Abstract:Myelodysplastic syndrome (MDS) is difficult to diagnose and classify because it has the potential to evolve into acute myeloid leukemia (AML). Raman spectroscopy and orthogonal partial least squares discrimination analysis (OPLS-DA) are used to systematically analyze peripheral blood serum samples from 33 patients with MDS, 25 patients with AML, and 29 control volunteers to gain insight into the heterogeneity of serum metabolism in patients with MDS and AML. AML patients show unique serum...

What problem does this paper attempt to address?
The problems this paper attempts to address are: 1. **Distinguishing Myelodysplastic Syndromes (MDS) from Acute Myeloid Leukemia (AML)**: Utilizing Raman spectroscopy and Orthogonal Partial Least Squares Discriminant Analysis (OPLS-DA) to systematically analyze serum samples from MDS, AML patients, and healthy controls to reveal the metabolic heterogeneity between these diseases. 2. **Identifying Potential Biomarkers**: By analyzing the differences in Raman peak intensities of proteins, nucleic acids, lipids, collagen, and carbohydrates in the serum, potential biomarkers for diagnosing MDS and AML are screened. 3. **Validating the Diagnostic Model's Effectiveness**: Establishing an OPLS-DA model and evaluating its ability to distinguish between different MDS subtypes and AML through cross-validation, thereby improving the speed and accuracy of early identification and diagnosis of MDS and AML. 4. **Exploring Gene Expression Differences**: Screening for differential expression of MDS and AML-related genes based on the Gene Expression Omnibus (GEO) database and analyzing their main signaling pathways, such as Th17 cell differentiation, pertussis, and cytokine receptor interaction. In summary, this study aims to provide new means and important information for the rapid early diagnosis of MDS and AML through the combination of Raman spectroscopy and multivariate analysis methods.
