Effect of mild hypothermia on the expression N-methyl-D-aspartate receptor 1 in hippocampus of rats after traumatic brain injury

Abstract:Objective To investigate the effect of mild hypothermia on the expression of N-methyl-D-aspartate receptor 1 (NMDAR1) after fluid percussion traumatic brain injury (TBI) in rats. Methods A total of 54 adult male Sprague-Dawley ratswere randomly divided into three groups: TBI with hypothermia treatment ([33.0±0.5] ℃, n=24), TBI with normothermia ([37.0±0.3] ℃, n=24), and sham injured group (n=6). TBI model was induced by fluid percussion TBI device. Mild hypothermia was achieved by partial immersion in awater bath (0 ℃) under general anesthesia for 4 h. All the rats were killed at4, 6, 12 and 24h after TBI, respectively. The rats in the sham injured group were killed 24 h after treatment. The mRNA and protein levels of NMDAR1 in hippocampus were measured by realtime-polgmerase chain reaction (RT-PCR) and Western-blot techniques, respectively. Results Compared with sham injured group, NMDAR1 mRNA levels in injured hippocampus of normothermia and hypothermia groups were significantly increased at4, 6 and 12h after TBI (all P<0.05), but there were no significant differences in NMDAR1 mRNA between these three groups at24 h after TBI (all P>0.05). Comparedwith normothermia group, NMDAR1 mRNA levelswere significantly decreased at4, 6 and 12h after TBI in hypothermia group (all P<0.05),while there were no significant differences at 24 h after TBI (P>0.05). The expression of NMDAR1 protein in injured hippocampus of normothermia and hypothermia groups were significantly increased at 4, 6, 12 and 24h after TBI as compared with that of sham injured group (all P<0.05),while NMDAR1 protein levels in hypothermia group were significantly lower than that in normothermia group (all P<0.05). Conclusion Mild hypothermia can efficiently suppress NMDAR1 expression in the rats with TBI induced by fluid percussion and have good neuroprotection effect. (Shanghai Med J,2013,36:361-365)
