Abstract:OBJECTIVE To improve the dissolution rate of empagliflozin by hot-melt granulation technology. METHODS The compositions of empagliflozin were prepared by hot-melt granulation, with microcrystalline cellulose (MCC) as filler, the ratio empagliflozin to MCC in composition 1, 2 and 3 was 1∶2,1∶3 and 1∶4, respectively . Appearances were observed by microscope, the physical states of compositions were investigated by differential scanning calorimetry (DSC) and X-ray powder diffraction analysis (XRPD). The dissolution rate of empagliflozin with different particle sizes and different compositions were also compared. RESULTS The DSC and XRPD spectrum of the compositions showed that empagliflozin existed in amorphous form and crystal form. The dissolution rate of compositions have been improved by hot-melt granulation. The dissolution rate of composition 2 and 3 were similar to that of micronized empagliflozin. CONCLUSION Dissolution rate of empagliflozin has been improved by hot-melt granulation.
Enhancing Stability and Formulation Capability of Fungicides by Cocrystallization Through a Novel Multistep Slurry Conversion Process

Abstract:Solid agrochemicals with poor thermal stability pose a huge challenge to their production and application. Herein, two low-melting-point fungicides, pyraclostrobin (ZJ) and flusilazole (FZ), were designed to form cocrystals with the high melting point fungicide of thiophanate-methyl (TM), respectively. An infrequent multistep slurry conversion crystallization process was discovered and studied, which accelerated the formation of cocrystals, and the mechanism was revealed. Oiling-out of FZ and ZJ was the key process to multistep slurry conversion crystallization. The heterogeneous system possessed the advantage of high energy, which accelerated the synthesis of cocrystals. Furthermore, the variations of physicochemical properties containing thermal stability, tap density, formulation ability, and storage stability of both the parent fungicides and cocrystals were analyzed and compared. Packing coefficient and crystal structure analysis revealed the structure-property relationships of cocrystals. The single-crystal structure showed the strong N-H center dot center dot center dot N hydrogen bonds between the FZ and TM, and N-H center dot center dot center dot O and N-H center dot center dot center dot S interactions in the ZJ-TM cocrystal. Strong hydrogen bonds stabilize the structures of cocrystals, which enables ZJ-TM and FZ-TM to be made into a greener water suspension concentrate, instead of an emulsifiable concentrate with pollution.

Abstract:Objective. To optimize the preparation formula and technology of Chuilian Jianpi granules. Methods. The formulation and preparation process were optimized by single factor experiment and response surface analysis, with the molding rate, hygroscopicity, and fluidity of particles as the comprehensive evaluation indexes, and the comprehensive score as the response value according to different weights. To further evaluate the stability and feasibility of the preparation formulation and technology, the physical fingerprint of the granules was constructed by seven indicators including particle tap density, bulk density, hygroscopicity, water content, angle of repose, Hausner ratio, and relative homogeneity index. Results. The optimum formula conditions of Chuilian Jianpi granules were as follows: the dosage ratio of drug to adjuvant was 1 : 0.8, lactose:mannitol = 1.5 : 1, and the amount of wetting agent (90% ethanol) was 25% of the granules, with high-speed stirring granulation. The similarity of the physical fingerprints of the 10 batches of Chuilian Jianpi granules was high, which is above 98.5%. Conclusion. The optimized preparation technology was stable and feasible, which can provide reference for the development of Chuilian Jianpi granules and other Chinese medicine granules.

Preparation and Characterization of N-succinyl-chitosan

Abstract:Poisonous solvents were widely used in the preparation of N-succinyl-chitosan,which were easy to cause environmental pollution and difficult to remove completely from the products.These disadvantages limited their uses in biomedical materials.In the present work,a novel method was proposed to successfully synthesize N-succinyl-chitosan,which was safe and environmentally friendly.A series of N-succinyl-chitosan with different degrees of substitution were prepared in absolute ethanol by controlling the reaction time.The different degrees of substitution,molecular weight and polydispersity index of the products were measured.The products were characterized by Fourier transform infrared(FTIR) spectroscopy,X-ray diffractometry(XRD) and thermogravimetry(TG).Furthermore,the water-solubility and biocompatibility were studied.When the reaction time is 15 h,the degree of substitution and molecular weight of the product are highest.Moreover,polydispersity index of the product is smallest among all the products(10 h,12 h,15 h,18 h,20 h).In addition,the results indicate N-succinyl-chitosan has been synthesized with decreased degree of crystallization and thermostability.Water-solubility of modified chitosan is improved due to the decrease of crystallization.N-succinyl-chitosan prepared in absolute ethanol has good biocompatibility,which has great potential in biomedical fields.
Preparation and Characterization of Body Granules for Direct Manufacturing of Longquan Celadon

