Abstract:There is a delicate relationship between nutrition and reproduction of mitten crab (Eriocheir sinensis). The crabs store significant amounts of energy in hepatopancreas, which is prepared for significant energy output and expenditure during reproduction, but the internal molecular mechanism has never been known. Here we present the first relationship between hepatopancreas and testis of E. sinensis. We acquired 6287 high quality expressed sequence tags (EST), representing 3829 unigenes totally, from healthy male mitten crabs of first grade. We investigated the Gene Ontology and the main metabolism processes of hepatopancreas and testis from E. sinensis. Genes most likely expressed more frequently and localized in hepatopancreas, and abundant genes from testis for multiple functions. Many genes important for the nutrition regulation are in the EST resource, including arginine kinase, leptin receptor-like protein, seminal plasma glycoprotein 120, and many kinds of zinc finger proteins. The EST data also revealed genes such as heat shock protein 70, testis enhanced gene transcript (TEGT), Cyclin K, etc. predicted to play important roles in regulation of reproduction mechanisms. Among these genes, alignment of leptin receptor-like protein and vasa-like protein from E. sinensis and other species showed even more genomic information on E. sinensis. We identified seventeen genes relevant to control of nutrition mechanisms and eleven genes involved in regulation of reproduction. And this study provides insights into the genetic and molecular mechanisms of nutrition and reproduction in the crab. Such information would facilitate the optimization of breeding in the aquaculture of mitten crabs.

Abstract:The oriental river prawn, Macrobrachium nipponense, is an important commercial aquaculture resource in China. During breeding season, short ovary maturation cycles of female prawns cause multi-generation reunions in ponds and affect the growth of females representing individual miniaturization (known as autumn -propagation). These reproductive characteristics pose problems for in large - scale farming. To date, the molecular mechanisms of reproduction regulation of M. nipponense remain unclear. To address this issue, we performed transcriptome sequencing and gene expression analyses of eyestalk and cerebral ganglia of female M. nipponense during breeding and non-breeding seasons. Differentially expressed gene enrichment analysis results revealed several important reproduction related terms and signaling pathways, such as "photoreceptor activity", "structural constituent of cuticle" and "G-protein coupled receptor activity". The following six key genes from the transcriptome were predicted to mediate environmental factors regulating reproduction of M. nipponense: neuroparsin, neuropeptide F II, orcokinin II, crustacean cardioactive peptide, pigment-dispersing hormone 3 and tachykinin. These results will contribute to a better understanding of the molecular mechanisms of reproduction of oriental river prawns. Further detailed functional analyses of the candidate reproduction regulation related neuropeptides are needed to shed light on the mechanisms of reproduction of crustacean.

Abstract:Reproductive biology is a uniquely important topic since it is about germ cells, which are central for transmitting genetic information from generation to generation. In this review, we discuss recent advances in mammalian germ cell development, including preimplantation development, fetal germ cell development and postnatal development of oocytes and sperm. We also discuss the etiologies of female and male infertility and describe the emerging technologies for studying reproductive biology such as gene editing and single-cell technologies.

Abstract:Mechanisms of the animal reproductive development are an important research field in life sciences. The study of the reproductive development and regulatory mechanisms in fishes is important for elucidating the mechanisms of animal reproduction. This paper summarizes recent advances in the mechanisms of fish sex determination and differentiation, of fish gonad development and maturation, and of fish germ cell development, as well as the according regulating strategies. Fishes comprise an evolutionary stage that links invertebrates and higher vertebrates. They include diversiform species, and almost all vertebrate types of reproduction have been found in fishes. All these will lead to important advances in the regulatory mechanisms of animal reproduction by using fishes as model organisms. It will also enable novel fish breeding techniques when new controllable on-off strategies of reproduction and/or sex in fishes have been developed.

