Expression and clinical significance of human telomerase reverse transcriptase mRNA in peripheral blood of patients with hepatobiliary malignancies

Abstract:Objective To explore the expression and clinical significance of human telomerase reverse transcriptase (hTERT) mRNA in peripheral blood of patients with hepatobiliary malignancies.Methods Thirty cases of hepatic carcinoma, 30 cases of gallbladder carcinoma, and 30 cases of cholangiocarcinoma, who were diagnosed pathologically in oncology department of our hospital from January 2013-December 2013, were included.Peripheral blood was drawn before operation, at 3rd month and recurrence after operation.The expression of hTERT mRNA was determined by reverse transcription-polymerase chain reaction (RT-PCR) after mononuclear cells were isolated.Thirty healthy subjects who received body check were included as controls in the same term.Results (1) In 30 cases of healthy subjects, 30 cases of liver cancer, 30 cases of gallbladder carcinoma, and 30 cases of cholangiocarcinoma preoperation, hTERT mRNA levels were 3.42 ± 1.75, 8.42 ± 2.42, 8.47 ± 2.39, and 8.17 ± 2.63 respectively;(2) The hTERT mRNA expression in peripheral blood in patients with hepatobiliary malignancies were related to tumor stage, differentiated degree and lymph node metastasis (P ＜ 0.01), but not to sex, years old and tumor category (P ＞ 0.05);(3) In 30 cases of liver cancer, 30 cases of gallbladder carcinoma, and 30 cases of cholangiocarcinoma, hTERT mRNA expression levels were 5.06 ± 1.58, 5.16 ± 1.57 and 5.13 ± 1.60 respectively postoperation;(4) Recurrence occurred in 54 cases during follow-up term, and the hTERT mRNA expression level in peripheral blood of recurrent patients increased before and at 3rd month after operation as compared with that in un-recurred patients (P ＜ 0.01).(5) In patients with tumor recurrence, hTERT mRNA expression level in peripheral blood was significantly increased at 3rd month after operation (P ＜0.01).Conclusion The hTERT mRNA expression in peripheral blood in patients with hepatobiliary malignancies significantly increases, and is related to the occurrence and development of tumor and clinical prognosis.
