Abstract:Abstract Dietary restriction (DR) delays aging, but the mechanism remains unclear. We identified polymorphisms in mtd , the fly homolog of OXR1 , which influenced lifespan and mtd expression in response to DR. Knockdown in adulthood inhibited DR-mediated lifespan extension in female flies. We found that mtd / OXR1 expression declines with age and it interacts with the retromer, which regulates trafficking of proteins and lipids. Loss of mtd / OXR1 destabilized the retromer, causing improper protein trafficking and endolysosomal defects. Overexpression of retromer genes or pharmacological restabilization with R55 rescued lifespan and neurodegeneration in mtd -deficient flies and endolysosomal defects in fibroblasts from patients with lethal loss-of-function of OXR1 variants. Multi-omic analyses in flies and humans showed that decreased Mtd/OXR1 is associated with aging and neurological diseases. mtd/OXR1 overexpression rescued age-related visual decline and tauopathy in a fly model. Hence, OXR1 plays a conserved role in preserving retromer function and is critical for neuronal health and longevity.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is: How does dietary restriction (DR) delay brain aging and the development of neurodegenerative diseases by maintaining the function of the retromer complex? Specifically, the study focused on the mechanism of action of the oxidative stress response gene OXR1 (mtd in Drosophila) in this process. ### Main research questions: 1. **The role of OXR1/mtd in DR**: - The study found that the expression of OXR1/mtd increases under DR conditions, and this expression is crucial for DR - mediated lifespan extension. - Genetic variation affects the expression of OXR1/mtd, thereby affecting the effect of DR. 2. **The relationship between OXR1/mtd and retromer**: - OXR1/mtd interacts with the retromer complex and maintains its function. - The deletion of OXR1/mtd leads to the instability of the retromer complex, which in turn causes abnormal protein transport and endolysosomal defects. 3. **The impact of OXR1/mtd on nerve health**: - The expression of OXR1/mtd in neurons is crucial for delaying brain aging and the development of neurodegenerative diseases. - Over - expression of OXR1 can improve the lifespan shortening and neurodegenerative disease phenotypes caused by mtd deletion. 4. **The roles of DR and OXR1/mtd in different genetic backgrounds**: - Drosophila with different genetic backgrounds respond differently to DR. Among them, specific alleles of mtd show higher expression levels under DR conditions, which is related to lifespan extension. ### Key findings: - **The expression regulation of OXR1/mtd**: - Traffic jam (TJ) up - regulates the expression of mtd under DR conditions. - CTCF inhibits the expression of mtd under ad libitum (AL) conditions. - **The stability of the retromer complex**: - OXR1/mtd interacts with key proteins of the retromer complex (such as VPS26, VPS35) and maintains its stability. - The deletion of OXR1/mtd leads to a decrease in the protein level of the retromer complex, causing endolysosomal dysfunction and an increase in autophagosome formation. - **The role of OXR1/mtd in neurodegenerative diseases**: - The over - expression of OXR1/mtd can improve the neurodegenerative disease phenotypes caused by tau protein pathology. - The drug R55 can restore the phenotypes caused by OXR1/mtd deletion by stabilizing the retromer complex. ### Conclusion: This study reveals the mechanism by which OXR1/mtd delays brain aging and the development of neurodegenerative diseases by maintaining the function of the retromer complex under DR conditions. These findings not only help to understand the protective effect of DR, but also provide new targets for the development of treatment strategies for neurodegenerative diseases.
