Abstract:A new strategy for the modification of cyclophosphamide was carried out and three series of new cyclophosphamide piperazinium salts 10,11 and 13 were prepared.These compounds,based on compound 9i scaffold,were evaluated for their in vivo anticancer activities against hepatocyte sarcoma 22 (H22).The structure-activity relationship study reveals that 1) the conformation and the substituent at N-3 of cyclophosphamide spiropiperazinium salts 10 greatly affect the activity;2) different kinds of cyclophosphamide non-spiropiperazinium derivatives 11 show different activities;3) for 1,2-benzisoxazole phosphoropiperazinium salts 13,it is possible to obtain anticancer drug candidates with suitable quaternary ammonium moiety.These results would help to further design and synthesize analogs of mustard anticancer drugs.

Abstract:A novel class of cyclophosphamide spiropiperaziniums was synthesized and evaluated for their in vivo anti-cancer activities against S180 and H22. Most of them exhibited definite activities. Especially, compounds 8b and 8k showed good anti-cancer activities, meanwhile, 8k also showed much lower toxicity than CP. Several interesting structure-activity relationships were revealed.

Abstract:Pyrazole compounds are one kind of heterocyclic compounds with extensive bioactivity,and lots of pyrazole compounds have been commercialized both in the region of medicament and pesticide.In the process of finding out some novel pyrazole compounds,a~d four novel pyrazole intermediates were synthesized.Taking diethyl oxalate as starting substance,the novel compounds can be synthesized via condensation,cyclic condensation,alkylation,(bromination) and hydrolysis reactions.The yield of products was about 78%-90%.Their structures were confirmed by elementary analysis,1H-NMR,MS and IR.

Abstract:A new easy-to-run route for synthesizing some novel pendant-armed benzene-containing cyclen derivatives was proposed.The titled compounds were synthesized by the reaction of 1,4,7,10-tetraazacyclododecane(cyclen)and the electrophilic reagent with potassium carbonate as the proton trapper and were characterized by MS and 1HNMR.The method features a lot of advantages,such as simplicity in operation,mild reaction conditions,high yield and environmental friendliness with the use of potassium carbonate instead of triethylamine and(N,N)-diisopropylethylamine.

Abstract:Several novel derivatives of mono- and bis(1,2,4-diazaphosphol-1-yl)pyridine (or pyrazine) as well as the first 2,6-bis(1,2,4-diazaphosphol-1-yl)pyridine (bdppy) and 2,6-bis(1,2,4-diazaphosphol-1-yl)pyrazine (bdppz) metal complexes [Zn(bdppy)2]2+·2ClO4− and [(bdppz)Cu(NCMe)]2+·2ClO4− were prepared and structurally characterized. The complex [Zn(bdppy)2]2+·2ClO4− has a higher local concentration of low-valent phosphorus (σ2λ3) on the periphery of the molecule. All the compounds were characterized using 1H, 13C{1H}, 31P{1H} NMR and IR spectroscopy, and X-ray crystallography. The properties of the compound [(bdppz)Cu(NCMe)]2+·2ClO4− were studied using EPR spectroscopy and magnetic susceptibility measurements.

Abstract:Seven new 1, 1-dialkyl-4-(3-bromopropionyl) piperazinium bromides were synthesized. Their structures were elucidated by IR, 1H NMR and elemental analysis. The compounds were screened for antitumor, analgesic and antioxide SOD activities. Though none of them showed antitumor activity, most of them showed distinct analgesic activity and antioxide SOD activity.

Abstract:Piperazine and morpholine are common modules in drugs. Design and synthesis of their surrogates may help to explore the chemical and patent space in medicinal chemistry. In this article, through [3+2] cycloaddition, the improved synthesis of 6-benzyl-2,6-diazaspiro[3.4]octane oxalate and 2-oxa-6-azaspiro[3.4]octane as substitutes of piperazine and morpholine was provided, respectively. Multi-gram quantities of the compounds could be easily obtained in relatively high yields.

Abstract:An efficient and facile synthetic approach towards a variety of novel 1,2,3-triazole derivatives containing a 1-hydroxyalkane-1,1-bisphosphonate moiety has been developed through the copper-catalyzed ultrasound-assisted 1,3-dipolar cycloaddition reaction and the following hydrolysis and phosphorylation reaction. Abstract:Cyclic phosphoramidate compounds are widely used as a type of anti-cancer medicines and their stereostructures are directly related to their activities and effects. A series of cyclic phosphoramidates -N-substituted -5, 5-dimetnyl-4 (1-methylethyl)-1, 3, 2-dioxaphosphorinane-2-oxide, were synthesized by the reactions of cyclic phosphite VI with amino compounds. The results showed that the reactions were high stereo-specific and nearly all the produced cyclic phosphoramidates were transisomers. The usefulness of this methodology also lies in the fact that the reactions were proceeded under mild conditions and the intermediate-cyclic phosphite VI need not be separated and purified.

