Effect of Drying Method and Extracting Temperature on Contents of Bioactive Constituents in Radix Isatidis and Folium Isatidis

Abstract:Objective To study the effect of different drying methods and extracting temperatures on the bioactive constituents in Radix Isatidis and Folium Isatidis. Methods Radix Isatidis and Folium Isatidis were dried in the shade, in the sun, and baked out with different temperatures; the bioactive constituents in Radix Isatidis and Folium Isatidis were extracted by Soxhlet extracting method under different temperatures; the contents of indigo and indirubin were determined by HPLC. Results Bioactive constituents of Radix Isatidis and Folium Isatidis lost less under the drying temperature of 60 ℃. The contents of the bioactive constituents reduced 40%—60% in Radix Isatidis and 30%~60% in Folium Isatidis at high drying temperature (over 90 ℃) and also reduced by dried in the shade compared with that at drying temperature of 60 ℃. The contents of the bioactive constituents were also reduced by dried in the shade; the contents of indigo and indirubin were the highest under the extracting temperature of 80—85 ℃ by using Soxhlet extracting method with chloroform solvent, the lowest, 95 ℃. Conclusion The suitable methods for drying Radix Isatidis and Folium Isatidis are to be dried in the sun and baked under 50—80 ℃. The temperature of water bath has significant impact on the extraction rate of the bioactive constituents while using Soxhlet extracting methods, 80—85 ℃ are suitable.
