Abstract:Some factors of the barium chloride-gelation method and the phenol-H2SO4 method which were used to determine the total sulfate and polysaccharide of fucoidan,and analyze the fucoidan extracted by ultrasonic wave assisted extraction method which optimized by orthogonal experimental design.The total sulfate and polysaccharide reached 22.49% and 36.97%,respectively.

Abstract:Fucoidan was isolated from the brown seaweed by multienzyme enzymolysis method,and the method was optimized by orthogonal experimental design.After removing the protein in the crude fucoidan using Sevage method,we got two components from crude fucoidan by DEAE-Sepharose FF ion exchange chromatogram,and analyzed their structure and antioxidation.The total sulfate and polysaccharide reached 21.25% and 30.24% respectively.

Abstract:One-factor-at-a-time and orthogonal array methods were used to investigate the optimal ultrasonic-assisted extraction process of fucoidan from Laminaria japonica in this work. The optimum extraction conditions were ultrasonic treatment power of 250 W, ultrasonic treatment time of 25 min, material/liquid ratio of 1:70 (g/mL) and extraction temperature of 60℃. Under this optimum condition the extraction rate , the contents of fucose and sulfate group were 5.58%, 20.93%and 26.87%, respectively, higher than obtained by hot water extraction. Neither protein nor nucleic acid was found in fucoidan obtained using hot water extraction or ultrosonic-assisted extraction method, and less pigment was found in fucoidan obtained using ultrosonic-assisted extraction method.

Abstract:Due to antitumor and antioxidant activities, there has been increasing interest in fucoidans, sulfated polysaccharides, from brown algae. Currently, the classic hot water extraction and acid extraction are time-consuming, and can cause the partial cleavage of sulfate esters, leading to bioactivity loss. The present study developed a novel process for extracting fucoidans from Laminaria japonica with 0.5% ethylenediaminetetraacetic acid at 70 °C. Crude fucoidans (CFs) obtained with a yield (∼16%) was purified to different fractions of F1, F2 and F3. The fractions displayed similar anticancer activity against Hela cells. Due to the differences in sulfate content, molar ratio of sulfate to fucose and monosaccharide composition, F1 and F2 exhibited stronger antioxidant ability than CFs and F3. This novel extraction provides an efficient procedure for extracting fucoidans from brown seaweeds with high extraction yield and removing pigments with one single step.

Abstract:The application of an ultrasound-assisted extraction method, combined with ion-exchange chromatography, has allowed separation of the total fucoidan fraction (F1) from the brown alga Fucus evanescens into two fractions, F2 and F3, in the ratio 1:0.2. The traditionally extracted F1 differs from F3 only by the acetate groups at C3 being somewhat predominant. F2 is described as having acetate groups located chaotically, and contains galactose and xylose residues, in addition to fucose. F3 consists of fucose residues acetylated almost exclusively at C3. Therefore, with ultrasound-assisted extraction from the brown alga Fucus evanescens, a new regular 1 → 3;1 → 4-α-L-fucan was obtained. It is sulphated at C2 and acetylated at C3 of the fucose residue and showed anticancer activity in vitro towards human colon carcinoma cells comparable to the total fraction F1.

Abstract:Fucoidan has attracted considerable attention from scientists and pharmaceutical companies due to its antioxidant, anticoagulant, anti-inflammatory, anti-tumor, and health-enhancing properties. However, the extraction of fucoidan from seaweeds often involves the use of harsh chemicals, which necessitates the search for alternative solvents. Additionally, the high viscosity and low cell permeability of high molecular weight (Mw) fucoidan can limit its effectiveness in drug action, while lower Mw fractions exhibit increased biological activity and are also utilized as dietary supplements. The study aimed to (1) extract fucoidan from the seaweed Fucus vesiculosus (FV) using an environmentally friendly solvent and compare it with the most commonly used extraction solvent, hydrochloric acid, and (2) assess the impact of ultrasound-assisted depolymerization on reducing the molecular weight of the fucoidan extracts and examine the cytotoxic effect of different molecular weight fractions. The findings indicated that the green depolymerization solvent, in conjunction with a brief ultrasound treatment, effectively reduced the molecular weight. Moreover, a significant decrease in cell viability was observed in selected samples, indicating potential anticancer properties. As a result, ultrasound was determined to be an effective method for depolymerizing crude fucoidan from Fucus Vesiculosus seaweed.

