Abstract:BACKGROUND: Regulatory B cells (Bregs) play an important role in maintaining immune homeostasis and have the potential to induce tolerance. Previous work has found that Breg cells are involved in heart transplantation tolerance. However, the effect of Breg on the transplantation tolerance and the underlying mechanisms remain to be clarified. METHODS: Using a within-species heart transplantation model, we aimed to investigate the role of CD19(+) CD5(+) CD1d(high) Bregs isolated from transplanted mice in preventing transplant rejection in vivo. We also explored the effects of CD40 and tumor necrosis factor receptor-associated factor 6 (TRAF6) ubiquitin ligase on Breg-mediated prolongation of survival in heart transplant (HT) mice, and the regulatory effects of downstream Cdk4 and Cdk6 proteins on dendritic cells (DCs), which clarified the function and molecular mechanism of Breg cells in HT mice. RESULTS: Our data suggest that adoptive transfer of the transplanted Bregs served as an effective tolerance-inducing mechanism in HT mice and was involved in the CD40-TRAF6 signaling pathway in DCs. Moreover, DCs collected from the Breg treated HT mice also prolonged the survival of HT mice. Furthermore, DC-specific knockout of TRAF6 diminished Breg-mediated prolongation of survival in HT mice. Interestingly, gut microbes from donors increased the survival of cardiac allografts both in both the absence and presence of Bregs but were not implicated in CD40-TRAF6 signaling. CONCLUSIONS: These findings reveal a role of Breg cells in the induction of transplantation tolerance through the blockade of the CD40-TRAF6 signaling pathway, which might be used in the treatment of HT in the clinic. Prevention of kidney allograft rejection in rat by immune tolerance i nduction

Abstract:Objective　To investigate a clinically persp ective way of inducing transplantation tolerance.　Methods　Kidney tansplantation model was established using Lewis (RT1l) rats as recipients and DA (RT1a) rats as donor. Before transplantation the recipient rats were condi tioned with anti-lymphothyte serum (ALS) injected for three times, followed by 108 donor bone marrow cells transfusion (BMT) and injection of 100 mg/kg cyclo pho sphamide (CP), survival time of kidney allografts, mixed lymphocyte reaction (MLR ) and delayed-type hypersensitivity (DTH) of recipient to donor spleen cells we re observed and studied.　Results　5 of the 7 kidney allo grafts survived 73 to 90 days without sign of rejection. The donor specific MLR and DTH were suppressed.　Conclusions　Induction of trans plantation tolerance is a prospective way for the prevention of allograft reject ion.
Donor specific transfusion of splenocytes combined with delayed low dosage tacrolimus administration induce immune tolerance in allogene rat heart transplantation

Abstract:Objective To observe the effects of donor specific transfusion of splenocytes combined with delayed low dose tacrolimus(FK506)administration on the induction of immune tolerance in allogene rat heart transplantation.Methods Wistar rats and SD rats were divided into 7 groups.In A-F groups,Wistar and SD rats were respectively chosen as donors and recipients.And the G group was selected as homogenic control group,in which both the donors and recipients were SD rats.No treatment was done in A group.Donor specific splenocytes were transfused to recipients in B group.FK506[0.5 mg/(kg·d)] was respectively used in early or late time after transplantation in C and D groups.Donor specific splenocytes were transfused to recipients via portal vein in E and F groups.And FK506 was respectively used post-transplantation in early or late time in E and F groups.The improved celiac heterotopic rat heart transplantation was performed.The survival time and pathologic changes of transplanted heart were observed after transplantation.Results The transplanted hearts survived for a long time in G group(100 d).The survival time of A-F groups were(7.4±0.9)d,(22.1±3.6)d,(8.0±0.8)d,(7.9±1.3)d,(24.4±4.5)d,(95.5±6.0)d respectively.Seven eighths of transplanted hearts survived for a long time in F group.There was no significant difference in survival time of transplanted hearts between C,D group and A group(all in P>0.05).Compared with A group,survival time of transplanted hearts was significantly longer in B,E and F group(P<0.05-P<0.01).And survival time of transplanted hearts in F group was significantly longer than that in B and E group.The pathological examination showed no rejection appearance in the cardiac muscle of transplanted heart in G group.The transplanted hearts in A group stopped beating at 9 d after transplantation and pathological examination showed Grade 3-4 rejection.And no rejection was observed in the rats which survived for 100 days in F group.Conclusions Transfusion of donor specific splenocytes via recipient portal vein during transplantation combined with late administration of FK506 can first activate and then restrain immune system in allogene rat at early stage post-transplantation.And it also helps to induce immune tolerance.
Cardiac Allograft Tolerance Induced by Isogeneic CD4+CD25+ Regulatory T Cells.

