Abstract:High salinity poses a significant threat to arable land globally and contributes to desertification. Growth-promoting rhizobacteria assist plants in mitigating abiotic stresses and enhancing crop productivity through the production of siderophores, exopolysaccharides (EPS), solubilisation of phosphate, indole-3-acetic acid (IAA), and other secondary metabolites. This study aimed to isolate, identify, and characterise bacteria that exhibit robust growth-promoting properties. A total of 64 bacterial isolates from the rhizosphere of Miscanthus sinensis were evaluated for plant growth-promoting (PGP) traits, including IAA, EPS, siderophores, and solubilisation of phosphate. Among them, five isolates were selected as plant growth-promoting rhizobacteria (PGPR) based on their PGP features and identified via 16S rRNA sequencing: Enterococcus mundtii strain INJ1 (OR122486), Lysinibacillus fusiformis strain INJ2 (OR122488), Lysinibacillus sphaericus strain MIIA20 (OR122490), Pseudomonas qingdaonensis strain BD1 (OR122487), and Pseudomonas qingdaonensis strain MIA20 (OR122489), all documented in NCBI GenBank. BD1 demonstrated a higher production of superoxide dismutase (SOD) (17.93 U/mg mL), catalase (CAT) (91.17 U/mg mL), and glutathione (GSH) (0.18 U/mg mL), along with higher concentrations of IAA (31.69 μg/mL) and salicylic acid (SA) (14.08 ng/mL). These isolates also produced significant quantities of amino and organic acids. BD1 exhibited superior PGP traits compared to other isolates. Furthermore, the NaCl tolerance of these bacterial isolates was assessed by measuring their growth at concentrations ranging from 0 to 200 mM at 8-h intervals. Optical density (OD) measurements indicated that BD1 and INJ2 displayed significant tolerance to salt stress. The utilisation of these isolates, which enhances plant growth and PGP traits under salt stress, may improve plant development under saline conditions.

Abstract:In most in vitro run-off transcription reactions with T7 RNA polymerase, transcripts with heterogeneous ends are commonly obtained. Towards the goal of finding a simple and effective procedure for correct processing of their 3' ends we propose the use of trans-acting antigenomic delta ribozyme. We demonstrate that the extension of nascent transcripts with only seven nucleotides complementary to the ribozyme's recognition site, and subsequently, the removal of those nucleotides with the ribozyme acting in trans, is an efficient procedure for generating transcripts with homogenous 3' ends. This approach was tested on two model RNA molecules: an in vitro transcript of yeast tRNA(Phe) and a delta ribozyme, which processed itself during transcription. The proposed procedure is a simple alternative to the use of ribozymes as cis-cleaving autocatalytic cassettes attached to transcript 3' ends. As there is little possibility that the required additional stretch, only seven nucleotides long, enters into stable interactions with other parts of the transcripts, it can be cleaved off with high efficacy.
Plant growth promoting characteristics of halophilic and halotolerant bacteria isolated from coastal regions of Saurashtra Gujarat

Abstract:Halophiles are a class of microorganisms that thrive in environments with very high salt concentrations. The coastal regions of Saurashtra Gujarat host a diverse group of microorganisms including halophilic and halotolerant bacteria that may have plant growth promoting characteristics. Microorganisms with plant growth promoting characteristics are of immense importance in the field of agriculture and the present study was conducted to investigate the characteristics of halophilic and halotolerant bacteria isolated from agricultural soils of coastal regions of Junagadh and Porbandar districts of Saurashtra, Gujarat. A total of 15 isolated strains showed indole acetic acid production, solubilization of phosphate and potash, and nitrogen fixing capacity ranging from 18.77-33.48 μg ml-1, 50.10-106.10%, 180.42-239.92% and 0.170-0.480 g kg-1 of Jensen's agar medium, respectively, while two isolates were also found positive for siderophore production. Besides, nine out of fifteen isolates also showed positive ACC deaminase activity ranging from 0.92-5.78 µM α-ketobutyrate mg-1 h-1. The isolates were further characterized by physiological, microscopic, and biochemical tests. The halophilic and halotolerant bacterial isolates were identified by 16S rRNA gene sequencing as belonging to Halomonas pacifica, H. stenophila, and Bacillus haynesii, B. licheniformis and Oceanobacillus aidingensis respectively. The 16S rRNA partial gene sequence of two isolates belonging to H. pacifica and H. stenophila were submitted to NCBI with accession number MK955347 and MK961217 respectively. The findings of the present investigation showed that isolated bacterial halophiles possess promising plant growth promoting characteristics. Their potential as bioinoculants to alleviate salinity stress in crops and for bioremediation deserves further investigation.

