Abstract:In this study, GO modified with sulfhydryl group was prepared by thiolation on the surface of GO, which makes a meaningful material. GO with sulfhydryl group combined with gold nanoparticles, and then combined with peptide sequence using Au-S bond. According to the change of absorbance before and after adding chymotrypsin, chymotrypsin was detected. It was label-free detection. The functional peptide (EKEKEKEPPPPC) was used to optimize the experiment to remove the non-specific adsorption in the system that was reaction mixture. The detection limit of chymotrypsin was 0.25 pg/mL. This can improve the sensitivity of detection. And the method can be applied in a real sample, which has a good development and application prospect.

Abstract:Thrombin is an important kind of proteases that can cleavage specific peptide bonds and plays a crucial role in numerous diseases, which leads to large amounts of efforts on its detection. In this study, a novel peptide self-assembled biosensor was developed based on gold nanoparticles functionalized graphene oxide (GO) for specific detection of thrombin. Through sulfhydryl design, peptides containing thrombin cleavage sites could self-assemble with the nanocomposites of the graphene material and gold particles, which brought the biosensor unique optoelectronic properties and high biological sensitivity to trans duce bio-molecular interactions into optical signals. Spectroscopic measurements were carried out in the presence of thrombin to observe an increase of the absorption peak at similar to 528 nm anda decrease of the absorbance at wavelength larger than 600 nm. The biosensor showed a linear dose-dependence and stable response to thrombin. And with a 528-680 nm absorbance ratio as the sensitive detection parameter, the self-assembled biosensor exhibited high sensitivity and specificity for the thrombin detection. Thus, it revealed convenient and sensitive detection abilities for protease of thrombin, indicating a promising prospect for other related proteases, such as matrix metalloproteinases to cancer related diseases. Abstract:Nanocomposites of graphene oxide and gold nanoparticles (GO/GNPs) were synthesized for label-free detections of amino acids. Interactions between the composites and amino acids were investigated by both naked-eye observation and optical absorption spectroscopy. The GO/GNPs composites displayed apparent color changes and absorption spectra changes in presences of amino acids including glutamate, aspartate, and cysteine. The interaction mechanisms of the composites and amino acids were discussed and explored with sulfhydryl groups and non-α-carboxylic groups on the amino acids. Sensing properties of the composites were tested, while pure gold particles were used as the control. The results suggested that the GO/GNPs composites had better linearity and stability in dose-dependent responses to the amino acids than those of the particles, especially in detections for acidic amino acids. Therefore, the nanocomposites platform can provide a convenient and efficient approach for label-free optical detections of important molecules such as amino acids.
Graphene oxide-based optical biosensor functionalized with peptides for explosive detection.

Abstract:A label-free optical biosensor was constructed with biofunctionalized graphene oxide (GO) for specific detection of 2,4,6-trinitrotoluene (TNT). By chemically binding TNT-specific peptides with GO, the biosensor gained unique optoelectronic properties and high biological sensitivity, with transducing bimolecular bonding into optical signals. Through UV absorption detection, increasing absorbance responses could be observed in presence of TNT at different concentrations, as low as 4.40×10(-9) mM, and showed dose-dependence and stable behavior. Specific responses of the biosensor were verified with the corporation of 2,6-dinitrotoluene (DNT), which had similar molecular structure to TNT. Thus, with high sensitivity and selectivity, the biosensor provided a convenient approach for detection of explosives as miniaturizing and integrating devices.

Abstract:Graphene oxide is a flexible nanoscale building block, which is contributed to improve the electrical, optical, and chemical properties. Here, gold nanoparticles on graphene oxide substrate were synthesized as sensitive nanoprobes for rapid L-cysteine detection. The smartphone-based system with multimode analysis of red-green-blue (RGB), hue-saturation-value (HSV), hue-saturation-lightness (HSL), and cyan-magenta-yellow-black (CMYK), were proposed for the investigation of interactions between the nanoprobes and L-cysteine. Then, optical absorption spectra were further examined to confirm the concentration characteristics of L-cysteine. Results suggested that gold nanoparticles on graphene oxide substrate had satisfying linearity and stability in dose-dependent responses to L-cysteine detection in the multimode analysis with the limit of detection at 0.06 mu M. Therefore, the smartphone-based nanoprobes system not only equipped with a high resolution camera and a powerful processor, but also was a fully automated device without additional label or complex detection. These features make it to be a convenient and efficient analysis platform for multiple fields, such as clinical application, industrial quality control, environment monitoring, and food assessment.