Abstract:Crustacean aquaculture, especially monosex culture, requires a comprehensive knowledge of the mechanisms regulating sex determination and differentiation. Decapod crustaceans display either female heterogamety (ZW/ZZ) or male heterogamety (XY/XX) as their mode of genotypic sex determination. However, some decapod species exhibit sexual plasticity via removing the androgenic gland, knockdown insulin receptor transcripts, administrating exogenous sex steroids, and altering environmental factors. Although the master sex‐determining gene in decapods remains elusive, various sexual genes, hormones, and neurotransmitter are emerging as important factors in sex differentiation. This review looks into recent advancements in studies related to the genetic sex determination system and factors that trigger sex reversal. Moreover, the elucidation of the role of genetic factors in sex differentiation in both females and males, as well as their underlying interrelated regulatory mechanisms, provides justification for three levels of application: the induction of monosex populations, detection of precocious sex and genotype, and prevention of uncontrolled reproduction. Finally, we proposed future directions of bioinformatics analysis and genome editing coupled with sex control to increase the yield for economic decapods. In conclusion, this review provides a foundation for future research on the reproductive biology and sexual regulation of decapods, with the aim of aiding practitioners in advancing the crustacean aquaculture industry.

Abstract:Background: The Chinese mitten crab (Eriocheir sinensis) is widely distributed in the coastal and coastal water of China and has important economic values. Salinity is the key environmental factor affecting the mating and spawning of E.sinensis. During the mating and spawning of E. sinensis, the hormone changes in the body are usually accompanied, but there is no report about the effects of salinity on related hormones in the process of breeding of E. sinensis. Methods: Mating salinity including 0‰, 2‰, 4‰, 6‰ and spawning salinity such as 3‰, 6‰, 9‰, 12‰, 15‰, 18‰, 21‰ were set up, three parallel groups are set for each salinity, 10 female and 5 male crabs are put in each tank. 5 samples were gathered to analyze the contents of 17α,20β-dihydroxy-4-pregnene-3-one (DHP), Gonadotropin (GTH), Prostaglandins (PG), Estradiol-17β (E2) and Testosterone (T) in the hemolymph of female E. sinensis after mating and spawning. Result: The contents of DHP, E2, PG and T in female crabs hemolymph decreased first and then added with the increase of salinity before spawning. DHP, PG and E2 added first and then declined with the increase of salinity after spawning. When salinity was 6‰, the contents of DHP, PG, E2 and T in the hemolymph of female crabs before spawning reached the highest, the contents of DHP and PG decreased and E2 and T increased after spawning. The comprehensive study indicated that salinity had a certain effect on five hormones in the hemolymph of female E.sinensis during reproduction, five hormones didn’t change significantly in the process of mating, DHP, GTH and PG were involved during the spawning of E.sinensis.

Abstract:Ovarian development is a key physiological process that holds great significance in the reproduction of the Chinese mitten crab ( Eriocheir sinensis ), which is an economically important crab species for aquaculture. However, there is limited knowledge for the regulatory mechanisms of ovarian development. To study the molecular mechanisms of its ovarian development, transcriptome analysis was performed in the ovary and hepatopancreas of E. sinensis during ovarian stages I (oogonium proliferation), II (endogenous vitellogenesis), and III (exogenous vitellogenesis). The results showed that 5,520 and 226 genes were differentially expressed in the ovary and hepatopancreas, respectively. For KEGG enrichment analysis, the differentially expressed genes in the ovary were significantly clustered in phototransduction-fly, phagosome, and ECM-receptor interaction. Significantly enriched pathways in the hepatopancreas included fatty acid biosynthesis, fatty acid metabolism, and riboflavin metabolism. Further analysis showed that 25 genes and several pathways were mainly involved in oogenesis, including the ubiquitin-proteasome pathway, cyclic AMP-protein kinase A signaling pathway, and mitogen-activated protein kinase signaling pathway. Twenty-five candidate genes involved in vitellogenesis and endocrine regulation were identified, such as vitellogenin, vitellogenin receptor, estrogen sulfotransferase, ecdysone receptor, prostaglandin reductase 1, hematopoietic prostaglandin D synthase and juvenile hormone acid O-methyltransferase. Fifty-six genes related to nutritional metabolism were identified, such as fatty acid synthase, long-chain-fatty-acid-CoA ligase 4, 1-acyl-sn-glycerol-3-phosphate acyltransferase 4, fatty acid-binding protein, and glycerol-3-phosphate acyltransferase 1. These results highlight the genes involved in ovarian development and nutrition deposition, which enhance our understanding of the regulatory pathways and physiological processes of crustacean ovarian development.