Abstract:Halotolerant plant growth promoting rhizobacteria (PGPR) are beneficial microorganisms utilized to mitigate the biotic and abiotic stresses in plants. The areas of Sundarban mangroves of West Bengal, India have been reported to be rich in halotolerant microflora, yet major area remains unexplored. The present study, therefore, aims to map down the region-specific native microbial community potent of salt tolerance, plant growth promoting (PGP) activity and antagonistic activity against fungal pathogens. Bacterial samples were isolated from the saline soil of the Sundarban mangroves. A total of 156 bacterial samples were isolated and 20 were screened for their salt tolerance potential. These isolates were characterised using morphological, biochemical, and molecular approaches. Based on 16s rRNA sequencing, they were classified into 4 different genera, including Arthrobacter sp. (01 isolate), Pseudomonas plecoglossicida (01 isolate), Kocuria rosea (01 isolate), and Bacillus (17 isolates). The halotolerant isolates which possessed plant growth promoting traits including phosphate, and zinc solubilization, indole acetic acid production, siderophore, and ammonia generation were selected. Further, the effect of two halotolerant isolates GN-5 and JR-12 which showed most prominent PGP activities was evaluated in pea plant under high salinity conditions. The isolates improved survival by promoting germination (36 to 43%) and root-shoot growth and weight of pea plant in comparison to non-inoculated control plants. In a subsequent dual culture confrontation experiment, both these halo-tolerant isolates showed antagonistic activities against the aggressive root rot disease-causing Macrophomina phaseolina (Tassi) Goid NAIMCC-F-02902. The identified isolates could be used as potential bioagents for saline soils, with potential antagonistic effect on root rot disease. However, further studies at the physiological and molecular level would help to delineate a detail mechanistic understanding of broad-spectrum defence against salinity and potential biotic pathogen.

Abstract:Plant growth promoting rhizobacteria (PGPR) are alternative strategy for mitigating the negative impacts of salinity stress on plants. The aim of this study was to isolate native PGPR with high plant growth promoting traits from Egyptian soils which could be used as an environmentally eco-friendly technique for alleviating salinity stress. Twenty-five Azotobacter spp. were isolated from Most Probable tubes, then all isolates were tested to grow in different concentrations of NaCl for salt tolerance. The highest tolerant Azotobacter spp. were seven isolates which then were tested for their ability to fix nitrogen invitro using acetylene reduction technique. The most active Azotobacter isolate was (No. AZ 1) which recorded 27.70 nmoles C2H4/ml/h. On the other side, twenty-five phosphate solubilizing bacteria were isolated from Pikovskaya’s agar plates. All isolates were tested for salt tolerance and four isolates were selected for further understanding the mechanism of phosphate solubilization and its relationship to pH in liquid Pikovskaya’s medium supplemented with tri-calcium phosphate. The highest quantity of released phosphorus in the culture (16.31 mg p /100 ml) was for the isolate PSB 14 which selected for further studies. As well as isolates AZ 1 and PSB 14 showed high effectiveness in the production of indole acetic acid (67.87 and 8.99 µg/ml), respectively. Based on the 16S rRNA sequence analysis, AZ 1 and PSB 14 were identified as Azotobacter salinstris ON809700 and Bacillus amyloliquefaciens ON818999. Based on this study, these salt-tolerant isolates could add benefits to plant grown in saline conditions.

