Abstract:More than half of flowering plants have a sophisticated mechanism for self-pollen rejection, named self-incompatibility. In the Brassicaceae family, the recognition specificity of a self-incompatibility system is achieved by the interaction of the stigmatic S-receptor kinase and its ligand S-locus cysteine-rich protein, which are encoded by two tightly linked polymorphic genes. During the last two decades, many studies have explored their functions, although their origin and evolutionary history have still not been elucidated clearly. In the present study, an extensive survey in nine whole-genome sequenced plants, including one moss, one fern and seven flowering plants, was conducted to clarify these issues. The data obtained showed that S_locus_glycop domain-related genes, which are land plant specific, have an ancient origin that can be traced back to early land plants and also have a significantly expansion in flowering plants. In the four predominant domain architectures (Types I to IV) of these proteins, Type III genes had absolute predominance and appeared to be raw materials for diversification of the S_locus_glycop domain-related genes by frequent domain re-organizations. S-receptor kinase-like sequences (Type IV) might have originated from Type III genes by domain gains, and S-locus glycoprotein-like sequences (Type I) might have arisen by partial duplication of its linked S-receptor kinase genes. Although similar topologies were detected in S-receptor kinase and S-locus cysteine-rich protein trees, their physical linkage were found only in Brassicaceae, suggesting that the strong linkage disequilibrium and their co-evolution may be a key factor for the origin and maintenance of the S-receptor kinase-based self-incompatibility system in Brassicaceae.

Abstract:In order to avoid species degradation caused by spontaneous pollen fertilization,flowering plants evolved out of many mechanisms,self-incompatibility was one of them.It could enhance the diversity of plants and the ability to adapt to the natural environment to effectively promote hybridization.It could be divided into different types according to different genetic methods.In this review,we based on the results of study of sporophytic self-incompatibility in recent years,reviewed the molecular mechanism of sporophytic self-incompatibility,and the function of S-locus and S-locus related gene in Brassica and other plants.There is less studies of sporophytic self-incompatibility in plants besides Brassica,these studies lacked of comprehensive understanding and research,and the signal transmission path was not entirely clear,therefore it needed further study.

Abstract:Most plants in the Brassicaceae evolve self-incompatibility (SI) to avoid inbreeding and generate hybrid vigor. Self-pollen is recognized by the S-haplotype-specific interaction of the pollen ligand S-locus protein 11 (SP11) (also known as S-locus cysteine-rich protein [SCR]) and its stigma-specific S-locus receptor kinase (SRK). However, mechanistically much remains unknown about the signaling events that culminate in self-pollen rejection. Here, we show that self-pollen triggers high levels of reactive oxygen species (ROS) in stigma papilla cells to mediate SI in heading Chinese cabbage (Brassica rapa L. ssp. pekinensis). We found that stigmatic ROS increased after self-pollination but decreased after compatible(CP)- pollination. Reducing stigmatic ROS by scavengers or suppressing the expression of respiratory burst oxidase homologs (Rbohs), which encode plant NADPH oxidases that produce ROS, both broke down SI. On the other hand, increasing the level of ROS inhibited the germination and penetration of compatible pollen on the stigma, mimicking an incompatible response. Furthermore, suppressing a B. rapa FERONIA (FER) receptor kinase homolog or Rac/Rop guanosine triphosphatase (GTPase) signaling effectively reduced stigmatic ROS and interfered with SI. Our results suggest that FER-Rac/Rop signaling-regulated, NADPH oxidase-produced ROS is an essential SI response leading to self-pollen rejection.

Abstract:Self-incompatibility (SI) is a widespread mechanism in flowering plants which prevents self-fertilization and inbreeding. In Brassica , recognition of the highly polymorphic S -locus cysteine-rich protein (SCR; or S -locus protein 11) by the similarly polymorphic S -locus receptor kinase (SRK) dictates the SI specificity. Here, we report the crystal structure of the extracellular domain of SRK9 (eSRK9) in complex with SCR9 from Brassica rapa . SCR9 binding induces eSRK9 homodimerization, forming a 2:2 eSRK:SCR heterotetramer with a shape like the letter “A”. Specific recognition of SCR9 is mediated through three hyper-variable (hv) regions of eSRK9. Each SCR9 simultaneously interacts with hvI and one-half of hvII from one eSRK9 monomer and the other half of hvII from the second eSRK9 monomer, playing a major role in mediating SRK9 homodimerization without involving interaction between the two SCR9 molecules. Single mutations of residues critical for the eSRK9-SCR9 interaction disrupt their binding in vitro . Our study rationalizes a body of data on specific recognition of SCR by SRK and provides a structural template for understanding the co-evolution between SRK and SCR.