Abstract:Chirality is a fundamental phenomenon of nature, and the enantioselective recognition of amino acids isomers is especially important for life science. In this study, chiroptical system based on chiral assembly graphene quantum dots (GQDs) was developed for visual testing of D-phenylalanine (D-Phe). Here, GQDs were used as the fluorescent element, and chiral functional moieties of 1,3,5-triformylphloroglucinol-functionalized chiral ( +)-diacetyl-L-tartaric anhydride (TPTA) were used as the chiral recognition elements. Based on the formed chiral microenvironment, the fluorescence intensity of TPTA-assembled GQDs had a good linear relationship with D-Phe in the concentration range of 0.1–5 μM, and the detection limit was 0.023 μM. According to the variation in luminance of TPTA-assembled GQDs, visual testing to D-Phe was realized using a smartphone-assisted chiroptical system with a detection limit of 0.050 μM. The spiked recoveries of both chiroptical sensing methods based on TPTA-assembled GQDs from the food matrix ranged from 86.20 to 110.0%. Furthermore, TPTA-assembled GQDs were successfully applied to intracellular chiroptical imaging in response to D-Phe in vitro. The developed chiral nanomaterial TPTA-assembled GQDs with excellent photochemical stability, optical properties, and bioimaging capabilities provide a promising technique for the visual detection of amino acid isomers in the field of smart devices.

Abstract:A graphene quantum dots (GQDs)–chitosan (CS) composite film was prepared via successive electrodeposition of GQDs and CS on the surface of a glassy carbon electrode (GCE). The strong interactions between GQDs and CS resulted in the formation of a regular and uniform film, which can be applied in the electrochemical chiral recognition of tryptophan (Trp) enantiomers. CS in the composite film provides a chiral microenvironment, meanwhile, GQDs can amplify the electrochemical signals and improve the recognition efficiency. Due to the synergetic effect of GQDs and CS, chiral recognition of Trp enantiomers is achieved successfully. Compared with previous reports utilizing GQDs in photoluminescent research, this work opens a new avenue for broadening the applications of GQDs in the electrochemically chiral sensors.

Abstract:The detection of proteins is of great biological significance as disease biomarkers in early diagnosis, prognosis tracking and therapeutic evaluation. Thus, we developed a simple, sensitive and universal protein-sensing platform based on peptide and graphene oxide (GO). The design consists of a fluorophore (TAMRA, TAM), a peptide containing eight arginines and peptide ligand that could recognize the target protein, and GO used as a quencher. To demonstrate the feasible use of the sensor for target detection, Bcl-xL was evaluated as the model target. The sensor was proved to be sensitive and applied for the detection of the target proteins in buffer, 2% serum and living cells.

Abstract:Herein, a hydrophilic graphene composite functionalized with glutathione (GSH) and L(+)-Cysteine (Cys) was prepared via a simple and fast synthesis route, which was named G@S@Au@GC. The combination attack with two different zwitterionic polymers resulted in enhanced adsorption sites for glycopeptides. The obtained G@S@Au@GC exhibited excellent performance on a low limit of detection (0.2 fmol), a high selectivity (HRP: bovine serum albumin = 1:1500), a good load capacity (250 mu g center dot mg(-1)) and recovery rate (93%), which was also evaluated with IgG. Subsequently, 60 glycopeptides from complex biological sample (human saliva) were identified by Nano-LC-MS/MS. The advantages of combination attack, low-cost, simple and fast synthesis, and superior enrichment performance make G@S@Au@GC composite a bright future on glycoproteomics analysis and related diseases. Abstract:The development of a general approach for the nondestructive chemical and biological functionalization of graphene could expand opportunities for graphene in both fundamental studies and a variety of device platforms. Graphene is a delicate single-layer. two-dimensional network of carbon atoms whose properties can be affected by covalent modification. One method for functionalizing materials without fundamentally changing their inherent. structure is using biorecognition moieties. In particular, oligopeptides are molecules containing a broad chemical diversity that can be achieved within a relatively compact size. Phage display is a dominant method for identifying peptides that possess enhanced selectivity toward a particular target. Here, we demonstrate a powerful yet, benign approach for chemical functionalization of graphene via comprehensively screened phage displayed peptides. Our results show that graphene can be selectively recognized even in nanometer-defined strips. Further, modification of graphene with bifunctional peptides reveals both the ability to impart selective recognition of gold nanoparticles and the development of an ultrasensitive graphene-based TNT sensor. We anticipate that these results could open exciting opportunities in the use of graphene in fundamental biochemical recognition studies, as well as applications ranging from sensors to energy storage devices.
A Graphene Oxide-Peptide Fluorescence Sensor for Proteolytically Active Prostate-Specific Antigen.

