Abstract:Self-incompatibility(SI) prevents inbreeding and abroad exists a genetic barrier form any flowering plants.Therefire,it plays an important role in breeding programme.SI is genetically controlled by a single polymorphic locus,termed the S-locus.The article reviews certain progrees in Brassica Oleracea signaling about some self-incompatibility-related genes such as SRK,SCR/SP(11),SLG,ARC(11),THL(1),THL(2) and so on in recent years.SRK gene and SP(11)/SCR gene are thought to be two key molecules in SI.The protein of SP(11)/SCR encoding specifically expressed in the anther tapetum and in pollen.SLG may play a role in enhancing the SI response.Meanwhile,discussed the brilliant prospect about the model of action of dowm protein in signal ransduction pathway.

Abstract:The self-incompatibility (SI) system inBrassica oleracea is genetically controlled by a single polymorphicS-locus that encodes proteins initiating a process of SI signaling transduction. This process involves eight protein elements including SLG, SCR, SRK, MLPK, THL, ARC1, Exo70A1, and MIP-MOD. Here, based on their corresponding gene’s architecture, we summa-rized these elements on their advances both in structure and function of their genes and proteins, as well as their interaction along the whole SI signal transduction process. Furthermore, we put forward some insights into unknown areas of SI, hoping to provide a few of clues for further exploration of SI mechanism inB. oleraceaor otherBrassicaspecies.

Abstract:In order to avoid species degradation caused by spontaneous pollen fertilization,flowering plants evolved out of many mechanisms,self-incompatibility was one of them.It could enhance the diversity of plants and the ability to adapt to the natural environment to effectively promote hybridization.It could be divided into different types according to different genetic methods.In this review,we based on the results of study of sporophytic self-incompatibility in recent years,reviewed the molecular mechanism of sporophytic self-incompatibility,and the function of S-locus and S-locus related gene in Brassica and other plants.There is less studies of sporophytic self-incompatibility in plants besides Brassica,these studies lacked of comprehensive understanding and research,and the signal transmission path was not entirely clear,therefore it needed further study.

Abstract:Self-incompatibility (SI) is a widespread mechanism in flowering plants that promotes outbreeding and thereby increases genetic diversity. Recognition specificity in Brassica is achieved by the interaction of the female determinant S -receptor kinase (SRK) and its ligand, the male determinant S -locus protein 11 (SP11). The interaction between SP11 and SRK triggers the signaling cascade in an S -haplotype-specific manner and results in the rejection of self-pollen, but the signal components involved are still not well characterized. S haplotypes are widespread in self-compatible amphidiploid B. napus , and the interaction of heterozygous S haplotypes causes the loss of SI. This review highlights the recent advances made towards understanding the genetic analysis, distribution, and evolution of S haplotypes, the signal factors, and the potential of SI in B. napus hybrid breeding program.
Transcriptomic Analysis Identifies Critical Signaling Components Involved in the Self-Incompatibility Response in Chinese Cabbage

Abstract:Self-incompatibility (SI) is a genetic mechanism in plants to prevent inbreeding, promote outcrossing and thereby increase genetic diversity. Although current studies have well revealed the pathway of SI recognition specificity underlying interaction of female and male determinants in Brassica, the inhibition mechanism of incompatible pollen germination and tube growth is still deficient. In this study, histological analysis showed that the pollen germination and tube growth were obviously suppressed after self-pollination in self-incompatible Chinese cabbage. Furthermore, RNA-seq and gene expression analysis were performed to identify the differentially expressed genes (DEGs) involved in SI response of Chinese cabbage. A total of 59 DEGs belonging to known SI signaling factors and Ca2+-binding proteins were identified in Chinese cabbage after incompatible self-pollination. QRT-PCR analysis found that several genes were significantly differentially expressed during SI response. Chromosomal localization and potential duplicated genes were also predicted. Additionally, spatial temporal expression profiling revealed that 15 DEGs were specifically expressed in the floral organ, some of which exhibited higher expressions in the anther and style. These findings will provide insights into the functional exploration of SI signaling factors in Chinese cabbage, and promote the further understanding of SI signaling networks in Brassica.