Abstract:Fucoidan, a sulphated polysaccharide, is found exclusively in brown seaweeds and has been reported to possess a wide range of biological functionalities. Fucoidans are found within the cell wall of brown seaweeds, which is composed of recalcitrant cellulose and hemicellulose. This hampers the recovery of fucoidans. In addition, fucoidans are found within a network of viscous hydrocolloids, alginates, further complicating their recovery. Traditionally, the hot water extraction method is used to recover fucoidans from brown seaweed, however, this is characterized by low yields and long extraction time. To combat these issues, several novel extraction technologies have been introduced, these include ultrasound assisted extraction and enzyme-assisted extraction. Thus, the main aim of this study was to investigate and optimize fucoidan recovery from Ecklonia maxima based on ultrasound-assisted enzymatic extraction. The impact of temperature (40–65 °C), ultrasound intensity (0–118 W·cm −2 ), enzyme dosage (0–0.05 ml·g −1 ) and pH (4.5–6) on total dissolved, total carbohydrates and inorganic sulphates yields was studied. The application of ultrasound-assisted enzymatic extraction mainly improved the extraction of total carbohydrates. Ultrasound significantly improves the kinetics and extraction of fucoidan, but there was no merit when it was applied with enzymes. Results reveal that at optimized conditions, the fucoidan extracted 79.13 mg.g −1 (7.9 % w/w) of algal dry weight. The present study provides insight into the extraction potentials of enzyme-assisted extraction, ultrasound-assisted extraction, and ultrasound-assisted enzymatic extraction.

Abstract:Fucoidan, a sulfated polysaccharide known for its anticancer, immunomodulant, antioxidant and antimicrobial activity is abundant in brown seaweeds. Sargassum ilicifolium , a brown alga, was not extensively explored for fucoidan extraction using different methods and assessments of their impact on its structural and therapeutic characteristics. This study aimed to compare the structural features, biological activity, solubility, and emulsification characteristics of fucoidan extracted from Sargassum ilicifolium using conventional and ultrasonic bath extraction. The ultrasonic bath extraction method was 2.5 times higher than the conventional method to extract fucoidan. The structural integrity of fucoidan obtained by both methods was compared using FTIR, 1 HNMR, XRD, FESEM, EDX, and DSC. The monosaccharide composition of fucoidan was determined by TLC. The IC 50 values for antioxidant, anti-inflammatory and antitumor effects on oral cancer cells were found to be significantly lesser for crude fucoidan of S. ilicifolium obtained by ultrasonic bath method than for conventional extraction. Furthermore, fucoidan from the alga has emulsifying and water-holding characteristics which supports its potential use in the food processing industries.

Abstract:The effects of ultrasound on the molecular weight, structure, and antioxidant potential of a fucoidan found in Isostichopus badionotus were investigated. The results showed the molecular weight (Mw) of fucoidan decreased obviously after ultrasound treatment. Higher ultrasonic intensity, lower temperature, and lower fucoidan concentrations led to a more effective sonochemical effect. The kinetic model for fucoidan degradation fitted to 1/M(wt)-1/M(w0) = kt at the tested temperature. The optimized degradation conditions by response surface methodology (RSM) were temperature, 12 °C, and intensity, 508 W/cm². Structural analysis by FTIR and NMR indicated the fucoidan kept the linear tetrasaccharide repeating units as the original polysaccharides after the ultrasound treatment, with only slight destruction of the middle nonsulfated fucose units. Antioxidant activity assay showed the antioxidant activity was slightly improved by the ultrasound treatment. The results suggested that ultrasound treatment is an effective approach to decrease the M(w) of fucoidan with only minor structural destruction.