Abstract:Objectives: To evaluate the effect of isogeneic CD4+CD25+ regulatory T cells on cardiac allograft tolerance in heterotopic heart transplant from Balb/c to C57BL/6 mice.Materials and Methods: Isogeneic and allogeneic CD4+CD25+ regulatory T cells were obtained from pregnant C57BL/6 mice crossed with male Balb/c mice and from regular Balb/c mice. Recipient C57BL/6 mice were treated with sublethal radiation (2 Gy) and an infusion of isogeneic CD4+CD25+ regulatory T cells, allogeneic CD4+CD25+ regulatory T cells, or phosphate-buffered saline alone 1 day before a Balb/c-to-C57BL/6 heterotopic heart transplant. At 10 days after the transplant, cardiac allografts and the sera of recipients were evaluated with histology and cytokine analysis. Splenocytes of recipients were collected to determine chimerism on the day of the cessation of allograft heartbeat.Results: Mice that received an infusion of isogeneic CD4+CD25+ regulatory T cells had significantly greater mean median survival time, greater degree of chimerism, decreased levels of cytokines (monokine induced by interferon gamma, interleukin 6, interleukin 10, and regulated upon activation, normal T cell expressed and secreted protein), and decreased lymphocytic infiltration than did mice that received phosphate-buffered saline alone. The effects on allograft tolerance were stronger in mice that received the isogeneic than the allogeneic CD4+CD25+ regulatory T cells.Conclusions: Isogeneic CD4+CD25+ regulatory T cells may establish cardiac allograft tolerance by inducing mixed chimerism and suppressing immune responses. Infusion with isogeneic CD4+CD25+ regulatory T cells combined with radiation (sublethal dose, 2 Gy) may establish allograft tolerance in mice. Therefore, further study is warranted because isogeneic CD4+CD25+ regulatory T cells may have therapeutic benefits.

Abstract:Antibody-mediated rejection (ABMR) is a major cause of dysfunction and loss of transplanted kidney. The current treatments for ABMR involve nonspecific inhibition and clearance of T/B cells or plasma cells. However, the prognosis of patients following current treatment is poor. T follicular helper cells (Tfh) play an important role in allograft-specific antibodies secreting plasma cell (PC) development. Tfh cells are therefore considered to be important therapeutic targets for the treatment of antibody hypersecretion disorders, such as transplant rejection and autoimmune diseases. Tacrolimus (Tac), the primary immunosuppressant, prevents rejection by reducing T cell activation. However, its administration should be closely monitored to avoid serious side effects. In this study, we investigated whether Tac delivery to helper T (CD4 + ) cells using functionalized mesoporous nanoparticles can block Tfh cell differentiation after alloantigen exposure. Results showed that Tac delivery ameliorated humoral rejection injury in rodent kidney graft by suppressing Tfh cell development, PC, and donor-specific antibody (DSA) generation without causing severe side effects compared with delivery through the drug administration pathway. This study provides a promising therapeutic strategy for preventing humoral rejection in solid organ transplantation. The specific and controllable drug delivery avoids multiple disorder risks and side effects observed in currently used clinical approaches.