Abstract:Self-incompatibility plays a vital role in angiosperms, by preventing inbreeding depression and maintaining genetic diversity within populations. Following polyploidization, many angiosperm species transition from self-incompatibility to self-compatibility. Here, we investigated the S-locus in Brassicaceae and identified distinct origins for the sRNA loci, SMI and SMI2 (SCR Methylation Inducer 1 and 2), within the S-locus. The SMI loci were found to be widespread in Cruciferae, whereas the SMI2 loci were exclusive to Brassica species. Additionally, we discovered four major S-haplotypes (BnS-1, BnS-6, BnS-7, and BnS-1300) in rapeseed. Overexpression of BnSMI-1 in self-incompatible Brassica napus ('S-70(S1300S6)') resulted in a significant increase in DNA methylation in the promoter regions of BnSCR-6 and BnSCR-1300, leading to self-compatibility. Conversely, by overexpressing a point mutation of BnSmi-1 in the 'S-70(S1300S6)' line, we observed lower levels of DNA methylation in BnSCR-6 and BnSCR-1300 promoters. Furthermore, the overexpression of BnSMI2-1300 in the 'SI-326(S7S6)' line inhibited the expression of BnSCR-7 through transcriptional repression of the Smi2 sRNA from the BnS-1300 haplotype. Our study demonstrates that the self-compatibility of rapeseed is determined by S-locus sRNA-mediated silencing of SCR after polyploidization, which helps to further breed self-incompatible or self-compatible rapeseed lines, thereby facilitating the utilization of heterosis.

Abstract:In the Brassicaceae, self-incompatibility(SI) is controlled by the S-locus receptor kinase(SRK) and the S-locus cysteine-rich(SCR) protein. These two proteins are the SI specificity determinants in stigma and pollen, respectively. Recently, the Arabidopsis thaliana with SI phenotype has been achieved by transferring SRK/SCR allelic pairs from self-incompatible relative species to wild type A. thaliana. The new model plant was applied in the mechanism and evolution research of SI in Brassicaceae. This review summarizes the advances in utilizing transgenic A. thaliana SI model, including screening of new SI genes, function analysis of candidate genes and investigation of evolutionary switches to self-compatibility(SC).

Abstract:Self-incompatibility (SI) is a widespread mechanism in flowering plants which prevents self-fertilization and inbreeding. In Brassica , recognition of the highly polymorphic S -locus cysteine-rich protein (SCR; or S -locus protein 11) by the similarly polymorphic S -locus receptor kinase (SRK) dictates the SI specificity. Here, we report the crystal structure of the extracellular domain of SRK9 (eSRK9) in complex with SCR9 from Brassica rapa . SCR9 binding induces eSRK9 homodimerization, forming a 2:2 eSRK:SCR heterotetramer with a shape like the letter “A”. Specific recognition of SCR9 is mediated through three hyper-variable (hv) regions of eSRK9. Each SCR9 simultaneously interacts with hvI and one-half of hvII from one eSRK9 monomer and the other half of hvII from the second eSRK9 monomer, playing a major role in mediating SRK9 homodimerization without involving interaction between the two SCR9 molecules. Single mutations of residues critical for the eSRK9-SCR9 interaction disrupt their binding in vitro . Our study rationalizes a body of data on specific recognition of SCR by SRK and provides a structural template for understanding the co-evolution between SRK and SCR.

