Improving Callus Induction and Differentiation Efficiency of In-vitro Immature Embryo of Spring Wheat (Triticum aestivum L.)

Abstract:Effects of inoculation direction, genotypes, embryo ages and plant growth regulators on callus induction and differentiation efficiency of in-vitro immature embryo of seven local spring wheat cultivars of Gansu were investigated in this research. The results indicated: inoculation direction had no significant effects on callus induction, but had significant effects on differentiation with up-shield inoculation having higher differentiation efficiency; the effective embryo ages for in-vitro immature embryo culture ranged from 14 days to 20 days, the optimal embryo age was 16 days; Callus induction had no significant difference among genotypes, but Callus differentiation and regeneration had great difference among genotypes. Differentiation rate presented single-curved trend with the increase of 2,4-D concentration, varying among genotypes, the suitable 2,4-D concentration for most of the cultivars was 1~6 mg/L, 0.2 mg/L NAA和0.2 mg/L IAA induced rooting promptly, with rooting rate up to 83.8% and 55.1%, respectively.