Abstract:The pursuit of transplantation tolerance is the holygrail in clinical organ transplantation. It has been established that regulatory T cells (Tregs) can confer donor-specific tolerance in mouse models of transplantation. However, this is crucially dependent on the strain combination, the organ transplanted and most importantly, the ratio of Tregs to alloreactive effector T cells. The ex vivo expansion of Tregs is one solution to increase the number of alloantigen specific cells capable of suppressing the alloresponse. Indeed, ex vivo expanded, alloantigen specific murine Tregs are shown to preferentially migrate to, and proliferate in, the graft and draining lymph node. In human transplantation it has been proposed that depletion of the majority of direct pathway alloreactive T cells will be required to tip the balance in favour of regulation. Ex vivo expansion of alloantigen specific, indirect pathway human Tregs, which can cross regulate the residual direct pathway has been established. Rapid expansion of these cells is possible, whilst they retain antigen specificity, suppressive properties and favourable homing markers. Furthermore, considerable progress has been made to define which immunosuppressive drugs favour the expansion and function of Tregs. Currently a series of clinical trials of adoptive Treg therapy in combination with depletion of alloreactive T cells and short term immunosuppression are underway for human transplantation with the aim of minimizing immunosuppressive drugs and completely withdrawal.

Immune Tolerance Induced by Immature Dendritic Cells and Bone Marrow Transplantation in Rat Renal Allogeneic Graft Model and Its Mechanisms

Abstract:Purpose To study immune tolerance induced by immature dendritic cells transfusion and bone marrow transplantation in rat renal transplantation model,and to explore a possible strategy for renal allograft protection in clinic. Methods Immature dendritic cells (imDC) and fresh bone marrow cells (BMC) from DA rat were prepared for use before transplantation. Renal allograft from DA rat was transplanted to Lewis rat. Forty recipient rats were randomly divided into 5 groups: (1) negative group; (2) imDC group; (3) BMT group; (4) imDC+ BMT group; (5) cyclophosphamide (CP) group. Anti-donor cytotoxic T lymphocyte activity was assayed by 51Cr release test. Cytokine in the supernatant of splenic cells culture were measured by ELISA,including IL-4,IFN-γ and TGF-β. Results The median survival time (MST) was (7.12±1.25) days in negative group,(24.36±3.20) days in imDC group, (7.87±2.10) days in BMT group,(18.13±2.16) days in Cp group. When combined imDC transfusion and BMT,the MST was prolonged to (80.75±16.88) days, which had significant difference compared with negative group, imDC group,BMT group and CP group (P0.01).CTL from tolerant rats had decreased cytotoxicity to cells from donor rats (4.3±1.0)% vs (39.6±4.4)%,P0.01),but showed normal cytotoxicity to cell from the third party rat (9.8±3.2) %.IFN-γ in the supernatant of tolerant rat splenic cells culture was (532.78±34.54) ng/L,which was lower than normal and rejection rat (P0.01). IL-4 and TGF-β in the supernatant of tolerant rat splenic cells culture was (185.44±32.17) ng/L and (716.82±197.62) ng/L, which was higher than normal or rejection rat (P0. 01). Conclusions Combined used of imDC transfusion and bone marrow transplantation is an effective way to induce immune tolerance in rat renal transplantation model. Its possible mechanisms may due to immune deviation of cytokine net and rat donor-specific CTL anergy.
The Mixed Lymphocyte Reaction and the Ratio of Cd4(+)Cd25(+)Foxp3(+)/Th17 T Cell in Long Term Graft and Recipents Survival of Kidney Transplantation with Living-Related Donor

