The Metabolic Flux Analysis of Candida Krusei at Osmotic Stress

Abstract:Metabolic flux analysis was used to study the cell physiology of Candida krusei at osmotic stress. Glycerol and trehalose are the most prominent compatible solutes in Candida krusei cell. Under osmotic condition (NaCl as the osmoticum), the glycerol concentration is doubled and the trehalose content increased 7% than that of the control ones. In contrast, the extracellular pyruvate production decreases nearly half and the CO2 export declines correspondingly. The metabolic flux analysis indicates that the carbon flux flowed toward trehalose synthesis from glucose-6-phosphate node increases 1.6-fold under osmotic condition. At the same time, the carbon flux flowed toward glycerol-3-phosphate (G3P) to form glycerol from dihydroxyacetone phosphate (DHAP) and glyceraldehydes-3-phosphate (GAP) node increases 67.2%, while the carbon flux flowed toward TCA cycle decreases 26.9%. Energy metabolic flux analysis also suggests that the metabolism of Candida krusei under hyperosmotic condition is more efficient for the glycerol and trehalose production. The carbon flux flowed toward NADH anabolism is one third compared to that of the normal cultivation.
