Abstract:Rationally constructing single-atom enzymes (SAEs) with superior activity, robust stability, and good biocompatibility is crucial for tumor therapy but still remains a substantial challenge. In this work, we adopt biocompatible carbon dots as the carrier material to load Ru single atoms, achieving Ru SAEs with superior multiple enzyme-like activity and stability. Ru SAEs behave as oxidase, peroxidase, and glutathione oxidase mimics to synchronously catalyze the generation of reactive oxygen species (ROS) and the depletion of glutathione, thus amplifying the ROS damage and finally causing the death of cancer cells. Notably, Ru SAEs exhibit excellent peroxidase-like activity with a specific activity of 7.5 U/mg, which surpasses most of the reported SAEs and is 20 times higher than that of Ru/C. Theoretical results reveal that the electrons of the Ru 4d orbital in Ru SAEs are transferred to O atoms in H2O2 and then efficiently activate H2O2 to produce •OH. Our work may provide some inspiration for the design of SAEs for cancer therapy. The Supporting Information is available free of charge at https://pubs.acs.org/doi/10.1021/acsami.1c12706.TEM and HRTEM images and hydrodynamic diameter of CDs; TEM and HRTEM images and size distribution of Ru SAEs; zeta potential of CDs and Ru SAEs in water; AFM phase image and corresponding height information of Ru SAEs; XRD patterns and FTIR spectra of CDs and Ru SAEs; XPS survey scan, C 1s, N 1s, and O 1s spectra of CDs and Ru SAEs; O–K-edge NEXAFS spectra of CDs and Ru SAEs; EPR spectra of CDs and Ru SAEs; Ru 3p XPS spectra of Ru SAEs; EXAFS fitting curve for Ru SAEs; GSH depletion with different concentrations of Ru SAEs; OXD-like activity at different concentrations of TMB and the catalyst; POD-like activity at different concentrations of TMB, H2O2, and the catalyst; corresponding Michaelis–Menten kinetic curves and Lineweaver–Burk plots; OXD- and POD-like activities of various nanomaterials; catalytic stability of Ru SAEs and Ru SAEs incubated with different buffers; TEM and HAADF-STEM images and XRD patterns of Ru SAEs after incubation in different buffers for 7 days and reacting with 1 mM H2O2 for 12 h; deformation charge density of Ru-N2C2; temperature changes of solutions of different concentrations of Ru SAEs; IR images and heating and cooling curves of Ru SAEs; vis/NIR absorption spectra and temperature changes of CDs and Ru SAEs; 4T1 tumor cell viability assays; fluorescence imaging with DCFH-DA and intracellular GSH levels of 4T1 cells under different treatments; fluorescence imaging of 4T1 cells stained with FDA and PI; JC-1 analysis of 4T1 cells as a measure of mitochondrial depolarization; photos of mice under different treatments; body weight variation of the mice; H&E-stained images of organ slices; EXAFS fitting parameters at the Ru K-edge for various samples; and kinetic parameters of different SAEs and Ru-based POD mimics (PDF)This article has not yet been cited by other publications.
