Formation of Type Ⅰ Collagen in Nude Mice Through Osteo-inductive Culture of Cryopreserved Bone Marrow Stromal Cells

Abstract:Objective To explore the effects of osteo-inductive culture on the synthesis of type Ⅰ collagen by bone marrow stromal cells(BMSCs) in nude mice.Methods BMSCs from beagle dogs were harvested and cryopreserved for 12 months.Recovered BMSCs were combined with collagen scaffold in vitro in osteo-inductive or non-osteo-inductive culture medium.Osteo-inductive conditioned collagen scaffold alone,osteo-induced and non-induced BMSCs-scaffold complex were respectively implanted into the subcutaneous tissues of nude mice for 4 weeks.Type Ⅰ collagen formation was evaluated by H&E staining and immunohistochemistry(IHC).Results Scarce collagen fibers were observed in the group implanted with collagen scaffold alone,moderate collagen fibers in non-induced BMSCs-scaffold group,and large quantities of newly formed collagen fibers in osteo-induced BMSCs-scaffold group.Enhanced formation of type Ⅰ collagen was observed in osteo-induced BMSCs-scaffold group(0.1963±0.0126),with lesser formation in non-induced BMSCs-scaffold group(0.1489±0.0037) and scaffold alone group(0.0775±0.0058)(P<0.05).Conclusion Osteo-inductive culture can enhance type Ⅰ collagen formation in nude mice by cryopreserved BMSCs.
