Development and Validation of RP-HPLC Method for Quantification of Resveratrol in a Resveratrol-Solid Lipid Nanoparticles Loaded Microneedles Patch

Abstract:The objective of this study was to design and subsequently validate a precise and distinctive reversed-phase high-performance liquid chromatography (RP-HPLC) method for the accurate measurement of resveratrol (RSV), a polyphenol compound in the nanoformulations loaded into a microneedles (MNs) patch. The method was developed, optimized, and validated following International Council for Harmonisation guidelines (ICH) Q2 (R1), utilizing a reverse phase C18 column and a straightforward mobile phase. Samples were analyzed at ambient temperature, employing a flow rate of 1 mL/min and detecting wavelengths at 306 nm. The optimized HPLC method showcased remarkable linearity, selectivity, and precision. Specifically, it displayed exceptional linearity (R 2 = 1.0) within a range of 4–20 μg/mL, with corresponding limits of detection (LOD) and quantification (LOQ) at 0.014 and 0.043 μg/mL, respectively. The methodology showed robustness with satisfactory intra- and interday precision. The developed stability-indicating HPLC method was applied to analyze RSV content and assess its loading in nanoformulations, and was proven to be selective, accurate, sensitive, reliable and consistent with regulatory guidelines. The RP-HPLC approach proved successful in assessing the drug content of RSV in novel formulations.
