Abstract:A new indole alkaloidal glucoside together with three known compounds aurantiamide acetate (2), eleutheroside E (3) and 1-O-caffeoyl-β-D-glucopyranoside (4) has been isolated from ethanol extract of the aerial parts of Clematis terniflora DC. On the basis of their spectroscopic and chemical evidence, the new compound was elucidated as (6-O-β-D-glucopyranosyl-1H-indol-3-yl) carboxylic acid methyl ester (1). Compounds 1 and 3 showed significant cytotoxicity against human ECA-109.

Abstract:Two new monoterpenoid indole alkaloids, named taberibogines E and F (1 and 2), together with three known ones (3-5) were isolated from the stems of Tabernaemontana bovina Lour (Apocynaceae). Their structures including absolute configurations were elucidated from a combination of NMR and HRESIMS data and NMR calculations as well as DP4+ probability analyses. Compounds 1 and 2 exhibited inhibitory effects on LPS-induced nitric oxide (NO) production in RAW 264.7 macrophages.

Abstract:Three new indole alkaloids, named talatensindoids A‐C (1‐3), together with two known biogenetically related indole alkaloids tryptamine (4) and L‐tryptophan (5) were isolated from the Talaromyces assiutensis JTY2 based on the guidance of OSMAC approach. The structures of these indole alkaloids were determined by comprehensive spectroscopic analyses. The absolute configuration of 3 was confirmed by X‐ray crystallographic analysis. Compound 1 represent the rare example of a chlorine‐substituted indole alkaloid from natural products. The inhibitory activity of compounds 1‐5 against two phytopathogenic fungi and three phytopathogenic bacteria was evaluated. Compound 1 exhibited broad spectrum antibacterial activities.

Abstract:Three new indole alkaloids, N(4)-demethyl-12-methoxyalstogustine (1), 17-carboxyl-N(4)-methylechitamidine chloride (2), and 17-carboxyl-12-methoxy-N(4)-methylechitamidine chloride (3), along with 15 known alkaloids, were isolated from the ethanolic extract of the stem bark of Winchia calophylla. The structures of 1-3 were elucidated on the basis of spectroscopic means and chemical methods. The determination of relative configurations at C-19 and C-20 for 1-3 was aided by 13C NMR spectroscopic data. The absolute configurations of alkaloids 1-3 were determined by direct comparison of their CD spectra with those of known alkaloids. All the alkaloids were tested in cytotoxic assays against P-388 and A-549 tumor cell lines, and only two of them showed weak activity against the A-549 cell line.

Abstract:Three novel monoterpenoid indole alkaloids gardflorine A (1), gardflorine B (2), and gardflorine C (3) were isolated from the leaves of Gardneria multiflora. Their structures, including absolute configurations, were established on the basis of spectroscopic methods (MS, UV, IR, 1D and 2D NMR) and circular dichroism experiments. All the compounds were evaluated for their vasorelaxant and acetylcholinesterase (AChE) inhibitory activities. Compound 1 exhibited potent vasorelaxant activity, with an EC50 value of 8.7 μM, and compounds 2 and 3 showed moderate acetylcholinesterase (AChE) inhibitory activities, with IC50 values of 26.8 and 29.2 μM, respectively.

Abstract:Two new monoterpenoid indole alkaloids, melaxillines A (1) and B (2), together with four known alkaloids (3–6), were isolated from the roots of Melodinus axillaris. The structures were determined by extensive spectroscopic methods, including 1D and 2D-NMR spectroscopy, high resolution mass spectrometric data, and by comparison to literature data. New compounds 1–2 might be derived from a common precursor (3) by proposing their biogenetic route, and compound 1 represents the first Melodinus alkaloid featuring an ethyl acetate group skeleton at C-18. The anti-inflammatory effects of all the isolated compounds were evaluated by measuring the inhibitory ratios of β-glucuronidase release in rat polymorphonuclear leukocytes (PMNs) induced by platelet-activating factor. Compounds 1–4 showed significant anti-inflammatory activities with IC50 values ranging from 1.51 to 21.56μM, respectively.

Abstract:Four new zwitterionic monoterpene indole alkaloids, rhynchophyllioniums A-D (1-4), together with eight known alkaloids (5-12), were isolated from the hook-bearing stems of Uncaria rhynchophylla. Their structures were elucidated by extensive spectroscopic data analysis of MS, 1D and 2D NMR, and ECD, and the zwitterionic forms and absolute configurations of 1 and 2 were unambiguously confirmed by single crystal X-ray diffraction analysis. All the isolates, including the monoterpene indole alkaloids with free C-22 carboxyl group and those with C-22 carboxyl methyl ester, were proved to be naturally coexisting in the herb by LC-MS analysis. This is the first report of monoterpene indole alkaloids that exist in the form of zwitterion. Additionally, the cytotoxic activities of all isolates against A549, HepG2, and MCF-7 cell lines are reported.