Abstract:This paper presents a novel sensor to detect proteolytically active prostate-specific antigen (PSA) by assembling a purpose-designed FITC-labeled peptide with graphene oxide (GO). The fluorescence of the dye-labeled peptide was quenched in the presence of GO. Reaction of the sensor with PSA cleaves the peptide, leading to the release of the dye moiety and a great increase in fluorescence intensity in a dose- and time-dependent manner, and PSA can be quantified accordingly. This approach is simple compared to existing methods since the GO-peptide-based sensor is easily assembled and detection can be achieved without the involvement of complicated procedures. Moreover, the applicability of the method has been demonstrated by detecting PSA in spiked urine samples.
Highly Soluble PEGylated Pyrene-Gold Nanoparticles Dyads for Sensitive Turn-on Fluorescent Detection of Biothiols.

Abstract:Highly soluble fluorescent pyrene derivative with substantially improved fluorescence intensity in aqueous buffer was obtained via PEGylation strategy. The highly soluble PEGylated pyrene (PEO-Py) non-covalently adsorbed onto the surface of gold nanoparticles (Au NPs) to form dyads with quenched fluorescence due to highly efficient energy transfer between PEO-Py and Au NPs. The PEO-Py/Au NPs dyads were used for the sensitive turn-on fluorescent detection of biothiols. The fluorescence of PEO-Py was restored by the addition of cysteine (Cys), indicating that Cys can modulate the energy transfer between PEO-Py and Au NPs. This phenomenon then allowed for the sensitive detection of Cys with a limit of detection (LOD) of 11.4 nM. The linear range of determination of Cys was from 1.25 x 10(-8) to 2.25 x 10(-7) M. None of the other amino acids found in proteins showed obvious interference with the determination. It was important to note that the detection sensitivity of the PEO-Py/Au NPs system was more than 5-fold improved compared with the Py/Au NPs system. In addition, other biothiol molecules, such as glutathione, could also be detected by this sensor system. The method was also successfully applied to the determination of the total content of aminothiols in human plasma. Therefore an easily prepared, inexpensive, high solubility fluorescent probe has been realized and is also expected to detect other biological analytes of interest.

Abstract:We report on a novel nanoarchitecture for use in highly bioactive electrochemical biosensors. It consists of multilayers of nanostructured plasma-polymerized pyrrole (ppPY) and nanosheets of electrically conductive graphene. The ppPY films were deposited by plasma-enhanced chemical vapor deposition on a graphene surface to form nanostructured composites (G-ppPY). The G-ppPY films were then coated with protein (BSA as a model) by adsorption, and then with DNA. The adsorption of protein and DNA on the nanocomposite was studied by electrochemical impedance spectroscopy and with a quartz crystal microbalance. Results demonstrated that the adsorption of biomolecules on G-ppPY films causes a higher variation in its electrochemical properties and adsorbed amount than that on a plain ppPY surface. This indicates that the presence of graphene can enhance the electrochemical activity of ppPY without reducing the sensitivity of biomolecular adsorption. Figure A novel nanoarchitecture is developed for use in highly bioactive electrochemical biosensors, which is composed of multilayers of nanostructured plasma-polymerized pyrrole and electrically conductive graphene nanosheets. The presence of graphene can enhance the electrochemical activity of ppPY without reducing the sensitivity of biomolecular adsorption.

Abstract:In this work, a dispersible graphene-based material with a characteristic of aggregation-induced emission (AIE) was prepared by wet chemical reduction of graphene oxide (GO). During the GO reduction process, a conjugated molecule TPEP containing tetraphenylethylene (TPE) and pyrene was employed as a stabilizer because of the π –π interactions and the wrapping effect. The as-prepared rGO-TPEP not only has good dispersion in solution but also processes the AIE feature. Its fluorescence intensity is 2.23 times higher than that of TPEP at the same condition. The unique optical properties and AIE effect enable the rGO-TPEP as a chemical sensor for highly sensitive explosive detection in aggregated state and solid state. In the aggregated state, trace 2,4-dinitrotoluene (DNT) can be detected by the rGO-TPEP even when the concentration is as low as 0.91 ppm, and the quenching constant is as high as 2.47 × 104 M -1 .