Abstract:The celadon body granules were prepared by combining aqueous slurry with lfuidization spray drying for direct manufacturing of Longquan celadon. The dispersibility and stability and spray drying behavior of aqueous slurry were studied, and the effect of solid content on the rheological behaviour of slurry and particle size distribution and molding properties of granulated powder pellets were also discussed. The results show that the slurry is Bingham lfow behaviour, the slurry with solid content of 40 wt.%has a lower viscosity and shear stress, while the slurry with solids content of 50 wt.%possesses good stability and dispersibility. After spray drying of slurry with solids content of 50 wt.%, the granules show regular spherical shape, wide particle size distribution, and good lfowability, compaction and molding properties.
Study on Preparation of Shengmaiyin of Traditional Chinese Medicine Granules Using Freeze-Drying and High Shear Mixing Technique

Abstract:To improve the traditional preparation of Chinese medicine Shengmaiyin for patient to take medicine conveniently,the liquid extract of Shengmaiyn as a model was dried by freezing-drier to form extract dryness.The extract dryness was formulated with micronization silica gel in different ratios and granulated with high shear mixer granulator.It was feasible that the extract dryness was mixed with excipients including silica gel and micron silica gel in ratio of 3 to 1 with 4.0 mL of 10%HPMC of 80% ethanol solution.The process of granulation was going smoothly,the uniform granules was obtained,the moisture absorption was lower.The freezing-drying and high shear mixing techniques could be used to prepare the granules of the traditional Chinese medicine for lowering the moisture absorption of extract dryness and improving the stability of granules.

Abstract:Objective To determine the molding process of Qizhi Yifei granules, and establish the quality standard of Qizhi Yifei granules. Methods The flowability of extract powder and granule were de-termined by resting angle meter respectively. The percentage of moisture absorption, particle formation rate, shape of soft wood and degree of difficulty of granulation were taken as the indexes to determine the excipients, the ratio of drugs and additives and the concentration of wetting agents. The solubility, bulk density and critical relative humidity of the particles were determined respectively. Qualitative identification of Radix Astragali seu Hedysari, alcohol processed Rhizoma Polygonati, processed Hirudo, Rhizoma Pinelliae Preparata, Bulbus Fritillariae Thunbergii, Radix Angelicae Sinensis, Exocarpium Citri Grandis and Radix Peucedani in the granules were performed by thin layer chromatography ( TLC). The high performance liquid chromatography-evaporative light scattering detector method (HPLC-ELSD) was used to determine the contents of astragaloside IV in Qizhi Yifei granules. Results The flowability of extract powder was improved by granulation, and the best excipient was dextrin, with the best ratio of extract to powder ∶ dextrin ( 3 ∶ 2 ). The wetting agent was 90% ethanol. The TLC spots were clear without interference of the negative control. The linear range of astragaloside IV was 0. 02502 ~0. 70056 mg/mL (r=0. 9994), with the average recovery of 96. 76% and RSD 0. 52%. Conclusion The molding method has high molding rate, good melting property and critical relative humidity. The method is accurate, reliable, specific and reproducible, and can be used for the quality control of Qizhi Yifei granules.

