 21.25% and 30.24% respectively.

Abstract:Some factors of the barium chloride-gelation method and the phenol-H2SO4 method which were used to determine the total sulfate and polysaccharide of fucoidan,and analyze the fucoidan extracted by ultrasonic wave assisted extraction method which optimized by orthogonal experimental design.The total sulfate and polysaccharide reached 22.49% and 36.97%,respectively.

Abstract:Fucoidans are multifunctional marine macromolecules that are subjected to numerous and various downstream processes during their production. These processes were considered the most important abiotic factors affecting fucoidan chemical skeletons, quality, physicochemical properties, biological properties and industrial applications. Since a universal protocol for fucoidans production has not been established yet, all the currently used processes were presented and justified. The current article complements our previous articles in the fucoidans field, provides an updated overview regarding the different downstream processes, including pre-treatment, extraction, purification and enzymatic modification processes, and shows the recent non-traditional applications of fucoidans in relation to their characters.

Abstract:The marine acid polysaccharide fucoidan has attracted attention from both the food and pharmaceutical industries due to its promising therapeutic effects. Fucoidan is a polysaccharide that mainly consists of L-fucose and sulphate groups. Its excellent biological function is attributed to its unique biological structure. Classical activities include antitumor, antioxidant, anticoagulant, antithrombotic, immunoregulatory, antiviral and anti-inflammatory effects. More recently, fucoidan has been shown to alleviate metabolic syndrome, protect the gastrointestinal tract, benefit angiogenesis and bone health. This review focuses on the progress in our understanding of the biological activities of fucoidan, highlighting its benefits for the treatment of human disease. We hope that this review can provide some theoretical basis and inspiration for the product development of fucoidan.

Abstract:A low molecular weight fucoidan (DFPS), obtained from the brown seaweed Laminaria japonica, was separated into three fractions by anion-exchange column chromatography. All three fractions (DF1, DF2, DF3) predominantly contained variety fucose, galactose and sulfate group. The antioxidant and anticoagulant activities of these fractions were investigated. The results showed that all fractions possessed considerable antioxidant activities, DF1 was most active. All samples inhibited coagulant in APTT, TT and PT assays obviously. Available data presented the content of sulfate group, the molar ratio of sulfate/fucose and sulfate/total sugar, and the molecular weight played an important role on antioxidant and anticoagulant activity.

Abstract:Oligosaccharides derived from natural resources are attracting increasing attention as both food and nutraceutical products because of their beneficial health effects and lack of toxicity. During the past few decades, many studies have focused on the potential health benefits of fucoidan. Recently, new interest has emerged in fucoidan, partially hydrolysed into fuco-oligosaccharides (FOSs) or low-molecular weight fucoidan, owing to their superior solubility and biological activities compared with fucoidan. There is considerable interest in their development for use in the functional food, cosmetic, and pharmaceutical industries. Therefore, this review summarises and discusses the preparation of FOSs from fucoidan using mild acid hydrolysis, enzymatic depolymerisation, and radical degradation methods, and discusses the advantages and disadvantages of hydrolysis methods. Several purification steps performed to obtain FOSs (according to the latest reports) are also reviewed. Moreover, the biological activities of FOS that are beneficial to human health are summarised based on evidence from in vitro and in vivo studies, and the possible mechanisms for the prevention or treatment of various diseases are discussed.

For the past decade fucoidan has been extensively studied due to its numerous interesting biological activities. Recently the search for new drugs has raised interest in fucoidans. In the past few years, several fucoidans' structures have been solved, and many aspects of their biological activity have been elucidated. This review summarizes the research progress on the structure and bioactivity of fucoidan and the relationships between structure and bioactivity.

Abstract:Due to antitumor and antioxidant activities, there has been increasing interest in fucoidans, sulfated polysaccharides, from brown algae. Currently, the classic hot water extraction and acid extraction are time-consuming, and can cause the partial cleavage of sulfate esters, leading to bioactivity loss. The present study developed a novel process for extracting fucoidans from Laminaria japonica with 0.5% ethylenediaminetetraacetic acid at 70 °C. Crude fucoidans (CFs) obtained with a yield (∼16%) was purified to different fractions of F1, F2 and F3. The fractions displayed similar anticancer activity against Hela cells. Due to the differences in sulfate content, molar ratio of sulfate to fucose and monosaccharide composition, F1 and F2 exhibited stronger antioxidant ability than CFs and F3. This novel extraction provides an efficient procedure for extracting fucoidans from brown seaweeds with high extraction yield and removing pigments with one single step.

Abstract:Recent studies have demonstrated that fucoidans and their degradation products possess various biological activities. Thus, the enzymes to hydrolyze fucoidans attract more attention. This review summarizes the recent achievements in the discovery, detection of activity, properties and application of fucoidan-degrading enzymes derived from marine microorganisms. Also, the approaches to understand fucoidan-degrading enzymes from marine microorganisms are introduced. It is expected that the rapid development of modern omics and structural biology will promote the research of novel fucoidan-degrading enzymes derived from marine microorganisms.