Abstract:The self-incompatibility (SI) system of the Brassicaceae is based on allele-specific interactions among haplotypes of the S locus. In all tested self-incompatible Brassicaceae, the S haplotype encompasses two linked genes, one encoding the S-locus receptor kinase (SRK), a transmembrane kinase displayed at the surface of stigma epidermal cells, and the other encoding its ligand, the S-locus cysteine-rich (SCR) protein, which is localized in the pollen coat. Transfer of the two genes to self-fertile Arabidopsis thaliana allowed the establishment of robust SI in several accessions, indicating that the signaling cascade triggered by this receptor–ligand interaction and the resulting inhibition of “self” pollen by the stigma have been maintained in extant A. thaliana. Based on studies in Brassica species, the membrane-tethered kinase MLPK, the ARM repeat-containing U-box protein ARC1, and the exocyst subunit Exo70A1 have been proposed to function as components of an SI signaling cascade. Here we tested the role of these molecules in the SI response of A. thaliana SRK-SCR plants. We show that the A. thaliana ARC1 ortholog is a highly decayed pseudogene. We also show that, unlike reports in Brassica, inactivation of the MLPK ortholog AtAPK1b and overexpression of Exo70A1 neither abolish nor weaken SI in A. thaliana SRK-SCR plants. These results do not support a role for these molecules in the SI response of A. thaliana.

Abstract:Self-incompatibility (SI) in plants genetically prevents self-fertilization to promote outcrossing and genetic diversity. Its hybrids in Brassica have been widely cultivated due to the propagation of SI lines by spraying a salt solution. We demonstrated that suppression of Brassica napus SI from edible salt solution treatment was ascribed to sodium chloride and independent of S haplotypes, but it did not obviously change the expression of SI-related genes. Using the isobaric tags for relative and absolute quantitation (iTRAQ) technique, we identified 885 differentially accumulated proteins (DAPs) in Brassica napus stigmas of un-pollinated (UP), pollinated with compatible pollen (PC), pollinated with incompatible pollen (PI), and pollinated with incompatible pollen after edible salt solution treatment (NA). Of the 307 DAPs in NA/UP, 134 were unique and 94 were shared only with PC/UP. In PC and NA, some salt stress protein species, such as glyoxalase I, were induced, and these protein species were likely to participate in the self-compatibility (SC) pathway. Most of the identified protein species were related to metabolic pathways, biosynthesis of secondary metabolites, ribosome, and so on. A systematic analysis implied that salt treatment-overcoming SI in B. napus was likely conferred by at least five different physiological mechanisms: (i) the use of Ca2+ as signal molecule; (ii) loosening of the cell wall to allow pollen tube penetration; (iii) synthesis of compatibility factor protein species for pollen tube growth; (iv) depolymerization of microtubule networks to facilitate pollen tube movement; and (v) inhibition of protein degradation pathways to restrain the SI response.

Abstract:Self-incompatibility (SI) is a mechanism in plants that prevents self-fertilization and promotes outcrossing. SI is also widely utilized in the breeding of Brassicaceae crops. Understanding the regulatory mechanisms of SI is essential but has been greatly restrained in most Brassicaceae crops due to inefficient transformation. In this study, we developed methods for examining signaling pathways and genes of pollen-stigma interactions in Brassicaceae crops lacking an efficient genetic transformation system. We pretreated excised stigmas of Brassica rapa (B. rapa L. ssp. Pekinensis) in vitro with chemicals to modify signaling pathways or with phosphorothioate antisense oligodeoxyribonucleotides (AS-ODNs) to modify the expression of the corresponding genes involved in pollen-stigma interactions. Using this method, we first determined the involvement of reactive oxygen species (ROS) in SI with the understanding that the NADPH oxidase inhibitor diphenyleneiodonium chloride, which inhibits ROS production, eliminated the SI of B. rapa. We further identified the key gene for ROS production in SI and used AS-ODNs targeting BrRBOHF (B. rapa RESPIRATORY-BURST OXIDASE HOMOLOGF), which encodes one of the NADPH oxidases, to effectively suppress its expression, reduce stigmatic ROS, and promote the growth of self-pollen in B. rapa stigmas. Moreover, pistils treated in planta with the ROS scavenger sodium salicylate disrupted SI and resulted in enlarged ovules with inbred embryos 12 d after pollination. This method will enable the functional study of signaling pathways and genes regulating SI and other pollen-stigma interactions in different Brassicaceae plants. Treating stigmas with chemical and antisense oligodeoxyribonucleotides enables the study of signaling pathways and genes in pollen-stigma interactions in Brassicaceae crops lacking a transgenic system.