Abstract:Objective To optimize the preparation technology of Xiasangju Effervescent Granules.Methods The aqueous extraction and alcohol precipitation were adopted for preparing Xiasangju Effervescent Granules.Polyethylene glycol was used to coat sodium bicarbonate as the effervescent agent and the orthogonal design was adopted to optimize the use amount of citric acid and sodium bicarbonate.To judge whether to adopt polyethylene glycol for coating.The content of ursolic acid in Xiasangju Effervescent Granules and the effervescent time were used as the evaluation indicators.The 3-month stability test was performed.Results The optimum conditions for Xiasangju effervescent granules were as follows:using 11%citric acid,8%sodium bicarbonate as the effervescent agent and 5%polyethylene glycol for coating sodium bicarbonate to produce Xiasangju Effervescent Granules.Conclusion This technological process is simple with stable quality and suitable for the industrial production.
Development of Self Nano-dispersible Granules and Tablet Using Dry Nanosuspension Technique for Poorly Water-soluble Drugs

Abstract:The “dry nanosuspension” granules and tablet, which could be spontaneously dispersed to submicrom-sized drug particles, were developed using wet milling and sequential fluidized-bed drying technique. Poorly water-soluble drug particles were beads-milled in the aqueous solution of dispersing agents using simple and general-purpose apparatus in laboratory. The milled nanosuspension was sprayed to the carrier particles of lactose in the fluidized-bed granulator to obtain the granulated product. It was found that the polymeric agent loaded as a milling promotor could play a role of binder to support the granulation and that the nano-sized drug particles were uniformly fixed on the surface of granulated particles. The resultant granules could re-construct the original drug nanosuspension within a few minutes in water, resulting in rapid dissolution behavior. Further, it was also found that the tablet prepared by compressing the granules kept their own rapid dispersible and release properties. The present technique via wet milling and fluidized-bed granulation/drying processes would be a novel dissolution-enhanced technology for poorly water-soluble drugs.

Abstract:OBJECTIVE: To study the preparation technique of tanshinone dispersible tablets.METHODS: Taking the disintegration time - limit, in vitro dissolubility and suspensibility as indices, the formula of tanshinone dispersible tablets was screened by orthogonal design.RESULTS The dispersible tablets could completely disintegrate within 30 seconds and pass through 710m seive mesh, which all conformed to the requiremtes of BP(1993) .The in vitro dissolubility of this product was superior to that of ordinary tablets obviously .CONCLUSION: The preparation technique of tanshinone dispersible tablets is mature and the quality is reliable.

Abstract:Objective:To establish the method for improving quality standard of Zhisoudingchuan Pills.Methods:Ephedrine Hydrochloride in pills was determined by RP-HPLC,TLC was used to identify Bitter Apricot Seed,chemical reaction is used to identify gypsum;。The samples were separated by Allsphere ODS-2.5 μm(4.6×250 mm)with the mobile phase consisting of acetonitrile-water(0.1% phosphoric acid and 0.1% triethylamine)(3:97);The detection wavelength was at 207 nm.Results:For Ephedrine Hydrochloride,linear range was 0.2246～1.1240μg,and the average recovery was 98.03%.Bitter Apricot Seed in Zhisoudingchuan Pills could be identified by TLC,The reaction to identify gypsum is evident.Conclusion:The method to determine content of Ephedrine is precise and the operation of extraction is simple、The specificity of this method is reliable,The specificity to identify Bitter Apricot Seed using TLC is good,The reaction to identify gypsum is acurate,simple,specialized,reproducible and suitable for quality control of Zhisoudingchuan Pills.
Melt granulation using a twin-screw extruder: a case study

Abstract:The purpose of this study was to use a twin-screw extruder for melt granulation. Polyethylene glycols (PEG 400 and 4000) were used as binders for the development of a drinking water formulation with immediate drug release. The effect of drug content, PEG 400/4000-ratio, surfactant (type and concentration) and granulation temperature on granule properties and dissolution characteristics was determined. The granulation temperature had an important influence on the granule formation. High yield (95% of the granules <1400 microm) was obtained only at a temperature near the melting point of PEG 4000. During granulation the drug of BCS class II was finely dispersed in the PEGs, creating a micro-environment around the drug particles enhancing the dissolution rate. To obtain complete drug release within 10 min for a formulation containing 10% drug, the addition of 2% (w/w) surfactant (polysorbate 80 or Cremophor RH40) was required. At a higher drug content (20%), the PEG 4000 concentration had to be increased to 20% to improve granule properties and 4% polysorbate 80 was required to obtain 100% drug release. X-ray diffractograms showed distinct peaks of crystalline drug, the crystallinity of the drug did not change after 50 days, independent of the storage conditions.
[Preparation and Evaluation of Enteric-Coated and Taste Masking Clarithromycin Granules].