Abstract:Salinity stress is a barrier to crop production, quality yield, and sustainable agriculture. The current study investigated the plant growth promotion, biochemical and molecular characterization of bacterial strain Enterobacter cloacae PM23 under salinity stress (i.e., 0, 300, 600, and 900 mM). E. cloacae PM23 showed tolerance of up to 3 M NaCl when subjected to salinity stress. Antibiotic-resistant Iturin C (ItuC) and bio-surfactant-producing genes (sfp and srfAA) were amplified in E. cloacae PM23, indicating its multi-stress resistance potential under biotic and abiotic stresses. Moreover, the upregulation of stress-related genes (APX and SOD) helped to mitigate salinity stress and improved plant growth. Inoculation of E. cloacae PM23 enhanced plant growth, biomass, and photosynthetic pigments under salinity stress. Bacterial strain E. cloacae PM23 showed distinctive salinity tolerance and plant growth-promoting traits such as indole-3-acetic acid (IAA), siderophore, ACC deaminase, and exopolysaccharides production under salinity stress. To alleviate salinity stress, E. cloacae PM23 inoculation enhanced radical scavenging capacity, relative water content, soluble sugars, proteins, total phenolic, and flavonoid content in maize compared to uninoculated (control) plants. Moreover, elevated levels of antioxidant enzymes and osmoprotectants (Free amino acids, glycine betaine, and proline) were noticed in E. cloacae PM23 inoculated plants compared to control plants. The inoculation of E. cloacae PM23 significantly reduced oxidative stress markers under salinity stress. These findings suggest that multi-stress tolerant E. cloacae PM23 could enhance plant growth by mitigating salt stress and provide a baseline and ecofriendly approach to address salinity stress for sustainable agriculture.
Genomic characterization of the salt-tolerant Bacillus and Jeotgalicoccus strains reveals a diverse metabolism relevant to plant growth promotion and salt stress tolerance

Abstract:Previous research leads have affirmed the crucial role of plant growth-promoting rhizobacteria (PGPR) strains Bacillus safensis (NBRI 12 M), Bacillus subtilis (NBRI 28B, NBRI 33N), and Jeotgalicoccus huakuii (NBRI 13E) in salt stress amelioration and plant growth promotion. In the present study, whole-genome analysis unveiled the underlying molecular mechanisms accounting for phyto-beneficial and stress-mitigating traits of the selected PGPR strains. The genomic characterization has revealed that NBRI 12 M, NBRI 28B, NBRI 33N, and NBRI 13E possessed a single circular chromosome of 3.73 Mb, 4.07 Mb, 4.10 Mb, and 2.17 Mb size, respectively. The genome analysis of these strains demonstrated varied genes such as mrp and yfiY for plant growth promotion, nutrient metabolism, and other secondary metabolites biosynthesis. High salinity tolerance genes (yicL, ydhP_1, spoIIQ, and spoIIID), encoding for membrane transporter, dormancy, and sporulation, were also identified. In addition, several chemotaxis (cheA, cheY, and cheW) and motility gene clusters (motB_1, motB_2) were found in the PGPR strains for successful rhizosphere colonization. Further, NBRI 12 M has significantly increased the shoot and root length and dry weight by 14.13%, 20.63%, and 9.63%, respectively, under salt stress. In addition, NBRI 12 M inoculation reduced defense enzymes by 79.77%, 84.75%, 74.11%, 70.77%, and 57.75% for SOD, APX, GPX, CAT, and PPO, respectively. Overall findings from this study offered a detailed comparative genomic analysis of salt stress ameliorating PGPR of Bacillus genera towards enhancing the deep insights for host-PGPR association.