Abstract:Plants have evolved diverse self-incompatibility (SI) systems for outcrossing. Ever since Darwin's time, considerable progress has been made toward elucidating this unrivaled reproductive innovation. Recent advances in interdisciplinary studies and the applications of biotechnology have given rise to major breakthroughs in understanding the molecular pathways leading to SI, particularly the strikingly different SI mechanisms operating in Solanaceae, Papaveraceae, Brassicaceae and Primulaceae. These best understood SI systems, together with the discoveries from other "non-model" SI taxa such as Poaceae, suggest a complex evolutionary trajectory of SI with multiple independent origins and prevalent and irreversible losses. The extensive exploration of self/non-self-discrimination signaling cascades has revealed a comprehensive catalog of the male and female identity genes and modifier factors controlling SI. These findings also enable the characterization, validation and manipulation of SI-related factors to be applied in a crop improvement context, addressing the challenges associated with the development of inbred lines. Here, we review the current knowledge on the evolution of SI systems, summarize the key achievements in the molecular basis of pollen‒pistil interactions, discuss potential prospects in breeding SI crops, and raise several unsolved questions that require further investigation.

Abstract:To avoid inbreeding and promote out-crossing,many flowering plants have adopted self-incompatibility(SI) systems,through which incompatible(self or genetically related) pollen is recognized and rejected,whereas compatible(non-self) pollen is allowed to grow in the style to deliver the germ cells to the ovary for double fertilization.Among various SI systems,gametophytic SI in Solanaceae,Scrophulariaceae and Rosaceae appears to be the most common whereby the specificity of SI response is controlled by a single polymorphic S-locus.Recent studies have shown that the S-locus is organized in a haplotype fashion and carries at least two genes determining the recognition specificity:S-ribonucleases expressed in the pistil(pistil-S) and S-locus F-box(SLF) genes in the pollen(pollen-S).Here we discuss recent data on the possible molecular mechanisms eliciting the S-RNase-based self-incompatibility response.

Abstract:Self-incompatibility (SI) is a mechanism in plants that prevents self-fertilization and promotes out-crossing. SI is also widely utilized in the breeding of Brassicaceae crops. Understanding the regulatory mechanisms of SI is essential but has been greatly restrained in most Brassicaceae crops due to inefficient transformation. Here, we developed methods for studying signaling pathways and genes of pollen-stigma interactions in Brassicaceae crops lacking an efficient genetic transformation system. We pretreated excised stigmas of Brassica rapa (Brassica rapa L. ssp. Pekinensis) in vitro with chemicals to modify signaling pathways or with phosphorothioate antisense oligodeoxyribonucleotides (AS-ODNs) to modify the expression of the corresponding genes involved in pollen-stigma interactions. Using this method, we firstly determined the involvement of reactive oxygen species (ROS) in SI with the understanding that the NADPH oxidase inhibitor diphenyleneiodonium chloride (DPI), which inhibits ROS production, eliminated SI of B. rapa. We further identified the key gene for ROS production in SI and used AS-ODNs targeting BrRBOHF (Brassica rapa RESPIRATORY-BURST OXIDASE HOMOLOGF), which encodes one of the NADPH oxidases, to effectively suppress its expression, reduce stigmatic ROS, and promote the growth of self–pollen in B. rapa stigmas. Moreover, pistils treated in planta with the ROS scavenger sodium salicylate (Na-SA) disrupted SI and resulted in enlarged ovules with inbred embryos 12 days after pollination. This method will enable the functional study of signaling pathways and genes regulating SI and other pollen-stigma interactions in different Brassicaceae plants.