Abstract:Plants have evolved diverse self-incompatibility (SI) systems for outcrossing. Ever since Darwin's time, considerable progress has been made toward elucidating this unrivaled reproductive innovation. Recent advances in interdisciplinary studies and the applications of biotechnology have given rise to major breakthroughs in understanding the molecular pathways leading to SI, particularly the strikingly different SI mechanisms operating in Solanaceae, Papaveraceae, Brassicaceae and Primulaceae. These best understood SI systems, together with the discoveries from other "non-model" SI taxa such as Poaceae, suggest a complex evolutionary trajectory of SI with multiple independent origins and prevalent and irreversible losses. The extensive exploration of self/non-self-discrimination signaling cascades has revealed a comprehensive catalog of the male and female identity genes and modifier factors controlling SI. These findings also enable the characterization, validation and manipulation of SI-related factors to be applied in a crop improvement context, addressing the challenges associated with the development of inbred lines. Here, we review the current knowledge on the evolution of SI systems, summarize the key achievements in the molecular basis of pollen‒pistil interactions, discuss potential prospects in breeding SI crops, and raise several unsolved questions that require further investigation.

Abstract:Self-incompatibility (SI) refers to the characteristic that the pistil of a plant can recognize its own pollen and allogeneic pollen so as to inhibit the germination of its own pollen. PUB (Plant U-Box) proteins play an important role in plant stress resistance and signal transduction. In this study, through the analysis of stigma transcriptome data of Brassica oleracea self-pollination at 0-60 minute(s), a PUB protein coding gene BoPUB9 was screened and up-regulated by self-pollination. The BoPUB9 cDNA sequence was 1368 bp, gDNA of 1720 bp in length, containing one arm repeat domain and one U-box domain. The relative expression levels showed that BoPUB9 gene was expressed in different tissues of Brassica oleracea, and the relative expression level was the highest in sepals, followed by petals, stamens and stigmas, and relatively lower in pollen buds, which was consistent with the results of GUS staining. The relative expression level of BoPUB9 gene was up-regulated rapidly in self-pollination 0-30 minute(s), the expression level after 15 min was more than 20 times that after cross-pollination, and the difference was the biggest after 30 min, the expression level after self-pollination was more than 40 times that after cross-pollination. Subcellular localization revealed that BoPUB9 protein was expressed in both nucleus and cytoplasm, and BoPUB9 protein was successfully induced in E. coli, and the relative molecular weight of 51 kD was consistent with the predicted results. The results of yeast two-hybrid and poll-down indicated that the intracellular domain of SRK interacted with PUB9 protein. It was speculated that BoPUB9 might be a novel gene involved in the process of self-incompatibility in Brassica oleracea, which providing a new content for further research and utilization of self-incompatibility in Brassica oleracea.

Abstract:Self-incompatibility (SI) is a genetic mechanism to promote outcrossing and prevent self-fertilization in plants. Brassicaceae crops have a typical SI response that exhibits the inhibition of incompatible pollen germination and pollen tube growth. Calcium (Ca2+) is a necessary regulator of pollen germination and pollen tube growth. However, the signaling processes by which Ca2+ regulates the SI response are not clear in Chinese cabbage. In this study, exogenous CaCl2 and EGTA spraying was performed to alter the Ca2+ level of the stigma and detect the correlation between Ca2+ and SI response in Chinese cabbage. The decrease in Ca2+ levels in the stigma caused by EGTA treatment could promote pollen germination and break the SI response. Moreover, transmission electron microscopy showed that the accumulation and distribution of cytoplasmic Ca2+ in the stigma were related to the occurrence of the SI response. Additionally, eight BrCaM family genes were identified in Brassica rapa by genome-wide analysis. Expression analysis suggested that the expression of several BrCaM genes was coincident with Ca2+ level alterations and the SI response. These results facilitate understanding the putative roles of BrCaM genes in regulating pollen germination and pollen tube growth and provide valuable references for uncovering the molecular regulatory mechanism of the SI response in Brassicaceae crops.
Characterization of S haplotype in a new self-compatible Brassica rapa cultivar Dahuangyoucai