Abstract:Introduction: Regulatory T cells (Treg) have been shown to prevent allograft rejection and plays an important role in promote donorspecific antigenic tolerance in solid organ transplantation. In this study, we analyzed the ratio of CD4+CD25+Foxp3+/Th17 T cell in the peripheral blood of kidney recipients with living-related donor in different immunosuppressive therapy, and analyzed the relationship between the ratio of CD4+CD25+Foxp3+/Th17 T cell and patients who have long-term survival. Methods: The study was approved by the local ethical committee. Twelve renal transplant recipients with living-related donor received combined donor-specific blood transfusion and kidney transplantation more than six years were enrolled, and ten healthy volunteers as healthy control (HC). Posttransplant patients were divided into two groups depending on their immunosuppressive therapy: calcineurin inhibitors (CNI) and rapamycin (Rapa). Analyzed the expression of CD4+CD25+Foxp3+T cells and Th17 T cells in the peripheral blood under the stimulate condition (phorbol myfismte acetate and ionomycin) and no-stimulate condition for all objects by flow cytometry, and the mixed lymphocyte reaction (MLR) in recipient with their donors and independent third party by methyl thiazolyl tetrazolium (MTT). Results: The proportion of CD4+CD25+Foxp3+ T cells in the peripheral blood under the stimulate condition and no-stimulate condition in group of Rapa were 2.24% and 2.14%, respectively, significant higher than group of CNI (P< 0.05, Table 1), and, there was no significant difference for the proportion of Th17 T cells between the group of Rapa and CNI. The ratio of CD4+CD25+Foxp3+/Th17 T cells in group of Rapa under the stimulate condition and no-stimulate condition were 3.42 and 8.92, respectively, significant higher than group of CNI (P< 0.05, Table 1). In all groups, the result of MLR with independent third party higher than their donors, and the MLR with independent third party in group of Rapa was higher than group of CNI.[Table 1 The expression of Treg and Th17 T cells]Conclusion: This study showed in living-related donor kidney recipients who received combined donor-specific blood transfusion and kidney transplantation obtain better long-term graft and patient survival. Increased numbers of CD4+CD25+Foxp3+ T cells and the ratio of CD4+CD25+Foxp3+/Th17 T cells and higher MLR with independent third party in group of Rapa compared with group of CNI. Therefore, monitoring of Treg in the peripheral blood as a biomarker of tolerance may detect unresponsiveness to the graft.
Transfer of Regulatory T Cells Generated Ex Vivo Modifies Graft Rejection Through Induction of Tolerogenic CD4+CD25+ Cells in the Recipient

Abstract:Certain CD4+CD25+ T cells can induce and maintain T-cell non-responsiveness to donor alloantigens and have therapeutic potential in solid organ transplantation. Peripheral CD4+CD25- cells alloactivated with IL-2 and transforming growth factor beta (TGF-beta) ex vivo express the transcription factor FoxP3, and become potent antigen-specific CD4+CD25- suppressor cells. Here we report that the transfer of TGF-beta-induced regulatory CD4+ and CD8+ T cells (Tregs) co-incident with transplantation of a histoincompatible heart resulted in extended allograft survival. To account for this result, we injected non-transplanted mice with a single dose of CD4+ and CD8+ Tregs and transferred donor cells every 2 weeks to mimic the continuous stimulation of a transplant. We observed increased splenic CD4+CD25+ cells that were of recipient origin. These cells rendered the animals non-responsive to donor alloantigens by an antigen-specific and cytokine-dependent mechanism of action. Both the increased number of CD4+CD25+ cells and their tolerogenic effect were dependent on continued donor antigen boosting. Thus, Tregs generated ex vivo can act like a vaccine that generates host suppressor cells with the potential to protect MHC-mismatched organ grafts from rejection.

Abstract:Objective　 To compare the effects of different donor-specific antigens, through different pathways, on the inducement of immunological tolerance. Methods　 After the establishment of a stable rat cervical heterotopic heart transplantation model, donor specific transfusion (DST), donor specific spolenocyte (DSSL) and donor specific bone marrow (DSBM) were given to recipients 3-14 days prior to the transplantation through peripheral vein, portal vein and intrathymic injection respectiviely. Low dose CsA 5mg/(kg*d) was given from the 3rd-14th day pre-transplantation to the 28th day post-transplantation. The survival time of cardiac grafts was monitored, meanwhile, the donor specific mixed lymphocyte reaction (MLR) and the subsets of T lymphocytes CD8+ were determined. Results　 DSBM and the administrative pathway of intrathymic injection achieved the best effects on the inducement of immunological tolerance and the improvement of graft survival. Conclusion　 The donor specific antigens can induce the donor-specific immunological tolerance. It may be a practicable approach for the clinical experiments in the future.