Abstract:The study is to prepare taste masking and enteric-coated clarithromycin granules by melting and fluid bed coating technology. Clarithromycin and matrix materials were melted at a certain temperature, and then made into particles by fluidized bed coating. X-ray powder diffraction and scanning electron microscopy were used to identify the crystal and morphology of drug loading granules. In vitro dissolution method was used for the observation of the drug release behavior. The results showed that the drug particles size range was 0.2 - 0.6 mm; the crystal form of clarithromycin in the granule did not change; enteric-coated granules accumulated release in 0.1 mol L(-1) hydrochloric acid in 2 h was less than 10%, while in pH 6.8 phosphate buffer in 1 h was more than 80%. The taste masking and enteric-coated clarithromycin granules not only have good taste masking effect, but also have a good release behavior. It is expected to have better clinical application.

Abstract:Drug solubilization studies are continuously being conducted. Febuxostat (FBX) has a low solubility in water. This study aims to develop a stable FBX-solid dispersion (SD) formulation using a solvent evaporation method. The solubilization strategy of FBX is to develope an optimal FBX-SD formulation by selecting a solubilizer and carrier through the screening method. The final selected solubilizer, macrogol 15 hydroxystearate and polyoxyl 15 hydroxystearate (Kolliphor® HS-15), is widely used in the pharmaceutical industry as a nonionic solubilizing and emulsifying agent and has low toxicity. Especially when commonly used in developing lipophilic drug formulations, it dissolves well in water and ethyl alcohol. The optimal composition ratio of the formulation (SD4) was FBX:HS-15®:granular dicalcium phosphate dehydrate (DCP-D): A synthetic magnesium aluminometasilicate (Neusilin®UFL2):chitosan = 1:3:3:1:1 (w/w) and showed 3.0-, 2.3-, and 1.1-fold higher dissolution (%) of FBX compared to that of the Feburic tab® in pH 1.2 media, distilled water (DW), and pH 6.8 buffer, respectively. Also, in vitro release and in vitro permeability in SD4 formulation showed higher than that of Feburic tab®. Based on its stability over 6 months, it was confirmed that chitosan acted as a stabilizer. Moreover, due to weak intermolecular interactions, FBX in the SD4 formulation was considered to exist in a mixed state of amorphous and crystalline FBX. In conclusion, the improved dissolution (%) and stability of FBX in SD4 formulation were secured through the synergistic effect of excipients.

Abstract:SiC suspended slurry was prepared with organic additives and with deionized water as liquid media.The dispersibility mechanism and spray drying behavior of aqueous slurry were studied.The effects of the organic additives and solid content on the rheological behavior of slurry and the particle size distribution and molding properties of granulated powder pellets were also dis-cussed.The results show that the aqueous SiC slurry has well dispersed behavior and is stable at pH≥10.The slurry exhibited non-Newton flow with slight shear thinning rheology,and the viscosity of aqueous suspensions was very low when the organic addi-tives of 0.2%(mass fraction,the same below) tetramethylammonium hydroxide,2.0% polyvinyl alcohol and 1.0% polyethylene gly-col were added.After spray drying of the slurry with solid content of 50%,the flowability,compaction mechanism and molding properties of granulated powder pellets were obviously improved,while spherical pellets in the green body could be completely broken at a high molding pressure.
Synthesis and Performance Evaluation of a Novel Nanoparticle Coupling Expanded Granule Plugging Agent