Abstract:As important aquaculture species worldwide, shrimps and crabs are thermophilic animals with a feeble thermoregulation ability. Changes in environmental factors are the main reason for the decrease in the immunity and disease resistance ability of cultured organisms. Water temperature is one of the most common abiotic stress factors for aquatic ectotherms. It influences nearly all biochemical and physiological processes in crustaceans, resulting in an imbalance in ion and water homeostasis, neuromuscular function loss, cellular dehydration, and altered metabolic pathways. The present review summarizes the current knowledge on the effects of low temperature on the physiological response, and the behavior, development, and growth of shrimp and crab. We suggest a deeper research to understand the physiological processes involved in thermoregulation; this knowledge could be used to reduce the adverse effects in the shrimps and crabs during the culture.

Reproduction Performances of Mud Crab (Scylla olivacea) Broodstocks with Different Feeds

Abstract:Broodstock maturation diets is important research in order to increase the reproduction performance of spawning females. This research was conducted to determine the appropriate feed for mud crab broodstock spawned and as information in support of efforts to provide quality stem in a controlled manner. The study was conducted at the Installation Research Marana Institute For Coastal Aquaculture (RICA). The test animals that used were 15 ind. mud crab broodstock having of size 200±15 g, carapace length 6±0,5cm, carapace width 12±0,5 cm with gonadal maturity level (TKG I) obtained from crab fishing locations. Research using completely randomized design which consists of 3 treatments and 5 replications. The treatments tested were: A (trash fish), B (squid) and C (golden apple snail). Variable measured were the rate of gonadal maturity, egg diameter, the degree of spawning, embryo incubation period, the number of larvae, hatching rate, larval amino acid and proximate analyze content and water quality variables. The results of proximate analyze showed that trash fish have the highest fat content (7.85 %) while the squid had a high protein content (73.72 %). Amino acids contained in squid is alanine, cystine, and leucine are thought to have an important role in the maturation of the gonads crab. The feed squid gives the best performing for mud crab S.olivacea broodstockin the achievement of gonadal maturity stage, fecundity and hatching rate.

Abstract:China is the major distribution center of cranes in the world. Several species of rare cranes breed and winter in （China. The study on Chinese cranes is an important part of the world crones＇ protection. Ahhough, the research of cranes protection in China wasn＇t carried out until the early 1980s, it has been developing fast in the past twenty years. In this paper, we present a review on the study of Cranes in captivity in China, with focuses on ecological studies, propagation technology, studies on cranes＇ behavior in breeding season, as well as releasing of cranes. A brief introduction to related technologies widely in use abroad is also offered.
Mating behaviour and cholesterol nutritional strategies promoted ovarian development of female swimming crab ( Portunus trituberculatus )

Abstract:Female crabs enter a stage of rapid ovarian development after mating, and cholesterol is a substrate for steroid hormone synthesis. Therefore, in this experiment, an 8-week feeding trial was conducted to investigate the effects of mating treatments (mated crab and unmated crab) and three dietary cholesterol levels (0·09 %, 0·79 % and 1·40 %) on ovarian development, cholesterol metabolism and steroid hormones metabolism of adult female swimming crab (Portunus trituberculatus). The results indicated that crabs fed the diet with 0·79 % cholesterol significantly increased gonadosomatic index (GSI) and vitellogenin (VTG) content than other treatments in the same mating status. Moreover, mated crabs had markedly increased GSI and VTG content in the ovary and hepatopancreas than unmated crabs. The histological observation found that exogenous vitellogenic oocytes appeared in the mated crabs, while previtellogenic oocytes and endogenous vitellogenic oocytes were the primary oocytes in unmated crabs. The transmission electron microscopy analysis showed that when fed diet with 0·79 % cholesterol, the unmated crabs contained more rough endoplasmic reticulum and mated crabs had higher yolk content than other treatments. Furthermore, mating treatment and dietary 0·79 % cholesterol level both promoted cholesterol deposition by up-regulation of the mRNA and protein expression levels of class B scavenger receptors 1 (Srb1), while stimulating the secretion of steroid hormones by up-regulation of the mRNA and protein expression of steroidogenic acute regulatory protein (Star). Overall, the present results indicated that mating behaviour plays a leading role in promoting ovarian development, and dietary 0·79 % cholesterol level can further promote ovarian development after mating.