Abstract:Background Allogeneic transplant rejection is currently a major problem encountered during organ transplantation. The dendritic cell (DC) is the most effective powerful known professional antigen-presenting cell, and recent studies have found that DCs can also induce immune tolerance, and avoid or reduce the degree of transplant rejection. The aim of this study was to evaluate the effect of transfused immature CD4(+) DCs on renal allografts in the rat model.Methods In this study, we induced CD4(+) immature DCs from rat bone marrow cells by a cytokine cocktail. The immature CD4(+) DCs were identified by morphological analysis and then the suppressive activity of these cells conditioned with donor kidney antigen was evaluated in vitro and in vivo.Results Immature CD4(+) DCs conditioned with donor kidney antigen possessed immunosuppressive activity in vitro and they were able to prolong renal transplant survival in an allograft rat model in vivo.Conclusions Our study provides new information on efficacious renal transplantation, which might be useful for understanding the function of immature CD4(+) DCs in modulating renal transplant rejection and improving clinical outcome in future studies. Adenovirus-mediated CTLA4-Ig and FasL Gene Transfer Induces Long-Term Survival of Renal Allografts in Rats

Abstract:Objective To investigate the potential role of adenovirus-CTLA4-Ig and adenovirus-FasL recombinant in inducing transplantation tolerance using renal-graft model and its related mecha-nisms. Methods Allogeneic kidney transplants were performed between SD donors and Wistar recipients. The experimental rats were divided into 4 groups. In Ad-CTLA4-Ig group and Ad-CTLA4-Ig + Ad-FasL group, the donor kidney of the SD rats was locally transfected by Ad-CTLA4-Ig and Ad-CTLA4-Ig + Ad-FasL with the dose of 1 × 109-5 × 109 PFUml respectively and then transplanted to the recipient Wistar rats. In control group, the kidneys of the SD rats were directly transplanted to Wistar rats without any thera-py. The rats treated with Ad-EGFP served as empty vector group. After kidney transplantation, the survival time and the kidney function in each group were observed. Kidney allografts were evaluated by HE staining and immunohistochemical staining. The pathological features and ultrastructures of the grafts were ob-served. Results The survival time of allografts were prolonged significantly in recipients receiving Ad-CT-LA4-Ig + Ad-FasL with a mean survival time of (64.67 ± 6.41) days ,significantly longer than that in Ad-CTLA4-Ig treated group (31.33±6.77) days,control group (8.17 ± 1.17) days and empty vector group (8.00 ± 1.55) days (P ＜ 0.01). After transplantation, the levels of creatinine in serum were significantly higher in control group and empety vector group than in Ad-CTLA4-Ig + Ad-FasL treated group and Ad-CTLA4-Ig treated group. Conclusion Adenoviral vectors can be successfully transduced into rat kidneys with the CTLA4-Ig and FasL cDNA. Ad-mediated transduction of the CTLA4-Ig and FasL gene can signifi-cantly prolong the survival of rat renal allograft. Abstract:Naturally occurring CD4(+)CD25(+) regulatory T cells (Tregs) have been shown to play a key role in the control of autoimmunity. Interestingly, they are also capable of mediating transplantation tolerance and they can have indirect allospecificity for donor antigens. An increasing body of evidence in experimental studies has indicated that adoptive transfer of in vitro expanded CD4(+)CD25(+) Tregs with indirect antidonor allospecificity can induce long-term donor-specific transplantation tolerance. Thus, adoptive cell therapy using patient-specific CD4(+)CD25(+) Tregs as individualised medicine to promote clinical transplantation tolerance is promising.

