Determination of Teniposide in Rat Plasma by HPLC-UV

Abstract:AIM To improve a HPLC-UV method for the determination of teniposide in rat plasma.METHODS The concentration of teniposide was determined by using HPLC-UV method with the RP-C_(18)column(250 mm×4.60 mm, 5 tan,Phenomenex).The mobile phase consisted of methanol-distilled water-acetic acid(38:60:2,V/V)with a flow rate of 1.0 mL·min~(-1),whileγ= 230 nm.RESULTS The linear range was 0.5-25.0 mg·L~(-1)(r=0.9999).The limit of detection was 0.06mg·L~(-1).RSD of inter-day and intra-day were all less than 5%.The concentration of tenipo- side in rat plasma at 30 min time point was (4.21±0.37)mg·L~(-1)(n = 4)after intravenous administration of teniposide solution.CONCLUSION The analytical method in this study is simple,sensitive,accurate,which can be easily applied to teniposide pharmacokinetic and pharmacological studies.