Abstract:The phytochemical investigation of the stem barks of Voacanga africana resulted in the isolation of fifteen known monoterpenoid indole alkaloids ( 1 – 15 ). The structures of the alkaloids were determined by comparison of their physical and spectroscopic data with those reported in literature. Twelve of these metabolites ( 4 – 15 ) were previously unreported within the genus Voacanga , thus they can serve as chemotaxonomic markers for V. africana to differentiate between V. africana from other species of the genus Voacanga . The alkaloids 3-oxoconopharyngine ( 6 ), ervahainanmine ( 10 ), crassanine ( 14 ) and conolobine A ( 15 ) hold the potential to serve as chemical markers for distinguishing V. africana from other family species. Furthermore, compounds 3 , 6 , 10 , 12 showed potential antimicrobial activities against Candida albicans and Bacillus subtilis with MIC values of 6.25–12.50 μg/mL.

Abstract:In a previous paper, the isolation and identification of six known indole alkaloids form Alstonia Yunnanensis Diels was reported. This paper reports the isolation of nine other indole alkaloids and a nonalkaloidal component. They are lochnerinine (Ⅰ), 11-methoxy-19-hydroxytabersonine (Ⅱ), pseudoakuammigine (Ⅳ), picrinine (Ⅴ), deacetylpicraline 3, 4, 5- trimethoxybenzoate (Ⅵ), corynanthine (Ⅸ), yohimbine (Ⅹ), 17-acetylsarpagine (Ⅺ), 16-episarpagine (Ⅻ) and methyl 3, 4, 5-trimethoxycinnamate. Their structures were elucidated by spectroeopic analysis (UV, IR, NMR, MS), physical constants and preparation of derivatives. It has been shown that Ⅵ, Ⅺ and Ⅻ are new indole alkaloids.

Abstract:Melodicochinines A - D (1-4), four new monoterpene indole alkaloids (MIAs), along with 21 known ones, were isolated from the stems and twigs of Melodinus cochinchinensis. Their structures were elucidated on the basis of extensive spectroscopic analysis. A ubiquitin-rhodamine 110 assay showed that 11-methyloxytabersonine had potential inhibitory effect against ubiquitin-specific protease 7 (USP7).

Abstract:One new monoterpene alkaloid (1) and eight known compounds (2-9), belonging to two monoterpene alkaloids (1-2), one pyridine alkaloid (3), four phenylpropanoid derivatives (4-7), and two matrine-type alkaloids (8-9), were isolated from Rauvolfia vomitoria Afzel. The structure of new compound 1 was determined by extensive spectroscopic analysis and electronic circular dichroism (ECD) calculation. Notably, this is the first reported monoterpene alkaloids from R. vomitoria. Besides, compounds 3, 5, and 7-9 were first discovered in the Apocynaceae family and compounds 4 and 6 were first reported in the Rauvolfia genus. This is also the first example of matrine-type alkaloids reported in the Apocynaceae family, indicating that matrine-type alkaloids are not unique to the Leguminosae/Fabaceae family. All isolated compounds were evaluated for their anti-acetylcholinesterase, anti-alpha-glucosidase, and antioxidant activities. Compound 6 showed significant alpha-glucosidase inhibitory activity and remarkable DPPH radical scavenging capacity, both of which are superior to the positive controls. Molecular docking of compound 6 with alpha-glucosidase was further performed, suggesting that compound 6 could form hydrogen bonds with residues ALA-292, ASN-259, and ARG-263.

Abstract:Four new indole alkaloids, daturametelindoles A-D (1–4) were isolated from the EtOAc soluble partition of the ethanol extract of the Datura metel seeds. The structures of the new compounds were determined based on spectroscopic evidence, including their 1D- and 2D-NMR spectra and mass spectrometry. In particular, compounds 1–4 were all racemes, confirmed by the optical rotations and CD spectra. Unfortunately, the chiral monomers were not obtained due to the amount, but the developments of their chiral separation and chiral resolution were completed. All isolated compounds were evaluated for cytotoxic effects against human gastric adenocarcinoma cells (SGC-7901), human hepatoma (Hepg2), and human breast cancer (MCF-7).