Abstract:Objective To monitor the kinetic level of CD4+CD25+ regulatory T cells in peripheral blood of rat with renal allograft,the value of predicting rejection or tolerance in renal transplantation.Methods The kinetic level of CD4+CD25+ regulatory T cells in peripheral blood of rat with renal allograft was monitorred by FCM(flow cytometry).Results During the process of acute rejection,the level of CD4+CD25+ regulatory T cells in peripheral blood of rat with renal allograft was significantly reduced than that in the syngraft control group.the level of CD4+CD25+ regulatory T cells in CsA group was significantly higher than that in the syngraft control group.Conclusion CD4+CD25+ regulatory T cells played an important role in suppressing rejection of renal transplantation.Monitoring the kinetic level of CD4+CD25+ regulatory T cells in peripheral blood of renal allograft could predict rejection occurring and recognize immune reaction or tolerance status existing in recipient against renal allograft.
Biological Features of Intrahepatic CD4 + CD25 + T Cells in the Naturally Tolerance of Rat Liver Transplantation

Abstract:The biological features of intrahepatic CD4+ CD25+ T regulatory cells in the naturally tolerance of rat liver transplantation were explored. Orthotopic liver transplantation was performed in two allogeneic rat strain combinations, one with fatal immunosuppression despite a complete major histocompatibility complex mismatch. The subjects were divided into three groups according to different donors and recipients [Tolerance group: LEW-to-DA; Rejection group: DA-to-LEW; Syngegnic group (control group): DA-to-DA]. The proportion of intrahepatic CD4+CD25+ T cells from three groups was determined by flow cytometry (FCM) in different time. The intrahepatic CD4+CD25+ T cells were isolated by magnetic activated cell sorting (MACS) method and identified by FCM. The Foxp3 mRNA was detected by reverse transcriptase polymerase chain reaction (RT-PCR). And their suppression on the proliferation of CD4+CD25− T effector cells was analyzed by cell proliferation assay in vitro. Beginning immediately after transplantation, the proportion of Treg cells increased over time in both allogeneic groups but was significantly greater in the Rejection group. The proportion of Treg cells declined after day 5, and such reduction was more dramatic in the Rejection group than in the Tolerance group. Animals in the Tolerance group showed a second increase in the proportion after day 14. Intrahepatic CD4+ CD25+ T cells isolated from spontaneous tolerance models inhibited the proliferation of mixed lymphocyte reaction. The purity of CD4+CD25+ T cells sorted by MACS was 86%–93%. The CD4+CD25+ T cells could specifically express the Foxp3 gene compared with CD4+CD25− T cells. In vitro, the spleen cells from LEW rats can irritate the proliferation of CD4+CD25+ T cells more obviously than the syngegnic spleen cells. CD4+CD25+ Tr cells could suppress the proliferation of CD4+CD25− T cells, but the inhibition was reversed by exogenous IL-2 (200 U/mL). The CD4+CD25+ T regulatory cells specifically express the Foxp3 gene, which may play an important role in the induction of liver transplantation tolerance by suppressing the reaction of effective T cells.

Abstract:Background: Inducing donor-specific tolerance in renal transplant patients could potentially prevent allograft rejection and calcineurin inhibitor nephrotoxicity. Combined kidney and hematopoietic stem cell transplant from an HLA-matched donor is an exploratory and promising therapy to induce immune tolerance. Investigation of molecular mechanisms involved in the disease is needed to understand the potential process of cell therapy and develop strategies to prevent this immunologic rejection. Methods: We enrolled nine patients in a clinical study in which cryopreserved donor hematopoietic stem cells were infused on days 2, 4, and 6 after kidney transplantation. One month post-transplant, 4 plasma samples were collected from combined transplants (C + Tx), and 8 plasma samples from patients with kidney transplantation alone (Tx). High abundance proteins in plasma were depleted and the two-dimensional liquid chromatography-tandem mass spectrometry coupled with iTRAQ labeling was utilized to identify the protein profiling between the two groups. Clusters of up- and down-regulated protein profiles were submitted to MetaCore for the construction of transcriptional factors and regulation networks. Results and discussion: Among the 179 identified proteins, 65 proteins were found in C + Tx with at least a 2-fold change as compared with Tx. A subset of proteins related to the complement and coagulation cascade, including complement C3a,complement C5a, precrusors to fibrinogen alpha and beta chains,was significantly downregulated in C + Tx. Meanwhile, Apolipoprotein-A1(ApoA1), ApoC1, ApoA2, ApoE, and ApoB were significantly lower in Tx compared to C + Tx. Gene ontology analysis showed that the dominant processes of differentially expressed proteins were associated with the inflammatory response and positive regulation of plasma lipoprotein particle remodeling. Conclusions: Thus, our study provides new insight into the molecular events in the hematopoietic stem cell-induced immunologic tolerance.
Mechanism of Donor Antigen-Specific CD4+CD25+regulatory T Cells on Prolonging Islet Grafts Survival