Abstract:Plant growth-promoting rhizobacteria (PGPR) are considered as bio-ameliorators that confer better salt resistance to host plants while improving soil biological activity. Despite their importance, data about the likely synergisms between PGPR and halophytes in their native environments are scarce. The objective of this study was to assess the effect of PGPR ( Glutamicibacter sp. and Pseudomonas sp.) inoculation on biomass, nutrient uptake, and antioxidant enzymes of Suaeda fruticosa , an obligate halophyte native in salt marshes and arid areas in Tunisia. Besides, the activity of rhizospheric soil enzyme activities upon plant inoculation was determined. Plants were grown in pots filled with soil and irrigated with 600 mM NaCl for 1 month. Inoculation (either with Pseudomonas sp. or Glutamicibacter sp.) resulted in significantly higher shoot dry weight and less accumulation of Na + and Cl – in shoots of salt-treated plants. Glutamicibacter sp. inoculation significantly reduced malondialdehyde (MDA) concentration, while increasing the activity of antioxidant enzymes (superoxide dismutase; catalase; ascorbate peroxidase; and glutathione reductase) by up to 100%. This provides strong arguments in favor of a boosting effect of this strain on S. fruticosa challenged with high salinity. Pseudomonas sp. inoculation increased shoot K + and Ca 2+ content and lowered shoot MDA concentration. Regarding the soil biological activity, Pseudomonas sp. significantly enhanced the activities of three rhizospheric soil enzymes (urease, ß-glucosidase, and dehydrogenase) as compared to their respective non-inoculated saline treatment. Hence, Pseudomonas sp. could have a great potential to be used as bio-inoculants in order to improve plant growth and soil nutrient uptake under salt stress. Indole-3-acetic acid concentration in the soil increased in both bacterial treatments under saline conditions, especially with Glutamicibacter sp. (up to +214%). As a whole, Glutamicibacter sp. and Pseudomonas sp. strains are promising candidates as part of biological solutions aiming at the phytoremediation and reclamation of saline-degraded areas.

Abstract:In the present study halotolerant bacteria were isolated from saline soil (EC ~ 11.9). Based on salt tolerance and plant growth promoting characteristics isolate AF7 was selected for further study. It was identified as Alcaligenes sp. on the basis of protein profiling and 16S rRNA sequence homology. Interestingly, AF7 showed diverse PGP characters at different salinity levels. While phosphate solubilization activity was expressed up to 300 mM NaCl, siderophore production was shown up to 700 mM, zinc solubilization up to 1000 mM and indole acetic acid (IAA), gibberellic acid (GA) and exopolysaccharides (EPS) production were depicted till 1400 mM. Correlative and regression analysis suggested positive relation between IAA, GA, EPS, siderophore production and zinc solubilization capability of AF7 and salinity up to 300 mM NaCl. EPS was found to be the most significant response and there was 263% increment in presence of 300 mM NaCl when compared to non-saline control. Analysis also showed that while growth promoting attributes were significant up to a threshold salinity level, further increasing the stress deviates the mechanism towards survival involving proline, antioxidant and hydroxyl scavenging activities. Combination of halotolerant AF7 and EPS showed more than twofold increase in the vegetative growth parameters of rice at ~ 170 mM NaCl (EC 9 dS/m). The study shows the mechanisms/metabolites of the plant growth promoting bacterium (PGPB) AF7 prominently involved during the salinity stress. Study also proves that novel bioformulations can be developed by integrative use of EPS and salt tolerant-PGPB which can be effective for saline soils.