Abstract:Self-incompatibility in the Brassicaceae is controlled by multiple haplotypes encoding the pollen ligand (S-locus protein 11, SP11, also known as S-locus cysteine-rich protein, SCR) and its stigmatic receptor (S-receptor kinase, SRK). A haplotype-specific interaction between SP11/SCR and SRK triggers the self-incompatibility response that leads to self-pollen rejection, but the signalling pathway remains largely unknown. Here we show that Ca(2+) influx into stigma papilla cells mediates self-incompatibility signalling. Using self-incompatible Arabidopsis thaliana expressing SP11/SCR and SRK, we found that self-pollination specifically induced an increase in cytoplasmic Ca(2+) ([Ca(2+)]cyt) in papilla cells. Direct application of SP11/SCR to the papilla cell protoplasts induced Ca(2+) increase, which was inhibited by D-(-)-2-amino-5-phosphonopentanoic acid (AP-5), a glutamate receptor channel blocker. An artificial increase in [Ca(2+)]cyt in papilla cells arrested wild-type (WT) pollen hydration. Treatment of papilla cells with AP-5 interfered with self-incompatibility, and Ca(2+) increase on the self-incompatibility response was reduced in the glutamate receptor-like channel (GLR) gene mutants. These results suggest that Ca(2+) influx mediated by GLR is the essential self-incompatibility response leading to self-pollen rejection.
Exo84c-regulated degradation is involved in the normal self-incompatible response in Brassicaceae

Abstract:The self-incompatibility system evolves in angiosperms to promote cross-pollination by rejecting self-pollination. Here, we show the involvement of Exo84c in the SI response of both Brassica napus and Arabidopsis. The expression of Exo84c is specifically elevated in stigma during the SI response. Knocking out Exo84c in B. napus and SI Arabidopsis partially breaks down the SI response. The SI response inhibits both the protein secretion in papillae and the recruitment of the exocyst complex to the pollen-pistil contact sites. Interestingly, these processes can be partially restored in exo84c SI Arabidopsis. After incompatible pollination, the turnover of the exocyst-labeled compartment is enhanced in papillae. However, this process is perturbed in exo84c SI Arabidopsis. Taken together, our results suggest that Exo84c regulates the exocyst complex vacuolar degradation during the SI response. This process is likely independent of the known SI pathway in Brassicaceae to secure the SI response.

Abstract:Self-incompatibility (SI) is a genetically controlled system adopted by many flowering plants to avoid inbreeding and thus to maintain species diversity. Generally, self-pollen rejection Occurs through active pollen and pistil recognition and subsequent signaling responses. So far, three different molecular controls of pollen and pistil recognition have been characterized and are exemplified by three families: the Solanaceae, the Papaveraceae, and the Brassicaceae. With more components involved in these SI systems coming to light, recent studies have provided intriguing insights into the downstream reactions that follow the initial SI signal perception. The process of pollen rejection is closely associated with rapid and effective proteolytic events, including the ubiquitin-proteasome pathway and the vacuolar sorting pathway. Here, we review our current understanding of the roles of proteolysis in SI responses of flowering plants.
Genome-wide Identification and Characterization of SnRK Family Genes in Brassica Napus

Abstract:Background Sucrose non-fermenting 1 related protein kinases ( SnRK) play crucial roles in responding to biotic and abiotic stresses through activating protein phosphorylation pathways. However, little information of SnRK genes was available in Brassica napus , one of important oil crops. Recently, the released sequences of the reference genome of B.napus provide a good chance to perform genome-wide identification and characterization of BnSnRK gene family in the rapeseed. Results Totally 114 SnRK genes distributed on 19 chromosomes were identified in the genome of B.napus and classified into three subfamilies on the basis of phylogenetic analysis and the domain types. According to gene structure and motif composition analysis, the BnSnRK sequences showed obvious divergence among three subfamilies. Gene duplication and synteny between the genomes of the rapeseed and Arabidopsis were also analyzed to provide insights into the evolutionary characteristics of BnSnRK family genes. C is- element analysis revealed that BnSnRKs may response to diverse environmental stresses. Moreover, the expression patterns of BnSnRKs in various tissues and under diverse abiotic stresses were distinct difference. Besides, Single Nucleotide Polymorphisms (SNP) distribution analysis suggests the function disparity of BnSnRK family genes in different genotypes of the rapeseed. Conclusion We examined genomic structures, evolution features, expression patterns and SNP distribution of 114 BnSnRKs . The results provide valuable information for functional characterization of BnSnRK genes in future studies.
Gene coexpression analysis reveals key pathways and hub genes related to late-acting self-incompatibility in Camellia oleifera