Abstract:Fucoidan refers to a type of polysaccharide which contains substantial percentages of L-fucose and sulfate ester groups, mainly derived from brown seaweed. For the past decade fucoidan has been extensively studied due to its numerous interesting biological activities. Recently the search for new drugs has raised interest in fucoidans. In the past few years, several fucoidans' structures have been solved, and many aspects of their biological activity have been elucidated. This review summarizes the research progress on the structure and bioactivity of fucoidan and the relationships between structure and bioactivity.

Abstract:Fucoidan is a sulphated polysaccharide with diversified biological activities commonly found in brown seaweeds. In our present study, fucoidan was extracted from Sargassum polycystum collected from Saint Martin's Island in three different extraction methods. The composition of the fucoidans was determined by measuring the total carbohydrate, sulfate, uronic acid, fucose, and protein contents. Fourier-transform infrared spectroscopy (FT-IR) and Proton nuclear magnetic resonance ( 1 H NMR) were studied to determine the structural groups of extracted fucoidans. Functional activities were analyzed by antioxidant activity and anti-diabetic activity assay. Fucoidan yield was obtained highest in the acid extraction method (0.061 ± 0.007 g/g), followed by the water extraction method (0.058 ± 0.005 g/g) and salt extraction method (0.041 ± 0.005 g/g). Water extracted fucoidan showed the highest carbohydrate content (62.97 %), uronic acid content (6.46 %), and protein content (2.62 %). Sulphate content (28.56 %) was highest in salt extracted fucoidan, whereas fucose content (24.0 %) was highest in water extracted fucoidan. FT-IR and 1 H NMR analysis confirm several characteristic picks for the extracted fucoidans. Extracted fucoidans presented comparable antioxidant activity, but salt-extracted fucoidan presented the highest DPPH radical scavenging assay (86.85 ± 0.74 %, IC 50 = 0.03 mg/mL), ABTS radical scavenging assay (65.13 ± 0.92 %, IC 50 = 1.94 mg/mL), Phosphomolybdenum assay (4.356 ± 0.11) and Nitric Oxide (NO) radical scavenging assay (57.08 ± 0.52 %, IC 50 = 3.73 mg/mL) in dose-dependency manner. Further anti-diabetic activity assay resulted in the highest α-amylase inhibition activity (62.82 ± 1.36 %, IC 50 = 2.30 mg/mL) and α-glucosidase inhibition activity (55.73 ± 0.84 %, IC 50 = 3.71 mg/mL) by salt-extracted fucoidan. These functional potentials of seaweed may add significant value to the development of functional food and pharmaceutical materials.

Purification and Molecular Characterization of Fucoidan Isolated from Ascophyllum nodosum Brown Seaweed Grown in Ireland

Abstract:The present study investigates the molecular characteristics of fucoidan obtained from the brown Irish seaweed Ascophyllum nodosum, employing hydrothermal-assisted extraction (HAE) followed by a three-step purification protocol. The dried seaweed biomass contained 100.9 mg/g of fucoidan, whereas optimised HAE conditions (solvent, 0.1N HCl; time, 62 min; temperature, 120 °C; and solid to liquid ratio, 1:30 (w/v)) yielded 417.6 mg/g of fucoidan in the crude extract. A three-step purification of the crude extract, involving solvents (ethanol, water, and calcium chloride), molecular weight cut-off filter (MWCO; 10 kDa), and solid-phase extraction (SPE), resulted in 517.1 mg/g, 562.3 mg/g, and 633.2 mg/g of fucoidan (p < 0.05), respectively. In vitro antioxidant activity, as determined by 1,1-diphenyl-2-picryl-hydrazyl radical scavenging and ferric reducing antioxidant power assays, revealed that the crude extract exhibited the highest antioxidant activity compared to the purified fractions, commercial fucoidan, and ascorbic acid standard (p < 0.05). The molecular attributes of biologically active fucoidan-rich MWCO fraction was characterised by quadruple time of flight mass spectrometry and Fourier-transform infrared (FTIR) spectroscopy. The electrospray ionisation mass spectra of purified fucoidan revealed quadruply ([M+4H]4+) and triply ([M+3H]3+) charged fucoidan moieties at m/z 1376 and m/z 1824, respectively, and confirmed the molecular mass 5444 Da (~5.4 kDa) from multiply charged species. The FTIR analysis of both purified fucoidan and commercial fucoidan standard exhibited O-H, C-H, and S=O stretching which are represented by bands at 3400 cm−1, 2920 cm−1, and 1220–1230 cm−1, respectively. In conclusion, the fucoidan recovered from HAE followed by a three-step purification process was highly purified; however, purification reduced the antioxidant activity compared to the crude extract.

