Bioprospecting rhizosphere fungi endowed with multifarious plant growth-promoting potential to enhance finger millet growth under salinity stress

Abstract:Rhizosphere fungi in the proximity of plant roots have unique metabolic abilities that aid plants in handling environmental stressors like salinity and boost sustainable agriculture. The present study focused on isolating fungal isolates from the rhizosphere of saline and non-saline regions in India for their endurance of high NaCl levels. Microscopic methods were used to identify fungal isolates cultured on potato dextrose agar (PDA) medium, and diversity indices were determined. Further, the screening was carried out for plant growth-promoting (PGP) traits, production of extracellular enzymes, and their potential to enhance finger millet growth under normal and NaCl-stressed conditions in vitro, followed by molecular identification (18S rRNA). In the present study, 110 PGPF isolates were identified based on morphological characteristics into six genera. A total of seven isolates were found to have the highest potential for NaCl stress tolerance, ranging from 0 to 1200 mM. Of the 110 isolates, 94.54% of isolates produced IAA and siderophore, whereas phosphate-solubilization and ACC deaminase activity were observed in 90 and 80% of isolates respectively. Ammonia and HCN synthesis were detected in 66.36 and 27.27% of isolates, respectively. Additionally, extracellular enzyme production was observed. 53 isolates showed 100% germination in finger millet seeds under distilled water in vitro. Whereas 33 exhibited 90–100% germination, with increased root and shoot length compared to the control, under 400 mM NaCl. The Bonitur scale and principal component analysis (PCA) supported shortlisting the most promising isolates based on their diverse PGP traits and seed germination ability. Thus, this study emphasizes the potential of these isolates that could be developed as biostimulants and biofertilizers suitable for specific geographical areas affected by salinity stress to enhance salinity stress tolerance in finger millet.