Abstract:Introduction: Self-incompatibility (SI) is an important strategy for plants to maintain abundant variation to enhance their adaptability to the environment. Camellia oleifera is one of the most important woody oil plants and is widely cultivated in China. Late acting self-incompatibility (LSI) in C. oleifera results in a relatively poor fruit yield in the natural state, and understanding of the LSI mechanism remains limited. Methods: To better understand the molecular expression and gene coexpression network in the LSI reaction in C. oleifera , we conducted self- and cross-pollination experiments at two different flower bud developmental stages (3–4 d before flowering and 1 d before flowering), and cytological observation, fruit setting rate (FSR) investigation and RNA-Seq analysis were performed to investigate the mechanism of the male −female interaction and identify hub genes responsible for the LSI in C. oleifera . Results: Based on the 21 ovary transcriptomes, a total of 7669 DEGs were identified after filtering out low-expression genes. Weighted gene coexpression network analysis (WGCNA) divided the DEGs into 15 modules. Genes in the blue module (1163 genes) were positively correlated with FSR, and genes in the pink module (339 genes) were negatively correlated with FSR. KEGG analysis indicated that flavonoid biosynthesis, plant MAPK signaling pathways, ubiquitin-mediated proteolysis, and plant-pathogen interaction were the crucial pathways for the LSI reaction. Fifty four transcription factors (TFs) were obtained in the two key modules, and WRKY and MYB were potentially involved in the LSI reaction in C. oleifera . Network establishment indicated that genes encoding G-type lectin S-receptor-like serine (lecRLK), isoflavone 3'-hydroxylase-like (CYP81Q32), cytochrome P450 87A3-like (CYP87A3), and probable calcium-binding protein (CML41) were the hub genes that positively responded to the LSI reaction. The other DEGs inside the two modules, including protein RALF-like 10 (RALF), F-box and pectin acetylesterase (MTERF5), might also play vital roles in the LSI reaction in C. oleifera . Discussion: Overall, our study provides a meaningful resource for gene network studies of the LSI reaction process and subsequent analyses of pollen−pistil interactions and TF roles in the LSI reaction, and it also provides new insights for exploring the mechanisms of the LSI response.

Abstract:SUMMARYThe angiosperms are the largest vascular plant group. However, the mechanisms for their rapid expansions in the Cretaceous and Paleogene remain unclear. Here, we find that self-incompatibility (SI) appears to be one central component contributing to their expansions. SI is a genetic barrier to prevent inbreeding widely found in the angiosperms and controlled by the polymorphic S -locus. So far, five molecular types of S -loci have been identified with the first defined by Lolium perenne domain of unknown function 247 ( LpS-DUF247 ), the second S-RNase and S-locus F-box ( SLF s), the third S-locus cysteine-rich protein ( SCR ) and S-locus receptor kinase ( SRK ), the fourth Papaver rhoeas stigma S ( PrsS ) and Papaver rhoeas pollen S ( PrpS ) and the fifth a hemizygous S -locus supergene of Primula . Phylogenomics and functional analyses showed that SI evolved twice in the Cretaceous and around the Cretaceous-Paleogene boundary, respectively. The first evolution involved the origins of the S genes of types 1 and 2 in the ancestors of Poales and eudicots, respectively. Then, the S -locus of type 2 underwent divergent evolution with losses in several ancestral eudicot lineages. The second involved the S -loci of types 3, 4 and 5 in the ancestors of Brassicaceae, Paperveraceae and Primulaceae, respectively. Interestingly, the times of the two evolutions of SI systems coincided with early angiosperm expansions. Furthermore, we find that neofunctionalization and de novo gene formation contributed to the S gene origins. Taken together, our results reveal a molecular evolutionary framework of SI in the angiosperms and its promoting role in their expansions.
Cytological Observation and Transcriptome Comparative Analysis of Self-Pollination and Cross-Pollination in Dendrobium Officinale