Abstract:Fucoidan is a bioactive compound of brown seaweed with antioxidant characteristics. This study examined the aftermath of the extraction method on the yield, fucose content, xylose content, sulfate content, total sugar, antioxidant activity, and functional groups of fucoidan from Sargassum hystrix. The brown seaweed was extracted using 4 methods, namely, A (0.1 N HCl, room temperature, 24 h), B (2% CaCl2, 85°C, 4 h), C (85% ethanol, room temperature, 12 h), and D (0.5% EDTA, 70°C, 3 h). The antioxidant activity testing was carried out using the 2,2-diphenyl-1-picrylhydrazyl (DPPH), Ferric-Reducing Antioxidant Power (FRAP), and Hydroxyl Radical Scavenging Activity (HRSA). The yield for methods of A, B, C, and D was 2.46 ± 0.30 , 0.68 ± 0.34 , 1.18 ± 0.15 , and 0.62 ± 0.25 % , with fucose content of 39.97 ± 4.82 , 26.72 ± 3.38 , 41.08 ± 9.49 , and 40.62 ± 8.59 % , xylose content of 8.07 ± 0.92 , 5.63 ± 0.40 , 6.80 ± 0.83 , and 7.83 ± 1.83 % , and the sulfate content of 11.47 ± 2.20 , 15.31 ± 2.47 , 30.62 ± 2.76 , and 27.80 ± 3.59 % . The result indicated the occurrence of a sulfate ester group in the functional group analysis with numerous similarities with the commercial fucoidan. The highest antioxidant activity of fucoidan from S. hystrix was found in method C, which was influenced by sulfate levels. Therefore, the extraction method of fucoidan from S. hystrix affects the characteristics and antioxidant activity.
Ultrasound-Assisted Extraction of Alginate from Fucus vesiculosus Seaweed By-Product Post-Fucoidan Extraction

Abstract:The solid phase byproduct obtained after conventional fucoidan extraction from the brown seaweed Fucus vesiculosus can be used as a source containing alginate. This study involves ultrasound-assisted extraction (UAE) of alginate from the byproduct using sodium bicarbonate. Response surface methodology (RSM) was applied to obtain the optimum conditions for alginate extraction. The ultrasound (US) treatments included 20 kHz of frequency, 20–91% of amplitude, and an extraction time of 6–34 min. The studied investigated the crude alginate yield (%), molecular weight, and alginate content (%) of the extracts. The optimum conditions for obtaining alginate with low molecular weight were found to be 69% US amplitude and sonication time of 30 min. The alginate extracts obtained were characterized using Fourier transform infrared (FTIR) spectroscopy, thermogravimetric analysis (TGA), and differential scanning calorimetry (DSC). Ultrasound-assisted extraction involving a short treatment lasting 6–34 min was found to be effective in extracting alginate from the byproduct compared to the conventional extraction of alginate using stirring at 415 rpm and 60 °C for 24 h. The US treatments did not adversely impact the alginate obtained, and the extracted alginates were found to have similar characteristics to the alginate obtained from conventional extraction and commercial sodium alginate.