Abstract:Herein, we describe the synthesis of pyridinium, sulfonium and ammonium bicyclo-[1.1.1]-pentane salts as potential aryl isosteres. Arylsulfonium, ammonium and pyridinium salts are central motifs in several natural products, pharmaceuticals, and high value commodities within the industrial chemical sector. The reaction proceeds by a nucleophilic substitution on a bench stable precursor 1,3-diodobicyclo-[1.1.1]-pentane (DIBCP). The transformation displays broad substrate scope, good to excellent yield profile, with several of the BCP products being fully characterised by single crystal X-ray crystallography, highlighting the unique 3-dimensional structure of these potential isosteres. Finally, the pyridinium and quinolinium BCP salts can be cleanly converted into piperidinone and quinolone BCP analogues.

Abstract:Racemic diastereoisomeric spiro-phosphonate derivatives 5 similar to 6 were obtained by tandem double Michael addition and intramolecular nucleophilic substitution of 3-bromo-5-methoxy-5H-furan-2-one and alpha-hydroxylbenzylphosphonate under a warm condition. The target compounds 5 similar to 6 were identified by the elemental analysis, IR, H-1 NMR, C-13 NMR and MS techniques. The stereochemistry and configuration of 5c and 6c were confirmed further by X-ray crystallography. This result could lead to a new strategy for the synthesis of some complicated compounds containing a phosphorus functional group.

Abstract:Synthesis of new functional organic molecules is a critical path that may greatly accelerate the evolution of organic optoelectronic materials. We have achieved a facile synthesis strategy to produce unique saddle-shaped multi-functional triazepinium salts that exhibit excellent solid-state fluorescence. Their fluorescence performance could be easily regulated by adjusting the dihedral angle between the main skeleton and the substituted moiety, giving a large Stokes shift, non-aggregation quenching, long lifetime, and multilevel-redox characteristics.

Abstract:The bicyclic 1-aza-2-azoniaallenium salt intermediates, generated from the azoester species upon treatment with a Lewis acid, have been demonstrated to participate in Huisgen-type cycloaddition with nitriles to result in the formation of fused 6,7,8,9-tetrahydro-5 H-[1,2,4]triazolo[1,5- d][1,4]diazepinium salts. This transformation is interpreted as a regular [3++2] cycloaddition between intermediates as the reactive 1,3-monopole reactants and nitriles as the nucleophilic reagents followed by spontaneous [1,2]-cationic rearrangement. The azoester precursors were easily accessible via oxidation of the corresponding hydrazones using hypervalent iodine oxidant PhI(OAc)2 under mild conditions. The [1,2,4]triazolodiazepine compounds represent a class of N-containing biologically important heterocycles with a new type of scaffold.

Abstract:A new, one-pot and three-component synthesis of 4H-pyrido[1,2-a]pyrimidines, 4H-pyrimido[1,2-a]pyrimidines, and 4H-pyrazino[1,2-a]pyrimidines is described. The reactive 1:1 zwitterionic intermediate, formed by the addition of isocyanides to dialkyl acetylenedicarboxylates, was trapped by N-(2-heteroaryl)amides to yield a ketenimine intermediate, which was cyclized and then rearranged under the reaction conditions to afford the title compounds under mild reaction conditions in good yields. Single-crystal X-ray analysis conclusively confirms the structure of the obtained bridgehead bicyclic 6–6 heterocyclic compounds.

Abstract:A convenient and practical method for the synthesis of 2-alkylthio-4-amino-5-cyano-6-aryl(alkyl)pyrimidines has been developed via a three-component, one-pot reaction from aldehydes, malononitrile and S-alkylisothiouronium salts in water at room temperature. A series of polysubstituted pyrimidines were prepared by this method in moderate to excellent yields. In addition, two kinds of pyrimidine-fused heterocyclic derivatives with potential pharmacological activity were constructed from our 2-alkylthio-4-amino-5-cyano-6-arylpyrimidines.