Abstract:Dendrobium officinale is a rare and traditional medicinal plant with high pharmacological and nutritional value. The self-incompatibility mechanism of D. officinale reproductive isolation was formed in the long-term evolution process, but intraspecific hybridization of different germplasm resources leads to a large gap in the yield, quality, and medicinal value of D. officinale. To investigate the biological mechanism of self-incompatibility in D. officinale, cytological observation and the transcriptome analysis was carried out on the samples of self-pollination and cross-pollination in D. officinale. Results for self-pollination showed that the pollen tubes could grow in the style at 2 h, but most of pollen tubes stopped growing at 4 h, while a large number of cross-pollinated pollen tubes grew along the placental space to the base of ovary, indicating that the self-incompatibility of D. officinale may be gametophyte self-incompatibility. A total of 63.41 G basesum of D. officinale style samples from non-pollinated, self-pollination, and cross-pollination by RNA-seq were obtained, and a total of 1944, 1758, and 475 differentially expressed genes (DEGs) in the comparison of CK (non-pollinated) vs. HF (cross-pollination sample), CK vs. SF (self-pollination sample) and SF vs. HF were identified, respectively. Forty-one candidate genes related to self-incompatibility were found by function annotation of DEGs, including 6 Ca2+ signal genes, 4 armed repeat containing (ARC) related genes, 11 S-locus receptor kinase (SRK) related genes, 2 Exo70 family genes, 9 ubiquitin related genes, 1 fatty acid related gene, 6 amino acid-related genes, 1 pollen-specific leucine-rich repeat extensin-like protein (LRX) related gene and 1 lectin receptor-like kinases (RLKs) related gene, showed that self-incompatibility mechanism of D. officinale involves the interaction of multiple genes and pathways. The results can provide a basis for the study of the self-incompatibility mechanism of D. officinale, and provide ideas for the preservation and utilization of high-quality resources of D. officinale.

Abstract:Summary Receptor‐like kinases (RLKs) are evolved for plant cell–cell communications. The typical RLK protein contains an extracellular and hypervariable N‐terminus to perceive various signals, a transmembrane domain to anchor into plasma membrane, and a cytoplasmic, highly conserved kinase domain to phosphorylate target proteins. To date, RLKs have manifested their significance in a myriad of biological processes during plant reproductive growth, especially in male fertility. This review first summarizes a recent update on RLKs and their interacting protein partners controlling anther and pollen development, pollen release from dehisced anther, and pollen function during pollination and fertilization. Then, regulatory networks of RLK signaling pathways are proposed. In addition, we predict RLKs in maize and rice genome, obtain homologs of well‐studied RLKs from phylogeny of three subfamilies and then analyze their expression patterns in developing anthers of maize and rice to excavate potential RLKs regulating male fertility in crops. Finally, current challenges and future prospects regarding RLKs are discussed. This review will contribute to a better understanding of plant male fertility control by RLKs, creating potential male sterile lines, and inspiring innovative crop breeding methods.

Abstract:Self-incompatibility (SI) is a genetic mechanism that promotes out-breeding and maintains genetic diversity in plant species. The inhibition of pollen germination and pollen tube growth is one of the major characteristics of SI response. The involvement of Ca 2+ signaling mediated by Ca 2+ sensors in the SI response has been demonstrated to affect the germination of incompatible pollen and the normal elongation of pollen tubes. In this study, the calmodulin-like 49 ( BrCML49 ) gene in Chinese cabbage ( Brassica rapa L. ssp. pekinensis ) was down-regulated expression during the SI process, indicating that BrCML49 may participate in the regulation of SI response. The transient suppression of BrCML49 gene induced the abnormal germination of pollen in vitro, which showed the putative positive roles of BrCML49 in regulating pollen germination. The inhibited germination of self- and cross-pollen in BrCML49 -suppressed stigma was also observed, which revealed the activated SI response. After BrCML49 suppression, the increases in Ca 2+ level in stigmatic papilla cells were consistent with the increases in ROS/H 2 O 2 contents in the stigma, which may be essential for the occurrence of SI response. Furthermore, heterologous overexpression of BrCML49 in Arabidopsis validated the positive regulation of BrCML49 in pollen germination and pollen tube growth. In addition, BrCML49 overexpressed Arabidopsis exhibited an enhanced tolerance of pollen germination to high Ca 2+ condition. These results will provide valuable information for understanding the roles of BrCML49 in regulating pollen germination and pollen tube growth during SI process in Chinese cabbage.

S-RNase Evolution in Self-Incompatibility: Phylogenomic Insights into Synteny with S-Like Genes

Abstract:S-RNases are essential in the gametophytic self-incompatibility (GSI) system of many flowering plants, where they act as stylar-S determinants. Despite their significance, the syntenic genomic origin and evolutionary trajectory of S-RNase genes in eudicots have remained largely unclear. Here, we performed large-scale phylogenetic and microsynteny network analyses of RNase T2 genes across 130 angiosperm genomes, encompassing 35 orders and 56 families. Based on the connections observed in the synteny network, particularly the persisting synteny block in none-SI Cucurbitaceae species, we infer that well-characterized S-RNase genes (Class III-A RNase genes) were duplicated and evolved from Class I S-like RNase genes, possibly as a result of the gamma triplication event shared by core eudicots. Additionally, we identified frequent lineage-specific gene transpositions of S-RNase genes across diverse angiosperm lineages, including Rosaceae, Solanaceae, and Rutaceae families, accompanied by a significant increase in transposable element (TE) activity near these genes. Our findings delineate the precise genomic origin and evolutionary path of eudicot S-RNase genes, enhancing our understanding of the evolution of the S-RNase-based GSI system.