Reproductive Ability Disparity in the Pacific Whiteleg Shrimp (Penaeus vannamei): Insights from Ovarian Cellular and Molecular Levels

Abstract:The Pacific whiteleg shrimp (Penaeus vannamei) is a highly significant species in shrimp aquaculture. In the production of shrimp larvae, noticeable variations in the reproductive capacity among female individuals have been observed. Some females experience slow gonadal development, resulting in the inability to spawn, while others undergo multiple maturations and contribute to the majority of larval supply. Despite numerous studies that have been conducted on the regulatory mechanisms of ovarian development in shrimp, the factors contributing to the differences in reproductive capacity among females remain unclear. To elucidate the underlying mechanisms, this study examined the differences in the ovarian characteristics between high and low reproductive bulks at different maturity stages, focusing on the cellular and molecular levels. Transmission electron microscopy analysis revealed that the abundance of the endoplasmic reticulum, ribosomes, mitochondria, and mitochondrial cristae in oocytes of high reproductive bulk was significantly higher than that of the low reproductive bulk in the early stages of ovarian maturation (stages I and II). As the ovaries progressed to late-stage maturation (stages III and IV), differences in the internal structures of oocytes between females with different reproductive capacities gradually diminished. Transcriptome analysis identified differentially expressed genes (DEGs) related to the mitochondria between two groups, suggesting that energy production processes might play a crucial role in the observed variations in ovary development. The expression levels of the ETS homology factor (EHF) and PRDI-BF1 and RIZ homology domain containing 9 (PRDM9), which were significantly different between the two groups, were compared using qRT-PCR in individuals at different stages of ovarian maturation. The results showed a significantly higher expression of the EHF gene in the ovaries of high reproductive bulk at the II and IV maturity stages compared to the low reproductive bulk, while almost no expression was detected in the eyestalk tissue of the high reproductive bulk. The PRDM9 gene was exclusively expressed in ovarian tissue, with significantly higher expression in the ovaries of the high reproductive bulk at the four maturity stages compared to the low reproductive bulk. Fluorescence in situ hybridization further compared the expression patterns of EHF and PRDM9 in the ovaries of individuals with different fertility levels, with both genes showing stronger positive signals in the high reproductive bulk at the four ovarian stages. These findings not only contribute to our understanding of the regulatory mechanisms involved in shrimp ovarian development, but also provide valuable insights for the cultivation of new varieties aimed at improving shrimp fecundity.

Effects of dietary phospholipid and highly unsaturated fatty acid on the gonadal development, tissue proximate composition, lipid class and fatty acid composition of precocious Chinese mitten crab, Eriocheir sinensis

Abstract:Precocious puberty is a major constraint to the economical rearing Eriocheir sinensis farming. Although dietary phospholipid (PL) and highly unsaturated fatty acids (HUFA) reportedly enhanced ovarian development in normal adult E. sinensis, it had opposite effects of reducing precocity in juveniles. This study investigated the effects of dietary PL and HUFA on survival, gonadal development and biochemical composition of precocious E. sinensis. Two diets with PL and HUFA supplementation (diet A) and deficient (diet B) were formulated and fed to precocious E. sinensis. Although no significant differences were found on survival and gonadosomatic index of crabs fed both diets, crabs fed diet B had significantly higher hepatosomatic index and hepatopancrean lipid content than crabs fed diet A. The percentages, 14:0, 18:1n-9, 20:5n-3 and 22:6n-3, in hepatopancreas, muscle and gonads were positively correlated to the diets. Interestingly, the crabs fed diet B accumulated significantly higher 18:2n-6 in gonads and muscle than those fed diet A. Furthermore, substantially higher 20:4n-6 was detected in the gonad and muscle than in the diets. These results suggested that dietary PL and HUFA had limited effects on male gonad biochemical composition. Meanwhile, the precocious E. sinensis may be capable of negating the negative effects of HUFA deficiency by enhanced tissue 18:2n-6 accumulation and converting 18:2n-6 to 20:4n-6.

Abstract:The swimming crab Portunus trituberculatus is one of the most important economic species in China and its mature ovary often determines its commercial value and production. Although the ovary maturation of crustaceans is generally affected by exogenous nutrition, the specific nutritional needs of ovary maturation of P. trituberculatus are poorly understood. To this end, we collected the P. trituberculatus samples with five ovarian maturation stages and measured their biochemical composition of the ovary, hepatopancreas, and muscle at each ovarian developmental stage. We further analyzed their relation to the ovarian developmental stage of P. trituberculatus by principal components analysis (PCA). We found the levels of branched-chain amino acids, long-chain polyunsaturated fatty acids (LC-PUFA), and monounsaturated fatty acids (MUFAs) in the ovary and hepatopancreas increased during the ovary maturation process, and also passively correlated with ovarian developmental stage, which highlights the necessity of these specific nutrients for oogenesis and for improving the nutrient quality of crabs. In addition, we found an increasing tendency of carotenoid content and phosphatidylcholine in phospholipid in the ovary from the pre-developmental stage to the proliferative stage, but not in the hepatopancreas and muscle, which highlights the possible involvement of carotenoids during the rapid oocyte development process. Our study may provide valuable information for developing a suitable broodstock diet that promotes the ovarian maturation of adult P. trituberculatus and ensures high-quality larval production.