Abstract:Fucoidans from brown macroalgae (brown seaweeds) have different structures and many interesting bioactivities. Fucoidans are classically extracted from brown seaweeds by hot acidic extraction. Here, we report a new targeted enzyme-assisted methodology for fucoidan extraction from brown seaweeds. This enzyme-assisted extraction protocol involves a one-step combined use of a commercial cellulase preparation (Cellic®CTec2) and an alginate lyase from Sphingomonas sp. (SALy), reaction at pH 6.0, 40 °C, removal of non-fucoidan polysaccharides by Ca2+ precipitation, and ethanol-precipitation of crude fucoidan. The workability of this method is demonstrated for fucoidan extraction from Fucus distichus subsp. evanescens (basionym Fucus evanescens) and Saccharina latissima as compared with mild acidic extraction. The crude fucoidans resulting directly from the enzyme-assisted method contained considerable amounts of low molecular weight alginate, but this residual alginate was effectively removed by an additional ion-exchange chromatographic step to yield pure fucoidans (as confirmed by 1H NMR). The fucoidan yields that were obtained by the enzymatic method were comparable to the chemically extracted yields for both F. evanescens and S. latissima, but the molecular sizes of the fucoidans were significantly larger with enzyme-assisted extraction. The molecular weight distribution of the fucoidan fractions was 400 to 800 kDa for F. evanescens and 300 to 800 kDa for S. latissima, whereas the molecular weights of the corresponding chemically extracted fucoidans from these seaweeds were 10–100 kDa and 50–100 kDa, respectively. Enzyme-assisted extraction represents a new gentle strategy for fucoidan extraction and it provides new opportunities for obtaining high yields of native fucoidan structures from brown macroalgae.

Abstract:Laminaria digitata is a high-quality seaweed resource that is widely cultured and has good application prospects. In this study, Laminaria digitata fucoidan (LF) was extracted from Laminaria digitata, and purified using DEAE-Sepharose Fast Flow gel column to obtain four different grades. Among those, LF4 (Mw:165 kDa), mainly composed of fucose(56.80 %), had the highest total sugar (66.91 %) and sulfate (17.07 %) content. FT-RT and NMR results showed that LF4 was mainly composed of galactosylated galactofucose, and has a sulfate group attached to fucose C4. With the animal experimentation, it was revealed that hyperlipidaemic mice had significantly higher levels of TC (5.52 mmol/L), TG (2.28 mmol/L) and LDL-C (5.12 mmol/L) and significantly lower levels of HDL-C (2 mmol/L). However, LF had the efficacy in modulating the lipid metabolism disturbances induced by hyperlipidemia, as well as the ability to regulate cholesterol transport in serum. Moreover, it regulated AMPK/ACC, PPAR-α/LAXRa, Nrf2/Nqo1, TLR4/NF-κB signaling pathway genes and proteins expression in the liver. In addition, it promoted the production of beneficial short-chain fatty acids (SCFAs) while improving the composition and structure of gut microbiota, including balancing the abundance of Bacteroidota, Firmicutes, Muribaculaceae, Alloprevotella, Escherichia-Shigella, Prevotella and NK4A136. The results clearly indicated that LF4 could significantly ameliorate hyperlipidemia, suggesting its prospective application as a functional food.