Potential of Halophilic/Halotolerant Bacteria in Enhancing Plant Growth Under Salt Stress

Abstract:In the current study, fourteen bacterial strains were obtained in salt contaminated soils. The identification and characterization of the bacterial strains were performed by conventional and molecular techniques. According to the results of 16S rRNA gene sequence analysis, five genera (Bacillus, Staphylococcus, Oceanobacillus, Exiguobacterium, and Halobacillus) were identified with a homology of equal to 99% or higher similarity. Afterward, these fourteen halotolerant/halophilic bacterial strains were investigated for their plant growth promoting (PGP) traits including production of indole-3-acetic acid (IAA) and siderophore, activity of 1-aminocyclopropane-1-carboxylate (ACC) deaminase, fixation of nitrogen, and phosphate solubilization potential. Five of the bacterial strains possessing PGP traits were tested for their effects on the growth of a salt sensitive plant (wheat) in a hydroponic system under salt stress (200 mM). Inoculation of five bacterial strains under salt stress significantly enhanced plant weight (Triticum aestivum) ranged from 71.18 to 89.04%. Salt stress amelioration potential of Oceanobacillus picturae and Staphylococcus succinus on T. aestivum has been shown for the first time in this study. In non-saline soil, the promising effect of plant growth bacteria is clear; however, in saline soil, the use of PGP halophilic and halotolerant bacteria can increase the productivity of salt sensitive plants. Therefore, the novel halophilic and halotolerant bacteria that promote plant growth can be developed for agricultural uses in saline soils.

Abstract:Salinity, a frequently occurring abiotic stress, is a major constraint for crop productivity worldwide. The present study was conducted to evaluate the ability of plant growth promoting rhizobacteria (PGPR) Bacillus cereus Pb25, isolated from soil irrigated with saline water, to promote Vigna radiate (mungbean) growth in the absence and presence of salt stress (9 dS m−1). Results demonstrated that B. cereus promoted V. radiate plant growth significantly even in the presence of salt. Inoculations with PGPR improved the plant growth, and increased the root, shoot fresh and dry biomass and yield as compared to plants with no bacterial treatment (control). Results showed that both chlorophyll content and plant growth were inhibited by saline stress and the salt-induced oxidative damage (measured by MDA, H2O2) was alleviated by PGPR inoculation. Furthermore, PGPR inoculation significantly increased the antioxidant enzymes (POD, SOD and CAT) activities and enhanced the accumulation of proline, potassium, nitrogen and phosphorus as well as decreased sodium accumulation in saline stressed plants. Regarding the soil biological activity, inoculated PGPR enhanced the activity of dehydrogenase, alkaline phosphatase, microbial biomass carbon, available phosphorus and total organic carbon under saline stress as compared to saline treatment alone. These results suggest that B. cereus can be used in salinized agricultural lands as bio-inoculant to increase crop productivity.

Abstract:An antibiotic weakly active against Candida was isolated as a colorless crystalline hydrochloride from a culture of a streptomyces which showed some resemblances with Streptomyces eurythermits. This antibiotic was unstable to acid and alkali, but was recovered in good yield by elution with 0.5M ammoniumchloride solution from the ion-exchange resin, Amberlite IRC-50 (Na+ type). It was determined to be acrylamidine by spectroscopic data, degradation and direct comparison with a synthetic sample. The LD50 (mice) of acrylamidine was 44 mg/kg intravenously.
Halotolerant bacteria mitigate the effects of salinity stress on soybean growth by regulating secondary metabolites and molecular responses