Abstract:As a well-known example of “living fossil,” horseshoe crabs are ecologically significant macroinvertebrates in coastal and estuarine ecosystems. The tri-spine horseshoe crab, Tachypleus tridentatus , has been widely utilized for Tachypleus amebocyte lysate production and food consumption since the 1980s, which led to considerable population declines along the west coast of the Pacific Ocean. The declining horseshoe crab population is expected to have ecological and social impacts. Stock enhancement through captive rearing of juveniles is cited as an important alternative to repopulate the native T. tridentatus , which in turn supports sustainable resource utilization and research activities. The hatchery production techniques for this species have gradually developed following the mass culture efforts in Japan since the late 1980s. However, the previous studies have primarily concerned the feed types and husbandry conditions to maximize the growth and survival of the juveniles. Little is known about the practicability and effectiveness of releasing large numbers of hatchery-bred individuals through releasing programs. In this review, we (1) summarize the available captive breeding and rearing techniques, (2) discuss the release strategies that could potentially improve the survival of released juveniles, and (3) identify the future opportunities and challenges in establishing technical frameworks to support responsible stock enhancement programs for T. tridentatus . The information should benefit future horseshoe crab fisheries management efforts in the attempt to restore the severely depleted populations.

Comparative Transcriptome Analysis Reveals Related Regulatory Mechanisms of Androgenic Gland in Eriocheir sinensis

Abstract:Chinese mitten crab (Eriocheir sinensis) is one of the most commercially important aquaculture species in China. The androgenic gland (AG) of crustaceans plays pivotal roles in the regulation of male differentiation and in maintaining the male sexual characteristics. In order to reveal related mechanisms in AG, we compared transcriptomes of AG between proliferation and secretion phase. A total of 72,000 unigenes and 4,027 differentially expressed genes were obtained. Gene ontology enrichment analysis indicated that biological processes and metabolic pathways related to protein synthesis and secretion such as transcription, translation, and signal transduction were significantly enriched. Critical genes such as IAG, SXL, TRA-2, SRY, FTZ-F1, FOXL2 , and FEM-1 were identified and potentially involved in maintaining the testis development and spermatogenesis. Ribosomes pathway revealed the cause of insulin-like androgenic gland hormone secretion increase. Three insulin-like receptors were thought to be associated with growth and spermatogenesis. In the neuroactive ligand-receptor interaction pathway, the expression of octopamine receptor, 5-HT receptor 1, and melatonin receptor was significantly changed, which revealed the key regulation mechanism of aggressive and mating behavior of males. Comparative transcriptome analysis provided new insights into the genome-wide molecular mechanisms of AG development and the regulatory mechanisms of male development.

Abstract:Crabs thrive in diverse ecosystems, from coral reefs to hydrothermal vents and terrestrial habitats. Here, we report a comprehensive genomic analysis of the mud crab using ultralong sequencing technologies, achieving a high-quality chromosome-level assembly. The refined 1.21 Gb genome, with an impressive contig N50 of 11.45 Mb, offers a valuable genomic resource. Gene family analysis shows expansion in development-related pathways and contraction in metabolic pathways, indicating niche adaptations. Notably, Investigation into Hox gene regulation sheds light on their role in pleopod development, with the Abd-A gene identified as a linchpin. Posttranscriptional regulation involving novel-miR1317 negatively regulates Abd-A levels. Furthermore, the fru gene's potential role in ovarian development and the identification of novel-miRNA-35 as a regulator of Spfru2 add complexity to gene regulatory networks. Comparative functional analysis across Decapoda species reveals neofunctionalization of the elovl6 gene in the synthesis of long-chain polyunsaturated fatty acids (LC-PUFA), suggesting its importance in environmental adaptation. These findings contribute significantly to our understanding of crab adaptability and evolutionary dynamics, offering a robust foundation for future investigations.
