Ethylene Biology Blooms from Fundamental Research to Postharvest Applications

Abstract:The 10th International Conference on the Plant Hormone Ethylene, co-chaired by Zhengguo Li (Chongqing University), Hongwei Guo (Peking University), Joseph Ecker (SALK, USA), Mondher Bouzayen (University of Toulouse), and Caren Chang (University of Maryland), was held on November 14–18, 2015, in Chongqing, China. This conference was organized locally by the School of Life Sciences, Chongqing University, the School of Life Sciences, Peking University, and the Postharvest Branch of Chinese Society for Horticultural Science. More than 300 delegates from 16 countries and areas attended this conference. There were 65 oral presentations and 61 posters, covering topics from ethylene physiological responses, ethylene signaling, to postharvest biology. The opening keynote speech was given by Dr. Xing Wang Deng (Peking University) on the topic of ethylene and light interactions during seedling emergence from soil. Deng's group found that mechanical stress from hard soil cover induces endogenous ethylene production and EIN3 protein accumulation, which is necessary for successful seedling emergence. They also discovered that COP1 is required for sensing the soil depth. COP1 directly targeted EBF1/EBF2 degradation and leading to EIN3 accumulation. When seedlings emerged close to the soil surface, the light intensity increased and COP1 activity was then inhibited for precise control of EIN3 stability. This work is a nice example to illustrate the roles of ethylene in the adaption of plants to the environment. The following individual session comprised talks from established scientists and selected talks from young investigators, postdocs, or students. Due to space limitations, we can only share some of the presentations in this conference with the readers, and discuss the trends in ethylene biology research. First, signal interactions with other hormones or stresses are hot research fields in the current ethylene research community. Similar to the opening keynote speech made by Xing Wang Deng on the topic of ethylene–light interactions, there were several talks on ethylene interactions with auxin, gibberellins, jasmonate, abscisic acid, salt stress, drought stress, etc. These studies are not only limited to Arabidopsis but also involve rose and fruits such as tomato and strawberry. Second, although the ethylene signaling framework has been established for more than a decade, there are still several interesting breakthroughs in this fundamental area. Hongwei Guo's group (Peking University) found that cytosolic EIN2 associates with EBF1/2 mRNA 3′ untranslated region (UTR) and tethers EBF1/2 mRNA into the processing body (P-body) for translational inhibition. This unexpected function of EIN2 adds another layer of regulation to ethylene signaling. Chi-Kuang Wen's team (Shanghai Institute of Plant Physiology and Ecology, CAS) identified two new genes in ethylene signaling (ECR1 and ECR2), which may act synergistically with CTR1 to inhibit ethylene signaling downstream of ethylene receptors but upstream of EIN2 and EIN3. Dominique Van Der Straeten (Ghent University) presented their work on cell-type-specific ethylene signaling. They used various cell-type-specific promoters to drive the expression of EBF1 or EBF2 in certain cell types to cause cell-type-specific ethylene irresponsive and then observed ethylene-related phenotypes in plants. They indeed found key cell types for ethylene-mediated growth inhibition. Third, transcriptional regulation is a major focus in postharvest biology for better understanding of the actions of ethylene during fruit development. Transcription factors ERFs are the central players for controlling fruit flavor, ripening, and texture. There were several talks on the characterization of ERF functions in a variety of plants. In addition, RNA-seq technology has been adopted as a method for exploring the transcriptional network in fruits. Fourth, new technology and large scale “-omics” experiments have been carried out in ethylene biology. Ning Li (Hong Kong University of Science and Technology) used the isotope metabolic labeling proteomics approach and identified multiple EIN2- and EIN3/EIL1-independent phosphor-relay-mediated post-translational modifications. Siling Zhong (The Chinese University of Hong Kong) talked about their findings on the global DNA methylation changes during tomato ripening and shed new light on the understanding of ethylene production in non-ripening tissues and climacteric fruit ripening. Last but not least, knowledge from fundamental research will boost postharvest biology. For example, Georg Groth (Heinrich-Heine University) found that the nuclear localization sequence (NLS) in EIN2 is necessary for EIN2-ETR1 physical interactions, and peptides mimicking the NLS can abrogate EIN2-ETR1 interactions and even cause reduction of ethylene responses. Their findings shed new light on the control of ethylene signaling for extending fruit shelf-life. Although the communications between signaling researchers and postharvest biologists are ever more intense, we still need to strengthen our collaborations. For example, how to deliver and transform the new knowledge in ethylene signaling into the fruit postharvest field? We believe that, as pointed out by both Jean-Claude Pech in his talk and Mondher Bouzayen in the closing remarks, an ethylene conference is the best platform for exchanging ideas and discussing collaborations. The wonderful talks and posters presented at this conference are at the forefront of ethylene research. Moreover, young scientists and students are our hope for boosting future ethylene studies. It is a pleasure to hear lots of interesting talks from the next generation if scientists at this conference. After a vote from all the attendees, the 11th International Conference on the Plant Hormone Ethylene will be held in the first week of June, 2018, located in Chania, Greece. Dr. Angelos Kanellis will be in charge of the local organization for this conference. Finally, we would like to pay tribute to our friend and colleague Prof. Michael McManus (Massey University, New Zealand), who passed away several months before conference opening.