Abstract:Abstract Background Salinity is a major threat to the agriculture industry due to the negative impact of salinity stress on crop productivity. In the present study, we isolated rhizobacteria and evaluated their capacities to promote crop growth under salt stress conditions. Results We isolated rhizospheric bacteria from sand dune flora of Pohang beach, Korea, and screened them for plant growth-promoting (PGP) traits. Among 55 bacterial isolates, 14 produced indole-3-acetic acid (IAA), 10 produced siderophores, and 12 produced extracellular polymeric and phosphate solubilization. Based on these PGP traits, we selected 11 isolates to assess for salinity tolerance. Among them, ALT29 and ALT43 showed the highest tolerance to salinity stress. Next, we tested the culture filtrate of isolates ALT29 and ALT43 for IAA and organic acids to confirm the presence of these PGP products. To investigate the effects of ALT29 and ALT43 on salt tolerance in soybean, we grew seedlings in 0 mM, 80 mM, 160 mM, and 240 mM NaCl treatments, inoculating half with the bacterial isolates. Inoculation with ALT29 and ALT43 significantly increased shoot length (13%), root length (21%), shoot fresh and dry weight (44 and 35%), root fresh and dry weight (9%), chlorophyll content (16–24%), Chl a (8–43%), Chl b (13–46%), and carotenoid (14–39%) content of soybean grown under salt stress. Inoculation with ALT29 and ALT43 also significantly decreased endogenous ABA levels (0.77-fold) and increased endogenous SA contents (6–16%), increased total protein (10–20%) and glutathione contents, and reduced lipid peroxidation (0.8–5-fold), superoxide anion (21–68%), peroxidase (12.14–17.97%), and polyphenol oxidase (11.76–27.06%) contents in soybean under salinity stress. In addition, soybean treated with ALT29 and ALT43 exhibited higher K + uptake (9.34–67.03%) and reduced Na + content (2–4.5-fold). Genes involved in salt tolerance, GmFLD19 and GmNARK , were upregulated under NaCl stress; however, significant decreases in GmFLD19 (3–12-fold) and GmNARK (1.8–3.7-fold) expression were observed in bacterial inoculated plants. Conclusion In conclusion, bacterial isolates ALT29 and ALT43 can mitigate salinity stress and increase plant growth, providing an eco-friendly approach for addressing saline conditions in agricultural production systems.

Abstract:As one of the major non–biological stresses, soil salinity negatively affects agricultural productivity, microbial communities, and agricultural economics in impacted areas and is a significant agrarian problem worldwide that has expanded in the last few decades. The use of glycophytes (salt–sensitive plants) is the primary limitation to such an agricultural approach. However, halophytes that are proficient at obtaining essential micro/macronutrients from exceedingly saline environments and thrive under conditions in which salinity–sensitive crops are either unproductive or inefficient are prime candidates for utilization in these circumstances. In addition, the bacterial microbiome associated with the roots of halophytic plants is a useful resource for the isolation of salt–tolerant bacteria with multiple plant growth–promoting characteristics, such as the production of different phytohormones, dissolution of insoluble inorganic phosphates, siderophore production, molecular nitrogen fixation, the activity of 1–aminocyclopropane–1–carboxylic acid (ACC) deaminase enzyme, and pathogen suppression, which enhance salinity tolerance of these plants as well as glycophytes and thereby stimulate the growth of plants under salinity–induced stress. The most dominant among reported halotolerant bacterial genera in most of the halophytic plants are Halomonas, Bacillus, Streptomyces, Oceanobacillus, and Pseudomonas. Here, it is shown that a wide range of halotolerant bacteria with a variety of plant growth-promoting characteristics may provide an important resource for augmenting crop tolerance to salinity and thereby boosting saline soil–based agriculture.

Phylogenetic analysis of halophyte-associated rhizobacteria and effect of halotolerant and halophilic phosphate-solubilizing biofertilizers on maize growth under salinity stress conditions