Abstract:2,2-Dichloro-4,4,6,6-bis[spiro(2′,2″-dioxy-1′,1″-biphenylyl)]cyclotriphosphazene was synthesised from the reaction of hexachlorocyclotriphosphazene with biphenyl-2,2′-diol. 2,2-Bis(4-formylphenoxy)-4,4,6,6-bis[spiro(2′,2″-dioxy-1′,1″-biphenylyl)]cyclotriphosphazene was obtained from the reaction of 2,2-dichloro-4,4,6,6-bis[spiro(2′,2″-dioxy-1′,1″-biphenylyl)] cyclotriphosphazene with 4-hydroxybenzaldehyde. 2,2-Bis[4-(4-quinazolinone)phenoxy]-4,4,6,6-bis[spiro(2′,2″-dioxy-1′,1″-biphenylyl)]cyclotriphosphazene was synthesised from the reaction of 2,2-bis(4-formylphenoxy)-4,4,6,6-bis[spiro(2′,2″-dioxy-1′,1″-biphenylyl)]cyclotriphosphazene with anthranilamide in N,N-dimethylformamide. The novel cyclophosphazene derivatives were synthesised under mild conditions from the reaction of 2,2-bis[4-(4-quinazolinone)phenoxy]-4,4,6,6-bis[spiro(2′,2″-dioxy-1′,1″-biphenylyl)]cyclotriphosphazene with 4-chlorobenzenemethanethiol, p-thiocresol, 4-aminothiophenol, and 4-chlorothiophenol in the presence of p-toluenesulfonyl chloride via C-OH bond activation under mild conditions. All products were generally obtained in high yields. Their structures were characterised by HRMS, IR and 1H, 13C spectroscopy.

Abstract:A novel four-component diastereoselective synthesis of piperidin-2-one salts containing a quaternized pyridine unit is reported. The Michael–Mannich cascade was conducted using Michael acceptors, pyridinium ylides, aromatic aldehydes, and ammonium acetate in methanol. It is a convenient approach to the synthesis of 1-((3 SR ,4 RS ,6 SR )-4,6-diaryl-5,5-dicyano-2-oxopiperidin-3-yl)pyridin-1-ium halides with three stereocenters in 48–84% yield or 1-[(3 SR ,4 RS ,5 RS ,6 SR )-4,6-diaryl-5-cyano-5-(methoxycarbonyl)-2-oxopiperidin-3-yl]-pyridin-1-ium halides with four stereocenters in 44–74%. This reaction is highly stereoselective. Only one diastereomer was formed. Ammonium acetate plays a dual role, acting as a base and as a nitrogen source. Structures of the synthesized compounds were confirmed by 1 H, 13 C NMR, IR, and mass spectra. The formation of a single diastereomer was confirmed by singe crystal X-ray diffraction studies. Products were obtained by simple filtration, and other purification methods as column chromatography were not necessary.

Abstract:A series of novel salts of heterocyclic polyamines with 5-sulfosalicylic acid (C4H7N4+)(C7H5O6S−)∙2(H2O) (1), (C4H6ClN4+)(C7H5O6S−)∙H2O (2), (C5H8N3+)(C7H5O6S−)∙H2O (3), (C5H7N6+)(C7H5O6S−)∙H2O (4), (C6H14N22+)(C7H4O6S2−)∙H2O (5), and (C14H19N2+)(C7H5O6S−) (6) have been successfully synthesized. Their crystal structures have been determined by single-crystal X-ray diffraction. Overall, compounds adopt a layered structure with aminium cations and 5-sulfosalicylic anions linked via water molecules. The solid-state architectures of these compounds are dominated by O(N,H)-H⋯O and N-H⋯N hydrogen bonds and stabilized by weak interconnects. C-Cl⋯π and S-O⋯π interactions, apart from π⋯π and C-H(O)⋯π, were reported. Diverse approaches were used to study the effect of substituents in the polyamines in solid-state arrangement. A Hirshfeld surface analysis, with associated 3D Hirshfeld surface maps and 2D fingerprint plots, molecular electrostatic potential, and energy frameworks were used to comprehensively investigate the nature and hierarchy of non-covalent interactions and inspect supramolecular differences. The contact enrichment ratio calculations provided deeper insight into the propensity of interconnects to influence crystal packing. The evaluation of the effects of H-bonding synthons resulting from different substituents in the polyamines on self-assemblies is also presented. In the context of crystal engineering, a specific intramolecular synthon via O-H⋯O observed in nearly all crystals can be employed in the pseudo-cyclic replacement strategy in the design of new molecules.