Abstract:A fucoidan derived from marine brown algae has great potential in biomedical filed. Herein, the fucoidan was successfully isolated and purified by using chitosan microspheres, resulting in the sulfate fucoidan (CF) with degree of sulfation (DS) of 0.94. CF was identified to be highly branched, consisting of fucose (77.4%) and galactose (13.9%), etc., supported by the results of GCMS and light scattering with the structure-sensitive parameter of 0.98 in 0.15 M aqueous NaNO3. The individual CF and its desulfated derivative chains adopted sphere-like conformation in water, observed by atomic force microscopy. CF exhibited higher antiangiogenesis than the desulfated one and strong antileukemia activities through inhibiting cell proliferation and inducing cell apoptosis via cell cycle arrest at G1 phase in vitro. This work provided important information that ester sulfate groups of polysaccharide played an important role in the enhancing of bioactivities of fucoidan, and put forward to a potential drug to treat acute myelocytic leukemia (AML) and tumors.

Abstract:The importance of fucoidan as a functional ingredient in food, health products, and pharmaceutics is well-recognized due to its beneficial biological effects. Fucoidan is usually extracted from brown seaweeds, including Undaria pinnatifida. Fucoidan exhibits beneficial bio-activity and has antioxidant, anticancer, and anticoagulant properties. This review focuses on the biological activity of U. pinnatifida-derived fucoidan and investigates its structure–activity or fraction–activity relationship. It also describes several fucoidan extracts, along with their claimed anticancer effects. It aims to provide information and thoughts for future research such as the development of fucoidan into functional foods or nutraceuticals.

Abstract:Fucoidans are valuable marine polysaccharides with various bioactivities and physicochemical properties. However, its digestive properties, mucoadhesive properties, and bioactivity in the gastrointestinal tract are still unclear. In this study, simulated digestion, fecal fermentation in vitro , and rheology models were utilized to investigate the chain conformation, influence on gut microbiota, and mucin adhesive properties of fucoidan from the sea cucumber Thelenota ananas ( Ta -FUC). The results showed that Ta -FUC was nondigestible with a temporary decrease in molecular weight in gastric conditions, accompanied by the chain conformation becoming more flexible. Moreover, Ta- FUC exhibited strong mucin adhesive function in the simulated intestinal environment, with supramolecular disulfide, hydrogen, and hydrophobic interactions in order of intensity. During fermentation, Ta -FUC was degraded by the intestinal flora to produce various short-chain fatty acids and promoted the relative abundance of Bacteroidota and Firmicutes , reducing the proportion of Proteobacteria . Therefore, these results indicate that Ta -FUC could be a potential prebiotic and ingredient for developing targeted delivery systems in the functional food and pharmaceutical industries.

Abstract:Fucoidan from Laminaria japonica became sterilized with an autoclave and ultraviolet (UV) radiation. Potential prebiotic and antibacterial activities of sterilized fucoidans (SF) were the subject of investigation. Molecular weight, monosaccharide composition, FTIR, and NMR spectra of SF underwent evaluations to elucidate the relationship between the structure and activities of SF. The growth of Lactobacillus rhamnosus GG and L. acidophilus with autoclave sterilized fucoidan (ASF) and the growth of L. plantarum, L. gasseri, L. paracasei, and L. reuteri with UV sterilized fucoidan (USF) increased significantly. Also, fucoidan was vastly more effective than fructooligosaccharides in improving the growth of L. gasseri, L. reuteri, and L. paracasei. The growth of Escherichia coli and Bacillus cereus decreased at each SF concentration. ASF was more effective against E. coli, B. cereus, and Staphylococcus aureus than the USF efficiency. However, USF exhibited more inhibitory effects on the growth of Enterobacteriaceae compared to the ASF efficiency. When comparing the ASF and USF, autoclave caused a considerable decrease in molecular weight and uronic acid content, increased fucose and galactose, and made no significant changes in NMR spectra. Fucoidan effectively promoted probiotic bacterial growth and reduced pathogenic outbreaks in the medium. Therefore, it can occur as a new algal prebiotic and antibacterial agent.

Abstract:Fucoidan is a polysaccharide largely made up of l-fucose and sulfate groups. Fucoidan is favorable worldwide, especially amongst the food and pharmaceutical industry as a consequence of its promising therapeutic effects. Its applaudable biological functions are ascribed to its unique biological structure. Classical bioactivities associated with fucoidan include anti-oxidant, anti-tumor, anti-coagulant, anti-thrombotic, immunoregulatory, anti-viral and anti-inflammatory effects. More recently, a variety of in vitro and in vivo studies have been carried out to further highlight its therapeutic potentials. This review focuses on the progress towards understanding fucoidan and its biological activities, which may be beneficial as a future therapy. Hence, we have summarized in vitro and in vivo studies that were done within the current decade. We expect this review and a variety of others can contribute as a theoretical basis for understanding and inspire further product development of fucoidan.