Abstract:Aims: The main objective of the present work was to evaluate plant growth-promoting abilities of bacterial strains from the rhizosphere of halophytes and their effect on maize growth under salinity stress. Methods and results: Halophilic bacteria were identified using 16S rRNA sequence analysis and their plant growth-promoting abilities were characterized. Phylogenetic analysis showed that bacterial strains belonging to Bacillus, Halobacillus and Pseudomonas were dominant in the rhizosphere of halophytes. More than 93% strains showed P-solubilization activity and IAA production. About 54% strains were able to produce ACC deaminase, 29% strains showed positive results for nitrogen fixation, 41 and 21% strains showed siderophores and HCN production ability respectively. More than 90% strains showed antifungal activity against more than two fungal pathogens and production of different hydrolytic enzymes. To study the plant growth-promoting effect on maize, five bacterial strains Bacillus safensis HL1HP11 and Bacillus pumilus HL3RS14, Kocuria rosea HL1RP8, Enterobacter aerogenes AT1HP4 and Aeromonas veronii AT1RP10 were used as inoculants; in the form of seed coat and enriched soil-based phosphate biofertilizers. All bacterial strains positively affected the maize growth as compared to non-inoculated control + NaCl plants. Plants inoculated with Bacillus HL3RS14-based soil biofertilizers showed maximum increase in dry weights of root (48-124%) and shoot (52-131%) as compared to control + NaCl (soil + rock phosphate, no inoculum). PGPR inoculations under salinity stress conditions showed high concentrations of proline, glycine betaine and malondialdehyde. Conclusion: These results indicated that under saline soil conditions, halophilic PGPR strains combined with carrier materials are promising candidates as biofertilizers.

Abstract:In the current study, the PGP (production of siderophore and IAA, fixation of nitrogen and solubilization of phosphate) potential of forty-five bacterial isolates comprising sixteen genera (Marinibacillus, Zhihengliuella, Bacillus, Nesterenkonia, Halobacillus, Promicromonospora, Terribacillus, Jeotgalibacillus, Staphylococcus, Oceanobacillus, Virgibacillus, Halomonas, Gracilibacillus, Planococcus, Thalassobacillus and Microbacterium) was investigated. Among these forty-five bacterial isolates, most of the isolates (40 isolates) possess plant growth promoting potential. Four bacterial isolates [Marinibacillus sp. (EM1), Zhihengliuella salsuginis (EM30), Planococcus sp. (EK9), and Promicromonospora sp. (EK12)] possessing PGP traits were applied on the barley grown under salt (NaCl) stress in a hydroponic system. Comparing to the un-inoculated group, all the four bacterial isolates significantly improved plant growth that inhibited with NaCl (200 mM) stress. The growth rate of the plants inoculated with Zhihengliuella salsuginis (EM30), Promicromonospora sp. (EK12), Planococcus sp. (EK9), and Marinibacillus sp. (EM1) was 81.86, 80.51, 70.62 and 69.22, respectively. As far as we know, this is the first report showing the PGP potential of Marinibacillus sp. (EM1), Bacillus horikoshii (EM9), Bacillus patagoniensis (EM12), Bacillus agaradhaerens (EK13), Oceanobacillus profundus (EM26) Thalassobacillus devorans (EM34) and Promicromonospora sp. (EK12).

Abstract:Salinity is one of the significant abiotic stresses that exert harmful effects on plant growth and crop production. It has been reported that the harmfulness of salinity can be mitigated by the use of salt-tolerant plant growth-promoting (PGP) bacteria. In this study, four bacteria were selected from a total of 30 cultures, based on salt-tolerant and PGP properties. The isolates were found to produce indole acetic acid (8.49-19.42 μg/ml), siderophore (36.04-61.77%), and solubilize potassium and inorganic phosphate. Identification based on 16S rRNA gene sequencing revealed that the isolates belonged to Cronobacter (two isolates) and Enterobacter (two isolates). Inoculation of PGP bacteria under 2 and 10% salinity stress showed enhanced plant growth parameters in Vigna radiata compared to both salinity and non-salinity control plants. The rate of germination (113.32-206.64%), root length (128.79-525.31%), shoot length (34.09-50.32%), fresh weight, and dry weight were 3-fold higher in bacteria-treated seeds than control plants. The estimation of chlorophyll (1-5-fold), carotenoids (1-4-fold), and proline content (3.65-14.45%) was also higher compared to control plants. Further, the bacterized seeds showed enhanced nitrogen and phosphorous uptake and mobilized sodium ions from roots to leaves. Overall the strains SS4 and SS5 performed well in both 2 and 10% salt-amended soils. These strains could be formulated as a bioinoculant to mitigate the salinity stress in salinized soils.