Abstract:Objective To investigate the potential mechanism of antigen-specific CD4+CD25+ regulatory T cells on the suppression of rejection for allogenetic islet transplantation in vivo.Methods Islets with 8×105 antigen-specific Treg were allogeneically transplanted under the kidney capsule of streptozotocin-induced diabetic BALB/cByJ mice.Mean survival time(MST),the ratio of CD4+/CD8+ T cells,cytokine expression in both tolerant and rejected mice was detected.Results In vivo,the mean survival time of recipients with islets and antigen-specific CD4+CD25+ Treg group(C group) was(34.57±17.15) days,whereas transplanted islets without Treg treatment(B group) survived a mean time of(10.60±1.82)d(P0.01).The ratio of CD4+/CD8+ T cells from antigen-specific CD4+CD25+ Treg transplantation group was significantly lower than that from islet transplantation group(P0.01).The mRNA expressions of IL-10,TGF-β were upregulated in antigen-specific CD4+CD25+ Treg transplantation group.However,themRNA expressions of IL-1β,IL-2,IFN-γ were similarly upregulated in islet B group.Conclusions Allogeneic antigen-specific CD4+CD25+ Treg cells can prolong islet graft survival by the mechanism of affecting Th2/Th1 cells responses.

Abstract:Kidney transplantation is the most common solid organ transplant and the best current therapy for end-stage kidney failure. However, with standard immunosuppression, most transplants develop chronic dysfunction or fail, much of which is due to chronic immune injury. Tregs are a subset of T cells involved in limiting immune activation and preventing autoimmune disease. These cells offer the potential to provide tolerance or to allow reduction in immunosuppression in kidney transplants. The importance of Tregs in kidney transplantation has been shown in a number of seminal mouse and animal studies, including those with T cell receptors (TCRs) transgenic Tregs (TCR-Tregs) or Chimeric Antigen Receptor (CAR) Tregs (CAR-Tregs) showing that specificity increases the potency of Treg function. Here we outline the animal and human studies and clinical trials directed at using Tregs in kidney transplantation and other tolerance settings and the various modifications to enhance allo-specific Treg function in vivo and in vitro .

Changes of CD4+CD25+ Regulatory T Cells in Orthotopic Liver Transplantation Model of Rats

Abstract:To evaluate the roles of CD4+CD25+ Treg in spontaneous immune tolerance,we established the orthotopic liver transplantation rat model and compared the proportion of CD4+CD25+ Treg in peripheral blood.Thirty one of inbred male BN rats and LEW rats were modified Kamada two-cuff technique,then randomly divided into 3 groups: rejection group(LEW→BN,n=9),tolerance group(BN→LEW,n=11),isogenic group(BN→BN,n=11).We collected blood serum of the recipients and randomly killed 2 recipients of each group on 5 days,10 days,30 days post-transplant,for measuring the proportion of CD4+CD25+Treg and observing the pathological changes in liver.Five recipients were left in each group to observe the survival time.We found that the median survival times of three groups were 14 days,112 days,and 129 days,respectively;and there was no rejection in tolerance group and isogenic group.The grafts Banff scales in rejection group were Ⅲ.Beginning immediately after transplantation,the ratio of Treg cells increased in both tolerance group and rejection group.Ten days after transplantation,CD4+CD25+Treg proportion in tolerance group was significant higher that that in isogenic group(P 0.01).And the tolerance group maintains the high level of Treg cells until 30 days post-transplant.The results show that the CD4+CD25+Treg is one of the mechanisms which can induce spontaneous immune tolerance following rat liver transplantation.