Abstract:This study focuses on the characteristics of fractured and vuggy high-temperature and high-salt reservoirs in the Tahe Oilfield. The Acrylamide/2-acrylamide-2-methylpropanesulfonic copolymer salt was selected as a polymer; the hydroquinone and hexamethylene tetramine was selected as the crosslinking agent with a ratio of 1:1; the nanoparticle SiO2 was selected, and its dosage was optimized to 0.3%; Additionally, a novel nanoparticle coupling polymer gel was independently synthesized. The surface of the gel was a three-dimensional network structure, with grids arranged in pieces and interlaced with each other, and the structure was very stable. The SiO2 nanoparticles were attached to the gel skeleton, forming effective coupling and enhancing the strength of the gel skeleton. To solve the problem of complex gel preparation and transportation, the novel gel is compressed, pelletized, and dried into expanded particles through industrial granulation, and the disadvantage of the rapid expansion of expanded particles is optimized through physical film coating treatment. Finally, a novel nanoparticle coupling expanded granule plugging agent was developed. Evaluation of the performance of the novel nanoparticle coupling expanded granule plugging agent. With an increase in temperature and mineralization, the expansion multiplier of granules decreases; aged under high-temperature and high-salt conditions for 30 days, the expansion multiplier of granules can still reach 3.5 times, the toughness index is 1.61, and the long-term stability of the granules can be good; the water plugging rate of granules is 97.84%, which is superior to other widely used particle-based plugging agents.

Abstract:<p>The quality control of drug products during manufacturing processes is important, particularly the presence of different polymorphic forms in active pharmaceutical ingredients (API) during production, which could affect the performance of the formulated products. In this study, Fexofenadine Hydrochloride (FXD) Form I (anhydrous) was used as the starting material for pharmaceutical formulation, while FXD exhibited a polymorphic conversion to hydrate Form II in presence of water. It is necessary to monitor phase transition from Form I to Form II during wet granulation. The aim of this paper was to quantify the polymorphic forms of FXD during wet granulation process by NIR and DSC technique. Calibration models were developed and validated in binary mixtures of FXD polymorphs forms and multicomponent mixtures of FXD formulation. The NIR and DSC can be successfully employed to evaluate and quantify Form I during wet granulation. The Form I % determined was similar by NIR and DSC. The relationship between phase transition of FXD and water content was determined. In addition, the effect of various excipients on the phase transition of FXD was also evaluated by NIR. The understanding of the phase transition in wet granulation process of pure FXD, formulation and FXD-excipient mixtures provided directions for optimization of FXD manufacturing processes.</p>

Abstract:Chitosan (CS)/cellulose (BC) blend films were successfully prepared using ZnCl2•3H2O as a solvent. Homogeneous structures without obvious phase separation between CS and BC for all blend films were observed by scanning electron microscope (SEM) analysis. The tensile strengths of CS/BC blend films decreased with the increase of chitosan content. The results of X-ray diffraction (XRD) analysis indicated that the crystal structures of BC and CS were disrupted during the processes of dissolving and regeneration. Also, the reformation of hydrogen bonds between CS and BC during dissolution and regeneration processes resulted in the shift of diffraction peaks. Fourier transforms infrared spectroscopy (FT-IR) and thermogravimetric analysis (TGA) analysis results confirmed this observation. Moreover, obvious antimicrobial capability of CS/BC blend films against E. coli has been observed, indicating that antibacterial activity of chitosan has not been significantly inactivated while using ZnCl2•3H2O as a solvent. Therefore, ZnCl2•3H2O can be regarded as a promising solvent to prepare degradable films with antibacterial properties.

Homogeneous structures without obvious phase separation between CS and BC for all blend films were observed by scanning electron microscope (SEM) analysis. The tensile strengths of CS/BC blend films decreased with the increase of chitosan content. The results of X-ray diffraction (XRD) analysis indicated that the crystal structures of BC and CS were disrupted during the processes of dissolving and regeneration. Also, the reformation of hydrogen bonds between CS and BC during dissolution and regeneration processes resulted in the shift of diffraction peaks. Fourier transforms infrared spectroscopy (FT-IR) and thermogravimetric analysis (TGA) analysis results confirmed this observation. Moreover, obvious antimicrobial capability of CS/BC blend films against E. coli has been observed, indicating that antibacterial activity of chitosan has not been significantly inactivated while using ZnCl2 center dot 3H(2)O as a solvent. Therefore, ZnCl2 center dot 3H(2)O can be regarded as a promising solvent to prepare degradable films with antibacterial properties.