Abstract:Sub-Kelvin scanning tunneling spectroscopy in the Chevrel phases SnMo6S8 and PbMo6S8 reveals two distinct superconducting gaps with Δ1=3 meV, Δ2∼1.0 meV and Δ1=3.1 meV, Δ2∼1.4 meV, respectively. The gap distribution is strongly anisotropic, with Δ2 predominantly seen when scanning across unit-cell steps on the (001) sample surface. The spectra are well fitted by an anisotropic two-band BCS s-wave gap function. Our spectroscopic data are confirmed by electronic heat capacity measurements, which also provide evidence for a twin-gap scenario.

Abstract:Soil salinity has emerged as a serious issue for global food security. It is estimated that currently about 62 million hectares or 20 percent of the world's irrigated land is affected by salinity. The deposition of an excess amount of soluble salt in cultivable land directly affects crop yields. The uptake of high amount of salt inhibits diverse physiological and metabolic processes of plants even impacting their survival. The conventional methods of reclamation of saline soil which involve scraping, flushing, leaching or adding an amendment (e.g., gypsum, CaCl2, etc.) are of limited success and also adversely affect the agro-ecosystems. In this context, developing sustainable methods which increase the productivity of saline soil without harming the environment are necessary. Since long, breeding of salt-tolerant plants and development of salt-resistant crop varieties have also been tried, but these and aforesaid conventional approaches are not able to solve the problem. Salt tolerance and dependence are the characteristics of some microbes. Salt-tolerant microbes can survive in osmotic and ionic stress. Various genera of salt-tolerant plant growth promoting rhizobacteria (ST-PGPR) have been isolated from extreme alkaline, saline, and sodic soils. Many of them are also known to mitigate various biotic and abiotic stresses in plants. In the last few years, potential PGPR enhancing the productivity of plants facing salt-stress have been researched upon suggesting that ST-PGPR can be exploited for the reclamation of saline agro-ecosystems. In this review, ST-PGPR and their potential in enhancing the productivity of saline agro-ecosystems will be discussed. Apart from this, PGPR mediated mechanisms of salt tolerance in different crop plants and future research trends of using ST-PGPR for reclamation of saline soils will also be highlighted.

Abstract:Salinity stress is one of the major abiotic stresses threatening sustainable crop production worldwide. The extent of salinity affected area is expected to cover about 50% of total agricultural land by 2050. Salinity stress produces various detrimental effects on plants' physiological, biochemical, and molecular features and reduces productivity. The poor plant growth under salinity stress is due to reduced nutrient mobilization, hormonal imbalance, and formation of reactive oxygen species (ROS), ionic toxicity, and osmotic stress. Additionally, salinity also modulates physicochemical properties and reduces the microbial diversity of soil and thus decreases soil health. On the other hand, the demand for crop production is expected to increase in coming decades owing to the increasing global population. Conventional agricultural practices and improved salt-tolerant crop varieties will not be sufficient to achieve the yields desired in the near future. Plants harbor diverse microbes in their rhizosphere, and these have the potential to cope with the salinity stress. These salinity-tolerant plant growth-promoting bacteria (PGPB) assist the plants in withstanding saline conditions. These plant-associated microbes produce different compounds such as 1-aminocyclopropane-1-carboxylate (ACC) deaminase, indole-3-acetic acid (IAA), antioxidants, extracellular polymeric substance (EPS), and volatile organic compounds (VOC). Additionally, the naturally associated microbiome of plants has the potential to protect the host through stress avoidance, tolerance, and resistance strategies. Recent developments in microbiome research have shown ways in which novel microbe-assisted technologies can enhance plant salt tolerance and enable higher crop production under saline conditions. This focused review article presents the global scenario of salinity stress and discusses research highlights regarding PGPB and the microbiome as a biological tool for mitigation of salinity stress in plants.
