Expression of MMP-1,MMP-2,uPA in oral squamous cell carcinoma

Abstract:Objective: To reveal the relation between tumor biologic behavior of oral squamous cell carcinoma (OSCC) and the expression of MMP-1, MMP-2 and uPA. Methods:The expression of MMP-1, MMP-2, uPA were analyzed in 80 cases of OSCC and epithelium immediately adjacent to carcinoma (EAC) by immunohistochemistry method (universal two-step method). Results:The expressions of MMP-1, MMP-2, uPA in OSCC were significantly higher than that in EAC(P<0.05). The expression of MMP-1 in moderate and low grade carcinoma was significantly higher than that in high grade carcinoma(P<0.05),that in OSCC was higher than that in EAC(P< 0.05 ); the expressions of MMP-2 and uPA in high, moderate and low grade carcinoma were not significantly different(P>0.05),but were higher in OSCC than in EAC(P<0.05). The expression of MMP-1 in OSCC was positively correlated with the growth mood, grade, clinical stage, lymph node status(r= 0.363 ,0.375,0.479, 0.506 ) and not with the growth locus and T stage; the expression of MMP-2 in OSCC was positively correlated with T stage, clinical stage, lymph node status(r=0.300,0.552,0.626) and not with the growth mood, growth locus and pathological grade; the expression of uPA in OSCC was positively correlated with the T stage, growth mood, clinical stage, lymph node status(r = 0.292 ,0.279,0.525,0.524) and not with the growth locus and pathological grade.Conclusion:The expressions of MMP-1, MMP-2, uPA in OSCC might be closely related with the invasion and lymph metastasis. MMP-1 is a risk factor of neck lymph node metastasis.