Abstract:Neutral and acidic polysaccharides and their protein complexes were fractionated and purified from the brown seaweed umitoranoo (Sargassum thunbergii) by fractional extraction, iron-exchange chromatography, and gel filtration. Thirty-one polysaccharide fractions were obtained and tested for antitumor activity in mice with Ehrlich carcinoma transplanted i.p. Two of the fractions, GIV-A ([alpha]25D -127 degrees and mol. wt., 19,000) and GIV-B ([alpha]25D -110 degrees and mol. wt., 13,500) had such activity. On the basis of chemical and spectral analyses, these compounds were found to be a fucoidan or L-fucan containing approx. 30% sulfate ester groups per fucose residue, about 10% uronic acid, and less than 2% protein.
Optimizing the fucoidan extraction using Box-Behnken Design and its potential bioactivity

Abstract:Fucoidan is a sulfated polysaccharide that occurs naturally in the cell wall of brown seaweeds and has substantial biological efficacy. Optimizing the extraction of fucoidan from different brown seaweeds was the primary goal of this research. The optimization of fucoidan extraction was applied on the brown macroalga Turbinaria turbinata using a Box-Behnken Design (BBD) to inspect the impacts of different pH (3, 5, 7), temperature (70, 80, 90 °C) and extraction duration (60, 120, 180 min) on both the yield and sulfate content of fucoidan. The optimized parameters recorded to maximize the fucoidan yield and its sulfate content were a pH of 3.44 and a temperature of 82.26 °C for 60 min. The optimal conditions obtained from BBD were used for fucoidan extraction from T. turbinata, Sargassum cinereum, Padina pavonica, and Dictyota dichotoma. The highest average of fucoidan yield was derived from P. pavonica (40.76 ± 4.04 % DW). FTIR, 1H NMR, and HPLC were used to characterize extracted fucoidan. The extracted fucoidan's Physical characteristics, biochemical composition, antioxidant potential, antitumor effect against breast cancer cells (MCF-7), and antimicrobial and anticoagulant activity were assessed. The extracted fucoidan from D. dichotoma, followed by that extracted from S. cinereum, which had the highest sulphate content, depicted the highest antioxidant, anticancer, and anticoagulant activities. Fucoidan has demonstrated a strong antimicrobial action against some pathogenic microorganisms; Bacillus subtilis, Staphylococcus aureus, Escherichia coli, Klebsiella pneumonia, and Candida albicans. The anticoagulant properties of fucoidan from D. dichotoma were stronger than those of fucoidan from S. cinereum, T. turbinata, and P. pavonica due to its higher sulphate content. These findings could be used for various biomedical applications to improve the pharmaceutical industry.

Abstract:<p>Fucoidans are sulfated polysaccharides of marine origin and they have distinguished biological applications due to their anti-viral, anti-inflammatory, anti-angiogenic, and anti-tumorigenic potential. In this study, fucoidan was extracted from a brown macroalga (<em>Sargassum siliquosum</em>) using microwave-assisted extraction (MAE) and ultrasound-assisted extraction (UAE). MAE under the condition of 750 W, 10 minutes, and a liquid/solid ratio of 15 mL/g, showed higher extraction efficiency with a total sugar yield of 6.94% per dry weight. The crude fucoidan obtained with ethanol precipitation was further purified by isoelectric point precipitation and calcium chloride addition (for protein and uronic acid removal), followed by anion-exchange chromatography (AEC) and dialysis. The purity of the polysaccharides content of crude fucoidan increased from 33.3% to 64.5% with a total sugar recovery of 50.3%. Based on the sugar and sulfate group (the major components of fucoidan), the purity of the final sulfated polysaccharides product is nearly 84.0%, and this purified fucoidan product was found to display biological activities linked to anti-lipogenesis with 28.9% decreased lipid synthesis in contrast to the free-fatty-acid-induced control.</p>

Abstract:A low molecular weight fucoidan (DFPS), obtained from the brown seaweed Laminaria japonica, was separated into three fractions by anion-exchange column chromatography. All three fractions (DF1, DF2, DF3) predominantly contained variety fucose, galactose and sulfate group. The antioxidant and anticoagulant activities of these fractions were investigated. The results showed that all fractions possessed considerable antioxidant activities, DF1 was most active. All samples inhibited coagulant in APTT, TT and PT assays obviously. Available data presented the content of sulfate group, the molar ratio of sulfate/fucose and sulfate/total sugar, and the molecular weight played an important role on antioxidant and anticoagulant activity.