Effects of UV/H2O2 Degradation on the Physicochemical and Antibacterial Properties of Fucoidan

Abstract:The applications of fucoidan in the food industry were limited due to its high molecular weight and low solubility. Moderate degradation was required to depolymerize fucoidan. A few studies have reported that fucoidan has potential antibacterial activity, but its antibacterial mechanism needs further investigation. In this study, the degraded fucoidans were obtained after ultraviolet/hydrogen peroxide treatment (UV/H2O2) at different times. Their physicochemical properties and antibacterial activities against Staphylococcus aureus and Escherichia coli were investigated. The results showed that the average molecular weights of degraded fucoidans were significantly decreased (up to 22.04 times). They were mainly composed of fucose, galactose, and some glucuronic acid. Fucoidan degraded for 90 min (DFuc-90) showed the strongest antibacterial activities against Staphylococcus aureus and Escherichia coli, with inhibition zones of 27.70 + 0.84 mm and 9.25 + 0.61 mm, respectively. The minimum inhibitory concentrations (MIC) were 8 mg/mL and 4 mg/mL, respectively. DFuc-90 could inhibit the bacteria by damaging the cell wall, accumulating intracellular reactive oxygen species, reducing adenosine triphosphate synthesis, and inhibiting bacterial metabolic activity. Therefore, UV/H2O2 treatment could effectively degrade fucoidan and enhance its antibacterial activity.

Abstract:Low molecular weight seaweed polysaccharides exhibit promising potential as novel therapeutics for the prevention of obesity and gut microbiota dysbiosis. The interplay between polysaccharides and gut microbiota may play crucial roles in their anti-obesity effects, but is largely unknown, including the impact of polysaccharides on the composition of the gut microbiota with polysaccharide-degrading capacity. The primary structure of a 5.1 kDa fucan (J2H) from Saccharina japonica was characterized and oral administration of J2H effectively suppressed high-fat diet-induced obesity, blood glucose metabolic dysfunction, dyslipidemia, and gut microbiota dysbiosis. Furthermore, the Jensen-Shannon divergence analysis demonstrated that J2H enriched at least four gut bacterial species with fucoidan-degrading potential, including Bacteroides sartorii and Bacteroides acidifaciens. Our findings suggest that the low molecular weight S. japonica fucan, J2H, is a promising potential agent for obesity prevention and its enrichment of gut bacteria with fucoidan-degrading potential may play a vital role in the anti-obesity effects.

Abstract:Laminaria digitata is a high-quality seaweed resource that is widely cultured and has good application prospects. In this study, Laminaria digitata fucoidan (LF) was extracted from Laminaria digitata, and purified using DEAE-Sepharose Fast Flow gel column to obtain four different grades. Among those, LF4 (Mw:165 kDa), mainly composed of fucose(56.80 %), had the highest total sugar (66.91 %) and sulfate (17.07 %) content. FT-RT and NMR results showed that LF4 was mainly composed of galactosylated galactofucose, and has a sulfate group attached to fucose C4. With the animal experimentation, it was revealed that hyperlipidaemic mice had significantly higher levels of TC (5.52 mmol/L), TG (2.28 mmol/L) and LDL-C (5.12 mmol/L) and significantly lower levels of HDL-C (2 mmol/L). However, LF had the efficacy in modulating the lipid metabolism disturbances induced by hyperlipidemia, as well as the ability to regulate cholesterol transport in serum. Moreover, it regulated AMPK/ACC, PPAR-α/LAXRa, Nrf2/Nqo1, TLR4/NF-κB signaling pathway genes and proteins expression in the liver. In addition, it promoted the production of beneficial short-chain fatty acids (SCFAs) while improving the composition and structure of gut microbiota, including balancing the abundance of Bacteroidota, Firmicutes, Muribaculaceae, Alloprevotella, Escherichia-Shigella, Prevotella and NK4A136. The results clearly indicated that LF4 could significantly ameliorate hyperlipidemia, suggesting its prospective application as a functional food.

Abstract:One-factor-at-a-time and orthogonal array methods were used to investigate the optimal ultrasonic-assisted extraction process of fucoidan from Laminaria japonica in this work. The optimum extraction conditions were ultrasonic treatment power of 250 W, ultrasonic treatment time of 25 min, material/liquid ratio of 1:70 (g/mL) and extraction temperature of 60℃. Under this optimum condition the extraction rate , the contents of fucose and sulfate group were 5.58%, 20.93%and 26.87%, respectively, higher than obtained by hot water extraction. Neither protein nor nucleic acid was found in fucoidan obtained using hot water extraction or ultrosonic-assisted extraction method, and less pigment was found in fucoidan obtained using ultrosonic-assisted extraction method.