Abstract:Self-incompatibility and recurrent transitions to self-compatibility have shaped the extant mating systems underlying the nonrandom mating critical for speciation in angiosperms. Linkage between self-incompatibility and speciation is illustrated by the shared pollen rejection pathway between self-incompatibility and interspecific unilateral incompatibility (UI) in the Brassicaceae. However, the pollen discrimination system that activates this shared pathway for heterospecific pollen rejection remains unknown. Here we show that Stigma UI3.1 , the genetically identified stigma determinant of UI in Arabidopsis lyrata × Arabidopsis arenosa crosses, encodes the S-locus-related glycoprotein 1 (SLR1). Heterologous expression of A. lyrata or Capsella grandiflora SLR1 confers on some Arabidopsis thaliana accessions the ability to discriminate against heterospecific pollen. Acquisition of this ability also requires a functional S-locus receptor kinase (SRK), whose ligand-induced dimerization activates the self-pollen rejection pathway in the stigma. SLR1 interacts with SRK and interferes with SRK homomer formation. We propose a pollen discrimination system based on competition between basal or ligand-induced SLR1–SRK and SRK–SRK complex formation. The resulting SRK homomer levels would be sensed by the common pollen rejection pathway, allowing discrimination among conspecific self- and cross-pollen as well as heterospecific pollen. Our results establish a mechanistic link at the pollen recognition phase between self-incompatibility and interspecific incompatibility.

Abstract:"Heteromorphic self-incompatibility" (HetSI) in plants is a mechanism of defense to avoid self-pollination and promote outcrossing. However, the molecular mechanism underlying HetSI remains largely unknown. In this study, RNA-seq was conducted to explore the molecular mechanisms underlying self-compatible (SC, "T × P" and "P × T") and self-incompatible (SI, "T × T" and "P × P") pollination in the two types of flowers of Plumbago auriculata Lam. which is a representative HetSI plant. By comparing "T × P" vs. "T × T", 3773 (1407 upregulated and 2366 downregulated) differentially expressed genes (DEGs) were identified, 1261 DEGs between "P × T" and "P × P" (502 upregulated and 759 downregulated). The processes in which these DEGs were significantly enriched were "MAPK (Mitogen-Activated Protein Kinases-plant) signaling pathway", "plant-pathogen interaction","plant hormone signal transduction", and "pentose and glucuronate interconversion" pathways. Surprisingly, we discovered that under various pollination conditions, multiple notable genes that may be involved in HetSI exhibited distinct regulation. We can infer that the HetSI strategy might be unique in P. auriculata . It was similar to "sporophytic self-incompatibility" (SSI) but the HetSI mechanisms in pin and thrum flowers are diverse. In this study, new hypotheses and inferences were proposed, which can provide a reference for crop production and breeding.

Abstract:Self-incompatibility (SI) is a pollen-stigma recognition system controlled by a single and highly polymorphic genetic locus known as the S-locus. The S-locus exists in all Brassica napus (B. napus, AACC), but natural B. napus accessions are self-compatible. About 100 and 50 S haplotypes exist in Brassica rapa (AA) and Brassica oleracea (CC), respectively. However, S haplotypes have not been detected in B. napus populations. In this study, we detected the S haplotype distribution in B. napus and ascertained the function of a common S haplotype BnS-6 through genetic transformation. BnS-1/BnS-6 and BnS-7/BnS-6 were the main S haplotypes in 523 B. napus cultivars and inbred lines. The expression of SRK in different S haplotypes was normal (the expression of SCR in the A subgenome affected the SI phenotype) while the expression of BnSCR-6 in the C subgenome had no correlation with the SI phenotype in B. napus. The BnSCR-6 protein in BnSCR-6 overexpressed lines was functional, but the self-compatibility of overexpressed lines did not change. The low expression of BnSCR-6 could be a reason for the inactivation of BnS-6 in the SI response of B. napus. This study lays a foundation for research on the self-compatibility mechanism and the SI-related breeding in B. napus.