Abstract:The most important Brassica species, B. rapa, is naturally self-incompatible. Self-compatible mutants would be useful for dissecting the molecular mechanism of self-incompatibility (SI), a process that promotes outcrossing by recognizing and refusing self-pollens. The S haplotype in a new self-compatible B. rapa cultivar, Dahuangyoucai, was characterized for the first time in this study. Sequence analysis of the S-locus genes, SLG (S-locus glycoprotein), SRK (S-locus receptor kinase) and SCR (S-locus cysteine-rich protein) revealed that Dahuangyoucai contained S haplotype highly similar to S-f2, a non-functional class I S haplotype identified in another self-compatible B. rapa cultivar, Yellow Sarson. Mutations of MLPK (M-locus protein kinase) and non-transcription of the male determinant, SCR, were observed in this cultivar, which is similar to the situation reported in Yellow Sarson. With respect to the female determinant, SRK, no transcript was detected in Yellow Sarson but two fragments were detected in Dahuangyoucai. One fragment was highly similar to SRK-f2, but the other fragment was different from the signal factors previously identified in the SI reaction. The results suggest that Dahuangyoucai and Yellow Sarson have the same origin and a similar mechanism of self-compatibility, but diverge after mutations in SRK, SCR and MLPK. Further studying the self-compatibility of Dahuangyoucai might identify novel factors involved in the SI signalling cascade and provide new insights into the mechanisms of SI in Brassicaceae.

Abstract:Self-incompatibility (SI) is a mechanism in plants that prevents self-fertilization and promotes out-crossing. SI is also widely utilized in the breeding of Brassicaceae crops. Understanding the regulatory mechanisms of SI is essential but has been greatly restrained in most Brassicaceae crops due to inefficient transformation. Here, we developed methods for studying signaling pathways and genes of pollen-stigma interactions in Brassicaceae crops lacking an efficient genetic transformation system. We pretreated excised stigmas of Brassica rapa (Brassica rapa L. ssp. Pekinensis) in vitro with chemicals to modify signaling pathways or with phosphorothioate antisense oligodeoxyribonucleotides (AS-ODNs) to modify the expression of the corresponding genes involved in pollen-stigma interactions. Using this method, we firstly determined the involvement of reactive oxygen species (ROS) in SI with the understanding that the NADPH oxidase inhibitor diphenyleneiodonium chloride (DPI), which inhibits ROS production, eliminated SI of B. rapa. We further identified the key gene for ROS production in SI and used AS-ODNs targeting BrRBOHF (Brassica rapa RESPIRATORY-BURST OXIDASE HOMOLOGF), which encodes one of the NADPH oxidases, to effectively suppress its expression, reduce stigmatic ROS, and promote the growth of self–pollen in B. rapa stigmas. Moreover, pistils treated in planta with the ROS scavenger sodium salicylate (Na-SA) disrupted SI and resulted in enlarged ovules with inbred embryos 12 days after pollination. This method will enable the functional study of signaling pathways and genes regulating SI and other pollen-stigma interactions in different Brassicaceae plants.

Abstract:In nature, most self-incompatible flowering plants (angiosperms) show gametophytic self-incompatibility. Although gametophytic self-incompatibility functions can ultimately prevent self-fertilization, flowering plants have adopted different signal transduction pathways to reject self pollen. At present, there are mainly two pathways of signal transduction on gametophytic self-incompatibility. One is the S-RNase-based signal transduction in Solanaceae, Scrophulariaceae, and Rosaceae. The other is the cytosolic free Ca2+ acting as a second messenger in pollen of Papaveraceae. This review highlights the recent progress made towards understanding the signal transduction on gametophytic self-incompatibility.
